Signalling molecules and microenvironment modulation in skin regeneration of chronic wound repair: A cellular perspective.

Tissue repair is an intricate biological process involving cellular and molecular mechanisms. These mechanisms coordinate the repair of damaged tissue, relying on the function of several signalling molecules. Growth factors, cytokines, and hormones perform a fundamental role in tissue regeneration, especially in skin regeneration. On the other hand, in case of diabetes or chronic wounds, the synthesis and regulation of these signalling molecules may be disrupted. Despite advances in medical science, chronic, non-healing wounds remain a continuous challenge, often characterized by reduced angio

Treatment of T2DM-related inflammation and vascular injury by regulating cellular crosstalk in the islet microenvironment.

Type 2 diabetes mellitus (T2DM), a complex systemic metabolic disorder caused by multiple factors, has been linked to numerous acute and chronic complications. T2DM pathogenesis includes glucotoxicity, lipotoxicity, inflammatory cytokines, and amyloid formation. Within the pancreatic islet microenvironment, the crosstalk among cell types plays a significant role in these pathogenic mechanisms. Islet β cells, macrophages, and endothelial cells, the three primary cell types, engage in intercellular communication under physiological and pathological conditions, critical to maintaining islet homeo

Immunophenotypic Characteristics of Bone Marrow Microenvironment Cellular Composition at the Biochemical Progression of Multiple Myeloma.

Multiple myeloma (MM) relapses are inevitable in the majority of patients, and in addition to genetic changes in the MM clone, the immune profile of the bone marrow (BM) plays a key role in this process. Biochemical progression or relapse (BR) precedes clinical relapse in a significant proportion of patients with MM. In the present study, we used flow cytometry to assess the cellular composition of the BM microenvironment in MM patients with confirmed BR. Fifteen distinct cells subsets in the BM were evaluated with the panel of antibodies used routinely for MRD monitoring in MM in 52 patients

Survey of cellular immune responses to human cytomegalovirus infection in the microenvironment of the uterine-placental interface.

Congenital human cytomegalovirus (HCMV) infection is a leading cause of birth defects, yet there are no established treatments for preventing maternal-fetal transmission. During first trimester, HCMV replicates in basal decidua that functions as a reservoir for virus and source of transmission to the attached placenta and fetal hemiallograft but also contains immune cells, including natural killer cells, macrophages, and T cell subsets, that respond to pathogens, protecting the placenta and fetus. However, the specific cellular and cytokine responses to infection are unknown, nor are the immun

Constitutional absence of epithelial integrin α3 impacts the composition of the cellular microenvironment of ILNEB keratinocytes.

Integrin α3β1, a major epidermal adhesion receptor is critical for organization of the basement membrane during development and wound healing. Integrin α3 deficiency leads to interstitial lung disease, nephrotic syndrome and epidermolysis bullosa (ILNEB), an autosomal recessive multiorgan disease characterized by basement membrane abnormalities in skin, lung and kidney. The pathogenetic chains from ITGA3 mutation to tissue abnormalities are still unclear. Although integrin α3 was reported to regulate multiple extracellular proteins, the composition of the extracellular compartment of integrin

Lysyl oxidase expression in cardiac fibroblasts is regulated by α2β1 integrin interactions with the cellular microenvironment.

Lysyl oxidase (LOX) catalyzes crosslink formation between fibrillar collagens and elastins and an increase in LOX activity has been associated with cardiac fibrosis following myocardial infarction (MI). It has been previously reported that LOX expression is regulated by growth factors and cytokines including transforming growth factor (TGF-β1); however, it is unclear how the biophysical and biochemical properties of the cellular microenvironment affect LOX expression. In this study, we isolated rat cardiac fibroblasts (CF) and infarct cardiac fibroblasts (ICF), from healthy and 1-week post-MI

Spatial analysis reveals cellular microenvironments and mechanisms of inflammation and injury in acute interstitial nephritis.

