Distribution and Catalytic Mechanism of a Novel Carboxylesterase Degrading Multiple Aryloxyphenoxypropionate Herbicides in Bacteria and Archaea.

Aryloxyphenoxypropionate (AOPP), a widely used class of herbicides, pose significant risks to ecosystems and human health. In this study, Bacillus amyloliquefaciens L1 was isolated as a novel degrader, completely degrading 97.42 mg/L cyhalofop-butyl within 12 h via ester bond hydrolysis to cyhalofop acid. A new carboxylesterase, PnbA, was identified from strain L1, showing substrate preference for cyhalofop-butyl > clodinafop-propargyl > haloxyfop-p-methyl > quizalofop-p-ethyl. Molecular docking and mutagenesis identified S190 as a key catalytic residue. Two mutants, K411A and T412A, exhibited

A highly sensitive and selective enzyme activated fluorescent probe for in vivo profiling of carboxylesterase 2.

Carboxylesterase 2 (CES2) is a serine-type hydrolase that plays important roles in xenobiotic detoxification and lipid metabolism. Its abnormal expression is highly associated with diseases such as diabetes and carcinoma. To date, intense attention has been attracted to CES2 targeted drug discovery and disease diagnosis. Thus, to further explore the physiological function of CES2 is of great importance. However, until now, most medical research on CES2 function and activity assays is still dependent on conventional methods, which could hardly specify CES2 activity. Therefore, there is an urgen

Carboxylesterase 1 is a core metabolic vulnerability of platinum-resistant high-grade serous ovarian carcinoma.

BACKGROUND: Platinum-resistant high-grade serous ovarian carcinoma (HGSOC) is a major clinical challenge, with high mortality rates and no effective therapeutic options. Metabolic adaptations-including enhanced lipid catabolism, oxidative phosphorylation (OXPHOS), and autophagy-are major drivers of chemoresistance; however, the molecular mechanisms underpinning this metabolic reprogramming remain poorly understood. Carboxylesterase 1 (CES1), an NF-κB-regulated neutral lipase, supports malignant cell survival under nutrient-depleted conditions in other cancers, but its role in HGSOC has not bee

Carboxylesterase 1 contributes to the human macrophage alternative activation response to Th2 cytokine interleukin-4.

Carboxylesterase 1 (CES1) is a hydrolytic enzyme with roles in xenobiotic and lipid metabolism. Our previous studies suggested a novel role for CES1 in the regulation of macrophage gene expression following exposure to inflammatory molecules. The nuclear receptor PPARγ has an important role in the M(IL-4) polarization response. Here, we investigated CES1 in human primary macrophages and THP-1 macrophages during M(IL-4) polarization. We hypothesized that ablating CES1's activity would decrease ligand signaling through PPARγ, thereby reducing the M2 polarization response. Inhibition of CES1's ac

Structural analysis of Sphingobium yanoikuyae P4 carboxylesterase reveals conformational and catalytic determinants for PET biodegradation.

The accumulation of poorly biodegradable polyethylene terephthalate (PET) in the environment has become an emerging ecological and health problem, with a strong need to develop environmentally friendly, biobased degradation methods. This in silico study demonstrates the structural and mechanistic characterization of a carboxylesterase from the phthalate-degrading Sphingobium yanoikuyae P4 for its potential to catalyze the hydrolysis of PET. Sequence analysis of the amino acid residues revealed the conserved α/β-hydrolase fold and the sequence motifs GXSXG and GGX, typical of PET-active enzymes

Distribution and Evolutionary Signatures of a Broad-Spectrum Pesticide-Degrading Carboxylesterase TriS From Strain S113.

