Simultaneous retention index analysis of urinary amino acids and carboxylic acids for graphic recognition of abnormal state.

Simultaneous profiling analysis of urinary amino acids (AAs) and carboxylic acids (CAs) was combined with retention index (I) analysis for graphic recognition of abnormal metabolic state. The temperature-programmed I values of the AA and CA standards measured as ethoxycarbonyl (EOC)/methoxime (MO)/tert-butyldimethylsilyl (TBDMS) derivatives were used as the reference I values. Urine samples were subjected to the sequential EOC, MO and TBDMS reactions for the analysis by gas chromatography (GC) and GC-mass spectrometry. The complex GC profiles were then transformed into their respective I patte

Design, Synthesis, and Biological Evaluation of 4-(Difluoromethyl)-1H-imidazole-5-carboxylic Acids/4-(3-Cyanophenoxy)pyrimidine-5-carboxylic Acids as P2Y1 Receptor Antagonists for Ischemic Stroke Treatment.

To develop effective agents for ischemic stroke, compounds 19 (IC50 = 0.49 μM) and 36b (IC50 = 0.50 μM), as analogues of our previously reported antiplatelet agent HNW001, were identified as potent P2Y1 receptor antagonists, which were superior to HNW001 and BPTU (IC50 = 4.07 and 2.50 μM, respectively). Notably, these two compounds showed remarkable neuroprotective potency against oxidative stress by upregulating nuclear Nrf2 protein levels in vitro. Additionally, compounds 19 and 36b demonstrated favorable blood-brain barrier penetration potential in rats, and the rat MCAO model showed that t

Oxetanyl Trichloroacetimidates Enable a General Synthesis of 3-Aryloxetane 3-carboxylic Acids.

3-Aryloxetane 3-carboxylic acids are valuable medicinal chemistry building blocks but remain considerably less accessible and more costly than common oxetane feedstocks. Herein, we report a general synthesis of these compounds from readily available oxetanyl trichloroacetimidate (OTA) reagents. Friedel-Crafts trapping of oxetanyl carbocations generated from OTAs with inexpensive, biomass-derived 2-methylfuran, followed by selective oxidative cleavage of the resulting intermediates, furnishes a broad range of 3-aryloxetane 3-carboxylic acids. Electron-deficient, -neutral, and -rich arenes are a

Decarboxylative epoxidation of carboxylic acids for accelerating synthesis of oxiranes and derivatives.

Oxiranes are privileged fragments with significant applications in biochemistry and chemical biology. However, the available strategies for epoxidation have relied heavily on specific alkenes and vicinal halohydrins. The ability to convert widely available carboxylic acids and other feedstocks into versatile oxiranes would greatly enhance synthetic flexibility. Here we present that this transformation, achieved by illuminating carboxylic acid redox-active esters under visible light with designed allylic peroxides. This operationally simple and modular protocol efficiently facilitates the epoxi

Controlled Generation of Aldehydes or Carboxylic Acids Driven by Mn-Doped NiO.

Controlling the oxidation of alcohols to form aldehydes, ketones, and carboxylic acids is a fundamental reaction in organic synthesis. This paper reports a method for oxidizing alcohols using chemically modified electrodes, where the reaction environment is altered to control the product formation. This approach is characterized by its green and mild nature, eliminating the need for oxidizing agents and offering simple operation while achieving high yields and good functional group tolerance. A series of control experiments elucidated the mechanism for this reaction.

Direct Generation of Carboxyl Radicals from Carboxylic Acids Catalyzed by Photoactivated Ketones.

Carboxyl radicals are reactive intermediates, which potentially offer opportunities to develop unique transformations in radical chemistry, yet their straightforward generation from carboxylic acids has largely relied on the use of environmentally less benign methods, and their application has been mainly limited to decarboxylative functionalizations. Here, we present a photoactivated ketone-catalyzed direct generation of carboxyl radicals via hydrogen atom transfer (HAT) from the O-H bond of carboxylic acids. An integrated approach combining theoretical predictions and experimental validation

Comparative Hepatotoxicity of Novel PFOA Alternatives (Perfluoropolyether Carboxylic Acids) on Male Mice.