Acute interstitial nephritis (AIN) is present in 15-20% of all acute kidney injury cases but lacks proven effective therapies beyond corticosteroids. Using high-resolution imaging mass cytometry and single-cell spatial transcriptomics to analyze human kidney biopsies with AIN, acute tubular injury (ATI), and reference tissue, we show that interferon-γ-induced CXCL9-CXCR3 signaling is markedly increased in AIN, with significantly more predicted interactions relative to ATI, dominated by homotypic T cell-T cell amplification networks concentrated in lymphoid aggregates. Tubular cells, predominan

Integrin β1 signaling at the crossroads of cellular plasticity, the tumor microenvironment, and therapy resistance in medulloblastoma.

Medulloblastoma (MB), the most common malignant pediatric brain tumor, remains associated with relapse and long-term treatment-related toxicities. Tumor-microenvironment interactions increasingly appear to contribute to progression, stemness, and therapy resistance in brain cancers. Integrin β1, a major mediator of cell-extracellular matrix interactions, regulates survival signaling, mechanotransduction, and cancer stem cell maintenance in several tumor types, particularly glioblastoma. This mini-review discusses how developmental programs, extracellular matrix remodeling, hypoxia, and cancer

Uncovering the Potential of 3D Spheroids for Modeling the Cellular Crosstalk Within the Tumor Microenvironment in HNSCC.

Head and neck squamous cell carcinoma (HNSCC) progression is profoundly influenced by dynamic cellular crosstalk within the tumor microenvironment (TME), where interactions between cancer cells, stromal components, and immune populations orchestrate therapeutic resistance, metastasis, and immune evasion. This review examines the utility of three-dimensional (3D) spheroid models as physiologically relevant platforms for investigating these cellular interactions compared with conventional two-dimensional (2D) cultures. By incorporating cancer-associated fibroblasts (CAFs), tumor-associated macro

Obesity modulates the cellular and molecular microenvironment in the peritoneal cavity: implication for ovarian cancer risk.

INTRODUCTION: Abdominal obesity increases the risk of developing ovarian cancer but the molecular mechanisms of how obesity supports ovarian cancer development remain unknown. Here we investigated the impact of obesity on the immune cell and gene expression profiles of distinct abdominal tissues, focusing on the peritoneal serous fluid (PSF) and the omental fat band (OFB) as critical determinants for the dissemination of ovarian metastases and early metastatic events within the peritoneal cavity. METHODS: Female C57BL/6 mice were fed a low-fat (LFD) or a high-fat diet (HFD) for 12 weeks until

The cerebrospinal fluid immune microenvironment in lung cancer leptomeningeal metastasis: cellular ecosystems and therapeutic possibilities.

Leptomeningeal metastasis (LM) is a devastating form of central nervous system (CNS) dissemination in lung cancer, particularly non-small cell lung cancer (NSCLC). Current treatment frameworks emphasize CNS drug penetration, oncogenic driver evolution, and intrathecal therapy. Emerging evidence supports viewing the cerebrospinal fluid (CSF) compartment as an active tumor-immune space, but the field remains early, with limited lung cancer-specific mechanistic and single-cell datasets. Lung cancer cells in CSF appear to adapt to low protein content, limited nutrients, fluid shear, weak matrix su

Single-cell RNA sequencing analysis reveals cellular heterogeneity and immune microenvironment dynamics in colorectal cancer.

BackgroundDespite treatment advances, prognoses of patients with advanced colorectal cancer remain poor. The limited effectiveness of programmed cell death protein 1 (PD-1) blockade immunotherapy in a subset of patients necessitates a deeper understanding of the colorectal cancer microenvironment. This study aimed to identify new therapeutic targets and potential biomarkers by investigating CD8 + CD101hiTim3+ (CCT) T cells and their progenitors in colorectal cancer, and their association with immunotherapy response at the transcriptomic level.MethodsWe used single-cell sequencing data and The

Myeloid-derived suppressor cells in the tumor microenvironment: cellular crosstalk, immunosuppressive effects, and therapeutic targets.

