Experimental investigation and thermodynamic correlation of chlordiazepoxide solubility in supercritical CO₂.

This study investigated the solubility of chlordiazepoxide in supercritical carbon dioxide (SCCO₂) at pressures ranging from 12 to 30 (MPa) and temperatures ranging from 308 to 338 (K). The measured solubilities ranged from 19.8 to 576 parts per million (ppm), or mole fractions of 0.0198 × 10⁻³ to 0.576 × 10⁻³. We used two equations of state (SRK and ECM-PR), enlarged liquid models, and semi-empirical analysis to assess the experimental data. Applying these models to the experimental solubility data of the chlordiazepoxide-SCCO₂ system revealed that Chrastil’s empirical models best aligned wit

Aetiology of pneumonia in patients in the intensive care unit treated with chlordiazepoxide.

INTRODUCTION: Research on pneumonia in patients undergoing treatment for alcohol withdrawal symptoms (AWS) is sparse. This study describes the microbiological aetiology in this patient group within an intensive care unit (ICU). METHODS: We conducted a retrospective, observational study at Copenhagen University Hospital, Bispebjerg, Denmark. Patients admitted to the ICU between 1 June 2017 and 31 December 2020 who had received at least 200 mg of chlordiazepoxide for AWS within the preceding days were identified through electronic health records. Admissions presenting with clinical symptoms of

Naltrexone potentiates both amnestic and anxiolytic effects of chlordiazepoxide in mice.

Studies have suggested that opioid antagonists potentiate the anxiolytic effect but not the amnestic action of chlordiazepoxide (CDZ). We investigated the effects of naltrexone (NAL) on the anxiolytic and amnestic effects of CDZ in mice tested in the plus-maze discriminative avoidance task (DAT). Mice are conditioned to choose between two enclosed arms (one of which aversive) while avoiding the two open arms of the apparatus. This task measures memory (time spent in the aversive vs. time in the non-aversive enclosed arms) and anxiety (time spent in the open arms). Mice treated with saline (SAL

Effects of chlordiazepoxide, buspirone and cocaine on behavior suppressed by timeout presentation.

Male Wistar rats were exposed to a two-component multiple schedule: a random-interval 30 s schedule of pellet presentation and a conjoint random-interval 30 s schedule of pellet presentation, random-interval 2 s schedule of timeout 10 s presentation. Once responding had stabilized subjects were injected intraperitoneally with vehicle, chlordiazepoxide (1-30 mg/kg), buspirone (0.1-4.2 mg/kg) or cocaine (1-30 mg/kg), 15 min before the start of the experimental session. Before drug administration, punished response rates were less than 30% of unpunished response rates for four of the six subjects

Presession noise increases sensitivity to chlordiazepoxide's discriminative stimulus in pigeons.

1. Pigeons were trained to discriminate chlordiazepoxide (CDP) from saline using two-key food reinforced drug discrimination procedures. Discriminative control by CDP was maintained despite extended training with vehicle-like doses of CDP, by using a modified "fading" procedure that provided for a mixture of drug discrimination training sessions preceded by an i.m. injection of either 8.0 mg/kg CDP, or a lower training dose of CDP (4.0, 2.8, 2.0, 1.4, 1.0, 0.7, or 0.5 mg/kg CDP), or saline. The lower training dose was decreased across blocks of sessions. 2. Four lower training doses (1.4, 1.0,

Tolerance/cross-tolerance to the discriminative stimulus effects of chlordiazepoxide and bretazenil.

Rats were trained to discriminate 10 mg/kg chlordiazepoxide (CDAP) from saline in a two-lever drug discrimination procedure. Both CDAP and the nonsedative benzodiazepine partial agonist, bretazenil, dose-dependently substituted for the training dose of CDAP. Training was then suspended and half of the rats were placed on chronic CDAP, while the other half received water. Tolerance to the discriminative stimulus effects of CDAP developed after 1 mo and a final dose of approximately 110 mg/kg/d, as evidenced by the fact that the training dose of CDAP no longer produced drug-appropriate respondin

Age and gender effects on chlordiazepoxide kinetics: relation to antipyrine disposition.

Twelve normal subjects aged 24-41 years, and 12 subjects aged 62-79 years, received single 50-mg doses of chlordiazepoxide hydrochloride by mouth and by intravenous injection on two occasions. Chlordiazepoxide volume of distribution was significantly correlated with body weight (r = 0.63, p less than 0.001), but was not related to age or sex. Among male subjects, elimination half-life was prolonged (20 vs. 8 h, p less than 0.025) and clearance reduced (20 vs. 43 ml/min, p less than 0.05) in elderly as opposed to young volunteers. Among women, there was no significant difference between elderly

Relationship of brain cyclic nucleotide levels and the interaction of ethanol with chlordiazepoxide.

The effects of combined administration of ethanol (4 g/kg) and chlordiazepoxide (CDP, 12.5 mg/kg) on mouse brain c-AMP and c-GMP levels were investigated in order to test the hypothesis that the supra-additive effect of CDP on ethanol sleep time may be related to a supra-additive alteration in brain cyclic nucleotide levels induced by the combined drugs. Ethanol alone or CDP by itself did not cause any change in brain c-AMP levels, except for a transient decrease in the cerebral cortex and midbrain at 0.5 hr after ethanol injection, as well as a transient increase in the cerebellum at 0.5 hr a

Effect of acute alcohol intoxication on the metabolism and plasma kinetics of chlordiazepoxide.

1. The metabolism and plasma kinetics of chlordiazepoxide have been determined in a group of volunteers and in a group of patients with acute alcohol intoxication. 2. Using the SAAM 26 non-linear least squares fitting programme, all chlordiazepoxide plasma concentration v time data following oral administration could be analysed in terms of a one-compartment open model with metabolic conversion of chlordiazepoxide to desmethylchlordiazepoxide. 3. Acutely intoxicated patients showed a prolonged elimination of chlordiazepoxide and a reduced clearance when compared with alcohol-free volunteers. T

Kinetics and mechanisms of hydrolysis of 1,4-benzodiazepines I: chlordiazepoxide and demoxepam.

Differential absorbance spectroscopy was successfully used to follow the hydrolysis kinetics of chlordiazepoxide and demoxepam from pH 1 to 11. Loss of the methylamino group from chlordiazepoxide produced demoxepam. Demoxepam degraded by a parallel consecutive reaction to 2-amino-5-chlorobenzophenone and a glycine derivative. Two intermediates were observed by TLC for demoxepam hydrolysis. One was assigned the open-ring structure resulting from amide hydrolysis, which kinetically appears to be the major mechanistic route leading to the benzophenone product. The other intermediate, representing

Schedule-induced polydipsic consumption of hypertonic NaCl solutions: effects of chlordiazepoxide.

Rats were exposed to daily, 3-h sessions of schedule-induced polydipsia (SIP) in which either water, hypertonic NaCl solution (1.5% or 2.2%), or concurrent water and 1.5% NaCl were available. Each condition was in effect for several, consecutive weeks. Presession subcutaneous injections of chlordiazepoxide (CDZP) produced dose-related increases in the polydipsic ingestion of both NaCl solutions but had smaller and less certain effects on water consumption. Under the concurrent-fluid presentation condition, CDZP primarily increased NaCl solution consumption. Conditions generating SIP may functi

The inability of CCK to block (or CCK antagonists to substitute for) the stimulus effects of chlordiazepoxide.

To further examine the relationship between cholecystokinin (CCK) and GABA, the present study assessed the ability of the CCK-A antagonist devazepide and the CCK-B antagonist L-365,260 to substitute for the stimulus effects of chlordiazepoxide (CDP), as well as the ability of CCK-8s to block these effects, in female Long-Evans rats within the conditioned taste aversion baseline of drug discrimination learning. Both devazepide and L-365,260 failed to substitute for the discriminative stimulus properties of CDP, and CCK-8s failed to block its stimulus effects. The benzodiazepine diazepam did sub

Isobolographic analysis of chlordiazepoxide and triazolam combinations in squirrel monkeys discriminating triazolam.

RATIONALE: The discriminative stimulus (DS) effects of chlordiazepoxide (CDP) differ from those of other typical benzodiazepine (BZ) agonists in that CDP does not always occasion full substitution for a BZ agonist DS. OBJECTIVES: The present study tested the hypothesis that the unusual DS effects of CDP may result from its relatively low intrinsic efficacy by examining the combinations of CDP and triazolam using isobolographic analysis in squirrel monkeys discriminating triazolam. METHODS AND RESULTS: Squirrel monkeys were previously trained to discriminate triazolam (0.03 mg/kg, i.v.) from

Inhibition by antimanic drugs of hyperactivity induced by methamphetamine-chlordiazepoxide mixture in mice.

The effects of lithium chloride and other antimanic drugs on locomotor hyperactivity induced by a mixture of methamphetamine (MAMP) and chlordiazepoxide (CDZP) were examined in mice, using an Animex activity meter. CDZP (12.5 mg/kg) given SC in combination with MAMP (1 mg/kg) caused a marked increase in locomotor activity, as compared with that in mice treated with MAMP alone. However, when CDZP (12.5 mg/kg) was administered together with 0.5 or 2.0 mg/kg of MAMP, no significant enhancement was observed. Lithium (2 and 3 mEq/kg, IP) and carbamazepine (4 and 8 mg/kg, IP) inhibited the hyperacti

Imipramine and chlordiazepoxide in depressive and anxiety disorders. I. Efficacy in depressed outpatients.

We randomly assigned 425 outpatients, independently classified as primarily depressed by two trained psychiatrists, to double-blind treatment with Imipramine hydrochloride, chlordiazepoxide hydrochloride, or placebo. Those patients who remained at least moderately depressed (following a two-week placebo washout period) were treated for an additional eight weeks. An endpoint analysis of 387 patients who completed two or more weeks of medication disclosed early therapeutic advantages of chlordiazepoxide. By week 4 of treatment, however, imipramine produced more improvement than did placebo and c
