PELI1 silencing delays intervertebral disc degeneration by impeding nucleus pulposus cell death.

PURPOSE: Intervertebral disc degeneration (IDD) is a spinal condition that causes low back pain. Pellino E3 ubiquitin protein ligase 1 (PELI1) expression reportedly correlates with inflammation and cell death. This study aimed to determine its potential role in IDD. METHODS: Cell counting kit-8 assay, 5-ethynyl-2'-deoxyuridine staining, senescence-associated β-galactosidase staining, morphological observation, lactate dehydrogenase (LDH) release assay, quantitative reverse transcriptase polymerase chain reaction, and western blotting were used to examine the effect of PELI1 on tumor necrosis

Anterior hypothalamic nucleus drives distinct defensive responses through cell-type-specific activity.

Innate defensive behaviors are essential for survival, allowing animals to appropriately respond to predatory threats. The anterior hypothalamic nucleus (AHN), a key region in the medial hypothalamic defense system, contains both GABAergic and glutamatergic neurons, reflecting a sophisticated balance between inhibitory and excitatory signaling. However, the specific behavioral functions of these neuronal populations have not been systemically examined. Here, we utilized fiber photometry and optogenetic stimulation to investigate the roles of AHN GABAergic, glutamatergic, and CaMKIIa+ neuronal

Dorsal Cochlear Nucleus Fusiform-cell Plasticity is Altered in Salicylate-induced Tinnitus.

Following noise overexposure and tinnitus-induction, fusiform cells of the dorsal cochlear nucleus (DCN) show increased spontaneous firing rates (SFR), increased spontaneous synchrony and altered stimulus-timing-dependent plasticity (StDP), which correlate with behavioral measures of tinnitus. Sodium salicylate, the active ingredient in aspirin, which is commonly used to induce tinnitus, increases SFR and activates NMDA receptors in the ascending auditory pathway. NMDA receptor activation is required for StDP in many brain regions, including the DCN. Blocking NMDA receptors can alter StDP timi

Cell-Type-Specific Afferent Innervation of the Nucleus Accumbens Core and Shell.

The nucleus accumbens (NAc) is clearly implicated in reward processing and drug addiction, as well as in numerous neurological and psychiatric disorders; nevertheless, the circuit mechanisms underlying the diverse functions of the NAc remain poorly understood. Here, we characterized the whole-brain and monosynaptic inputs to two main projection cell types - D1 dopamine receptor expressing medium spiny neurons (D1R-MSNs) and D2 dopamine receptor expressing medium spiny neurons (D2R-MSNs) - within the NAc core and NAc shell by rabies-mediated trans-synaptic tracing. We discovered that D1R-MSNs a

Nucleus pulposus cell-associated PLAU promotes intervertebral disc degeneration through HIPPO pathway.

Intervertebral disc degeneration (IDD) is the most common and critical pathological basis for various spinal disorders. In this study, we investigated how plasminogen activator urokinase (PLAU) promotes IDD progression through the HIPPO pathway. Bioinformatics analysis showed that PLAU is a hub gene in IDD. Both in vitro and in vivo experiments confirmed that PLAU can promote the apoptosis of nucleus pulposus chondrocytes. PLAU activates the HIPPO signaling pathway by increasing the phosphorylation levels of mammalian ste20-like kinases 1/2 (MST1/2), large tumor suppressor 1/2 (LATS1/2), and y

FrozONE: quick cell nucleus enrichment for comprehensive proteomics analysis of frozen tissues.

Subcellular fractionation allows for the investigation of compartmentalized processes in individual cellular organelles. Nuclear enrichment methods commonly employ the use of density gradients combined with ultracentrifugation for freshly isolated tissues. Although it is broadly used in combination with proteomics, this approach poses several challenges when it comes to scalability and applicability for frozen material. To overcome these limitations, we developed FrozONE (Frozen Organ Nucleus Enrichment), a nucleus enrichment and proteomics workflow for frozen tissues. By extensively benchmark

MINK1 deficiency stimulates nucleus pulposus cell pyroptosis and exacerbates intervertebral disc degeneration.

Intervertebral disc (IVD) degeneration, induced by aging and irregular mechanical strain, is highly prevalent in the elderly population, serving as a leading cause of chronic low back pain and disability. Evolving evidence has revealed the involvement of nucleus pulposus (NP) pyroptosis in the pathogenesis of IVD degeneration, while the precise regulatory mechanisms of NP pyroptosis remain obscure. Misshapen/Nck-interacting kinase (NIK)-related kinase 1 (MINK1), a serine-threonine protein kinase, has the potential to modulate the activation of NLRP3 inflammasome, indicating its pivotal role in

Human iPS cell-derived cartilaginous tissue spatially and functionally replaces nucleus pulposus.

The loss of nucleus pulposus (NP) precedes the intervertebral disk (IVD) degeneration that causes back pain. Here, we demonstrate that the implantation of human iPS cell-derived cartilaginous tissue (hiPS-Cart) restores this loss by replacing lost NP spatially and functionally. NP cells consist of notochordal NP cells and chondrocyte-like NP cells. Single cell RNA sequencing (scRNA-seq) analysis revealed that cells in hiPS-Cart corresponded to chondrocyte-like NP cells but not to notochordal NP cells. The implantation of hiPS-Cart into a nuclectomized space of IVD in nude rats prevented the de

G-protein-coupled receptors signalling at the cell nucleus: an emerging paradigm.

G-protein-coupled receptors (GPCRs) comprise a wide family of monomeric heptahelical glycoproteins that recognize a broad array of extracellular mediators including cationic amines, lipids, peptides, proteins, and sensory agents. Thus far, much attention has been given towards the comprehension of intracellular signaling mechanisms activated by cell membrane GPCRs, which convert extracellular hormonal stimuli into acute, non-genomic (e.g., hormone secretion, muscle contraction, and cell metabolism) and delayed, genomic biological responses (e.g., cell division, proliferation, and apoptosis). JAK3 Is Expressed in the Nucleus of Malignant T Cells in Cutaneous T Cell Lymphoma (CTCL).

Perturbation in JAK-STAT signaling has been reported in the pathogenesis of cutaneous T cell lymphoma (CTCL). JAK3 is predominantly associated with the intra-cytoplasmic part of IL-2Rγc located in the plasma membrane of hematopoietic cells. Here we demonstrate that JAK3 is also ectopically expressed in the nucleus of malignant T cells. We detected nuclear JAK3 in various CTCL cell lines and primary malignant T cells from patients with Sézary syndrome, a leukemic variant of CTCL. Nuclear localization of JAK3 was independent of its kinase activity whereas STAT3 had a modest effect on nuclear JAK

Qualitative and quantitative odour discrimation by mitral cells as compared to anterior olfactory nucleus cells.

The responses to odour stimulation of 61 single mitral cells and of 64 single anterior olfactory nucleus (AON) units were simultaneously recorded from rabbits. The test battery consisted of 6 chemical stimuli delivered at two intensity levels corresponding to a hundred-fold difference in concentration. The odour discrimination reaches its best for the mitral cells stimulated weakly; it decreased somewhere when the same cells are stimulated strongly. At AON level the odour discrimination already limited in the case of weak stimuli, almost disappears for strong stimuli. Multidimensional analysis

Endoplasmic Reticulum Stress Is Involved in Nucleus Pulposus Degeneration and Attenuates Low pH-Induced Apoptosis of Rat Nucleus Pulposus Cells.

The microenvironment of degenerative intervertebral disk (IVD) is characteristic of a high concentration of lactic acid and low pH levels, whereas the underlying mechanism has not been clearly defined. Endoplasmic reticulum (ER) is the hub of interactions between environmental signals and cellular biological functions, the malfunction of which is closely involved in the pathogenesis of multiple disorders, including IVD degeneration (IVDD). This research mainly aims at exploring what role ER stress plays in the natural process of IVDD and pH-induced apoptosis of rat nucleus pulposus (NP) cells

The cell body space occupied by the nucleus during the cell differentiation in human lymphocytic, granulocytic and erythroid cell lineages.