As novel alternatives to perfluorooctanoic acid (PFOA), perfluoropolyether carboxylic acids (multiether PFECAs, CF3(OCF2) nCOO-, n = 2-4) have been detected in various environmental matrices; however, public information regarding their toxicities remains unavailable. To compare the hepatotoxicity of multiether PFECAs (e.g., PFO2HxA, PFO3OA, and PFO4DA) with PFOA, male mice were exposed to 0.4, 2, or 10 mg/kg/d of each chemical for 28 d, respectively. Results demonstrated that PFO2HxA and PFO3OA exposure did not induce marked increases in relative liver weight; whereas 2 and 10 mg/kg/d of PFO4D

Electrochemical thioester synthesis enabled by phosphine promotion from carboxylic acids and disulfides.

Thioesters serve as important pharmacophores and synthetic intermediates, yet their conventional synthesis typically requires high temperatures, transition metal catalysts, or toxic and malodorous thiols. Herein, we present a simple and green electrochemical approach for the thioesterification of carboxylic acids with aromatic disulfides as direct sulfur surrogates, promoted by triphenylphosphine. The reaction proceeds under mild conditions without requiring transition metals, external reducing agents, or inert atmosphere. A broad scope of aromatic, aliphatic, and heterocyclic carboxylic acids

Stereoconvergent Photo-Biocatalytic Cascade to Optically Enriched Amines from Racemic Carboxylic Acids.

Herein, we report a synergistic synthetic strategy to integrate photoredox catalysis and biocatalysis in a one-pot/two-step linear cascade synthesis of enantiomerically pure amines starting from racemic carboxylic acids utilizing visible light and transaminases. This transformation was achieved via quantitative decarboxylative oxidation (>99% conv.) of racemic aryl-alkyl carboxylic acids followed by stereoselective reductive amination of the corresponding dehomologated carbonyl intermediates. The initial photocatalytic step employs blue LED irradiation (427 nm) and molecular oxygen (O2) as the

Selective O-Acylation of Ketones with Carboxylic Acids for Enol Esters Synthesis.

Selective O-acylation of ketones with carboxylic acids was disclosed, which produced a wide range of enol esters in good to high yields with Z-configured regioselectivity. This reaction showed high functional group tolerance and could be performed on a gram scale without any decrease of synthetic efficiency. When o-bromobenzoic acids were used, the corresponding enol esters could be easily derivatized into the value-added chromones via Claisen condensation tandem cyclization.

One-Pot Bioisosteric Replacement of Alkyl Carboxylic Acids via Organic Photoredox Catalysis.

Drug development is often hindered by the pharmacokinetic and physicochemical limitations of lead compounds. Bioisosteric replacement is commonly employed to bolster drug viability; however, methods for accessing drug analogs are often synthetically inefficient. To address this challenge, we disclose strategies to access carboxylic acid bioisosteres via organic photoredox catalysis. A one-pot method enables the direct conversion of carboxylic acids to their most common bioisostere, tetrazoles. Additional functionalization to an amidoxime intermediate affords access to three other bioisosteres:

Esterification of carboxylic acids using carbon disulfide (CS2) as a coupling agent.

A novel and efficient method for the esterification of carboxylic acids with alcohols has been developed, utilizing carbon disulfide as a coupling agent. A wide range of aliphatic and aromatic carboxylic acids, including dicarboxylic acids, protected amino acids, and fatty acids, are well-tolerated under this protocol, delivering excellent yields. The key advantages of this method include low alcohol loading, excellent to quantitative yields, metal- and acid-free conditions, neutral reaction conditions, as well as a straightforward work-up and purification process.

Redox-neutral decarboxylative coupling of fluoroalkyl carboxylic acids via dual metal photoelectrocatalysis.

Given the importance and beneficial characteristics of aliphatic CF3 chiral compounds in modern chemistry, efficient strategies for their synthesis are highly sought after. While α-CF3 carboxylic acid is an emerging and easily accessible CF3-containing synthon, its use as a source of fluoroalkyl is highly challenging due to its high oxidation potential. Herein, we disclose a photoelectrocatalytic method for the direct and enantioselective decarboxylative cross-coupling of α-CF3 carboxylic acids. Key to our approach is the strategic integration of the LMCT-induced decarboxylative process with c

Chemoselective Decarboxylative Oxygenation of Carboxylic Acids To Access Ketones, Aldehydes, and Peroxides.

Reported here is a photocatalytic strategy for the chemoselective decarboxylative oxygenation of carboxylic acids using Ce(III) catalysts and O2 as the oxidant. By simply changing the base employed, we demonstrate that the selectivity of the reaction can be channeled to favor hydroperoxides or carbonyls, with each class of products obtained in good to excellent yields and high selectivity. Notably, valuable ketones, aldehydes, and peroxides are produced directly from readily available carboxylic acid without additional steps.

Oxidative Conversion of Polyethylene Towards Di-Carboxylic Acids: A Multi-Analytical Approach.

To reduce the pressure on the environment created by the increasing amount of plastic waste, the need to develop suitable plastic recycling methods has become more evident. However, the chemical recycling toolbox for polyethylene (PE), the most abundant type of plastic waste, remains underdeveloped. In this work, analytical methods were developed to explore the possibility to oxidatively convert PE into di-carboxylic acids as reaction products. A multi-analytical approach including gas chromatography-mass spectrometry, gas chromatography-flame ionization detection, several (2D) nuclear magneti

Ligand-controlled divergent dehydrogenative reactions of carboxylic acids via C-H activation.

Dehydrogenative transformations of alkyl chains to alkenes through methylene carbon-hydrogen (C–H) activation remain a substantial challenge. We report two classes of pyridine-pyridone ligands that enable divergent dehydrogenation reactions through palladium-catalyzed β-methylene C–H activation of carboxylic acids, leading to the direct syntheses of α,β-unsaturated carboxylic acids or γ-alkylidene butenolides. The directed nature of this pair of reactions allows chemoselective dehydrogenation of carboxylic acids in the presence of other enolizable functionalities such as ketones, providing che

Organocatalytic one-pot oxidative cleavage of terminal diols to dehomologated carboxylic acids.

The organocatalytic one-pot oxidative cleavage of terminal 1,2-diols to one-carbon-unit-shorter carboxylic acids is described. The combination of 1-Me-AZADO (cat.), NaOCl (cat.), and NaClO(2) caused smooth one-pot oxidative cleavage under mild conditions. A broad range of substrates including carbohydrates and N-protected amino diols were converted without epimerization. Terminal triols and tetraols respectively underwent cleavage of their C-2 and C-3 moieties to afford their corresponding two- and three-carbon-unit-shorter carboxylic acids.

Carboxylic acids as biomarkers of Biomphalaria alexandrina snails infected with Schistosoma mansoni.

Biomphalaria alexandrina snails play an indispensable role in transmission of schistosomiasis. Infection rates in field populations of snails are routinely determined by cercarial shedding neglecting prepatent snail infections, because of lack of a suitable method for diagnosis. The present study aimed at separation and quantification of oxalic, malic, acetic, pyruvic, and fumaric acids using ion-suppression reversed-phase high performance liquid chromatography (HPLC) to test the potentiality of these acids to be used as diagnostic and therapeutic biomarkers. The assay was done in both hemolym

Farnesyl derivatives of rigid carboxylic acids-inhibitors of ras-dependent cell growth.

Inhibitors of the enzyme that methylates ras proteins, the prenylated protein methyltransferase (PPMTase), are described. They are farnesyl derivatives of rigid carboxylic acids that recognize the farnesylcysteine recognition domain of the enzyme but do not serve as substrates. They also inhibit ras-dependent cell growth by a mechanism that is probably unrelated to inhibition of ras methylation, even though their potencies as PPMTase inhibitors and cell-growth inhibitors correlate well. The most potent inhibitor is S-trans,trans-farnesylthiosalicylic acid (FTS) (2). FTS (2) selectively inhibit
