CALB2 mediates tumor progression and immune escape in colorectal cancer by shaping an inhibitory immune microenvironment.

BACKGROUND: Colorectal cancer (CRC) remains a leading cause of cancer-related mortality worldwide, characterized by a highly immunosuppressive tumor microenvironment (TME) that facilitates immune evasion and tumor progression. METHODS: Integrated analysis of TCGA and GEO datasets was performed to evaluate CALB2 expression and its prognostic significance in CRC. Functional experiments including proliferation, migration, colony formation, and apoptosis assays were conducted following CALB2 knockdown. Co-culture assays and multiplex immunohistochemistry (mIHC) were used to assess TME remodeling.

Downregulation of miR-92a in Decidual Stromal Cells Suppresses Migration Ability of Trophoblasts by Promoting Macrophage Polarization.

Preeclampsia (PE) is a severe pregnancy complication that accounts for about 14% of maternal deaths. Its clinical manifestations commonly include hypertension and proteinuria. However, it is largely limited in understanding its pathogenetic mechanism. In this study, we used bioinformatics to compare differential gene expressions in decidual stromal cells from PE patients and healthy donors. The result indicated that higher levels of CCL5 and CXCL2 were expressed in decidual stromal cells of PE patients compared with healthy pregnancy. The bioinformatics analysis confirmed that decidual stromal

Celastrol attenuates osteoclast-driven bone invasion in pituitary adenomas and suppresses HSP90AA1/TRAF6/NF-κB signaling.

Bone-invasive pituitary adenomas (BIPAs) are associated with skull base destruction, incomplete resection and postoperative recurrence, but pharmacological strategies targeting tumor-induced osteolysis remain limited. Here, we investigated whether celastrol, a bioactive triterpenoid from Tripterygium wilfordii, restrains BIPA progression by modulating macrophage-associated osteoclastogenesis. Clinical recurrence analysis, calvarial pituitary adenoma bone invasion models, micro-CT, histology, immunofluorescence, cell functional assays, osteoclast differentiation assays, RNA sequencing, molecula

Icm/Dot-dependent inhibition of phagocyte migration by Legionella is antagonized by a translocated Ran GTPase activator.

The environmental bacterium Legionella pneumophila causes a severe pneumonia termed Legionnaires' disease. L. pneumophila employs a conserved mechanism to replicate within a specific vacuole in macrophages or protozoa such as the social soil amoeba Dictyostelium discoideum. Pathogen-host interactions depend on the Icm/Dot type IV secretion system (T4SS), which translocates approximately 300 different effector proteins into host cells. Here we analyse the effects of L. pneumophila on migration and chemotaxis of amoebae, macrophages or polymorphonuclear neutrophils (PMN). Using under-agarose ass

Macrophage-derived CTSZ promotes prostate cancer progression via αVβ3 integrin-mediated activation of the AKT/FOXO1/JUNB signaling axis.

BACKGROUND: The tumor microenvironment (TME), especially tumor-associated macrophages (TAMs), plays a critical role in prostate cancer (PCa) progression. Cathepsin Z (CTSZ) is a lysosomal protease implicated in various malignancies, but its specific function and mechanism within the PCa-associated TAMs remain largely unexplored. METHODS: We analyzed single-cell RNA sequencing data (GSE141445, GSE137829) to identify CTSZ expression patterns. Its role in macrophage polarization was validated by immunofluorescence co-localization in human PCa tissues and in vitro experiments (qPCR, Western blot,

Mouse macrophages completely lacking Rho subfamily GTPases (RhoA, RhoB, and RhoC) have severe lamellipodial retraction defects, but robust chemotactic navigation and altered motility.

RhoA is thought to be essential for coordination of the membrane protrusions and retractions required for immune cell motility and directed migration. Whether the subfamily of Rho (Ras homolog) GTPases (RhoA, RhoB, and RhoC) is actually required for the directed migration of primary cells is difficult to predict. Macrophages isolated from myeloid-restricted RhoA/RhoB (conditional) double knock-out (dKO) mice did not express RhoC and were essentially "pan-Rho"-deficient. Using real-time chemotaxis assays, we found that retraction of the trailing edge was dissociated from the advance of the cell

Macrophage MerTK promotes profibrogenic cross-talk with hepatic stellate cells via soluble mediators.

BACKGROUND & AIMS: Activation of Kupffer cells and recruitment of monocytes are key events in fibrogenesis. These cells release soluble mediators which induce the activation of hepatic stellate cells (HSCs), the main fibrogenic cell type within the liver. Mer tyrosine kinase (MerTK) signaling regulates multiple processes in macrophages and has been implicated in the pathogenesis of non-alcoholic steatohepatitis-related fibrosis. In this study, we explored if MerTK activation in macrophages influences the profibrogenic phenotype of HSCs. METHODS: Macrophages were derived from THP-1 cells or di

Cross reactions among flaviviruses in macrophage migration inhibition assay.

Adult Swiss albino mice were immunized intravenously with flaviviviruses. Cellular immune response was studied by macrophage migration inhibition assay. Migration of cells from sensitized mice was significantly inhibited in the presence of homologous virus or antigen. Migration inhibition was also significant with serologically related viruses, thus establishing cross reaction in cellular immune response.

TIP60-mediated acetylation of UHRF1 is associated with tumor cell-dependent M2-like macrophage phenotypes and aggressiveness in gastric cardia adenocarcinoma.

Gastric cardia adenocarcinoma (GCA) is an aggressive malignancy in which immune microenvironment remodeling contributes to tumor progression. This study investigated whether Tat interactive protein, 60 kDa (TIP60)-mediated acetylation of ubiquitin-like with PHD and RING Finger domains 1 (UHRF1) regulates GCA progression and tumor cell-dependent M2-like macrophage polarization. Transcriptomic screening identified UHRF1 as a candidate oncogenic regulator, and UHRF1 was upregulated in GCA tissues and cell lines. Functional assays showed that UHRF1 promoted GCA cell proliferation, migration, invas

KLF4-induced upregulation of SOCS1 ameliorates myocardial ischemia/reperfusion injury by attenuating AC16 cardiomyocyte damage and enhancing M2 macrophage polarization.

Reperfusion strategies, the standard therapy for acute myocardial infarction (AMI), may result in ischemia/reperfusion (I/R) damage. Suppressor of cytokine signaling1 (SOCS1) exerts a cardioprotective function in myocardial I/R damage. Here, we investigated epigenetic modulators that deregulate SOCS1 in cardiomyocytes under hypoxia/reoxygenation (H/R) conditions. Human AC16 cardiomyocytes were exposed to H/R conditions to generate a cell model of myocardial I/R damage. Expression of mRNA and protein was detected by quantitative PCR and western blot analysis, respectively. Cell migratory and in

Salt-dependent chemotaxis of macrophages.

Besides their role in immune system host defense, there is growing evidence that macrophages may also be important regulators of salt homeostasis and blood pressure by a TonEBP-VEGF-C dependent buffering mechanism. As macrophages are known to accumulate in the skin of rats fed under high salt diet conditions and are pivotal for removal of high salt storage, the question arose how macrophages sense sites of high sodium storage. Interestingly, we observed that macrophage-like RAW264.7 cells, murine bone marrow-derived macrophages and peritoneal macrophages recognize NaCl hypertonicity as a chemo

Identification of CCL4-positive macrophages and their associated immune microenvironment in thymic epithelial tumors.

Thymic epithelial tumors (TETs) are one of the primary tumor types in the mediastinum, originating from thymic epithelial cells. Recent studies have highlighted the close association between specific macrophage subpopulations and the tumor immunosuppressive microenvironment. However, the characteristics and immunological functions of macrophages within TETs are still poorly understood. Single-cell transcriptome sequencing and the TCGA-THYM data were collected for integrated analysis of macrophage characteristics. The target cells were verified using multiplex immunofluorescence (mIF) staining.

Modulation of Inflamed Synovium Improves Migration of Mesenchymal Stromal Cells in Vitro Through Anti-Inflammatory Macrophages.

OBJECTIVE: Inflammation is known to negatively affect cartilage repair. However, it is unclear how inflammation influences the migration of mesenchymal stromal cells (MSCs) from the underlying bone marrow into the defect. We therefore aimed to investigate how synovial inflammation influences MSC migration, and whether modulation of inflammation with triamcinolone acetonide (TAA) may influence migration. DESIGN: Inflamed human osteoarthritic synovium, M(IFNγ+TNFα) pro-inflammatory macrophages, M(IL4) repair macrophages, M(IL10) anti-inflammatory macrophages, or synovial fibroblasts were cultur
