Harnessing Multiplexed Proteolysis-Targeting Chimera for Comprehensive Influenza A Virus Targeting.

Influenza A virus (IAV) remains a global health threat due to its high mutation rate and immune evasion. Current therapies, limited by single-target mechanisms and drug resistance, are insufficient. To address this, we developed a multiplexed PROTAC strategy that simultaneously targets and degrades multiple components of the viral ribonucleoprotein (vRNP) complex, the core machinery of IAV replication. Our PROTAC molecule leverages the highly conserved 5' untranslated region of viral RNA, integrating an oligonucleotide targeting ligand with an E3 ligase-recruiting unit to precisely bind and de

Proteolysis Targeting Chimera Degraders of the METTL3-14 m6A-RNA Methyltransferase.

Methylation of adenine N6 (m6A) is the most frequent RNA modification. On mRNA, it is catalyzed by the METTL3-14 heterodimer complex, which plays a key role in acute myeloid leukemia (AML) and other types of blood cancers and solid tumors. Here, we disclose the first proteolysis targeting chimeras (PROTACs) for an epitranscriptomics protein. For designing the PROTACs, we made use of the crystal structure of the complex of METTL3-14 with a potent and selective small-molecule inhibitor (called UZH2). The optimization of the linker started from a desfluoro precursor of UZH2 whose synthesis is mor

An unusal case of hermaphroditism--a 46,XX/69,XXY chimera.

A diploid/triploid karyotype is an uncommon but important cause of true hermaphroditism and ambiguous genitalia. Individuals have a recognisable phenotype and characteristic hydatidiform placental changes. We report a 46,XX/69,XXY chimeric hermaphrodite. This case highlights the typical features (large placenta, intrauterine growth retardation, asymmetric growth, cranio-facial anomalies, syndactyly and pigmentary dysplasia). It illustrates the importance of obtaining skin and gonadal karyotypes in the case of genital ambiguity, as the venous lymphocytic karyotype is usually diploid.

Specific receptor detection by a functional keratinocyte growth factor-immunoglobulin chimera.

Fibroblast growth factor receptors (FGFRs) are encoded by at least four distinct highly conserved genes, and alternative splicing generates multiple gene products. The close relationship among different FGFRs has greatly increased the difficulty in generating specific immunochemical probes. As an alternative strategy, we constructed a fusion protein comprising keratinocyte growth factor (KGF) and an IgG1 Fc domain (HFc). The chimeric molecule was efficiently secreted from transfectants as a disulfide-linked dimer that bound KGFRs with high affinity. Moreover, the KGF-HFc, like native KGF, indu

Nonrandom X-chromosome inactivation--an artifact of cell selection (mouse chimera).

The present study shows that a high degree of variability in the distribution of XX and XY cells exists among various tissues of artificially assembled or spontaneously occurring mouse chimeras. This variation from the expected equal distribution in various tissues of an animal may have resulted from random distribution, unequal segregation, or selective differences of such cells during development. This variation may also explain the observed intertissue and interanimal variations in the distribution of inactive paternal and maternal X-chromosomes in some mammalian females.

Research progress and future perspectives in proteolysis-targeting chimeras.

Proteolysis-targeting chimeras (PROTACs) have emerged as a transformative modality within the targeted protein degradation (TPD) landscape, inducing spatial proximity between E3 ubiquitin ligases and proteins of interest (POIs) to hijack the ubiquitin-proteasome system (UPS). Unlike traditional occupancy-driven inhibitors, this catalytic, event-driven mechanism enables the targeting of historically "undruggable" proteomes and circumvents acquired resistance. However, the field faces formidable challenges, including suboptimal pharmacokinetic profiles, the stoichiometric "hook effect," and a di

Enzyme Activation of Triggerable Polymeric Lysosome Targeting Chimeras.

Targeted protein degradation (TPD) has been highly effective for intracellular targets, but extending this approach to extracellular and membrane-bound proteins remains difficult because most extracellular TPD (eTPD) strategies depend on ligand-targeting receptors (LTRs) whose expression and recycling vary across tissues. Existing LTR-independent, multivalent platforms expand the scope of eTPDs by eliminating this dependence on LTRs and enabling cancer-selective designs. Layering in an additional degree of selectivity through endogenously activatable triggers, such as overexpressed enzymes, ca

Identification of Tumor Hypoxia-Activated Degradation-Targeting Chimeras.

Targeted protein degradation (TPD) has emerged as a promising therapeutic strategy for treating cancers. Nevertheless, enhancing selectivity toward tumors remains a critical challenge. In this study, we designed and synthesized a series of hypoxia-activated degradation-targeting chimeras (HDTACs) by incorporating tumor hypoxia-activated groups (HAGs)─which generate reactive oxygen species (ROS) under tumor hypoxia conditions─into EGFR-based protein of interest (POI) ligands through appropriate linkers, leveraging the ability of ROS to induce protein degradation. Among the synthesized compounds

Blastocyst complementation and interspecies chimeras in gene edited pigs.

The only curative therapy for many endstage diseases is allograft organ transplantation. Due to the limited supply of donor organs, relatively few patients are recipients of a transplanted organ. Therefore, new strategies are warranted to address this unmet need. Using gene editing technologies, somatic cell nuclear transfer and human induced pluripotent stem cell technologies, interspecies chimeric organs have been pursued with promising results. In this review, we highlight the overall technical strategy, the successful early results and the hurdles that need to be addressed in order for the

Interspecies chimeras as a platform for exogenic organ production and transplantation.

Chronic diseases are associated with considerable morbidity and mortality. Therefore, new therapeutic strategies are warranted. Here, we provide a brief review outlining the rationale and feasibility for the generation of intraspecies and interspecies chimeras, which one day may serve as a platform for organ transplantation. These strategies are further associated with consideration of scientific and ethical issues.

[The expression of the mutant gene mi in mouse aggregation chimeras].

Using aggregation of 8-cell embryos belonging to two mouse strains, we produced 17 chimeras, mi/mi<==>+/+ (3 organisms), mi/+<==>+/+ (9 organisms), and +/+<==>+/+ (5 organisms), that achieved the age of 30 days. Chimerism was identified by mosaic pigmentation of coat and retinal pigment epithelium (RPE). All mi/mi<==>+/+ chimeras suffered from microphthalmia. Disturbances of eye development in these mice were similar to those in mi/mi animals. In all three chimeras, pigmentation of RPE and choroid was represented with alternating pigmented and depigmented patches. Mosaic RPE pattern probably r

Translational modelling of human microglia from cultures to chimeras.

Microglia play a crucial role in maintaining homeostasis of the central nervous system, whereas their dysfunction is implicated in the pathogenesis of various neurological diseases. Nevertheless, interspecific genetic differences pose challenges to translating findings from conventional rodent models. Human pluripotent stem cell-derived microglia offer a genetically relevant alternative, but these cells undergo profound transcriptomic changes and lose key homeostatic features when isolated from physiological brain cues. Accordingly, research has increasingly focused on advanced modelling platf

Antibody-lectin chimeras for glyco-immune checkpoint blockade.

Despite the curative potential of checkpoint blockade immunotherapy, many patients remain unresponsive to existing treatments. Glyco-immune checkpoints, which involve interactions of cell-surface glycans with lectin, or glycan-binding, immunoreceptors, have emerged as prominent mechanisms of immune evasion and therapeutic resistance in cancer. Here, we describe antibody-lectin chimeras (AbLecs), a modular system for glyco-immune checkpoint blockade. AbLecs are bispecific antibody-like molecules comprising a cell-targeting antibody domain and a lectin 'decoy receptor' domain that directly binds

Assessment of lymphocyte development in radiation bone marrow chimeras.

Transplantation of marrow between mouse strains congenic for CD45 after lethal irradiation establishes hematopoiesis driven by genetically marked cells in recipient animals. After several weeks, peripheral blood or primary and secondary lymphoid organs of transplant recipients can be evaluated for the presence of donor-derived cells. Two- or three-color flow cytometry can be used to identify the progeny of transplanted cells, to document their cell-surface phenotypes, and to follow development of T, B, and myeloid lineages in vivo.

Synthesis, biophysical, and biochemical properties of PNA-DNA chimeras.

Three PNA-DNA chimeric dimer synthons (tT, upT and uhT, see Sch. 1) have been synthesized in solution and used to make T20-analogue chimeras applying standard solid-phase DNA synthesis protocol. Duplex forming ability of chimeras with dA20 and their hydrolyses by 3'- and 5'-exonucleases (snake venom and bovine spleen phosphodiesterase, respectively) have been investigated.

Assessment of lymphocyte development in radiation bone marrow chimeras.

The Basic Protocol utilizes congenic strains of mice that differ at the CD45 (common leukocyte antigen) locus. Bone marrow-derived cells from these strains of mice can be identified by monoclonal antibodies specific for the two allelic variants of the CD45 molecule. Because this antigen is expressed on all nucleated blood cells, it provides a convenient genetic marker for following the progeny of transplanted bone marrow cells.
