Sperm Selection and Embryo Development: A Comparison of the Density Gradient Centrifugation and Microfluidic Chip Sperm Preparation Methods in Patients with Astheno-Teratozoospermia.

In recent years, microfluidic chip-based sperm sorting has emerged as an alternative tool to centrifugation-based conventional techniques for in vitro fertilization. This prospective study aims to compare the effects of density gradient centrifugation and microfluidic chip sperm preparation methods on embryo development in patient populations with astheno-teratozoospermia. In the study, the semen samples of the patients were divided into two groups for preparation with either the microfluidic or density gradient methods. Selected spermatozoa were then used to fertilize mature sibling oocytes a

Subcellular structure of bovine thyroid gland. A study on bovine thyroid membranes by buoyant-density-gradient centrifugation in a B-XIV zonal rotor.

A combined mitochondrial and light mitochondrial fraction and a microsomal fraction were isolated from bovine thyroid gland and fractionated further in a B-XIV zonal rotor. A density gradient ranging from 20 to 50% (w/w) sucrose was used. The rotor was operated for 3 h at 45 000 rev./min. All manipulations were performed at 4 degrees C and at pH 7.4. 2. Membranous material was recovered in two zones: zone I, containing microsomal material derived from both smooth endoplasmic reticulum and plasma membranes and probably also from other smooth membranes; zone II, containing material from rough en

Density Gradient Centrifugation Versus Swim-Up for Cumulative Live Birth in IVF/ICSI Cycles With High Sperm DNA Fragmentation: A Propensity Score Matching Analysis.

BACKGROUND: A high sperm DNA fragmentation index (DFI) has been found to be associated with male infertility and suboptimal outcomes in assisted reproductive technology (ART). Although sperm preparation techniques can reduce DFI following processing, it is unclear whether density gradient centrifugation (DGC) or swim-up (SU) achieves better clinical outcomes, particularly in terms of cumulative live birth rates (cLBR), for patients with elevated DFI. OBJECTIVE: Using a propensity score-matched statistical approach, this study aims to compare the clinical effectiveness of DGC versus swim-up sp

Collection and Isolation of CD14+ Primary Human Monocytes Via Dual Density Gradient Centrifugation as a Model System to Study Human Cytomegalovirus Infection and Pathogenesis.

Human cytomegalovirus (HCMV) can cause severe disease in the immunocompromised. One of the hallmarks of HCMV infection of a human host is the targeted infection of peripheral blood monocytes (but not other leukocyte populations) that in turn serve as the key cell type for hematogenous dissemination and the establishment of persistence following primary infection. Monocytes are also a key cell type associated with viral reactivation and spread following viral reactivation. Because of their importance in the HCMV-host infection cycle and lifelong infection, it is critical to be able to study the

Diagnosis of recent rubella virus infection by demonstration of specific immunoglobulin M antibodies: comparison of solid-phase reverse immunosorbent test with sucrose density gradient centrifugation.

A solid-phase reverse immunosorbent test (SPRIST) based on the addition of an excess of rubella virus hemagglutinin was evaluated for the demonstration of rubella-specific immunoglobulin M (IgM), and the results were compared with those of the density gradient centrifugation technique. In a retrospective study in which 157 sera were tested, the two techniques yielded identical results (55 IgM-positive and 102 IgM-negative samples). In a prospective study, 592 sere were examined; 8 IgM-positive results by SPRIST corresponded to a recent rubella infection or vaccination. Neither rheumatoid facto

Cumulative live birth rates after IVF/ICSI cycles with sperm prepared by density gradient centrifugation vs. swim-up: a retrospective study using a propensity score-matching analysis.

BACKGROUND: Density gradient centrifugation (DGC) and swim-up (SU) are the two most widely used sperm preparation methods for in vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI). However, existing comparisons of IVF/ICSI outcomes following these sperm preparation methods are insufficient and controversial. METHODS: This retrospective study included all first autologous IVF and ICSI cycles performed between March 1, 2016, and December 31, 2020 in a single university-based center. A total of 3608 cycles were matched between DGC and SU using propensity score (PS) matching fo

Randomized non-inferiority within-patient trial of density gradient processing of sequential ejaculates versus centrifugation-free sperm separation device for ICSI in men with elevated sperm DNA fragmentation- study protocol.

BACKGROUND: Pathological sperm DNA fragmentation (SDF) has been associated with impaired fertilization, compromised embryo development, recurrent pregnancy loss, and lower pregnancy rates in assisted reproductive technology (ART). Two same-day strategies proposed to reduce SDF prior to in vitro fertilization/intracytoplasmic sperm injection (IVF/ICSI) are: (i) processing a second ejaculate obtained after a short abstinence interval using density gradient centrifugation (DGC), and (ii) selecting sperm from the first ejaculate using a centrifugation-free sperm separation device (CFSS). Randomised double-blind controlled study protocol of the cumulative live birth rate of in vitro fertilisation following sperm preparation by a microfluidic chip method versus a density gradient centrifugation method.

INTRODUCTION: Semen samples are commonly processed by a density gradient centrifugation method to isolate the most motile spermatozoa for fertilisation during in vitro fertilisation (IVF). Centrifugation can cause considerable damage to spermatozoa by the reactive oxygen species produced during the process, which has a negative effect on the IVF success. A microfluidic chip is an alternative sperm preparation technique using a miniaturised device containing channels and chambers in the microscale range for nanoparticle preparation. This technology enables the selection of mobile spermatozoa fr

Mass Cytometry Reveals Comparable Proportions of Leukocyte Subsets and Cell Surface P2X7 or CD39 on Human Peripheral Blood Mononuclear Cell Samples Isolated by Ficoll-Paque or SepMate Tube Density Gradient Centrifugation.

P2X7 is an adenosine 5'-triphosphate (ATP)-gated ion channel. CD39 (ectonucleotidase triphosphate diphosphohydrolase-1) hydrolyses ATP to reduce its extracellular concentration to limit P2X7 activation. Both P2X7 and CD39 are present on leukocytes. Ficoll-Paque density gradient centrifugation is widely used to isolate human peripheral blood mononuclear cells (PBMCs), yet the extent to which other density gradient centrifugation methods influence the expression of cell surface molecules remains unclear. Using mass cytometry, this study compared the proportions of mononuclear leukocyte subsets a

Rationale and study design of a double-blinded prospective randomized clinical trial comparing euploidy rates among embryos created from sibling oocytes injected with sperm processed by microfluidics or by density gradient centrifugation.

BACKGROUND: During the in vitro fertilization (IVF) process, sperm must be processed prior to insemination. While the most common method, density gradient centrifugation, can potentially damage sperm during centrifugation, a recent advancement in sperm processing uses a microfluidics system which selects for the most highly motile sperm. In selecting for these sperm which may be of higher quality, the euploidy rates of embryos created as a result may also be improved. The primary aim of this study is to compare the euploidy rates per mature oocyte between embryos created from sibling oocytes i

Cryptosporidium Oocyst Purification Using Discontinuous Gradient Centrifugation.

Many laboratory studies in cryptosporidial research require a source of purified oocysts. Sources can include experimentally infected laboratory animals or from samples collected from naturally infected animals and from clinical cases of human cryptosporidiosis. Purification of oocysts can be accomplished with readily available laboratory equipment including table-top centrifuges and microcentrifuges. Following purification, oocysts can be stored in antibiotic-supplemented buffers or in 2.5% aqueous potassium dichromate for over 6 months. Ultimately, oocyst viability and infectivity declines t

Sperm cleanup and centrifugation processing for cryopreservation.

Fertility rates with artificial insemination are highest with good-quality sperm samples. Therefore, nonviable sperm, cellular debris, and seminal plasma are preferably removed from semen samples prior to use or for preservation. Such compounds are sources where reactive oxygen species are generated during storage or upon cryopreservation, impairing sperm function. In this chapter we describe methods to remove seminal plasma and cellular debris from sperm samples, and for selecting morphologically normal motile sperm. The methods that are described here include: ordinary centrifugation, sperm

Removal of empty capsids from adeno-associated virus preparations by multimodal metal affinity chromatography.

Separation of empty and full adeno-associated virus capsids by multimodal metal affinity chromatography was investigated using a positively charged metal affinity ligand. A subpopulation of empty capsids eluted first, followed by full capsids, and later by more empty capsids and debris. Empty and full capsid composition of chromatography fractions was evaluated by cesium chloride density gradient centrifugation followed by stratigraphic flow analysis of the centrifuge tube contents, monitored by intrinsic fluorescence. Platelets in idiopathic thrombocytopenic purpura are increased in size but are of normal density.

To determine whether platelet size and volume are related to one another or to platelet age, subpopulations of platelets from patients with idiopathic thrombocytopenic purpura (ITP) have been produced on the basis of density using Percoll gradients. The density distribution of platelets from patients with ITP and from patients with other forms of thrombocytopenia (thought to be nonimmune in nature) was the same as in normal controls. However, the platelets in each density subpopulation from ITP patients were increased in size. beta-thromboglobulin (beta TG) content of platelets from each patie

Isolation and Analysis of Bacterial Ribosomes Through Sucrose Gradient Ultracentrifugation.

Ribosomes are large macromolecular complexes responsible for the translation process. During the course of ribosome biogenesis and protein synthesis, extra-ribosomal factors interact with the ribosome or its subunits to assist in these vital processes. Here we describe a method to isolate and analyze not only bacterial ribosomes but also their associated factors, providing insights into translation regulation. This detailed protocol allows the separation and monitoring of the ribosomal species and their interacting partners along a sucrose density gradient. Simultaneously, fractionation of the

Rapid Magnetic-microbead Method for Efficient Purification of Low-density Neutrophils.

Neutrophils, important leukocytes of innate immunity, have traditionally been considered a homogeneous cell population. Nevertheless, recent evidence has shown that neutrophils exist in several subpopulations. One such subpopulation is low-density neutrophils (LDN). LDN are found in small numbers in the blood of healthy individuals, but their numbers increase significantly in diseases such as systemic lupus erythematosus, autoimmune disorders, cancer, and infections. In these cases, LDN may participate in the pathogenesis of the disease. The only way to isolate LDN is through density-gradient
