[Nature of the restructuring of the T- and B-immunity systems in patients with cutaneous and visceral candidiasis].

The examination of 90 patients with superficial and visceral candidiasis, as well as Candida carriers, has revealed that the heavy contamination of the body with fungi of the genus Candida is accompanied by the specific transformation of the immune system, manifested by differences in the character of sensitization to Candida antigens: in the patients with superficial and visceral candidiasis the B-system of immunity is sensitized to a greater degree, while in the Candida carriers the sensitization of the T-system is more pronounced. Despite the fact that the immune system of patients with the

Gel Formulations with an Echinocandin for Cutaneous Candidiasis: The Influence of Azone and Transcutol on Biopharmaceutical Features.

Caspofungin is a drug that is used for fungal infections that are difficult to treat, including invasive aspergillosis and candidemia, as well as other forms of invasive candidiasis. The aim of this study was to incorporate Azone in a caspofungin gel (CPF-AZ-gel) and compare it with a promoter-free caspofungin gel (CPF-gel). An in vitro release study using a polytetrafluoroethylene membrane and ex vivo permeation into human skin was adopted. The tolerability properties were confirmed by histological analysis, and an evaluation of the biomechanical properties of the skin was undertaken. Antimic

Polymeric micelle gel with luliconazole: in vivo efficacy against cutaneous candidiasis in Wistar rats.

The objective of this research was focused on the design and development of luliconazole-loaded polymeric micelle hydrogel (LUL-PM-CHG) using quality by design (QbD) principle to improve the penetration and retention of LUL in the skin. The optimization of the formulation involved the utilization of a Box-Behnken design with three factors and three levels. The impact of specific formulation variables, namely the ratio of poloxamer P123 and F127, sonication time, and the quantity of drug, was investigated in terms of particle size, micellar incorporation efficiency, and polydispersity index. Cold Atmospheric Plasma Exerts Antimicrobial Effects in a 3D Skin Model of Cutaneous Candidiasis.

Cutaneous candidiasis is characterized by an overgrowth of Candida leading to skin inflammation and infection. Similar to bacteria, Candida can develop tolerance to common antifungal drugs. Cold atmospheric plasma (CAP), with its proven antimicrobial properties, offers a promising alternative to the prevailing methods. Because of plasma heterogeneity each new device must be tested individually for its effectiveness. Antimicrobial activity is usually studied using planktonic microorganisms or animal models, making it difficult to extrapolate the results to the human system. Therefore, a 3D skin

Dissolving microneedle-mediated dermal delivery of itraconazole nanocrystals for improved treatment of cutaneous candidiasis.

The administration of conventional dosage forms of itraconazole (ITZ) for cutaneous candidiasis treatment is limited by its poor aqueous solubility and the deep location ofCandida albicans(CA) in this disease. In the present work, we developed a nanocrystal (NC) form of ITZ, which was incorporated into dissolving microneedles (MNs) to facilitate skin delivery of ITZ into the infection site. The NCs were prepared by media milling with an ultra-small-scale device using Pluronic®F127 as a stabiliser. The antifungal activity of ITZ was enhanced by NC formulations (MIC value of 2.5 μg/ml), compared

Two cases of primary cutaneous candidiasis caused by Candida parapsilosis: a report and literature review.

Candida parapsilosis is a rare cause of primary cutaneous infection, with rare cases reported in the literature to date, most of the reports were immunocompromised individuals. Herein, we report two cases of primary cutaneous infection caused by C. parapsilosis in immunocompetent adults. C. parapsilosis were isolated and identified through morphological and molecular analyses, these isolates were found to be susceptible to itraconazole through in vitro antifungal susceptibility test. Both patients achieved clinical relief following oral itraconazole treatment. These two cases enriched the mani

Quinine Improves the Fungicidal Effects of Antimicrobial Blue Light: Implications for the Treatment of Cutaneous Candidiasis.

BACKGROUND AND OBJECTIVE: Candida albicans is an opportunistic fungal pathogen of clinical importance and is the primary cause of fungal-associated wound infections, sepsis, or pneumonia in immunocompromised individuals. With the rise in antimicrobial resistance, it is becoming increasingly difficult to successfully treat fungal infections using traditional antifungals, signifying that alternative non-traditional approaches must be explored for their efficacy. STUDY DESIGN/MATERIALS AND METHODS: We investigated the combination of antimicrobial blue light (aBL) and quinine hydrochloride (Q-HCL

Cutaneous candidiasis - an evidence-based review of topical and systemic treatments to inform clinical practice.

Cutaneous candidiasis is a common skin disease, and several treatments have been investigated within the last fifty years. Yet, systematic reviews are lacking, and evidence-based topical and systemic treatment strategies remain unclear. Thus, the aim of this review was to summarize efficacy and adverse effects of topical and oral therapies for cutaneous candidiasis in all age groups. Two individual researchers searched PubMed and EMBASE for 'cutaneous candidiasis' and 'cutaneous candidiasis treatment', 'intertrigo', 'diaper dermatitis' and 'cheilitis'. Searches were limited to 'English languag

The biological significance of TLR3 variant, L412F, in conferring susceptibility to cutaneous candidiasis, CMV and autoimmunity.