Ester-containing pesticides, such as diphenyl ether herbicides, aryloxyphenoxypropionate herbicides, and strobilurin fungicides, are widely applied in agriculture, but their residues pose significant risks to ecosystems. Functional microorganisms can degrade ester-containing pesticides in the natural environment. However, the microbial mechanisms responsible for their degradation in soils remain unclear. A novel hydrolase gene triS from the strain S113 encoded a broadly active carboxylesterase that hydrolyzes a wide range of ester-containing pesticides. TriS exhibited a maximum sequence simila

Lipopolysaccharide suppresses carboxylesterase 2g activity and 2-arachidonoylglycerol hydrolysis: A possible mechanism to regulate inflammation.

Inflammation is an important part of the innate immune response and is involved in the healing of many disease processes; however, chronic inflammation is a harmful component of many diseases. The regulatory mechanisms of inflammation are incompletely understood. One possible regulatory mechanism is the endocannabinoid system. Endocannabinoids such as 2-arachidonoylglycerol (2-AG) and anandamide (AEA) are generally anti-inflammatory via engagement of the cannabinoid receptor 2 (CB2) on innate cells; therefore, preventing the degradation of endocannabinoids by specific serine hydrolases such as

Global and local molecular dynamics of a bacterial carboxylesterase provide insight into its catalytic mechanism.

Carboxylesterases (CEs) are ubiquitous enzymes responsible for the detoxification of xenobiotics. In humans, substrates for these enzymes are far-ranging, and include the street drug heroin and the anticancer agent irinotecan. Hence, their ability to bind and metabolize substrates is of broad interest to biomedical science. In this study, we focused our attention on dynamic motions of a CE from B. subtilis (pnbCE), with emphasis on the question of what individual domains of the enzyme might contribute to its catalytic activity. We used a 10 ns all-atom molecular dynamics simulation, normal mod

Carboxylesterase Factors Influencing the Therapeutic Activity of Common Antiviral Medications Used for SARS-CoV-2 Infection.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the virus responsible for COVID-19, remains a major global health threat. The virus enters host cells by binding to the angiotensin-converting enzyme 2 (ACE2) receptor. Several small-molecule antiviral drugs, including molnupiravir, favipiravir, remdesivir, and nirmatrelvir have been shown to inhibit SARS-CoV-2 replication and are approved for treating SARS-CoV-2 infections. Nirmatrelvir inhibits the viral main protease (Mpro), a key enzyme for processing polyproteins in viral replication. In contrast, molnupiravir, favipiravir, and

Catalytic Hydrolysis Mechanism of Cocaine by Human Carboxylesterase 1: An Orthoester Intermediate Slows Down the Reaction.

Human carboxylesterase 1 (hCES1) is a major carboxylesterase in the human body and plays important roles in the metabolism of a wide variety of substances, including lipids and drugs, and therefore is attracting more and more attention from areas including lipid metabolism, pharmacokinetics, drug-drug interactions, and prodrug activation. In this work, we studied the catalytic hydrolysis mechanism of hCES1 by the quantum mechanics computation method, using cocaine as a model substrate. Our results support the four-step theory of the esterase catalytic hydrolysis mechanism, in which both the ac

Icaritin enhances the antitumor efficacy of irinotecan by dual-targeting carboxylesterase 2 and p53 in vitro and in vivo.

Carboxylesterase 2 (CES2), a member of the serine hydrolase superfamily, plays a crucial role in catalyzing the hydrolysis of numerous endogenous and exogenous compounds containing ester bonds. The commonly used clinical drug irinotecan (CPT-11) exerts its anti-tumor effect by being hydrolyzed by CES2 to generate SN-38. Epimedium, a widely used traditional Chinese herb with multiple pharmacological properties, has not yet been characterized for its effects on CES2. Our study systematically evaluated the three principal bioactive components of Epimedium for their effects on CES2 activity, revea

Dual-reporter fluorescent probe for precise identification of liver cancer by sequentially responding to carboxylesterase and polarity.

