ATP1A1/BCL2L1 predicts the response of myelomonocytic and monocytic acute myeloid leukemia to cardiac glycosides.

Myelomonocytic and monocytic acute myeloid leukemia (AML) subtypes are intrinsically resistant to venetoclax-based regimens. Identifying targetable vulnerabilities would limit resistance and relapse. We previously documented the synergism of venetoclax and cardiac glycoside (CG) combination in AML. Despite preclinical evidence, the repurposing of cardiac glycosides (CGs) in cancer therapy remained unsuccessful due to a lack of predictive biomarkers. We report that the ex vivo response of AML patient blasts and the in vitro sensitivity of established cell lines to the hemi-synthetic CG UNBS1450

Na+/K+-ATPase as a Target of Cardiac Glycosides for the Treatment of SARS-CoV-2 Infection.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), identified for the first time in Wuhan, China, causes coronavirus disease 2019 (COVID-19), which moved from epidemic status to becoming a pandemic. Since its discovery in December 2019, there have been countless cases of mortality and morbidity due to this virus. Several compounds such as chloroquine, hydroxychloroquine, lopinavir-ritonavir, and remdesivir have been tested as potential therapies; however, no effective treatment is currently recommended by regulatory agencies. Some studies on respiratory non-enveloped viruses such as

Modification of atrioventricular conduction under the combined influence of a cardiac glycoside and a calcium antagonist in the dog.

Conduction disorders may be logically expected from the digoxin-verapamil association, since each of these drugs is known to increase conduction time (CT) and effective refractory period (ERP) in the atrioventricular (AV) node. When AV conduction is considerably depressed by verapamil (1.27 mg/kg over 90 min) in the absence of vagal tone, digoxin, infused at 1 microgram/kg/min rate over 40 min, elicits a progressive but incomplete regression of the verapamil effects, as does hypercalcaemia up to 5.5 mmol/l. Infused at the 2.5 microgram/kg/min rate over 20 min, its antagonistic effects, like th

Studies on South African cardiac glycosides. II. Observations on the clinical and haemodynamic effects of cotyledoside.

In guinea-pigs, the oral and subcutaneous LD50 values were very similar (cf. 0,173 mg/kg over 48 h with 0,116 mg/kg over 24 and 48 h). When dosed subcutaneously, a cumulative effect was observed. Intravenous administration of cotyledoside to anaesthetized guinea-pigs resulted in: dyspnoea, increased heart rates and blood pressures, and electrocardiagraphic changes typical of cardiac glycoside poisoning. A positive cardiac inotropic effect was succeeded by a positive chronotropic one. In sheep, acute and subacute intoxication resulted in ruminal, respiratory and cardiac changes. The signs inclu

Study on differential distribution of cardiac glycoside extract from Periplocae cortex in normal and heart failure rat cardiac tissue.

Periplocae Cortex (PC) is the dried root bark of Periploca sepium Bunge, a plant belonging to the Asclepiadaceae family. Its cardiac glycoside compounds exhibit significant biological activity on the heart and demonstrate excellent therapeutic efficacy in treating chronic heart failure. This study employed ultra-performance liquid chromatography-triple quadrupole tandem mass spectrometry (UPLC-TQ-S-MS/MS) to establish a simultaneous analytical method for determining periplocin and its four metabolites (periplogenin, periplocymarin, gomphogenin, and 17α-asclepioside) in rat cardiac tissue, foll

Cardiac glycosides provide neuroprotection against ischemic stroke: discovery by a brain slice-based compound screening platform.

We report here the results of a chemical genetic screen using small molecules with known pharmacologies coupled with a cortical brain slice-based model for ischemic stroke. We identified a small-molecule compound not previously appreciated to have neuroprotective action in ischemic stroke, the cardiac glycoside neriifolin, and demonstrated that its properties in the brain slice assay included delayed therapeutic potential exceeding 6 h. Neriifolin is structurally related to the digitalis class of cardiac glycosides, and its putative target is the Na(+)/K(+)-ATPase. Other cardiac glycoside comp

[Acute electrophysiological effects of cardiac glycosides on sinus node function and ventricular repolarization under clinical conditions].

