Crystal structure of the dimeric extracellular domain of human carbonic anhydrase XII, a bitopic membrane protein overexpressed in certain cancer tumor cells.

Overexpression of the zinc enzyme carbonic anhydrase (CA; EC ) XII is observed in certain human cancers. This bitopic membrane protein contains an N-terminal extracellular catalytic domain, a membrane-spanning alpha-helix, and a small intracellular C-terminal domain. We have determined the three-dimensional structure of the extracellular catalytic domain of human CA XII by x-ray crystallographic methods at 1.55-A resolution. The structure reveals a prototypical CA fold; however, two CA XII domains associate to form an isologous dimer, an observation that is confirmed by studies of the enzyme i

Increase in cytosolic calcium maintains plasma membrane integrity through the formation of microtubule ring structure in apoptotic cervical cancer cells induced by trichosanthin.

This study investigates the role of dysregulated cytosolic free calcium ([Ca(2+)]c) homeostasis on microtubule (MT) ring structure in apoptotic cervical cancer (HeLa) cells induced by trichosanthin (TCS), a type I ribosome inactivating protein (RIP). The TCS-induced decrease in cell viability was significantly enhanced in combination with the specific calcium chelator, EGTA-AM. Sequestration of [Ca(2+)]c markedly disrupted the special MT ring structure. Furthermore, TCS tended to increase LDH release, whereas no significant differences were observed until 48 h of the treatment. In contrast, co

The Relationship of Omega 3 Polyunsaturated Fatty Acids in Red Blood Cell Membranes with Cognitive Function and Brain Structure: A Review Focussed on Alzheimer's Disease.

Significant research attention has focussed on the identification of nutraceutical agents for the prevention of cognitive decline as a natural means of cognitive preservation in the elderly. There is some evidence for a reduction of brain omega 3 polyunsaturated fatty acids (n-3 PUFAs) in normal aging and in Alzheimer's disease. n-3 PUFAs exhibit anti-inflammatory and anti-amyloidogenic properties as well as being able to reduce tau phosphorylation. Many observational studies have demonstrated a link between n-3 PUFAs and cognitive aging, and some, but not all, randomized controlled trials hav

Exogenous Proline Maintains Cell Wall Structure and Membrane Integrity in Rice Seedlings Under Cr(VI) Stress Associated with Regulation of Proline-Rich Proteins.

Cr(VI) pollution severely damages plant cell wall structure and membrane integrity. Proline-rich proteins (PRPs), key cell wall structural components, rely on proline as their biosynthetic precursor. Pro accumulation has been shown to positively correlate with PRP abundance, suggesting a direct biochemical link between exogenous Pro [Pro(exo)] application and PRP-mediated cell wall function. However, how Pro(exo) regulates PRP expression to maintain cell wall structural integrity and membrane stability in rice seedlings under Cr(VI) stress remains unclear. In this study, the subcellular distri

A partial structure for the gamma-aminobutyric acid (GABAA) receptor is derived from the model for the nicotinic acetylcholine receptor. The anion-exchange protein of cell membranes is related to the GABAA receptor.

Based on the nicotinic acetylcholine receptor model [(1987) Eur. J. Biochem. 168, 431-449], a partial model is constructed for the exobilayer portion of the GABAA receptor, an approach justified by the superfamily relationship of the two receptors [(1987) Nature 328, 221-227]. The model predicts successfully the excess positive charge on interior strands which constitute the ligand-responsive portion of the receptor. Binding to GABA expands the exobilayer portion of the receptor, opening a pathway to a chloride channel. Separate binding sites for antianxiolytics (benzodiazepines) and hypnotics

Mutant torsinA, which causes early-onset primary torsion dystonia, is redistributed to membranous structures enriched in vesicular monoamine transporter in cultured human SH-SY5Y cells.