Tumor microenvironment (TME)-induced immunosuppression is a major driver of cancer treatment failure and resistance to immunotherapy. Myeloid-derived suppressor cells (MDSCs) are not only pivotal suppressors of effector immune responses but also central organizers of tumor-supportive metabolic, stromal, and cellular crosstalk. This review systematically summarizes the origin and phenotypic characteristics of MDSCs and the mechanisms governing their recruitment, expansion, and activation within the TME. This review integrates current knowledge of MDSC biology through two complementary framework

Reprogramming the immunosuppressive breast cancer microenvironment: integrating cellular, metabolic, and stromal targets for rational immunotherapy.

Breast cancer, a highly heterogeneous malignancy and a leading cause of cancer-related mortality among women worldwide, is profoundly shaped in its initiation, progression, and therapeutic response by the tumor immune microenvironment (TIME). This review consolidates recent advances in deciphering the cellular, molecular, and metabolic complexity of breast cancer TIME and highlights mechanisms of immune suppression that impede durable treatment efficacy. We critically appraise current and emerging immunotherapeutic approaches, with a focus on strategies that aim to transform immunologically "c

Deciphering the cellular tumor microenvironment landscape in salivary gland carcinomas using multiplexed imaging mass cytometry.

BACKGROUND: To spatially characterize the single-cell tumor microenvironment (TME) of salivary gland carcinomas (SGC) and identify prognostic biomarkers. METHODS: SGC, including salivary duct carcinomas (SDC), acinic cell, mucoepidermoid, and secretory carcinomas, were analyzed using a 13-marker imaging mass cytometry panel. Multichannel image data from 54 primary cases and nodal metastases were processed to generate single-cell datasets. Cell phenotypes (tumor cells, cancer-associated fibroblasts (CAFs), endothelia, immune cells) were classified using a validated CAF algorithm, followed by s

HIV-associated non-Hodgkin lymphoma tumor-microenvironment axes differ by EBV status across cellular origins.

Non-Hodgkin Lymphoma (NHL) is the main cancer-related mortality for people living with HIV (PLWH). NHL genetic and molecular classifications have been intensely studied and correlated with clinical outcomes, but critical unanswered questions relevant to malignancy persist. For example, tumors positive for Epstein-Barr virus (EBV) are aggressive and account for 30-50% of HIV-associated NHLs, yet insights on the nature of EBV in NHL pathogenesis or potential therapeutic vulnerabilities have been limited. Here, we examined HIV-associated NHLs stratified by histopathologic classification, cell of

Extracellular biogenic nanoscale mitochondria reprogram the wound microenvironment via ROS scavenging independent of cellular uptake.

Mitochondria are nanoscale organelles essential for cellular metabolism and redox regulation, making them a compelling target for regenerative therapeutics. Analysis of wound-edge tissues from pediatric patients with chronic non-healing ulcers revealed marked metabolic insufficiency and impaired regenerative signaling, underscoring an unmet clinical need for mitochondrial-based interventions. Here, we show that topically applied mesenchymal stem cell-derived mitochondria (MSC-mt), functioning as naturally derived nanoscale organelles, markedly accelerate wound closure in a murine full-thicknes

Engineering the Cellular Microenvironment for Human Induced Pluripotent Stem Cell Cardiac Differentiation: Beyond Wnt Signaling.

Human induced pluripotent stem cells (hiPSCs) have revolutionized cardiovascular research by providing a renewable source of patient-specific cardiomyocytes for disease modeling, drug discovery, precision medicine, and regenerative therapies. Temporal modulation of canonical Wnt/β-catenin signaling has established the current gold standard for efficient and reproducible cardiac differentiation under chemically defined conditions. However, conventional Wnt-based protocols consistently generate cardiomyocytes with fetal-like structural, electrophysiological, metabolic, and contractile characteri

Cellular senescence-Related genes define the immune microenvironment and molecular characteristics in severe asthma patients.

Recent studies have shown that cellular senescence is involved in the pathogenesis of severe asthma (SA). The objective of this study was to investigate the role of cellular senescence-related genes (CSGs) in the pathogenesis of SA. Here, 54 differentially expressed CSGs were identified in SA patients compared to healthy control individuals. Among the 54 differentially expressed CSGs, 3 CSGs (ETS2, ETS1 and AURKA) were screened using the LASSO regression analysis and logistic regression analysis to establish the CSG-based prediction model to predict severe asthma. Moreover, we found that the p