The influence of digoxin, digitoxin and g-strophanthin (ouabain) on the sinus node function (recovery time, sinoatrial conduction time, PP-interval) and on ventricular repolarisation (corrected QT interval and QT during permanent atrial stimulation) was studied in 101 patients (36 with sick sinus syndrome-SSS, 34 with ischaemic heart disease-IHD, and 31 control patients without cardiac disease). Digoxin caused marked prolongation of sinoatrial conduction time (in SSS and in control patients), digitoxin prolonged the PP interval (in IHD and SSS patients); digitoxin and ouabain shortened the QTc

Ultrasonic extraction of cardiac glycosides from Adonis amurensis Regel et Radde roots: process optimization and content determination.

This study optimised the ultrasonic-assisted extraction of cardiac glycosides from Adonis amurensis Regel et Radde roots, increasing the extraction yield by 51.30% to 87.42 mg/g. The optimal conditions were ethanol 30%, liquid-to-solid ratio 25:1 (mL/g), ultrasonic time 58 min, temperature 67 °C, and power 250 W. For the first time, high-performance liquid conditions were established to separate four cardiac glycosides. After optimisation, the contents of apocynin and convallatoxin increased 3-4 times compared with the hot water bath method. Scanning Electron Microscope imaging confirmed that

Deciphering Multitarget Mechanisms of Cardiac Glycosides in Acute Myeloid Leukemia Using a Network Pharmacology and Molecular Docking.

PURPOSE: Acute myeloid leukemia (AML) remains the most aggressive form of leukemia, underscoring the urgent need for novel treatment strategies. Drug repurposing offers a promising approach to accelerate drug development process. Cardiac glycosides (CGs), traditionally used for heart conditions, have shown potential for AML therapy. As leukemic stem cells (LSCs) are key contributors to AML relapse and chemotherapy resistance, this study aims to elucidate the potential shared mechanisms of proscillaridin (PSN) and ouabain (OUA), two CGs with anti-LSC activity, focusing on their multitarget effe

Effect of digoxin and amino sugar cardiac glycoside (ASI-222) on plasma antidiuretic hormone activity.

Digoxin acts at central neural (CNS) as well as peripheral sites after intravenous administration. In contrast, the analog, 3-beta-O(4-amino-4,6-dideoxy-beta-D-galactopyranosyl)-digitoxigenin (ASI-222), cannot cross the blood-brain barrier so it acts only at sites outside the CNS. The effects of these two agents on plasma antidiuretic hormone activity (ADH) were investigated in conscious dogs. Despite previous evidence that digoxin produces reflex decreases in sympathetic nerve activity by activating ventricular receptors with vagal afferents, no decreases in ADH were detected when either digo

Dependence of the cardiac uptake of digitalis glycosides on the extracellular calcium concentration in guinea pig isolated hearts.

The purpose of the present study was to investigate, at the myocardial level, the divergent influences of Ca2+ on the action of various cardiotonic steroids. Therefore, the cardiac uptake of 3-H-digitoxin and 3-H-digitoxin was studied in experiments on guinea pig isolated hearts. The following results were obtained: with digitoxin the increase of the extracellular calcium concentration from 0.45 upto 7.2 mM resulted in a concomitant decrease of the myocardial glycoside uptake from about 1.8 nmoles/g wet weight down to about 1.2 nmoles/g wet weight. Similar results were obtained when digoxin up

Inhibition of the sodium pump in squid giant axons by cardiac glycosides: dependence on extracellular ions and metabolism.

1. The rate of inhibition of the Na pump by ouabain was examined both by direct measurement of the rate of decline of the Na efflux and by the binding of [(3)H]ouabain.2. The onset of inhibition of the Na efflux was concentration-dependent; but did not follow simple first order kinetics. The time course of inhibition was roughly exponential although in about 30% of the axons inhibition was preceded by a transient stimulation of the Na efflux.3. Inhibition of the Na efflux by both ouabain and strophanthidin was apparently irreversible.4. The onset of inhibition was slowed markedly at low temper

Targeting stress induction of GRP78 by cardiac glycoside oleandrin dually suppresses cancer and COVID-19.

BACKGROUND: Despite recent therapeutic advances, combating cancer resistance remains a formidable challenge. The 78-kilodalton glucose-regulated protein (GRP78), a key stress-inducible endoplasmic reticulum (ER) chaperone, plays a crucial role in both cancer cell survival and stress adaptation. GRP78 is also upregulated during SARS-CoV-2 infection and acts as a critical host factor. Recently, we discovered cardiac glycosides (CGs) as novel suppressors of GRP78 stress induction through a high-throughput screen of clinically relevant compound libraries. This study aims to test the possibility th
