A Cocktail of Three Virulent Phages Controls Multidrug-Resistant Salmonella Enteritidis Infection in Poultry.

Salmonella enterica is not only the most common pathogen of poultry and poultry-derived products but is also a significant foodborne pathogen. In recent years, many S. enterica isolates have exhibited multi-drug resistance, which places huge pressure on global economy and health. Since phages are an attractive alternative to biocontrol pathogens, we isolated a total of 15 Salmonella phages from sewage effluent, sediment, and chicken manure. The GRNsp1, GRNsp3, GRNsp6, GRNsp21, GRNsp27, GRNsp30, GRNsp50, and GRNsp51 phages exhibited a wide host range against S. enterica serovars Enteritidis and

Isolation, Characterization and Draft Genome Analysis of Bacteriophages Infecting Acidovorax citrulli.

Bacterial fruit blotch and seedling blight, caused by Acidovorax citrulli, is one of the most destructive diseases of melon and watermelon in many countries. Pathogen-free seed and cultural practices are major pillars of the disease control. However, use of bacteriophages as natural biocontrol agents might also contribute to the disease management. Therefore, we isolated 12 bacteriophages specific to A. citrulli, from phyllosphere and rhizosphere of diseased watermelon plants. The phage strains were characterized based on their host range, plaque and virion morphology, thermal inactivation poi

Comparative Genomic Studies of Salmonella Heidelberg Isolated From Chicken- and Turkey-Associated Farm Environmental Samples.

Salmonella is one of the leading causes of human foodborne gastroenteritis in the United States. In addition, Salmonella contributes to morbidity and mortality in livestock. The control of Salmonella is an increasing problematic issue in livestock production due to lack of effective control methods and the constant adaptation of Salmonella to new management practices, which is often related to horizontal acquisition of virulence or antibiotic resistance genes. Salmonella enterica serotype Heidelberg is one of the most commonly isolated serotypes in all poultry production systems in North Ameri

Isolation, characterization, therapeutic potential and depolymerase identification of a lytic bacteriophage Kpp-9 against Klebsiella pneumoniae with capsule serotype K2.

Klebsiella pneumoniae is one of the most threatening multidrug-resistant bacteria, and its inherent capsule and extensive biofilm formation pose a significant barrier to the treatment of infection. Bacteriophages (or phages) and phage-derived depolymerases are attracting attention as potential alternatives to antibiotics. Here, we isolated a lytic phage, named Kpp-9, using a capsule-type K2 strain, Kp09. Kpp-9 could inhibit bacterial biofilm formation, eradicate the mature biofilm in vitro, improve the survival rate of Kp09-infected mice, and alleviate the symptoms of pneumonia in mice. Morpho

[Biological and genome characteristics of a novel broad host-range phage ΦEP1 infecting enteroinvasive Escherichia coli].

We analyzed the biological and genome characteristics of a phage infecting enteroinvasive Escherichia coli (EIEC), aiming to provide resources and a reference for the prevention and treatment of EIEC. With the EIEC preserved in our laboratory as the host bacterium, one strain of phage was isolated from the effluent sample from a chicken farm in Huzhou, Zhejiang and named ΦEP1. The titer, optimal multiplicity of infection, one-step growth curve, temperature, pH value, chloroform and bile salt sensitivity of ΦEP1 were determined by the double-layer agar plate method. The morphology of the phage

Controlling Multidrug-Resistant Staphylococcus aureus by Combined Use of Antimicrobials and Phage STPX-6 with Broad Spectrum and High Efficiency.

OBJECTIVE: The emergence of antibiotic-resistant Staphylococcus aureus, especially methicillin-resistant S. aureus (MRSA), poses a great challenge for animal and public health. This study aimed to isolate a broad-spectrum and high-efficiency MRSA phage and explore the phage-antibiotic synergistic effect on MRSA. RESULTS: Phage STPX-6 belongs to Caudovirales, Podoviridae. It has a hexahedral head and a short tail. Its genome length was 17,007 bp, and it did not contain resistance genes and virulence genes. STPX-6 lysed 79.6% (133/167) of 167 S. aureus and 87.96% (95/108) of MRSA from different

Metagenomic community composition and resistome analysis in a full-scale cold climate wastewater treatment plant.

BACKGROUND: Wastewater treatment plants are an essential part of maintaining the health and safety of the general public. However, they are also an anthropogenic source of antibiotic resistance genes. In this study, we characterized the resistome, the distribution of classes 1-3 integron-integrase genes (intI1, intI2, and intI3) as mobile genetic element biomarkers, and the bacterial and phage community compositions in the North End Sewage Treatment Plant in Winnipeg, Manitoba. Samples were collected from raw sewage, returned activated sludge, final effluent, and dewatered sludge. A total of 2

Bacteriophages Isolated From Turkeys Infecting Diverse Salmonella Serovars.

Salmonella is one of the leading causes of foodborne illnesses worldwide. The rapid emergence of multidrug-resistant Salmonella strains has increased global concern for salmonellosis. Recent studies have shown that bacteriophages (phages) are novel and the most promising antibacterial agents for biocontrol in foods because phages specifically kill target bacteria without affecting other bacteria, do not alter organoleptic properties or nutritional quality of foods, and are safe and environmentally friendly. Due to the vast variation in Salmonella serotypes, large numbers of different and highl

Dysbiosis of gut microbiota in COVID-19 is associated with intestinal DNA phage dynamics of lysogenic and lytic infection.

This study compared intestinal DNA phage dynamics and gut microbiota changes observed at the onset of coronavirus disease 2019 (COVID-19). The study participants included 19 healthy individuals and 19 patients with severe acute respiratory syndrome coronavirus 2 infection. Significant differences were observed in the diversity of the intestinal DNA virome after the onset of COVID-19 compared with that in healthy individuals. Classification by their tail morphology resulted in the order Caudovirales, a double-stranded DNA phage, accounting for >95% of all participants. In classifying phages bas

Newly Isolated Bacteriophages from the Podoviridae, Siphoviridae, and Myoviridae Families Have Variable Effects on Putative Novel Dickeya spp.

Soft rot pathogenic bacteria from the genus Dickeya cause severe economic losses in orchid nurseries worldwide, and there is no effective control currently available. In the last decade, the genus Dickeya has undergone multiple changes as multiple new taxa have been described, and just recently a new putative Dickeya species was reported. This study reports the isolation of three bacteriophages active against putative novel Dickeya spp. isolates from commercially produced infected orchids that show variable host-range profiles. Bacteriophages were isolated through enrichment from Dickeya-infec

Phenotypic and genomic characterization of three lytic bacteriophages against MDR Escherichia coli from livestock in India.

BACKGROUND: Antimicrobial resistance (AMR) in Escherichia coli, particularly among animal-derived isolates, poses a major threat to public and veterinary health. With conventional antibiotics becoming increasingly ineffective against multidrug-resistant (MDR) strains, alternative solutions are urgently needed. Lytic bacteriophages, known for their host specificity and potent antibacterial activity, are promising therapeutic options. However, limited genomic data on phages from diverse ecological contexts hinder a comprehensive understanding of their diversity and functional potential. METHODS:
