In the present study we explored the mechanisms of carbachol-induced muscle atonia in the alpha-chloralose-anesthetized animal. We compared our findings to those that have been previously obtained in unanesthetized cats during muscle atonia occurring during natural active sleep. Accordingly, in cats anesthetized with alpha-chloralose, intracellular records were obtained from masseter motoneurons before and after carbachol-induced motor atonia. Following the induction of atonia, the membrane potential activity was dominated by high-frequency, discrete, hyperpolarizing potentials. These hyperpol

Diffusion tensor imaging (DTI) in animal models are essential for translational neuroscience studies. A critical step in animal studies is the use of anesthetics. Understanding the influence of specific anesthesia regimes on DTI-derived parameters, such as fractional anisotropy (FA) and mean diffusivity (MD), is imperative when comparing results between animal studies using different anesthetics. Here, the quantification of FA and MD under different anesthetic regimes, alpha-chloralose and isoflurane, is discussed. We also used a range of b-values to determine whether the anesthetic effect was

Evoked neural activity (ensemble single-unit activity and evoked field potential) and functional magnetic resonance imaging (fMRI) changes of the primary somatosensory cortex in response to electrical stimulation of the hind paw were studied in rats under anesthesia. The effects of stimulation frequency (ranging from 0.3 to 10 Hz) and types of anesthetics (alpha-chloralose and sodium pentobarbital) on blood oxygen level dependent (BOLD) activation and neural activation were compared. Both ensemble single-unit activity and BOLD signal changes achieved maximal activation at 3 Hz of stimulation a

Several general anesthetics and baclofen (Lioresal), an analog of gamma-aminobutyric acid, were tested for their effects on reflexes evoked by stimulation of afferent fibers in the carotid sinus nerve, aortic nerve and cervical vagus nerve. In both anesthetized and decerebrate unanesthetized cats, baclofen, barbiturates (methohexital and pentobarbital) and alpha-chloralose converted depressor responses evoked by afferent fiber stimulation to pressor episodes. The original depressor responses were reinstated temporarily by the gamma-aminobutyric acid antagonists bicuculline and picrotoxin, but

Epicardial coronary arteries dilate after release of a transient coronary occlusion in awake chronically instrumented dogs but not in anesthetized dogs studied acutely after surgery. To determine whether anesthesia or surgical trauma is responsible for this lack of reactive dilation, we evaluated the effect of anesthesia on reactive coronary dilation in six chronically instrumented dogs. Circumflex coronary diameter was measured with sonomicrometry. Response to release of a 20- or 30-s coronary occlusion was studied before and after sodium pentobarbital (20 +/- 2 mg/kg) and before and after al

The ability of captopril (SQ-14225), an angiotensin I-converting enzyme inhibitor, to reduce blood pressure in dogs anesthetized with either pentobarbital sodium or a combination of morphine plus chloralose was studied over a sixty minute period. In addition, since both of these anesthetic agents cause an increase in the plasma renin activity of these animals, the drug was used to further investigate whether the hypotensive action of this drug is dependent on the level of plasma renin activity. Captopril produced significant but similar decreases in blood pressure of both the anesthetized grou

1. Unilateral section of the nerves to an extensor muscle of the head, biventer cervicis, leads to an ataxia of the hind legs in cats.2. In chloralose anaesthetized cats, shocks to the central end of the cut biventer cervicis muscle nerves leads to direct discharges over lumbosacral ventral roots, to a facilitation of monosynaptic reflexes in flexor and extensor muscles and to a prolonged inhibition of spino-bulbo-spinal reflexes. Such effects are not unique, but follow stimulation of skin and muscle nerves of the forepaw.3. Supratentorial pathways are involved in descending spinal interaction

This study adopts and validates an anesthetic protocol designed for rat whisker stimulation experiments, achieving significant enhancements in the neural response of the barrel field cortex. By combining alpha-chloralose, low-dose Isoflurane (0.5%) and Dexdomitor, the protocol not only maintains a stable anesthetic state but also markedly improves the amplitude and latency of local field potential (LFP) signals. Experimental results reveal that LFP amplitudes in the barrel field under this protocol are twice as high as those achieved with Isoflurane and four times as high as those with Ketamin

The frequent accompaniment of hypertension by orthostatic circulatory disorders prompted us to investigate the effect of repeated and sustained head-up and head-down tilt positions on cardiovascular responses in spontaneously hypertensive rats vs. Wistar rats using radiotelemetric implants. Repeated orthostasis caused a transient elevation in blood pressure (7.3±1.7 mmHg) and heart rate (39.7±10.5 BPM), while repeated antiorthostasis led only to reversible tachycardia (85.6±11.7-54.3±16.8 BPM) in spontaneously hypertensive rats. In contrast to the Wistar rats, sustained tilt failed to affect t

We present a dual-imaging technique combining laser speckle contrast imaging and spectral-domain Doppler optical coherence tomography to enable quantitative characterization of local cerebral blood flow (CBF) changes in rat cortex in response to drug stimulus (e.g., cocaine) at high spatiotemporal resolutions. To examine the utility of this new technique, animal experiments were performed to study the influences of anesthetic regimes (e.g., isoflurane, alpha-chloralose) on the pharmadynamic effects of acute cocaine challenge. The results showed that cocaine-evoked CBF patterns (e.g., increases

The acid-base status in dogs anaesthetized with chloralose or combinations of chloralose and barbiturates was studied. No surgical procedures were used to elucidate whether the anesthesia per se changes in the acid-base status. Irrespective of the anaesthetic used, a progressive metabolic acidosis developed after 2-3 hours. This progressed for several hours despite normal arterial blood pressure and pulse rate. A theoretical model, based on the results, is presented. Even the induction of anesthesia caused an impairment of the peripheral circulation. This impairment, in combination with rising

Dose-response relationships for bronchoconstriction in response to aerosal histamine were assessed before and after vagotomy in 11 dogs anesthetized with barbiturates and in 9 dogs anesthetized with alpha-chloralose-urethan. The dose-response relationships following vagotomy were assessed during spontaneous ventilation and during muscular paralysis and mechanical ventilation with tidal volume (VT) similar to each animal's VT prior to vagotomy. After vagotomy the spontaneous VT of both groups increased but the VT of the alpha-chloralose-urethan group was significantly less than that of the barb

OBJECTIVE AND METHODS: Epicardial application of pharmacologic agonists has been used to study nociceptive and reflex responses to agents such as bradykinin. We utilized a model where intrapericardial bradykinin was administered in a closed-chest rat. The procedure allows for reproducible administration of microliter doses of pharmacologic agonists in both conscious and anesthetized animals.
RESULTS: Bradykinin (BK) has been shown to produce sympathoexcitatory reflexes when applied to the heart. BK typically produced a dose-dependent (0.001-10 micrograms) decrease in arterial blood pressure a

1 Some prostaglandins have been found to be capable of affecting respiration in anaesthetized cats.2 Prostaglandins E(1), E(2), F(2alpha), A(1) and A(2) all elicited increases in respiratory frequency when administered to cats anaesthetized with either pentobarbitone or alpha-chloralose. This effect was abolished by bilateral vagotomy.3 Prostaglandins of the E and A series, but not prostaglandin F(2alpha), elicited increases in tidal volume which were accompanied by falls in systemic blood pressure in cats anaesthetized with pentobarbitone. The changes in blood pressure were also obtained in c

The synthesis of new α,β-unsaturated furanuronic acid derivatives of α-gluco-, β-gluco- and β-manno-chloraloses via a convenient one pot procedure using the Knoevenagel-Doebner reaction approach are described. The dialdofuranose derivatives were reacted with malonic acid under Knoevenagel-Doebner reaction conditions and (E)-α,β-unsaturated furanuronic acid derivatives were obtained.

Antagonists of NMDA receptors can inhibit both the transmission of pain signals from the intestine and enteric reflexes. However, it is unknown whether doses of the NMDA antagonist, ketamine, that are used in anaesthetic mixtures suppress motility reflexes and visceromotor responses (VMRs). In fact, whether intestinal motility is affected by NMDA receptor blockers in vivo has been little investigated. We studied the effects of ketamine and memantine, administered intravenously or intrathecally. Rats were maintained under alpha-chloralose plus xylazine or pentobarbitone anaesthesia; VMR and jej

The influence of isoflurane on intestinal reflex vasoconstriction during hemorrhage was investigated in cats (n = 10) during basal chloralose-nitrous oxide anesthesia. Intestinal blood flow (IBF) was studied in a model with controllable intestinal perfusion pressures to exclude local myogenic vascular responses related to changes in intraluminal pressure. A jejunal segment, which was dissected free in situ, was perfused via an extracorporeal arterial circuit which included a roller pump and a variable arterio-venous shunt. Intestinal perfusion pressure was controlled by adjusting the shunt flo
