The D enantiomers of three naturally occurring antibiotics--cecropin A, magainin 2 amide, and melittin--were synthesized. In addition, the D enantiomers of two synthetic chimeric cecropin-melittin hybrid peptides were prepared. Each D isomer was shown by circular dichroism to be a mirror image of the corresponding L isomer in several solvent mixtures. In 20% hexafluoro-2-propanol the peptides contained 43-75% alpha-helix. The all-D peptides were resistant to enzymatic degradation. The peptides produced single-channel conductances in planar lipid bilayers, and the D and L enantiomers caused equ

OBJECTIVE: To evaluate how stress at the larval stage alters adult mosquito performance and susceptibility to viral infection.
METHODS: We used a model system consisting of Sindbis virus (SINV) and the yellow fever mosquito Aedes aegypti. Larvae were either reared under optimal conditions (control) or exposed to one of four types of stressors; suboptimal nutrients, starvation, elevated temperature, and a low dose of the insecticide malathion and adult females were fed SINV infectious blood meal. Differential expressions of stress, immune-specific and detoxification genes was measured in fourt

In our study of the cecropin locus in Drosophila we have found a gene for a new peptide, andropin, with antibacterial properties. Transcripts from this gene, Anp, could be detected in newly eclosed males and reached steady-state levels after 1 day. Transcription was strongly induced in response to mating and is strictly confined to the ejaculatory duct of adult males. The deduced peptide sequence reveals a hydrophobic amino terminus with striking similarity to the signal peptide of the cecropins. The sequence of the predicted mature andropin shows no direct homology with the cecropins, but the

Black soldier fly larvae meal (BSFM) is increasingly recognised as a sustainable and functional protein source in the livestock feed industry, owing to its high crude protein and rich fatty acid content. Beyond its nutritional value, BSFM has demonstrated antibacterial properties, enhancing resistance in animals to pathogens such as Salmonella species. These findings are critical as antimicrobial resistance continues to rise in the livestock sector. The black soldier fly (BSF) genome encodes antimicrobial peptides (AMPs) that can be classified under families such as cecropins, defensins, and a

Mosquito antimicrobial peptides (AMPs) contribute to innate defense against bacteria, fungi, parasites, and, in some experimental contexts, arboviruses. This narrative review evaluates endogenous mosquito AMP families across Aedes, Anopheles, and Culex species and clearly separates them from heterologous peptides introduced into mosquitoes for transmission-blocking studies. The best-supported endogenous families are cecropins, defensins, gambicin, attacin, and diptericin, although gene repertoires vary among mosquito lineages. Mechanistic evidence is strongest for membrane interaction by indiv

Immune responses have been described for many different insect species. However, it is generally acknowledged that immune systems must therefore differ from those of vertebrates. An effective humoral immune response has been found in pupae of the cecropia moth, Hyalophora cecropia. The expression of this multicomponent system requires de novo synthesis of RNA and proteins and its broad antibacterial activity is due to at least three independent mechanisms, the most well known of which is the insect lysozyme. However, this enzyme is bactericidal for only a limited number of Gram-positive bacter

The in-vitro activity of two vertebrate lytic peptides, two ion transport system inhibitors and two polyether ionophores was investigated against four clinical isolates of Pneumocystis carinii recovered from bronchoalveolar lavages of AIDS patients. The susceptibility tests were performed by inoculating the isolates on to cell monolayers and determining the parasite count after 72 h incubation at 37 degrees C. The culture medium was supplemented with Dulbecco's modified Eagle's medium containing serial dilutions of cecropin P1, magainin II, benzamil, 5-(Nmethyl-Nisobutyl)amiloride (MIBA), lasa

Different species of Leishmania are responsible for cutaneous, mucocutaneous or visceral leishmaniasis infections in millions of people around the world [14]. The adverse reactions caused by antileishmanial drugs, emergence of resistance and lack of a vaccine have motivated the search for new therapeutic options to control this disease. Different sources of antimicrobial molecules are under study as antileishmanial agents, including peptides with antimicrobial and/or immunomodulatory activity, which have been considered to be potentially active against diverse species of Leishmania[7,39]. Ceratitis capitata (medfly) is one of the most devastating crop pests worldwide. The Sterile Insect Technique (SIT) is a control method that is based on the mass rearing of males, their sterilization, and release in the field. However, the effectiveness of the technique depends on the quality of the released males and their fitness. We previously isolated and selected a probiotic bacteria (Enterobacter sp.), from wild-caught medflies, according to criteria that improved biological quality traits of reared medfly males. We firstly evaluated the impact of the irradiation on the expression of diff

Wolbachia are maternally transmitted symbiotic bacteria that can spread within insect populations because of their unique ability to manipulate host reproduction. When introduced to nonnative mosquito hosts, Wolbachia induce resistance to a number of human pathogens, including dengue virus (DENV), Plasmodium, and filarial nematodes, but the molecular mechanism involved is unclear. In this study, we have deciphered how Wolbachia infection affects the Aedes aegypti host in inducing resistance to DENV. The microarray assay indicates that transcripts of genes with functions related to immunity and

