Immunohistochemical analysis of procathepsin L and cathepsin B in cutaneous Kaposi's sarcoma.

BACKGROUND: The expression of the proteinases cathepsins L and B are induced in tumors by malignant transformation, growth factors, and tumor promoters suggesting they play an important role in tumor invasion and metastasis. METHODS: By immunohistochemistry, procathepsin L and cathepsin B were studied in patients with Kaposi's sarcoma (KS). Formalin-fixed and paraffin-embedded tissues from 29 cases of KS (AIDS-associated KS, n = 24; non-AIDS-associated KS, n = 5) were immunolabeled with the polyclonal antibody directed against procathepsin L and the antisera directed against cathepsin B. A Review of Small Molecule Inhibitors and Functional Probes of Human Cathepsin L.

Human cathepsin L belongs to the cathepsin family of proteolytic enzymes with primarily an endopeptidase activity. Although its primary functions were originally thought to be only of a housekeeping enzyme that degraded intracellular and endocytosed proteins in lysosome, numerous recent studies suggest that it plays many critical and specific roles in diverse cellular settings. Not surprisingly, the dysregulated function of cathepsin L has manifested itself in several human diseases, making it an attractive target for drug development. Unfortunately, several redundant and isoform-specific func

Screening and Identification of Inhibitors of Trypanosoma brucei Cathepsin L with Antitrypanosomal Activity.

Current treatment options for human African trypanosomiasis (HAT) are ineffective, and they have several well-known clinical limitations. In our continued efforts to identify chemotypes that can be developed into clinically useful drugs, we screened a targeted compound library against the major cathepsin L (rhodesain) in T. brucei. We report the antirhodesain activity and antitrypanosomal activity of the compounds in this letter. The identified compounds can serve as starting points for structure- and/or phenotype-based lead optimization strategy against Trypanosoma brucei.

Overexpression of cysteine cathepsin L is a marker of invasion and metastasis in ovarian cancer.

Cysteine cathepsins (CTSs) are involved in the degradation and remodeling of the extracellular matrix and are associated with cellular transformation, differentiation, motility and adhesion in cancer development. Previous studies indicate that CTSs may be involved in ovarian cancer invasion and metastasis. However, due to the lack of large sample clinical studies and direct experimental evidence for the relationship between the expression of CTSs and invasion and metastasis, the diagnostic and prognostic value of CTSs in ovarian cancer progression has not been elucidated. In the present study,

Effects of an inhibitor of cathepsin L on bone resorption in thyroparathyroidectomized and ovariectomized rats.

The process of bone resorption by osteoclasts involves the dissolution of mineral salts and enzymatic degradation of the mainly collagenous extracellular matrix. Cysteine proteinases, which can efficiently degrade collagen at acidic pH, have been suggested to play an important role in the bone resorptive process. The cysteine proteinase cathepsin L is secreted by osteoclasts, and inhibitors of this enzyme can prevent bone resorption in vitro. The activity of acetyl-leu-leu-norleucinol (ALLN), a selective inhibitor of cathepsin L, was investigated in two models of bone resorption in vivo. In th

Kinetic Characterization of Inhibition of Cathepsins L and S by Peptides With Anticancer Potential.

Cysteine cathepsins have been suggested as attractive therapeutic targets due to their critical role in several pathologies. In particular, inhibitors of cysteine cathepsins reduce the viability of tumor cells. The present study uses enzyme kinetics to characterize the interaction of human cathepsins L and S with their peptide substrate acetyl-QLLR-7-amino-4-methylcoumarin (Ac-QLLR-AMC) and peptide inhibitors with anti-tumor activity: FFSFGGAL (CS-PEP1) and acetyl-PLVE-fluoromethyl-ketone (Ac-PLVE-fmk). Due to multiple cellular locations of cathepsins, our study is conducted under different pH

Molecular characterization of EcCLP1, a new putative cathepsin L protease from Echinococcus canadensis.

Echinococcus granulosus sensu lato is a platyhelminth parasite and the etiological cause of cystic echinococcosis (CE), a zoonotic and neglected disease that infects animals and humans worldwide. As a part of the biological arsenal of the parasite, cathepsin L proteases are a group of proteins that are believed to be essential for parasite penetration, immune evasion, and establishment in the tissues of the host. In this work, we have cloned and sequenced a new putative cathepsin L protease from Echinococcus canadensis (EcCLP1). The bioinformatic analysis suggests that EcCLP1 could be synthesi

Glomerular Damage in Trichloroethylene-Sensitized Mice: Targeting Cathepsin L-Induced Hyperactive mTOR Signaling.

Trichloroethylene (TCE) is a serious health hazard for workers with daily exposure, causing occupational medicamentosa-like dermatitis due to TCE (OMDT) and glomerular damage. Recent studies suggest that mTORC1 signaling is activated in various glomerular disorders; however, the role of mTORC1 signaling in TCE-induced glomerular damage remains to be explored. In the present study, 6 OMDT patients were enrolled and a TCE-sensitized mouse model was established to investigate molecular mechanisms underlying the glomerular damage associated with OMDT. Glomerular damage was assessed by levels of ur

Molecular and immunological characterization of cathepsin L-like cysteine protease of Paragonimus pseudoheterotremus.

