Germinal centers (GCs) are specialized sites within secondary lymphoid organs where B cells expand, are selected, and mature to produce high-quality antibodies. Their structural complexity makes them difficult to model in vitro. Here, we developed a human 3D lymphoid culture system combining lymphoid and stromal cells to better mimic GC environments than conventional 2D cultures. Tonsil cells were cultured with or without fibroblastic reticular cells (FRCs) in 2D or 3D hydrogels and stimulated with viral antigens or vaccines. FRC-supported 3D cultures significantly improved B and T cell surviv

Drug-induced liver injury (DILI) is a major clinical and regulatory challenge, with cholestasis representing a frequent mechanism of hepatotoxicity. Conventional two-dimensional (2D) hepatocyte cultures often fail to predict such effects, underscoring the need for advanced models that better replicate liver architecture and function. In this study, a three-dimensional (3D) model was developed by encapsulating HepaRG cells within type I collagen hydrogels to enhance functional maturation and reduce differentiation time compared to standard 2D cultures. The 3D HepaRG model maintained long-term v

Cell-based aqueous two-phase system (ATPS) applications involve the construction of 3D cultures encapsulating cells in polyethylene glycol (PEG)-dextran (DEX) droplets due to their low interfacial tension. This technology has enabled cell patterning in a controlled-defined shape giving rise to specific cell microenvironments. The present work aims to encapsulate MCF-7 cancer cells in ATPS droplets evaluating four different compositions of PEG-DEX, construction Strategies A (droplet added), B (immersed), or C (covered), and two cell densities (5000 and 10,000 cells/µL) during a 24 h incubation

INTRODUCTION: Human trophoblastic cell lines, such as BeWo, are commonly used in 2D models to study placental Trypanosoma cruzi infections. However, these models do not accurately represent natural infections. Three-dimensional (3D) microtissue cultures offer a more physiologically relevant in vitro model, mimicking tissue microarchitecture and providing an environment closer to natural infections. These 3D cultures exhibit functions such as cell proliferation, differentiation, morphogenesis, and gene expression that resemble in vivo conditions.
METHODS: We developed a 3D culture model using

Hydrogel microfibers, which are characterized by flexible mechanical properties, a uniform spatial distribution, large surface areas, and excellent biocompatibility, hold great potential for various biomedical applications. However, the fabrication of heterogeneous hydrogel microfibers with high cell-loading capacity and the ability to carry multiple components via an environmentally friendly method remains challenging. In this study, we developed a novel pneumatic pump-assisted all-aqueous microfluidic system that enables the one-step fabrication of all-aqueous droplet-filled hydrogel microfi

Tumor spheroids are widely used to model the tumor microenvironment and evaluate immune-mediated cytotoxicity, but conventional methods such as ultralow-attachment and hanging-drop plates typically require 48-72 h to generate stable spheroids, limiting scalability and rapid testing. Here, we report a simple method for generating uniform tumor spheroids within 7-8 h using continuous rocking in standard multiwell plates. Computational fluid dynamics simulations showed that rocking produced mild oscillatory hydrodynamic conditions, with average shear stress ranging from 0.4 to 0.8 dyn/cm2 and X-d

Poly(ethylene glycol) (PEG)-based hydrogels are particularly challenging to degrade, which hinders efficient cell harvesting within the gel matrix. Here, highly branched copolymers of PEG methyl ether acrylate (PEGMA) and disulfide diacrylate (DSDA) (PEG-DS) with short primary chains and multiple pendent vinyl groups were synthesized by a "vinyl oligomer combination" approach. PEG-DS readily cross-links with thiolated gelatin (Gel-SH) to form hydrogels. Results demonstrate that shortening the primary chains of PEG-DS significantly enhances the viability of bone marrow mesenchymal stem cells (B

Cell culture techniques for producing a three-dimensional autologous epidermal autograft (cultured epidermal autograft) suitable for tissue grafting and wound healing procedures are described. This chapter commences with surgical biopsy of patient's skin tissue, further reduction of skin tissues to keratinocyte cells by enzymatic treatment, and recovery of viable adult keratinocytes in a new balanced buffered salt media supportive of the growth of clonally enriched isolated basal keratinocytes. Culture techniques required for the formation of a hole-free monolayer of undifferentiated basal ker