OBJECTIVE: Toll-like receptors, a major component of the innate immune system, play an important role in the initial response against pathogens. Genetic abnormalities in some receptors like TLR2, TLR3 and TLR4 have been associated with susceptibility to fungal and viral infections while other aberrations in TLR genes such as TLR3, TLR7 and TLR9 may predispose to autoimmunity. Recently we have shown an association of a TLR3 receptor variant, L412F, to susceptibility to chronic candidiasis, recurrent viral and bacterial infections and autoimmunity. We investigated here the biological implication

Enhanced antifungal therapy for cutaneous candidiasis: Development, evaluation, and optimization of luliconazole-loaded transethosomal gel.

Cutaneous candidiasis, a widespread opportunistic fungal infection primarily caused by Candida albicans, accounts for 1% of outpatient and 7% of inpatient dermatology consultations. The rise in antifungal resistance due to genetic mutations in Candida species has complicated therapeutic outcomes and increased mortality rates. This study aimed to develop a more effective topical therapy by formulating Luliconazole (LCZ), a potent imidazole antifungal limited by poor solubility and skin penetration, into a Transethosomal gel (LCZ-TEG) for enhanced delivery and efficacy. LCZ-loaded Transethosomes

Deep Cutaneous Candidiasis With Costal Osteomyelitis Following Pectoralis Major Myocutaneous Flap Reconstruction: A Case Report.

Candida is a yeast commonly found in various environments. It usually coexists with the skin without causing harm. It may exhibit pathogenicity when local or systemic immune defense mechanisms are compromised, which creates conditions favorable for its proliferation. This report discusses a case of invasive cutaneous candidiasis with rib osteomyelitis that developed at the donor site of a pectoralis major myocutaneous flap. The patient was a 58-year-old man who underwent reconstruction with a pectoralis major myocutaneous flap for adenoid cystic carcinoma in the midline floor of the mouth. Six

Atypical cutaneous candidiasis mimicking tinea corporis in an otherwise healthy young man: A case report.

Cutaneous candidiasis typically affects intertriginous areas and may be overlooked when occurring at non-intertriginous sites. We report a 28-year-old otherwise healthy man with a 7-month history of pruritic scaly erythematous lesions on both upper extremities and the upper abdomen. Before presentation, he had used a topical corticosteroid unsupervised for 2 months. The lesions were initially treated as tinea corporis. Direct microscopy showed pseudohyphae and spores, and two consecutive fungal cultures yielded Candida spp., with CHROMagar Candida supporting Candida albicans. The lesions resol

In vivo and in vitro Pathogenesis and Virulence Factors of Candida albicans Strains Isolated from Cutaneous Candidiasis.

BACKGROUND: The Candida albicans is one of the most important global opportunistic pathogens, and the incidence of candidiasis has increased over the past few decades. Despite the established role of skin in defense against fungal invasion, little has been documented about the pathogenesis of Candida species when changing from normal flora to pathogens of vaginal and gastrointestinal epithelia. This study was carried out to determine the in vivo and in vitro pathogenesis of clinical C. albicans strains isolated from skin lesions. METHODS: In this study, association of in vivo and in vitro pat

Itraconazole lipid nanocapsules gel for dermatological applications: In vitro characteristics and treatment of induced cutaneous candidiasis.

There is a growing clinical demand for topical itraconazole (ITC) delivery systems because of the expanding potential of the drug for topical fungal and non-fungal applications. Lipid-based nanocarriers offer great promise in this respect. In the present study, a new topical ITC gel based on lipid nanocapsules (LNC) was developed. ITC-LNC were compared to ITC-loaded nanostructured lipid carriers (ITC-NLC) with more established benefits as topical vectors. Both nanocarriers showed high entrapment efficiency (EE > 98%). Compared to ITC-NLC, the ITC-LNC showed a significantly smaller particle siz

Transdermal administration of farnesol-ethosomes enhances the treatment of cutaneous candidiasis induced by Candida albicans in mice.

UNLABELLED: Cutaneous candidiasis, caused by Candida albicans, is a severe and frustrating condition, and finding effective treatments can be challenging. Therefore, the development of farnesol-loaded nanoparticles is an exciting breakthrough. Ethosomes are a novel transdermal drug delivery carrier that incorporates a certain concentration (10-45%) of alcohols into lipid vesicles, resulting in improved permeability and encapsulation rates compared to conventional liposomes. Farnesol is a quorum-sensing molecule involved in morphogenesis regulation in C. albicans, and these ethosomes offer a pr

Candida species in cutaneous candidiasis patients in the Guilan province in Iran; identified by PCR-RFLP method.

Due to the epidemiological alteration in distribution of Candida species as well as significant increasing trend of either intrinsic or acquired in resistance of some of these fungi, the precise identification of Candida species is necessary for effective antifungal therapy and also for prevention of nosocomial infections. PCR-RFLP method is indicated to be a reliable, rapid and simple technique which is able to differentiate the Candida species. In the present study, we applied this method to evaluate the distribution of Candida species in patients affected with cutaneous candidiasis in the G