Cathepsin L is a cysteine protease belonging to the papain family. In parasitic trematodes, cathepsin L plays essential roles in parasite survival and host-parasite interactions. In this study, cathepsin L of the lung fluke Paragonimus pseudoheterotremus (PpsCatL) was identified and its molecular biological and immunological features characterized. A sequence analysis of PpsCatL showed that the gene encodes a 325-amino-acid protein that is most similar to P. westermani cathepsin L. The in silico three-dimensional structure suggests that PpsCatL is a pro-enzyme that becomes active when the prop

Substrate inhibition and allosteric regulation by heparan sulfate of Trypanosoma brucei cathepsin L.

The cysteine protease brucipain is an important drug target in the protozoan Trypanosoma brucei, the causative agent of both Human African trypanosomiasis and Animal African trypanosomiasis. Brucipain is closely related to mammalian cathepsin L and currently used as a framework for the development of inhibitors that display anti-parasitic activity. We show that recombinant brucipain lacking the C-terminal extension undergoes inhibition by the substrate benzyloxycarbonyl-FR-7-amino-4-methylcoumarin at concentrations above the K(m), but not by benzyloxycarbonyl-VLR-7-amino-4-methylcoumarin. [Activity of tissue cathepsin-L-like proteinases of women with womb body oncopathology].

Activity and optimal pH of cathepsin-L-like proteinases was studied in benign and malignant tumours of the womb body. In the benign tumors activity of cathepsin-L-like proteinases changes depending on the expansion and depth of extension benign tumour and is defined by proliferative potential of tumour cells of myometrium and endometrium. Activity of cathepsin-L-like proteinases in malignant epithelial tumour of endometrium--adenocarcinoma is inversely proportional to the level of differentiation of the tumour cells.

Development of multi-epitope Cathepsin L driven short peptide vaccine against Fasciola gigantica.

Fasciolosis is an important zoonotic disease caused by Fasciola species (Fasciola spp.). Fasciola spp. infection has the potential to affect the livestock economy. Furthermore, liver flukes have been found to present Triclabendazole resistance in many countries. Vaccines are used to prevent fasciolosis and are currently considered the best alternative. However, no liver fluke vaccine is commercially available at present. Fasciola gigantica Cathepsin Ls (FgCatLs) are vital enzymes for the liver fluke's survival. Therefore, this study aimed to design and investigate the immune response of multi-

Hyperglycemia induced cathepsin L maturation linked to diabetic comorbidities and COVID-19 mortality.

Diabetes, a prevalent chronic condition, significantly increases the risk of mortality from COVID-19, yet the underlying mechanisms remain elusive. Emerging evidence implicates Cathepsin L (CTSL) in diabetic complications, including nephropathy and retinopathy. Our previous research identified CTSL as a pivotal protease promoting SARS-CoV-2 infection. Here, we demonstrate elevated blood CTSL levels in individuals with diabetes, facilitating SARS-CoV-2 infection. Chronic hyperglycemia correlates positively with CTSL concentration and activity in diabetic patients, while acute hyperglycemia augm

Characterization and initial functional analysis of cathepsin K in turbot (Scophthalmus maximus L.).

Cathepsins are the best-known group of proteases in lysosomes, playing a significant role in immune responses. Cathepsin K (CTSK) is abundantly and selectively expressed in osteoclasts, dendritic cells and monocyte-derived macrophages, where it is involved in ECM degradation and bone remodeling. A growing body of evidences have indicated the vital roles of cathepsin K in innate immune responses. Here, one CTSK gene was captured in turbot (SmCTSK) with a 993 bp open reading frame (ORF). The genomic structure analysis showed that SmCTSK had 7 exons similar to other vertebrate species. The synten

Cloning and characterization of a cathepsin L-like cysteine protease from Taenia pisiformis.

Rabbit cysticercosis, caused by the larval stage of Taenia pisiformis, is a serious parasitic disease of rabbits. It was reported that some cysteine peptidases have potential roles in the pathogenesis of various parasitic infections. To investigate the biochemical characteristics and roles in the pathogenesis/host-invasion of cysteine peptidases, a cDNA sequence encoding for a cathepsin L-like cysteine protease (TpCP) was cloned and identified from the T. pisiformis metacestodes. This sequence was 1220 bp in its length, which included a 1017 bp open reading frame encoding a 339 amino acid pept

[Molecular cloning of two Rhipicephalus haemaphysaloides haemaphysaloides cathepsin L-like cysteine proteinase gene].

Ticks are obligate ectoparasites and vectors of arboviruses, vickettsiate, spirochetes and parasitil protozoa of humans and domestic animals. Immunological protection of mammalian hosts against tick infestation has been proposed as the most sustainable alternative tick control method to the current use of acaricides. The success of this method is dependent on the identification of key molecules for use as tick vaccine antigens. Proteolytic enzymes are involved in a wide range of cellular processes, thus they can be considered as good target antigens for a tick vaccine. In the present study, we

An AIE fluorescence sensor enables the discovery of Cathepsin L inhibitors in Xuebijing injection.

Cathepsin L (CTSL), a lysosomal cysteine protease implicated in various human pathological processes, including cancer and COVID-19, represents a promising therapeutic target. Herein, we present a simple and selective aggregation-induced emission (AIE)-based fluorescent sensor for CTSL inhibitor screening. The sensor was constructed by linking an AIE fluorophore with a CTSL-specific substrate peptide and comprehensively characterized using spectroscopic, microscopic, and microplate assay techniques. CTSL-mediated hydrolysis triggered fluorescence quenching via fluorophore deaggregation, wherea
