Iron-mediated protein-humic acid interactions under aerobic and anaerobic conditions: Implications for protein hydrolysis and wastewater treatment.

Proteins and carbohydrates are both major biodegradable fractions in wastewater. Complexation with coexisting compounds, such as iron (Fe) and humic acids (HA), which are both commonly present in wastewater, could influence the different degradation rates of proteins and carbohydrates. Depending on the redox conditions, Fe exists as Fe(II) or Fe(III), with differing binding affinities and chemical behaviour. This research aims to systematically assess the complex interaction between Fe, protein, and HA compounds under aerobic and anaerobic conditions. The results showed that the addition of Fe

Vitronectins produced by human cirrhotic liver and CCl4-treated rats differ in their glycosylation pattern and tissue remodeling activity.

Liver cirrhosis (LC) is a disease characterized by pathological accumulation and alteration of extracellular matrix (ECM) proteins; the interaction between two such proteins, collagen and vitronectin (VN), is considered to be the key to controlling ECM remodeling in liver cirrhosis. If it is possible to control the modification of oligosaccharides on VN, it may be possible to retard progression of liver cirrhosis. In this study, we examined the relationship between changes in VN glycosylation and activity related to the remodeling of hepatic tissue in human LC and a rat model of LC generated u

Complete Pathway Refactoring and Combinatorial Biosynthesis of Anthracyclines for Modulation of Histone Eviction and DNA Damage Bioactivities.

Anthracyclines including doxorubicin are cornerstone chemotherapeutic agents with high cytotoxic potency, but their use in cancer treatment is limited by irreversible dose-dependent cardiotoxicity. Recent investigations indicate that cardiotoxicity-free anthracyclines may be achieved through uncoupling DNA damage from chromatin damage. However, semi-synthetic derivatization is complicated by anthracycline structural complexity. Here, we report a modular synthetic biology platform for the complete refactoring and combinatorial biosynthesis of anthracyclines in Streptomyces coelicolor M1152ΔmatA

Glycosylated SARs Cov 2 interaction with plant lectins.

Lectins are non-immune carbohydrate-binding proteins/glycoproteins that are found everywhere in nature, from bacteria to human cells. They have also been a valuable biological tool for the purification and subsequent characterisation of glycoproteins due to their carbohydrate binding recognition capacity. Antinociceptive, antiulcer, anti-inflammatory activities and immune modulatory properties have been discovered in several plant lectins, with these qualities varying depending on the lectin carbohydrate-binding site. The Coronavirus of 2019 (COVID-19) is a respiratory disease that has swept t

Functional signatures of the gut microbiome in middle-aged regular runners: insights from a metagenomic study.

INTRODUCTION: Exercise influences host metabolism and inflammation, but its functional effects on the gut microbiome in middle-aged populations remain unclear. This study used shotgun metagenomics to investigate the associations between long-term endurance running and the gut microbial ecosystem and its functional potential in middle-aged adults. METHODS: We conducted a cross-sectional analysis comparing 33 middle-aged regular runners with 33 sedentary controls. No significant differences in age, BMI, dietary intake between groups. Fecal samples underwent metagenomic sequencing at an average

A comprehensive review of sulfated fucan from sea cucumber: Antecedent and prospect.

Sulfated fucan from sea cucumber is mainly consists of L-fucose and sulfate groups. Recent studies have confirmed that the structure of sulfated fucan mainly consists of repeating units, typically tetrasaccharides. However, there is growing evidence indicating the presence of irregular domains with heterogeneous units that have not been extensively explored. Moreover, as a key contributor to the nutritional benefits of sea cucumbers, sulfated fucan demonstrates a range of biological activities, such as anti-inflammatory, anticancer, hypolipidemic, anti-hyperglycemic, antioxidant, and anticoagu

Enzymatic Site-Selective Deglycosylation Enhances PPM1A Agonistic Activity of Dammarane Saponins for the Treatment of Hepatic Fibrosis.

Carbohydrate moieties profoundly influence target binding, drug-like properties, and mechanism of action, but achieving site-selective editing within complex oligosaccharide chains remains a challenge for conventional chemical methods. We herein report an enzymatic deglycosylation platform that enables precise, site-selective removal of sugar units from branched carbohydrate chains of dammarane-type saponins, the first reported PPM1A phosphatase agonists. SAR analysis confirms the distal xylose removal drastically improves PPM1A agonistic potency versus the parent saponins. Cellular and in viv

Metatranscriptomic Analysis Reveals Functional Features of the Gastrointestinal Microbiota in Tibetan Pigs.

