Metabolomics analyses and comparative insight to neuroprotective potential of unripe fruits and leaves of Citrus aurantium ethanolic extracts against cadmium-induced rat brain dysfunction: involvement of oxidative stress and akt-mediated CREB/BDNF and GSK3β/NF-κB signaling pathways.

Serious neurological disorders were associated with cadmium toxicity. Hence, this research aimed to investigate the potential neuroprotective impacts of the ethanolic extracts of Citrus aurantium unripe fruits and leaves (CAF and CAL, respectively) at doses 100 and 200 mg/kg against cadmium chloride-provoked brain dysfunction in rats for 30 consecutive days. HPLC for natural pigment content revealed that CAF implied higher contents of Chlorophyll B, while the CAL has a high yield of chlorophyll A and total carotenoid. Fifty-seven chromatographic peaks were identified by UPLC/MS/MS; 49 and 29 w

Arabidopsis thaliana defense response to the ochratoxin A-producing strain (Aspergillus ochraceus 3.4412).

OTA-producing strain Aspergillus ochraceus induced necrotic lesions, ROS accumulation and defense responses in Arabidopsis . Primary metabolic and defense-related proteins changed in proteomics. Ascorbate-glutathione cycle and voltage-dependent anion-selective channel proteins fluctuated. Mycotoxigenic fungi, as widespread contaminants by synthesizing mycotoxins in pre-/post-harvest infected plants and even stored commercial cereals, could usually induce plant-fungi defense responses. Notably, ochratoxin A (OTA) is a nephrotoxic, hepatotoxic, teratogenic, immunotoxic and phytotoxic mycotoxin.

Effects of loss of CO2-Responsive CCT Protein interaction with 14-3-3 proteins and its ectopic expression on starch synthesis in rice.

CO2-Responsive CCT Protein (CRCT) is a transcription factor that positively regulates the expression of starch synthesis-related genes in the vegetative organs of rice. CRCT interacts with 14-3-3 proteins. Yeast two-hybrid analysis showed that the CRCT with the S217A mutation was unable to interact with CRCT. This mutated CRCT was introduced into the CRCT knockout rice. The transcript of CRCT-S217A was detected significantly by qRT-PCR, but not by immunoblotting. Nevertheless, starch accumulation and the expression of starch synthesis-related genes in the leaf sheath of the transformants were

Psb28 protein is involved in the biogenesis of the photosystem II inner antenna CP47 (PsbB) in the cyanobacterium Synechocystis sp. PCC 6803.

The role of the Psb28 protein in the structure and function of the photosystem II (PSII) complex has been studied in the cyanobacterium Synechocystis sp. PCC 6803. The protein was localized in the membrane fraction and, whereas most of the protein was detected as an unassembled protein, a small portion was found in the PSII core complex lacking the CP43 antenna (RC47). The association of Psb28 with RC47 was further confirmed by preferential isolation of RC47 from the strain containing a histidine-tagged derivative of Psb28 using nickel-affinity chromatography. However, the affinity-purified fr

Nitric oxide delays tomato fruit carotenoid accumulation by inhibiting the transcriptional activation of the SlbZIP16-SlPol η module.

The massive accumulation of carotenoids concomitant with the degradation of chlorophyll constitutes a feature of tomato fruit ripening. Nitric oxide (NO) significantly inhibits carotenoid synthesis, yet the underlying molecular regulatory mechanism remains unclear. Here, we identified a DNA polymerase, SlPol eta (SlPol η), whose expression is positively correlated with carotenoid accumulation. Transient overexpression of SlPol η in tomato significantly upregulated the expression of carotenoid biosynthesis genes and accelerated chlorophyll degradation; Virus-induced gene silencing (VIGS) of SlP

Structural Dynamics and Optical Spectra of the Phycobiliprotein PC612 Complex.

Phycocyanin 612 (PC612) is a light-harvesting complex found in cryptophyte algae, characterized by its distinct open structure and flexible protein conformation, allowing the algae to survive in low-light aqueous environments. Although cryptophyte algae are known to show a diverse range of spectroscopic properties, the complex electronic structures and photophysical properties of their pigment molecules, known as bilins, are not yet fully understood, particularly in terms of how the surrounding protein environment influences them. In this study, we performed quantum mechanics/molecular mechani

Non-photochemical quenching in cryptophyte alga Rhodomonas salina is located in chlorophyll a/c antennae.