The present nuclear and cell body diameter measurements demonstrated size differences of the approximate cell space estimate occupied by the cell nucleus during the cell differentiation in lymphocytic, granulocytic and erythroid cell lineages. These lineages were used as convenient models because all differentiation steps were easily identified and accessible in diagnostic peripheral blood or bone marrow smears of blood donors (BDs), patients suffering from chronic lymphocytic leukemia (CLL), patients with chronic myeloid leukemia (CML) and refractory anemia (RA) of the myelodysplastic syndrom

Different surface sensing of the cell body and nucleus in healthy primary cells and in a cancerous cell line on nanogrooves.

Cancer cells are known to have alterations compared to healthy cells, but can these differences extend to the way cells interact with their environment? Here, the authors focused on the alignment on an array of grooves of nanometer depth using two cell types: healthy osteoprogenitor primary cells (HOP) and a cancerous osteosarcoma (SaOs-2) cell line. Another concern was how this alignment affects the cell's interior, namely, the nucleus. Based on the results, it is proposed that these two cell types respond to different size regimes: SaOs-2 cells are more sensitive to shallow grooves while HOP

Dynamic adaptive coassembled sericin protein orchestrating stem cell development for nucleus pulposus regeneration.

The zonal structure of natural extracellular matrix (ECM) creates a dynamic microenvironment that regulates mechanical and biochemical signals to support stem cell development. Here, we engineered a dynamic adaptive protein gel by coassembling bioactive sericin with an amphiphilic tripeptide, forming a nucleus pulposus (NP)-like viscoelastic network with rapid stress relaxation. This coassembled protein gel sequentially activated integrin β3-mediated mechanotransduction, promoted cytoskeletal remodeling and Yes-associated protein nuclear translocation, and subsequently up-regulated bioactive f

Cluster replicability in single-cell and single-nucleus atlases of the mouse brain.

Single-cell RNA sequencing has advanced our understanding of cellular heterogeneity. Ensuring the replicability of identified cell clusters across studies is essential for determining their biological robustness. We assess the replicability of cell clusters identified in two large mouse brain atlases, one generated using single-cell RNA sequencing and the other with single nuclei. Both profile over 4 million cells and group them into over 5000 clusters. Using transcriptome-wide neighbor voting, we identify 2009 reciprocally matched cluster pairs with consistent spatial localization and coordin

Comparative Transcriptomic Profiling of Corneal Compartments Using Single-Cell and Single-Nucleus Sequencing.

PURPOSE: Single-cell RNA sequencing (scRNA-seq) and single-nucleus RNA-sequencing (snRNA-seq) offer complementary advantages for corneal transcriptomics study. However, variations in cell-type distribution across platforms and the technical challenges in isolating corneal cells may lead to misinterpretation of biological conclusions. This study aimed to establish cell-type-specific recommendations for corneal transcriptomics. METHODS: We systematically evaluated scRNA-seq and snRNA-seq performance using matched human and mouse corneal datasets. The expression of Col8a1 and Kdr was measured by

Single-Nucleus RNA-Seq Reveals Spermatogonial Stem Cell Developmental Pattern in Shaziling Pigs.

Normal testicular development ensures the process of spermatogenesis, which is a complex biological process. The sustained high productivity of spermatogenesis throughout life is predominantly attributable to the constant proliferation and differentiation of spermatogonial stem cells (SSCs). The self-renewal and differentiation processes of SSCs are strictly regulated by the SSC niche. Therefore, understanding the developmental pattern of SSCs is crucial for spermatogenesis. The Shaziling pig is a medium-sized indigenous pig breed originating from central China. It is renowned for its superior