Early treatment significantly improves the survival rate of liver cancer patients, so the development of early diagnostic methods for liver cancer is urgent. Liver cancer can develop from viral hepatitis, alcoholic liver, and fatty liver, thus making the above diseases share common features such as elevated viscosity, reactive oxygen species, and reactive nitrogen species. Therefore, accurate differentiation between other liver diseases and liver cancer is both a paramount practical need and challenging. Numerous fluorescent probes have been reported for the diagnosis of liver cancer by detect

Potent, Irreversible Inhibition of Human Carboxylesterases by Tanshinone Anhydrides Isolated from Salvia miltiorrhiza ("Danshen").

The roots of Salvia miltiorrhiza ("Danshen") have been used in Chinese herbal medicine for centuries for a host of different conditions. While the exact nature of the active components of this material are unknown, large amounts of tanshinones are present in extracts derived from these samples. Recently, the tanshinones have been demonstrated to be potent human carboxylesterase (CE) inhibitors, with the ability to modulate the biological activity of esterified drugs. During the course of these studies, we also identified more active, irreversible inhibitors of these enzymes. We have purified,

Carboxylesterases 1 and 2 hydrolyze phospho-nonsteroidal anti-inflammatory drugs: relevance to their pharmacological activity.

Phospho-nonsteroidal anti-inflammatory drugs (phospho-NSAIDs) are novel NSAID derivatives with improved anticancer activity and reduced side effects in preclinical models. Here, we studied the metabolism of phospho-NSAIDs by carboxylesterases and assessed the impact of carboxylesterases on the anticancer activity of phospho-NSAIDs in vitro and in vivo. The expression of human liver carboxylesterase (CES1) and intestinal carboxylesterase (CES2) in human embryonic kidney 293 cells resulted in the rapid intracellular hydrolysis of phospho-NSAIDs. Kinetic analysis revealed that CES1 is more active

The carboxylesterase AtCXE12 converts volatile (Z)-3-hexenyl acetate to (Z)-3-hexenol in Arabidopsis leaves.

The green leaf volatiles (GLVs) (Z)-3-hexenal, (Z)-3-hexenol, and (Z)-3-hexenyl acetate play important roles in plant defense, deterring insect herbivores and attracting their natural enemies, while also serving as airborne signaling molecules capable of enhancing defenses in undamaged plant tissues. Almost all plants produce GLVs after wounding, beginning with the formation of (Z)-3-hexenal, which is subsequently converted to (Z)-3-hexenol and (Z)-3-hexenyl acetate. (Z)-3-hexenyl acetate can then be taken up by nearby plant tissues where it is predicted to be hydrolyzed to (Z)-3-hexenol, a pr

Genome-wide analysis of lipolytic enzymes and characterization of a high-tolerant carboxylesterase from Sorangium cellulosum.

Microorganisms are important sources of lipolytic enzymes with characteristics for wide promising usages in the specific industrial biotechnology. The cellulolytic myxobacterium Sorangium cellulosum is rich of lipolytic enzymes in the genome, but little has been investigated. Here, we discerned 406 potential lipolytic enzymes in 13 sequenced S. cellulosum genomes. These lipolytic enzymes belonged to 12 families, and most are novel with low identities (14-37%) to those reported. We characterized a new carboxylesterase, LipB, from the alkaline-adaptive So0157-2. This enzyme, belonging to family

Partial purification and characterization of a microsomal carboxylesterase specific for salicylate esters from guinea-pig liver.

Studies on liver carboxylesterases have predominantly involved the use of uncharged ester and amide substrates to monitor activity. A microsomal carboxylesterase (EC 3.1.1.1) from guinea-pig liver microsomes has been identified which specifically hydrolyses aspirin (White, K.N. and Hope, D.B. (1981) Biochem. J. 197, 771-773), a substrate which is negatively charged at physiological pH, and this work describes its partial purification and characterization. The enzyme is monomeric, it has a molecular weight of approx. 55 000 and is very sensitive to inhibition by the carboxylesterase inhibitor b