Photosynthesis uses light as a source of energy but its excess can result in production of harmful oxygen radicals. To avoid any resulting damage, phototrophic organisms can employ a process known as non-photochemical quenching (NPQ), where excess light energy is safely dissipated as heat. The mechanism(s) of NPQ vary among different phototrophs. Here, we describe a new type of NPQ in the organism Rhodomonas salina, an alga belonging to the cryptophytes, part of the chromalveolate supergroup. Cryptophytes are exceptional among photosynthetic chromalveolates as they use both chlorophyll a/c pro

Identification and characterization of Cd-induced peptides in Egeria densa (water weed): Putative role in Cd detoxification.

Egeria densa has ability to grow in heavy metal contaminated and polluted bodies of water. Shoots exposed to Cd at concentrations up to 300microM for 7 days showed a pronounced decrease in chlorophyll a and in total protein concentration. Thiol-containing compounds and low-molecular-weight polypeptides were detected in Cd-treated plant extracts by gel filtration chromatography. Two Cd-binding fractions, a thiol-enriched fraction and a non-thiol fraction with a lower molecular weight were identified in extracts by gel filtration. The main fraction of thiol-containing polypeptide, purified by ge

Inflorescence Photosynthesis Determines Resource Allocation, Seed Production and Senescence in an Agricultural Weed.

Agricultural weeds typically combine high reproductive output with fast vegetative growth. Here we analyzed the processes that overcome a potential trade-off between vegetative growth and reproduction in the weed Avena fatua. We determined photosynthetic function of inflorescences and analyzed the transcriptomes of individual inflorescence organs (palea, lemma, awn, and glume) compared to leaves and stems. Source-sink manipulations (defoliation, panicle shading, and de-awning) were conducted to assess the contribution of inflorescence photosynthesis to seed formation and to analyze feedback ef

Tandem duplication-driven expansion and UV-B stress adaptation of the LHC gene family in Artemisia annua L.

BACKGROUND: Artemisia annua L., is the primary natural source of the antimalarial drug artemisinin. In nature, fluctuating light is a major environmental stress that affects plant growth and artemisinin biosynthesis. Although the light-harvesting chlorophyll a/b-binding (LHC) superfamily plays a key role in mediating plant responses to fluctuating light, systematic research of this gene family in A. annua has not yet been conducted, limiting our understanding of light adaptation in this medicinally important species. RESULTS: This study investigated the evolutionary dynamics and functional ad

The out of phase 1 mutant defines a role for PHYB in circadian phase control in Arabidopsis.

Arabidopsis displays circadian rhythms in stomatal aperture, stomatal conductance, and CO(2) assimilation, each of which peaks around the middle of the day. The rhythmic opening and closing of stomata confers a rhythm in sensitivity and resistance, respectively, to the toxic gas sulfur dioxide. Using this physiological assay as a basis for a mutant screen, we isolated mutants with defects in circadian timing. Here, we characterize one mutant, out of phase 1 (oop1), with the circadian phenotype of altered phase. That is, the timing of the peak (acrophase) of multiple circadian rhythms (leaf mov

Synthesis and characterization of de novo designed peptides modelling the binding sites of [4Fe-4S] clusters in photosystem I.

Photosystem I (PS I) converts the energy of light into chemical energy via transmembrane charge separation. The terminal electron transfer cofactors in PS I are three low-potential [4Fe-4S] clusters named F(X), F(A) and F(B), the last two are bound by the PsaC subunit. We have modelled the F(A) and F(B) binding sites by preparing two apo-peptides (maquettes), sixteen amino acids each. These model peptides incorporate the consensus [4Fe-4S] binding motif along with amino acids from the immediate environment of the iron-sulfur clusters F(A) and F(B). The [4Fe-4S] clusters were successfully incor

LcSABP2, a salicylic acid binding protein 2 gene from Lycium chinense, confers resistance to triclosan stress in Nicotiana tabacum.

The triclosan (TCS) is one of the most commonly detected organic pollutants in the sewage sludge. TCS could induce phytotoxicity in plants. Salicylic acid (SA) is a phenolic compound capable of enhancing plant growth and development. It is well documented that abiotic stress tolerance could be enhanced by exogenous application of SA. However, the regulatory mechanisms for functions of endogenous SA in plants' responses to xenobiotics stress remains unclear. Our results indicated that TCS suppressed plant growth by restricting photosynthesis, decreasing chlorophyll contents and inducing over pr

Genome-wide identification of Lhcb gene family and functional characterization of BcLhcb1.2 for cold tolerance in non-heading Chinese cabbage.