A single GAG deletion in the DYT1 gene causes primary early-onset, generalized torsion dystonia. The DYT1 protein product, torsinA, belongs to the AAA+ family of proteins. When overexpressed, wild-type torsinA localizes mainly to the endoplasmic reticulum, whereas the mutant forms inclusions of unclear biogenetic origin. In this study, overexpressed wild-type torsinA in human neuroblastoma (SH-SY5Y) cell lines was distributed throughout the cell body and colocalized with a marker for the endoplasmic reticulum, confirming it is an endoplasmic reticulum protein. However, mutant torsinA showed pe

Differences between translation products of tick-borne encephalitis virus RNA in cell-free systems from Krebs-2 cells and rabbit reticulocytes: involvement of membranes in the processing of nascent precursors of flavivirus structural proteins.

Tick-borne encephalitis virus (TBEV) RNA was translated in extracts from Krebs-2 cells and in rabbit reticulocyte lysates. In the former system, two polypeptides, p53 and p13, corresponding to envelope (E) and core (C) proteins of the virion, respectively, were synthesized preferentially. In contrast, the major product in reticulocyte lysates was represented by a heterogeneous set of high-molecular-weight polypeptides which did not appear to include p53 or p13. The reticulocyte lysates, however, acquired the ability to produce structural proteins (p53 at least) after addition of purified membr

A novel function of WAVE in lamellipodia: WAVE1 is required for stabilization of lamellipodial protrusions during cell spreading.

When a cell spreads and moves, reorganization of the actin cytoskeleton pushes the cell membrane, and the resulting membrane protrusions create new points of contact with the substrate and generate the locomotive force. Membrane extension and adhesion to a substrate must be tightly coordinated for effective cell movement, but little is known about the mechanisms underlying these processes. WAVEs are critical regulators of Rac-induced actin reorganization. WAVE2 is essential for formation of lamellipodial structures at the cell periphery stimulated by growth factors, but it is thought that WAVE

A triarylboron-based binuclear Zn(II) complex as a two-photon fluorescent probe for simultaneous multicolor imaging of the cell membrane, endoplasmic reticulum, and nucleolus.

The visualization of subcellular structures is critical for understanding their intracellular function. We prepared two triarylboron-based multinuclear Zn2+ complexes, TAB-1-3Zn2+ and TAB-2-2Zn2+, which can be used as fluorescent probes for nucleoside polyphosphate (NPP) and RNA because their multi Zn2+ center can selectively combine with the phosphate side chain of NPP or RNA, accompanied by a corresponding fluorescence change. Among them, TAB-2-2Zn2+ is more suitable than TAB-1-3Zn2+ for live cell imaging because of its excellent cell membrane permeability resulting from amphiphilicity. Sinc

Trophoblast Cell Recovery from Angiogenesis-Tube Formation Assay for Differentiation Marker Expression Analysis.

During placenta development, extravillous trophoblast (EVT) cells invade the maternal decidua to remodel the uterine spiral arteries by a process of mesenchymal to endothelial-like transition. Traditionally, this process is evaluated by an in vitro tube-formation assay, where the cells organize themselves into tube-like structures when seeded over a polymerized basement membrane preparation. Although several structural features can be measured in photomicrographs of the structures, to assess the real commitment of EVT to the endothelial-type phenotype, biochemical analysis of cell extracts is

SARS-CoV-2 spike protein dictates syncytium-mediated lymphocyte elimination.

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) virus is highly contagious and causes lymphocytopenia, but the underlying mechanisms are poorly understood. We demonstrate here that heterotypic cell-in-cell structures with lymphocytes inside multinucleate syncytia are prevalent in the lung tissues of coronavirus disease 2019 (COVID-19) patients. These unique cellular structures are a direct result of SARS-CoV-2 infection, as the expression of the SARS-CoV-2 spike glycoprotein is sufficient to induce a rapid (~45.1 nm/s) membrane fusion to produce syncytium, which could readily

Appearance of membranocystic lesion (Nasu)-like changes in Behçet's syndrome. An electron microscopic study of erythema nodosum-like lesions.

