Effect of carnosine administration on certain metabolic parameters in bilharzial infected hamsters.

This study was designed to test the ability of carnosine to cure the metabolic disturbances induced by Schistosoma mansoni parasite. Results indicate that parasitic infection caused elevation of liver weight/body weight of S. mansoni infected hamsters, induced lipid peroxidation and reduced glycogen level. Moreover, the adenylate energy charge (AEC), ATP/ADP and ATP/AMP concentration ratios were markedly lower in infected hamsters. Administration of carnosine (10 mg/day) for 15 days either concurrent with infection, 2 and 4 weeks post exposure was effective in reducing worm burden and egg coun

An Engineered β-Aminopeptidase With Better Product Selectivity for L-Carnosine Production.

Efficient enzymaticsynthesis of L-carnosine (L-Car) is often limited by competing hydrolysis and side reactions that reduce substrate utilization and product yield. In this study, a β-aminopeptidase (CbAP) from Caulobacter sp. was systematically engineered to enhance catalytic selectivity toward L-Car using β-alaninamide and L-histidine as substrates. Analysis of β-alaninamide-derived byproducts revealed two major competing pathways, namely β-alaninamide hydrolysis and self-ligation into short oligopeptides. To evaluate the balance between L-Car synthesis and competing reactions, a composite a

Carnosine Potentiates a Compensatory Mitochondrial-Synaptic Proteomic Response in the ALS Cerebellum.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by progressive motor neuron degeneration and chronic neuroinflammation in the brain and spinal cord, involving complex interactions between neurons and immune cells. Carnosine (β-alanyl-L-histidine) has pathophysiological relevance due to its ability to detoxify reactive carbonyl species, including α,β-unsaturated aldehydes, scavenge free radicals, and chelate zinc, and has also been proposed to function in the central nervous system as a histidine reservoir for histamine synthesis. Here, we investigated the

Unfolding the mechanism of cisplatin induced pathophysiology in spleen and its amelioration by carnosine.

cis-Diamminedichloroplatinum (cisplatin) is an effective chemotherapeutic and is widely used for the treatment of various types of solid tumors. Bio-distribution of cisplatin to other organs due to poor targeting towards only cancer cells constitutes the backbone of cisplatin-induced toxicity. The adverse effect of this drug on spleen is not well characterized so far. Therefore, we have set our goal to explore the mechanism of the cisplatin-induced pathophysiology of the spleen and would also like to evaluate whether carnosine, an endogenous neurotransmitter and antioxidant, can ameliorate thi

Protective effects of L-carnosine on CCl4 -induced hepatic injury in rats.

The present study was undertaken to investigate the possible protective effect of L-carnosine (CAR), an endogenous dipeptide of alanine and histidine, on carbon tetrachloride (CCl4)-induced hepatic injury. Liver injury was induced in male Sprague-Dawley rats by intraperitoneal (i.p.) injections of CCl4, twice weekly for six weeks. CAR was administered to rats daily, at dose of 250 mg/kg, i.p. At the end of six weeks, blood and liver tissue specimens were collected. Results show that CAR treatment attenuated the hepatic morphological changes, necroinflammation and fibrosis induced by CCl4, as i

The effect of L-carnosine on the circadian resetting of clock genes in the heart of rats.

It is reported that the circadian timing system may be included in the mechanism by which L-carnosine (Car) affects multiple physiological alterations including blood glucose, cardiovascular functions etc. However, it is not clear whether Car would affect the circadian rhythm of clock genes in the heart and what is the possible mechanism underlying. To clarify these issues, we compared the effects of Car on the expression of circadian genes in the heart of normal and vagotomized rats under control and jet lag conditions. The normal and vagotomized (va) male Wistar rats were divided into three

Carnosine-like immunoreactivity in the olfactory bulb of the rat: an electron microscopic study.

Carnosine-immunoreactive primary olfactory nerve terminals are demonstrated in the glomerular layer of the rat olfactory bulb by immunoelectron microscopy. Asymmetrical synapses between dendrites of mitral/tufted cells and that of periglomerular cells could be observed. In the accessory olfactory system, carnosine-like immunoreactivity is also detected in the vomeronasal neurons.

Effects of carnosine on olfactory bulb EEG, evoked potentials and DC potentials.