Tibetan pigs exhibit remarkable adaptability to hypoxic high-altitude conditions. However, metatranscriptomics has not been used to investigate the transcriptional activity and functional responses of the gastrointestinal microbiota as part of their adaptation to high altitude. We collected 57 samples of the contents of the stomach, jejunum, cecum, colon, and rectum of six high-altitude Tibetan pigs (n = 6, elevation 3750 m) and six low-altitude black pigs (n = 6, elevation 500 m) for metatranscriptomic analysis. We identified 79 differentially abundant bacterial genera and 69 differentially a

Vatairea macrocarpa lectin (VML) induces depressive-like behavior and expression of neuroinflammatory markers in mice.

Lectins are proteins capable of reversible binding to the carbohydrates in glycoconjugates that can regulate many physiological and pathological events. Galectin-1, a β-galactoside-binding lectin, is expressed in the central nervous system (CNS) and exhibits neuroprotective functions. Additionally, lectins isolated from plants have demonstrated beneficial action in the CNS. One example is a lectin with mannose-glucose affinity purified from Canavalia brasiliensis seeds, ConBr, which displays neuroprotective and antidepressant activity. On the other hand, the effects of the galactose-binding le

Fusobacterium nucleatum Lipopolysaccharides Disrupt the Interaction of Siglec‑7 to Sialoglycans Expressed on Mammalian Cells.

Sialic acid-binding immunoglobulin-like lectins (Siglecs) are key immune receptors that bind to cell surface sialic acids, leading to modulation of the immune system. Interrupting the Siglec-sialoglycan binding in cancer has been proposed as a potential antitumor response strategy. We previously showed that colon-cancer-associated Fusobacterium nucleatum (Fn) ATCC 51191 interacts with Siglec-7 via its lipopolysaccharide (LPS), revealing Fn LPS as a new ligand for Siglec-7. Here, we used glycoengineered cells carrying sialic acid variants to investigate the capacity of LPS isolated from F. nucl

Structural and Functional Insights into Catalytic Efficiency and Stability Trade-Offs Induced by CBM Truncation in a GH5 Endoglucanase from Bacillus subtilis DLG.

Cellulose depolymerization into glucose requires the synergistic action of endoglucanase, exoglucanase, and β-glucosidase. In this study, Bacillus subtilis DLG endoglucanase (BsEGL) and its family 3 carbohydrate-binding module (CBM3)-truncated mutant (BsEGLCD) were heterologously expressed in Escherichia coli BL21(DE3) and characterized. Both enzymes displayed optimal activity at 50 °C and pH 4.5, with BsEGLCD exhibiting superior thermal stability and broader pH stability. Both enzymes maintained over 90% relative activity within 0.5-4.5 M NaCl. Notably, BsEGLCD exhibited superior ethanol tole

Integrative multi-omics reveals comprehensive gut-liver-adipose metabolic changes in peak laying hens with high or low egg production.

Eggs serve as an indispensable global nutritional resource, sustaining the economic foundation of the commercial poultry industry. To meet this continuous demand, egg formation involves an exceptionally energy-intensive biological process requiring continuous yolk precursor synthesis, which imposes a massive metabolic burden on laying hens. However, the comprehensive metabolic differences across the gut-liver-adipose axis between peak laying hens with high or low egg production remain incompletely characterized. A total of 180 healthy Hy-Line Brown laying hens (45-week-old) were continuously f

Enhancing the degradation of cellulose and hemicellulose in chili pepper straw waste using Cellulomonas iranensis 7-12, which was isolated from naturally decayed chili pepper straw.

The sustainable valorization of agricultural waste, such as chili pepper straw, is often challenged by the absence of effective microbes that can degrade cell wall components. In this study, metagenomic analysis found that Pseudomonadota was the dominant phylum in the carboxymethyl cellulose (CMC)-enriched microbial communities. In addition, a cellulolytic bacterial strain, designated as Cellulomonas iranensis 7-12, was isolated from naturally decayed chili pepper straw and identified by colony morphology, Gram staining, 16 S rRNA gene sequencing, and genome-based average nucleotide identity (

The development and excretion of Toxoplasma gondii oocyst manipulate the gut microbiota in its definitive host.