Membranocystic lesion (Nasu)-like changes were seen in the subcutaneous fat tissues of erythema nodosum-like lesions in three of 18 patients with Behçet's syndrome. The membranocystic structures, which were derived from degenerative fat cells with a loss of the cell membrane, were observed in the edematous connective tissues accompanied by fibrin deposition and diapedesis of erythrocytes. Electron microscopic evaluation revealed two different patterns of peculiar membranous structures in the cytoplasm of the degenerative fat cells. One was proliferating membrane structures such as well develop

Natural mechanisms and artificial PEG-induced mechanism that repair traumatic damage to the plasmalemma in eukaryotes.

Eukaryotic tissues are composed of individual cells surrounded by a plasmalemma that consists of a phospholipid bilayer with hydrophobic heads that bind cell water. Bound-water creates a thermodynamic barrier that impedes the fusion of a plasmalemma with other membrane-bound intracellular structures or with the plasmalemma of adjacent cells. Plasmalemmal damage consisting of small or large holes or complete transections of a cell or axon results in calcium influx at the lesion site. Calcium activates fusogenic pathways that have been phylogenetically conserved and that lower thermodynamic barr

Complex dynamic development of poliovirus membranous replication complexes.

Replication of all positive-strand RNA viruses is intimately associated with membranes. Here we utilize electron tomography and other methods to investigate the remodeling of membranes in poliovirus-infected cells. We found that the viral replication structures previously described as "vesicles" are in fact convoluted, branching chambers with complex and dynamic morphology. They are likely to originate from cis-Golgi membranes and are represented during the early stages of infection by single-walled connecting and branching tubular compartments. These early viral organelles gradually transform

A Salmonella protein causes macrophage cell death by inducing autophagy.

Salmonella enterica, the causative agent of food poisoning and typhoid fever, induces programmed cell death in macrophages, a process found to be dependent on a type III protein secretion system, and SipB, a protein with membrane fusion activity that is delivered into host cells by this system. When expressed in cultured cells, SipB caused the formation of and localized to unusual multimembrane structures. These structures resembled autophagosomes and contained both mitochondrial and endoplasmic reticulum markers. A mutant form of SipB devoid of membrane fusion activity localized to mitochondr

Motility voltage sensor of the outer hair cell resides within the lateral plasma membrane.

The outer hair cell (OHC) from the organ of Corti is believed to be responsible for the mammal's exquisite sense of hearing. A membrane-based motile response of this cell underlies the initial processing of acoustic energy. The voltage-dependent capacitance of the OHC, possibly reflecting charge movement of the motility voltage sensor, was measured in cells during intracellular dialysis of trypsin under whole cell voltage clamp. Within 10 min after dialysis, light and electron microscopic examination revealed that the subplasmalemmal structures, including the cytoskeletal framework and subsurf

Tau and Membranes: Interactions That Promote Folding and Condensation.

Tau misfolding and assembly is linked to a number of neurodegenerative diseases collectively described as tauopathies, including Alzheimer's disease (AD) and Parkinson's disease. Anionic cellular membranes, such as the cytosolic leaflet of the plasma membrane, are sites that concentrate and neutralize tau, primarily due to electrostatic interactions with tau's microtubule binding repeat domain (RD). In addition to electrostatic interactions with lipids, tau also has interactions with membrane proteins, which are important for tau's cellular functions. Tau also interacts with lipid tails to fac

Defects of basement membrane and hemidesmosome structure correlate with malignant phenotype and stromal interactions in HaCaT-Ras xenografts.

Benign and malignant HaCaT-ras clones, derived from immortalized HaCaT cells were grown as nude mouse surface transplants rendering a human tumor progression model. Searching for malignancy-related alterations, the deposition, localization and mRNA of basement membrane and hemidesmosome components were analysed by immunofluorescence, in situ hybridization and electron microscopy. Initially, at 1 week epithelia of benign and malignant cells revealed a similarly low polarity and an enlarged 'activated basal' compartment, reflected by partial dislocation and extended pericellular staining of the
