Reemerging of Encephalomyocarditis Virus in Pigs in Brazil: Pathological and Viral Characterization.

Encephalomyocarditis virus (EMCV) is a zoonotic disease caused by a highly pathogenic virus that affects wild and domestic animal species, with rodents as its reservoir. Most recently, in South America, this virus was detected in an outbreak affecting humans in Peru. In Brazil, EMCV infection was described in some wild species, in horses, and once in farrowing pigs. The aim of this study is to report the reemergence of EMCV in commercial growing-finishing pigs from two different farms that experienced sudden pig death in midwest Brazil. This aim was achieved through gross pathology, histologic

Extracellular vesicles engineered to directly target encephalomyocarditis virus ameliorates multi-organ viremia in a lethal infection model.

The outbreak and prevalence of encephalomyocarditis virus (EMCV) causes significant global mortality and morbidity to the pig industry. Though the current and most effective approach to control EMCV outbreak are done through inactivated vaccines, we have yet to see an effective antiviral agent that directly targets EMCV. Here, we present a molecular therapy consisting of extracellular vesicles (EVs) decorated with EMCV-specific single-chain variable fragment (scFv), engineered on the external loop of the EVS transmembrane domain CD63. These EMCV-scFv enriched EVs directly neutralizes infectiou

TDP-43 proteinopathy in Theiler's murine encephalomyelitis virus infection.

TDP-43, an RNA-binding protein that is primarily nuclear and important in splicing and RNA metabolism, is mislocalized from the nucleus to the cytoplasm of neural cells in amyotrophic lateral sclerosis (ALS), and contributes to disease. We sought to investigate whether TDP-43 is mislocalized in infections with the acute neuronal GDVII strain and the persistent demyelinating DA strain of Theiler's virus murine encephalomyelitis virus (TMEV), a member of the Cardiovirus genus of Picornaviridae because: i) L protein of both strains is known to disrupt nucleocytoplasmic transport, including transp

Detection of antibodies against Theiler's murine encephalomyelitis virus GDVII strain in experimental guinea pigs.

A disease affecting guinea pigs called 'guinea pig lameness' characterized by clinical signs of depression, lameness of limbs, flaccid paralysis, weight loss and death within a few weeks was first described by Römer in 1911. After a research group in our facility kept laboratory guinea pigs from two different origins together in one room, lameness was observed in two animals. Further investigations revealed a serological immune response against Theiler's murine encephalomyelitis virus (TMEV; GDVII strain) in these animals. Histopathology of the lumbar spinal cord of these animals showed mononu

Antigen-specific CD8 T cells accumulate and functionally reactivate in the bone marrow following brain infection.

BACKGROUND: Bone marrow is a primary immune organ responsible for hematopoiesis, yet its role in shaping adaptive immune responses remains incompletely understood. In particular, whether antigen-specific CD8 T cell responses can arise and persist within the bone marrow following brain infection is unclear. METHODS: We used intracranial infection with Theiler's Murine Encephalomyelitis Virus (TMEV) in C57BL/6 mice, along with complementary antigen delivery (OVA + Poly I:C) and systemic infection (LCMV) models. Antigen-specific CD8 T cells were quantified in the brain, secondary lymphoid organs

Occurrence of neuronal dysfunction in mice with virus- or toxic-induced lesions in the central nervous system.

Diseases of the central nervous system (CNS) often include neuronal damage. This study examined proteins crucial to apoptosis and neuronal signalling for identifying causes of neuronal dysfunction in two animal models: Theiler's murine encephalomyelitis virus-induced demyelinating disease (TMEV-IDD) and ethidium bromide (EtBr)-induced focal spinal cord injury. Transcriptomic analysis revealed increased levels of pro-apoptotic molecules including Caspase 8 (Casp8) and Cathepsin B (CTSB), but no significant changes in synaptic proteins including synaptophysin, synaptosomal-associated protein of

The Safety of Alcaligenes Lipid A in a Virus-Induced Immune Disease Model Associated with IgA, Th17 Cells, and Microbiota.

