In vitro Anti-Trypanosomal Activities of Indanone-Based Chalcones.

Human African trypanosomiasis is a neglected infectious disease that affects mostly people living in the rural areas of Africa. Current treatment options are limited to just four drugs that have been in use of four to nine decades. The life-threatening toxic side-effects associated with the use of these drugs are disconcerting. Poor efficacy, low oral bioavailability, and high cost are other shortcomings of current HAT treatments. Evaluating the potentials of known hits for other therapeutic areas may be a fast and convenient method to discover new hit compounds against alternative targets. A

Recent trends of chalcones potentialities as antiproliferative and antiresistance agents.

Chalcones are natural compounds found in plants, fruits and vegetables. This class of compounds has shown many biological activities including antioxidant, antimicrobial, anti-inflammatory, antifungal and antihypertensive, among others. In cancer, it has been reported that chalcones interfere in several points of the signal transduction pathways related to cellular proliferation, angiogenesis, metastasis, apoptosis and the reversal of multidrug resistance. The large number of research articles and patents related to chalcones is already an indication of their importance as a lead class of comp

Structural Dynamics and Susceptibility of Aminonaphthoquinone-Chalcone Hybrids Against Human Tyrosinase.

Tyrosinase is a key enzyme that catalyzes the rate-limiting step in melanogenesis. Although numerous mushroom-derived tyrosinase inhibitors have been identified, many exhibit limited efficacy against human tyrosinase. In this study, in silico approaches were employed to evaluate the inhibitory potential of aminonaphthoquinone-chalcone hybrids against human tyrosinase. Molecular docking analysis revealed that all 10 hybrids showed higher binding affinity than kojic acid. Among them, six hybrids (3, 4, 5, 7, 9, and 10) displayed higher binding affinity than the others and interacted with amino a

Conformationally adaptive benzimidazolium-chalcone hybrid salts as selective TGF-β1 inhibitors.

The transforming growth factor-β (TGF-β) signaling pathway is crucial in promoting tumor growth, enabling tumors to evade immune responses, and contributing to resistance against therapies. As a result, it is a significant target for cancer treatment. However, its full potential remains untapped because selectively inhibiting it without affecting normal cells is challenging. This study reports the design, synthesis, and comprehensive evaluation of novel benzimidazolium-chalcone hybrid salts (3a-3e) that strategically combine two privileged scaffolds with complementary anticancer mechanisms. Synthetic Progress and Antifungal Potential of Thiophene Chalcone Derivatives: A Review.

Fungal infections constitute a major global health concern, with annual mortality rates exceeding those of tuberculosis and malaria. This troubling situation has been exacerbated by the emergence of multidrug-resistant fungal strains, which have significantly undermined the effectiveness of current treatments. A number of antifungal medicines are now being utilised in clinical practice. Some of these treatments include azoles, polyenes, pyrimidines, and echinocandins. However, the therapeutic efficacy of these drugs is becoming increasingly limited due to the presence of unwanted side effects,

Molecular targets and anticancer activity of quinoline-chalcone hybrids: literature review.

α,β-Unsaturated chalcone moieties and quinoline scaffolds play an important role in medicinal chemistry, especially in the identification and development of potential anticancer agents. The multi-target approach or hybridization is considered as a promising strategy in drug design and discovery. Hybridization may improve the affinity and potency while simultaneously decreasing the resistance and/or side effects. The conjugation of quinolines with chalcones has been a promising approach to the identification of potential anticancer agents. Most of these hybrids showed anticancer activities thro

Synthesis and carbonic anhydrase inhibitory properties of novel chalcone substituted benzenesulfonamides.

Carbonic anhydrases (CAs, EC 4.2.1.1) are crucial metalloenzymes involved in many bioprocesses, through catalysis of the reversible hydration/dehydration process of CO2/HCO3-. The inhibition of human CA isoforms I and II with a new series of sulfonamide derivatives incorporating substituted chalcone moieties were studied in this study. All these newly synthesized sulfonamides demonstrated important inhibitory profiles to these CA isoforms with KIs in the range of 9.88 to 55.43nM, making these compounds interesting leads, with potential applications in medicinal chemistry.

Synthesis and Photophysical, Electrochemical, and DFT Studies of Piperidyl and Pyrrolidinyl Chalcones.

Small organic molecules with interesting optical and electrochemical properties find applications as organic luminescent materials. In this work, we report the synthesis of novel chalcones with D-A-D and D-A-D-A architecture, followed by their optical, electrochemical, and computational studies. The absorption band of these compounds occurs at 360-480 nm with emission maxima appearing around 513-552 nm. The large Stokes shifts (Δλ) for all compounds (90-132 nm) suggest intramolecular charge transfer (ICT) in the excited states. The molar absorptivity and fluorescence quantum yields were found