INTRODUCTION: Within adipose tissue (AT), different macrophage subsets have been described, which played pivotal and specific roles in upholding tissue homeostasis under both physiological and pathological conditions. Nonetheless, studying resident macrophages in-vitro poses challenges, as the isolation process and the culture for extended periods can alter their inherent properties.
METHODS: Stroma-vascular cells isolated from murine subcutaneous AT were seeded on ultra-low adherent plates in the presence of macrophage colony-stimulating factor. After 4 days of culture, the cells spontaneous

Three-dimensional (3D) cell spheroid technology has led to significant advances in cartilage injury repair, the construction of osteoarthritis (OA) models, and the development of platforms for screening therapeutics targeting cartilage diseases. Compared with conventional two-dimensional (2D) cell culture, this technology better mimics the in vivo microenvironment, thus providing stronger support for cell growth and viability. Furthermore, it effectively stimulates chondrocytes proliferation and differentiation, facilitating cartilage tissue regeneration and significantly improving the quality

The healing process of diabetic foot ulcers (DFUs) presents a slow pattern with an increased risk of infections, ischemia, and thrombosis correlated with high levels of reactive oxygen species production. Vascular injury is one of the factors contributing to the difficulty of wound healing in diabetic patients. Although the understanding of the pathophysiology of DFUs has significantly increased in recent years, associated treatments still have a high level of failure, leading to high morbidity rates, mortality, and amputations. Three-dimensional (3D) cell culture platforms offer a new approac

OBJECTIVE: To explore the promotion of dynamic distraction on osteogenic differentiation of human bone marrow mesenchymal stem cells (hBMMSCs) in three-dimensional culture.
METHODS: Dynamic stretching in a three-dimensional culture for hBMMSCs was achieved with proportions set at 5%, 10%, and 20%, a frequency of 0.5 Hz, and a dynamic stretching duration of 2 hours per day. Static culture was used as the control group. Deformation of hBMMSCs induced by dynamic stretching was observed through cytoplasmic fluorescence staining. After 7 days of dynamic stretching culture, the impact of dynamic st

The neurovascular unit (NVU) is a critical interface in the central nervous system that links vascular interactions with glial and neural tissue. Disruption of the NVU has been linked to the onset and progression of neurodegenerative diseases. Despite its significance the NVU remains challenging to study in a physiologically relevant manner. Here, a 3D cell triculture model of the NVU is developed that incorporates human primary brain microvascular endothelial cells, astrocytes, and pericytes into a tissue system that can be sustained in vitro for several weeks. This tissue model helps recapit

Human pluripotent stem cells (hPSCs) hold enormous potential for regenerative medicine and disease modeling applications. These cells, however, are commonly cultured on Matrigel, a 2D coating of mouse origin and significant complexity, hindering their clinical translation. These issues have motivated the development of (a) 2D natural alternatives, which can mimic the extracellular matrix niche, but result in cytoskeletal and epigenetic changes, insufficient cell-cell and cell-matrix interactions, lineage biases upon differentiation, and challenges with scale-up due to surface area constraints

Type 1 diabetes mellitus (T1D) is a global disease, and stem cell-derived insulin-producing cells show great promise for treatment; their in vitro development is limited. This suggests that the in vivo environment, particularly the native extracellular matrix (ECM), plays a crucial role in cell maturation. To address this, our study investigated this challenge through a two-part investigation aimed at enhancing the functional maturity of human Wharton's jelly-derived mesenchymal stem cells (WJ-MSCs). In the first part, we optimized a differentiation protocol by selectively identifying key smal

Induced pluripotent stem cell (iPSC)-derived T cells may provide future therapies for cancer patients, but those generated by current methods, such as the OP9/DLL1 system, have shown abnormalities that pose major barriers for clinical translation. Our data indicate that these iPSC-derived CD8 single-positive T cells are more like CD4+CD8+ double-positive T cells than mature naive T cells because they display phenotypic markers of developmental arrest and an innate-like phenotype after stimulation. We developed a 3D thymic culture system to avoid these aberrant developmental fates, generating a