Carnosine is a dipeptide found in great quantities in the primary olfactory nerve and has been suggested to be the neurotransmitter of the olfactory receptor axons. The aim of the present study was to describe some of its electrophysiological actions in the olfactory bulb (OB) of rabbits under anesthesia. Carnosine as a 10% solution in amounts of 2-5 microliter was injected to the OB at the level of the glomerular layer by means of a pipette attached to a Hamilton syringe. Average evoked potentials (AEPs) on the stimulation of the lateral olfactory tract (LOT-AEP), electroencephalographic (EEG

Reduced Muscular Carnosine in Proximal Myotonic Myopathy-A Pilot 1H-MRS Study.

OBJECTIVE: Myotonic dystrophy type 2 (proximal myotonic myopathy, PROMM) is a progressive multisystem disorder with muscular symptoms (proximal weakness, pain, myotonia) and systemic manifestations such as diabetes mellitus, cataracts, and cardiac arrhythmias. A hallmark feature is the selective degeneration of type-II fibers, likely driven by chronic myotonia and sustained metabolic stress. In this study, proton magnetic resonance spectroscopy (1H-MRS) was applied to assess intramuscular carnosine as a potential noninvasive marker of type-II fiber integrity, alongside extramyocellular lipids

Carnosine ameliorates postoperative cognitive dysfunction of aged rats by limiting astrocytes pyroptosis.

Postoperative cognitive dysfunction (POCD) is a common postoperative complication in elderly patients, and neuroinflammation is a key hallmark. Recent studies suggest that the NOD-like receptor family, pyrin domain containing 3 (NLRP3) inflammasome-mediated astrocytes pyroptosis is involved in the regulation of neuroinflammation in many neurocognitive diseases, while its role in POCD remains obscure. Carnosine is a natural endogenous dipeptide with anti-inflammatory and neuroprotective effects. To explore the effect of carnosine on POCD and its mechanism, we established a POCD model by explora

Effects of carnosine supplementation on physical endurance: a placebo-controlled randomized clinical trial.

BACKGROUND: Carnosine (β-alanine-L-histidine) is an endogenous dipeptide found in abundance in highly metabolic tissues such as skeletal muscle and the brain, where it is thought to play a role in intracellular buffering, and thereby, promote anaerobic glycolysis. Due to its perceived ability to support muscle energetics, carnosine, and its precursor, β-alanine, have found widespread use as performance-enhancing supplements. Nevertheless, the utility and efficacy of carnosine in enhancing physical performance have not been rigorously evaluated. METHODS: We conducted a placebo-controlled clini

Periodontitis-Associated Circulating EVs Promote Colorectal Cancer Progression via Carnosine-Mediated Acidosis Adaptation.

Colorectal cancer (CRC) is the third most common malignancy worldwide. Epidemiological studies have suggested a positive association between periodontitis (PD) and CRC risk; however, the mechanistic basis underlying this relationship remains unclear. Extracellular vesicles (EVs) represent an important mode of systemic communication and may mediate the distal effects between PD and CRC. PD model was established in Apc+/- mice with spontaneous intestinal tumorigenesis. Tumour onset, burden, and progression were evaluated in the colorectum and small intestine. Circulating EVs were isolated from t

Carnosine alleviates cisplatin-induced acute kidney injury by targeting Caspase-1 regulated pyroptosis.

Acute kidney injury (AKI) is a syndrome characterized by rapid loss of renal excretory function. Its underlying mechanisms remain unclear. Pyroptosis, a form of programmed cell death, plays an important role in AKI. It is characterized by cell swelling and membrane rupture, triggering the release of cellular contents and activating robust inflammatory responses. Carnosine, a dipeptide with antioxidant and anti-inflammatory properties, has therapeutic effects in AKI. However, the mechanism by which carnosine treats AKI-associated pyroptosis remains unexplored. In this study, we investigated the

Treatment with carnosine reduces hypoxia-ischemia brain damage in a neonatal rat model.

Perinatal hypoxia-ischemia brain damage (HIBD) is a major cause of mortality and morbidity in neonates, and there is currently no effective therapy for HIBD. Carnosine plays a neuroprotective role in adult brain damage. We have previously demonstrated that carnosine pretreatment protects against HIBD in a neonatal rat model. Therefore, we hypothesized that treatment with carnosine would also have neuroprotective effects. Hypoxia-ischemia was induced in rats on postnatal days 7-9 (P7-9). Carnosine was administered intraperitoneally at a dose of 250mg/kg at 0h, 24h, and 48h after hypoxia-ischemi