BACKGROUND: Oocysts serve as the primary source of Toxoplasma infection. Therefore, understanding oocyst development and exploring effective strategies to prevent oocyst excretion are crucial for controlling toxoplasmosis. METHODS: In this study, shotgun metagenomics was employed to characterize the functional and compositional changes in the gut microbiota of cats during oocyst development. The Spearman correlation test was utilized to analyze the correlation between differential Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways and carbohydrate-active enzymes (CAZymes) in key bacteria

CcpA and CodY Coordinate Acetate Metabolism in Streptococcus mutans.

In the dental caries pathogen Streptococcus mutans, phosphotransacetylase (Pta) and acetate kinase (Ack) convert pyruvate into acetate with the concomitant generation of ATP. The genes for this pathway are tightly regulated by multiple environmental and intracellular inputs, but the basis for differential expression of the genes for Pta and Ack in S. mutans had not been investigated. Here, we show that inactivation in S. mutans of ccpA or codY reduced the activity of the ackA promoter, whereas a ccpA mutant displayed elevated pta promoter activity. The interactions of CcpA with the promoter re

Glycan Profiling of Plant-Based Milk Alternatives by Antibody Microarrays.

The growing prevalence of lactose intolerance and cow's milk protein allergy has contributed to a significant increase in consumer demand for plant-based milk alternatives [ Food Res. Int. 2020, 131, 108972, 10.1016/j.foodres.2019.108972]. These products are marketed as nutritionally comparable to dairy milk, but limited data exist regarding their structural glycan composition, which may influence nutritional, functional, and immunological properties. This study aims to profile the glycan (polysaccharide) composition of nine widely consumed plant-based milk types: oat, almond, coconut, hazelnu

PACER: a novel 3D plant cell wall model for the analysis of non-catalytic and enzymatic responses.

BACKGROUND: Substrate accessibility remains a key limitation to the efficient enzymatic deconstruction of lignocellulosic biomass. Limited substrate accessibility is often addressed by increasing enzyme loading, which increases process and product costs. Alternatively, considerable efforts are underway world-wide to identify amorphogenesis-inducing proteins and protein domains that increase the accessibility of carbohydrate-active enzymes to targeted lignocellulose components. RESULTS: We established a three-dimensional assay, PACER (plant cell wall model for the analysis of non-catalytic and

Antiadhesive glycoconjugate metal complexes targeting pathogens Pseudomonas aeruginosa and Candida albicans.

Glycoconjugates are known to interact with carbohydrate-binding proteins involved in adhesion by pathogens, and offer opportunities to design antimicrobial agents. Metal complexes with Eu(III), Ni(II) and Zn(II) were prepared from glycoconjugate ligand 1Gal, which binds to P. aeruginosa's lectin LecA (Kd 9.6 ± 0.7 μM). In vitro anti-adhesive activity of these compounds was evaluated for both P. aeruginosa and C. albicans. Choice of metal ion played a crucial role in modulating anti-adhesive activity, with Eu(III) complexes most effective: [Eu·(1Gal)(H2O)6](CF3SO3)3 inhibits 60% biofilm formati

Akkermansia muciniphila exoglycosidases target extended blood group antigens to generate ABO-universal blood.

Matching donor and recipient blood groups based on red blood cell (RBC) surface ABO glycans and antibodies in plasma is crucial to avoid potentially fatal reactions during transfusions. Enzymatic conversion of RBC glycans to the universal group O is an attractive solution to simplify blood logistics and prevent ABO-mismatched transfusions. The gut symbiont Akkermansia muciniphila can degrade mucin O-glycans including ABO epitopes. Here we biochemically evaluated 23 Akkermansia glycosyl hydrolases and identified exoglycosidase combinations which efficiently transformed both A and B antigens and

Interactions that define the arrangement of sugar-binding sites in BDCA-2 and dectin-2 dimers.

The sugar-binding receptors dectin-2 and blood dendritic cell antigen 2 (BDCA-2) bind oligosaccharide ligands through extracellular carbohydrate-recognition domains (CRDs) and initiate intracellular signaling through Fc receptor γ adapters (FcRγ). Dectin-2 stimulates macrophages in response to pathogen binding while BDCA-2 modulates cytokine production in plasmacytoid dendritic cells. The oligomeric states of these receptors and the orientations of their CRDs have been investigated by analysis of a naturally occurring disulfide-bonded variant of BDCA-2 and by replacement of transmembrane domai