Lipid A is a component of lipopolysaccharide (LPS) of Gram-negative bacteria. Previously, we demonstrated that synthesized lipid A derived from Alcaligenes faecalis (ALA) could enhance antigen-specific immunoglobulin (Ig) A and T helper (Th) 17 responses, when ALA was co-administered experimentally with an antigen as a vaccine adjuvant. This raised concerns about the safety of the ALA usage, since IgA and Th17 responses have been suggested to play a pathogenic role in several immune-mediated diseases, including multiple sclerosis (MS). We investigated whether ALA administrations could exacerba

Transcriptome analysis following neurotropic virus infection reveals faulty innate immunity and delayed antigen presentation in mice susceptible to virus-induced demyelination.

Viral infections of the central nervous system cause acute or delayed neuropathology and clinical consequences ranging from asymptomatic courses to chronic, debilitating diseases. The outcome of viral encephalitis is partially determined by genetically programed immune response patterns of the host. Experimental infection of mice with Theiler's murine encephalomyelitis virus (TMEV) causes diverse neurologic diseases, including TMEV-induced demyelinating disease (TMEV-IDD), depending on the used mouse strain. The aim of the present study was to compare initial transcriptomic changes occurring i

[Isolation, identification and full-length genome sequence analysis of encephalomyocarditis virus from local aardvarks].

Encephalomyocarditis virus (EMCV) is a natural epidemic zoonotic pathogen. However, no reports have been published regarding the isolation, identification and full-length genome of EMCV from a local aardvark population. In present study, an EMCV isolate HNXX13 was isolated from aardvarks named Huainan-pig in Henan Province. The systematic identification, full-length genome sequencing and molecular characteristic analysis of the isolate HNXX13 were conducted. The result showed that the isolate was spherical with a diameter of 24-30 nm, neither heat- nor acid-resistant, sensitive to trypsin, ins

A comparison of the neurotropism of Theiler's virus and poliovirus in CBA mice.

Theiler's murine encephalomyelitis virus (TMEV) and poliovirus infect the central nervous system (CNS) and cause neurological damage. The exact route by which TMEV and polioviruses enter the CNS remains, for the most part, unknown, although the neural and/or the hematogenous pathway have both been postulated. To explore these hypotheses, this research focuses on both the site of entry and the pathway used to invade the CNS. Following different inoculation sites of the GDVII strain of Theiler's virus or Lansing Type 2 poliovirus in CBA mice, the incidence of paralysis and/or encephalitis was ev

Therapeutic effects of estrogens on inflammatory demyelination in a mouse model of multiple sclerosis.

Multiple Sclerosis (MS) is a neurodegenerative autoimmune disease in which the immune system targets the myelin sheath of nerve axons, leading to a variety of neurological signs. Sex hormones are thought to be important factors in MS since pregnancy and oral contraceptives reduce both symptoms and relapses. During pregnancy, remissions occur primarily in the third trimester when estradiol and estriol are elevated. We have investigated the role of these two hormones in an experimental model of virus-induced MS. Theiler's murine encephalomyelitis virus (TMEV) causes an inflammatory demyelinating

Diet composition and sterilization modifies intestinal microbiome diversity and burden of Theiler's virus infection-induced acute seizures.

OBJECTIVE: Brain infection with Theiler's murine encephalomyelitis virus (TMEV) in C57BL/6J mice can induce acquired epileptogenesis. Diet alters acute seizure incidence in TMEV-infected mice; yet it is unclear whether intestinal dysbiosis may also impact acute or chronic behavioral comorbidities. This study thus assessed the impact of diet formulation and sterilization on acute seizure presentation, gut microbiome composition, and epilepsy-related chronic behavioral comorbidities. METHODS: Baseline fecal samples were collected from male C57BL/6J mice (4- to 5-weeks-old; Jackson Labs) upon fa

Suppression of injuries caused by a lytic RNA virus (mengovirus) and their uncoupling from viral reproduction by mutual cell/virus disarmament.