Cecropin is a cationic antibacterial peptide composed of 35-39 residues. This peptide has been identified as possessing strong antibacterial activity and low toxicity against eukaryotic cells, and it has been claimed that some types of the cecropin family of peptides are capable of killing cancer cells. In this study, the host effect of cloning antibacterial peptide cecropinB2 was investigated. Three different host expression systems were chosen, i.e., Escherichia coli, Bacillus subtilis and Pichia pastoris. Two gene constructs, cecropinB2 (cecB2) and intein-cecropinB2 (INT-cecB2), were applie

A novel peptide, scorpine, was isolated from the venom of the scorpion Pandinus imperator, with anti-bacterial activity and a potent inhibitory effect on the ookinete (ED(50) 0.7 microM) and gamete (ED(50) 10 microM) stages of Plasmodium berghei development. It has 75 amino acids, three disulfide bridges with a molecular mass of 8350 Da. Scorpine has a unique amino acid sequence, similar only to some cecropins in its N-terminal segment and to some defensins in its C-terminal region. Its gene was cloned from a cDNA library.

Cecropins are antimicrobial peptides (30-35 amino acids) isolated from the hemolymph of the cecropia moth. Previous studies have demonstrated their antimicrobial efficacy against a variety of pathogens, including both gram-positive and -negative bacteria, fungi, protozoa, and enveloped viruses. To assess their therapeutic potential against ocular pathogens, we analyzed the in vitro antimicrobial activity of a synthetic cecropin analog, Shiva-11, against virulent microbial strains (Pseudomonas aeruginosa, Staphylococcus aureus, Streptococcus pneumoniae, and Candida albicans). Bacterial isolates

The characterisation of the antimicrobial activity of five antibacterial peptides, isolated from the venom of the neotropical wandering spider Cupiennius salei is reported here. The peptides have a molecular mass, determined by electrospray ionisation-mass spectrometry, between 3-4 kDa. Minimal inhibitory concentrations against five different bacteria species were determined by a liquid growth inhibition assay. All five peptides showed minimal inhibitory concentrations that are comparable to those of other known antibacterial peptides, like insect defensins and cecropins, found in the last yea

The prophenoloxidase (PPO) activation pathway and the Toll-mediated antimicrobial peptide (AMP) synthesis pathway constitute critical immune responses within the insect immune system. The activation of these pathways is driven by extracellular serine protease cascades, a process counterbalanced by the negative regulation of serpins. In this study, we identified a typical serpin, BmSerpin-7, and characterized its function in the silkworm, Bombyx mori. BmSerpin-7 expression was up-regulated in the hemocytes but down-regulated in the fat body after Micrococcus luteus and Yersinia pseudotuberculos

The black soldier fly (Hermetia illucens) is important for antimicrobial peptide (AMP) research due to its exposure to diverse microorganisms. However, the impact of different fungi on AMP abundance in H. illucens remains unexplored. We studied the induction of AMP expression under basal conditions and with three fungi: non-pathogenic Candida tropicalis, Saccharomyces cerevisiae, and pathogenic Beauveria bassiana, using RNA-sequencing and liquid chromatography with tandem mass spectrometry. Under naive conditions, most AMPs belonged to the lysozyme, cecropin, and defensin classes, with defensi

Insect-based bioactive components are emerging as novel sources of drugs, effective against various diseases. Inflammation is considered to be an innate immune response developed by different organisms against foreign pathogens and cellular stress. However, repetitive elevated inflammation is considered to be responsible for development of many other diseases including colitis and arthritis. Due to the limited activities and side effects of non-steroidal anti-inflammatory drugs, researchers are continuously looking for alternative sources of drug molecules to alleviate the inflammatory related

Venoms have evolved independently several times in Lepidoptera. Limacodidae is a family with worldwide distribution, many of which are venomous in the larval stage, but the composition and mode of action of their venom is unknown. Here, we use imaging technologies, transcriptomics, proteomics, and functional assays to provide a holistic picture of the venom system of a limacodid caterpillar, Doratifera vulnerans Contrary to dogma that defensive venoms are simple in composition, D. vulnerans produces a complex venom containing 151 proteinaceous toxins spanning 59 families, most of which are pep

The effects of anthropogenic chemical contaminants on aquatic organisms are largely influenced by underlying environmental conditions. This study evaluated how larval rearing temperature influences the impact of malathion on the fitness of the yellow fever mosquito Aedes aegypti. Larvae were exposed to water control, and low (0.03mg/L) or high (0.05mg/L) malathion dose at 20°C, 25°C and 30°C and emergence rate, time to emergence, female fecundity and expression of genes encoding two antimicrobial peptides (defensin, cecropin) and an iron-binding protein (transferrin) quantified.