The light-harvesting chlorophyll a/b binding protein (Lhcb) constitutes the antenna system of photosystem II (PSII), which also plays an important role in plant growth and responses to abiotic stress. However, the role of Lhcb genes in cold stress remains poorly characterized in non-heading Chinese cabbage. Here, 25 BcLhcb genes were identified and classified into 8 subfamilies, and these genes were mapped to nine chromosomes. Analysis indicated that BcLhcb genes may respond to hormonal and abiotic stress. Subcellular localization and expression pattern demonstrated that BcLhcb1.2, a chloropla

Transcription Factor McHB7 Improves Ice Plant Drought Tolerance through ABA Signaling Pathway.

As global climate change continues, drought episodes have become increasingly frequent. Studying plant stress tolerance is urgently needed to ensure food security. The common ice plant is one of the model halophyte plants for plant stress biology research. This study aimed to investigate the functions of a newly discovered transcription factor, Homeobox 7 (HB7), from the ice plant in response to drought stress. An efficient Agrobacterium-mediated transformation method was established in the ice plant, where ectopic McHB7 expression may be sustained for four weeks. The McHB7 overexpression (OE)

The breast cancer resistance protein protects against a major chlorophyll-derived dietary phototoxin and protoporphyria.

The breast cancer resistance protein (BCRPABCG2) is a member of the ATP-binding cassette family of drug transporters and confers resistance to various anticancer drugs. We show here that mice lacking Bcrp1Abcg2 become extremely sensitive to the dietary chlorophyll-breakdown product pheophorbide a, resulting in severe, sometimes lethal phototoxic lesions on light-exposed skin. Pheophorbide a occurs in various plant-derived foods and food supplements. Bcrp1 transports pheophorbide a and is highly efficient in limiting its uptake from ingested food. Bcrp1(-/-) mice also displayed a previously unk

Identification of distinct pH- and zeaxanthin-dependent quenching in LHCSR3 from Chlamydomonas reinhardtii.

Under high light, oxygenic photosynthetic organisms avoid photodamage by thermally dissipating absorbed energy, which is called nonphotochemical quenching. In green algae, a chlorophyll and carotenoid-binding protein, light-harvesting complex stress-related (LHCSR3), detects excess energy via a pH drop and serves as a quenching site. Using a combined in vivo and in vitro approach, we investigated quenching within LHCSR3 from Chlamydomonas reinhardtii. In vitro two distinct quenching processes, individually controlled by pH and zeaxanthin, were identified within LHCSR3. The pH-dependent quenchi

Supramolecular organization of photosystem II and its light-harvesting antenna in partially solubilized photosystem II membranes.

We present an extended analysis of the organization of green plant photosystem II and its associated light-harvesting antenna using electron microscopy and image analysis. The analysis is based on a large dataset of 16 600 projections of negatively stained PSII-LHCII supercomplexes and megacomplexes prepared by means of three different pretreatments. In addition to our previous work on this system [Boekema, E.J., van Roon, H., Calkoen, F., Bassi, R. and Dekker, J.P. (1999) Biochemistry 38, 2233-2239], the following results were obtained. Light-induced biogenesis of light-harvesting complex I (LHC I) during chloroplast development in barley (hordeum vulgare). Studies using cDNA clones of the 21- and 20-kilodalton LHC I apoproteins.

The light-harvesting complex (LHC) Ib pigment-proteins form the major component of the LHC I complex in higher plants. They comprise chlorophylls a and b, xanthophylls, and at least two polypeptide subunits of 21 and 20 kD in barley (Hordeum vulgare). We have identified two cDNA clones, LHC Ib-21 and LHC Ib-20, encoding the 21- and 20-kD LHC Ib apoproteins, respectively. N-terminal protein sequences of the purified LHC Ib polypeptides were used for the unequivocal correlation of these clones to their respective apoproteins. The cDNA clones encode two proteins that have strong sequence similari

Rational design and herbicidal activity of tenuazonic acid derivatives based on photosystem II D1 protein.

BACKGROUND: Natural products are an important source for the development of herbicides. Tenuazonic acid (TeA) is a natural phytotoxin produced by phytopathogenic fungi that strongly inhibits photosystem II (PS II) by preventing electron flow from the primary quinone acceptor (QA) of PS II to the secondary quinone acceptor (QB), thereby directly disrupting the key chain of plant photosynthesis. RESULTS: A series of TeA derivatives were rationally designed and synthesized based on the PS II D1 protein, and their herbicidal activities were evaluated. Compound C11 achieved > 90% control of Abutil