Viruses often elicit cell injury (cytopathic effect [CPE]), a major cause of viral diseases. CPE is usually considered to be a prerequisite for and/or consequence of efficient viral growth. Recently, we proposed that viral CPE may largely be due to host defensive and viral antidefensive activities. This study aimed to check the validity of this proposal by using as a model HeLa cells infected with mengovirus (MV). As we showed previously, infection of these cells with wild-type MV resulted in necrosis, whereas a mutant with incapacitated antidefensive ("security") viral leader (L) protein indu

Ido1 or Ido2 deficiency in myeloid-derived cells attenuates TMEV-induced ictogenesis.

Viral encephalitis is a serious medical condition that causes neuroinflammation, neurodegeneration, cognitive deficits and seizures (ictogenesis), predisposing patients to epilepsy. In a preclinical setting, intracranial infection of C57BL/6 mice with Theiler's murine encephalomyelitis virus (TMEV) is the best-characterized animal model of viral encephalitis resulting in ictogenesis and temporal lobe epilepsy. Macrophages play a critical yet unclear role during encephalomyelitis: macrophage depletion reduces TMEV-induced ictogenesis, whereas prevention of macrophage infiltration into the brain

Microbial contaminations of laboratory mice and rats in Taiwan from 2004 to 2007.

Limited data are available on the pathogen status of contemporary rodent colonies in Taiwan. Here we summarized the rodent pathogen diagnostic records of the Taiwan National Laboratory Animal Center during a 4-y period that representing approximately 10% of the rodent colonies in Taiwan. Demand for pathogen diagnostic service increased continuously from 2004 to 2007, with a 20% increase each year. In 2007, more than 20% of the mouse colonies were positive for mouse parvovirus, mouse hepatitis virus, Theiler murine encephalomyelitis virus, and Mycoplasma pulmonis, with fewer colonies diagnosed

Pathogenesis of encephalomyocarditis experimental infection in young piglets: a potential animal model to study viral myocarditis.

The pathogenesis of encephalomyocarditis (EMC) due to the EMC virus (EMCV) was studied in 24 piglets oro-nasally infected with the field isolate B279/95. Two pigs were kept as negative controls and were euthanised at hour 0. The remaining 24 were euthanised every 6 h up to 78-h post infection (hpi). Virus isolation, histological examination and EMCV immunodetection were performed on the spleen, intestine, pancreas, liver, kidneys, heart, lungs, lymph nodes, tonsils and brain. EMCV was isolated at 6-hpi from the intestine and lymph nodes and at 12-hpi from the heart. From 6 to 12-hpi, scattered

The "DDVF" motif used by viral and bacterial proteins to hijack RSK kinases mimics a short linear motif (SLiM) found in proteins related to the RAS-ERK MAP kinase pathway.

Proteins of pathogens such as cardioviruses, Kaposi sarcoma-associated herpes virus, varicella zoster virus and bacteria of the genus Yersinia were previously shown to use a common "DDVF" (D/E-D/E-V-F) short linear motif (SLiM) to hijack cellular kinases of the RSK (p90 ribosomal S6 kinases) family. Notably, the leader (L) protein of Theiler's murine encephalomyelitis virus (TMEV), a cardiovirus, and protein YopM of Yersinia species were shown to act as adapters to retarget RSKs toward unconventional substrates, nucleoporins and pyrin, respectively. Remarkable conservation of the SLiM docking

Genome Sequencing of Historical Encephalomyocarditis Viruses from South Africa Links the Historical 1993/4 Savanna Elephant (Loxodonta africana) Outbreak to Cryptic Mastomys Rodents.

From 1993 to 1994, 64 free-ranging elephants (Loxodonta africana) succumbed to encephalomyocarditis in the Kruger National Park, South Africa, of which 83% were adult bulls. Mastomys rodents were implicated as the reservoir host of the Encephalomyocarditis virus (EMCV) based on serology and RT-PCR. However, in the absence of sequence-confirmation of both the virus and the rodent host, definitive links between the elephant outbreak strains and rodent reservoir could not be established. In this study, we generate the first reference genome sequences for three historical EMCVs isolated from two M
