Separation and determination of pseudoephedrine, dextromethorphan, diphenhydramine and chlorpheniramine in cold medicines by nonaqueous capillary electrophoresis.

An easy, rapid and simple nonaqueous capillary electrophoresis (NACE) method was developed for the identification and determination of four basic nitrogenous compounds, i.e. pseudoephedrine (PE), dextromethorphan (DXM), diphenhydramine (DHM) and chlorpheniramine (CLP). The most suitable running buffer was composed of 40 mM ammonium acetate, 10% acetonitrile (ACN) in methanol with a fused-silica capillary column (47 cm x 75 microm i.d.), 25 kV applied voltage and 25 degrees C capillary temperature. The calibration curves revealed linear relationships between the peak area for each analyte and i

In vitro adsorption of phenobarbital, chlorpheniramine maleate, and theophylline by four commercially available activated charcoal suspensions.

Four commercially available antidotal activated charcoal suspensions (Actidose Aqua, InstaChar, LiquiChar, and CharcoAid 2000) were evaluated with respect to their abilities to adsorb three test drugs in vitro from simulated gastric fluid. Different volumes of each suspension were added to glass vials containing 20 mL of stock solutions of each drug. The vials were shaken for a time more than sufficient to ensure adsorption equilibrium, then the charcoal was removed from the samples by filtration. The filtrates were analyzed by ultraviolet spectroscopy to determine the residual drug concentrat

Inhibition of histamine-induced nasal symptoms by the H1 antihistamine chlorpheniramine maleate: demonstration of topical effect.

To study whether a histamine H1 receptor antagonist inhibits allergic rhinitis symptoms by a peripheral or a central mode of action 21 normal volunteers were challenged with 500 micrograms histamine chloride in the right nasal cavity on four occasions after bilateral pretreatment with an H1 antihistamine spray and a placebo spray in various combinations. The H1 antihistamine (chlorpheniramine maleate, 0.5 mg) significantly inhibited histamine-induced tickling (P less than 0.01), sneezing (P = 0.01) and discharge (P less than 0.01), but only when it was given on the same side as histamine. In Vitro Virucidal Effect of Intranasally Delivered Chlorpheniramine Maleate Compound Against Severe Acute Respiratory Syndrome Coronavirus 2.

Background The initial global outbreak of the novel coronavirus disease 2019 (COVID-2019) pandemic, which is responsible for the severe acute respiratory syndrome 2 (SARS-CoV-2), shares similarities with the severe acute respiratory syndrome coronavirus (SARS-CoV) and Middle East respiratory syndrome coronavirus (MERS-CoV) and behaves similarly to influenza with a high intranasal viral load. The genome sequence of COVID-19 opened the opportunity for multiple in vitro and clinical trials, but we still do not have a clear path to treatment. Chlorpheniramine maleate (CPM) is a safe and effective

Muricidal suppression by chlorpheniramine and changes in brain levels following dietary-induced thiamine deficiency in rats.

The effects of thiamine deficiency on pharmacological and pharmacokinetic activities of chlorpheniramine were investigated in rats. Chlorpheniramine (5-10 mg/kg) showed a dose-dependent suppressive effect on muricide induced by thiamine deficiency. The ED50 value for muricidal suppression at 1 hr was approximately 7.1 mg/kg (95% confidence limits, 5.4-9.3 mg/kg) after oral administration. Using a high-performance liquid chromatographic (HPLC) method, chlorpheniramine was detectable at 10 min in the blood and brain of rats. The present pharmacokinetic data suggest that chlorpheniramine can easi

The effects of chlorpheniramine and antiallergic drugs on Ascaris suum antigen-induced active cutaneous anaphylaxis in dogs.

Effects of chlorpheniramine and two antiallergic drugs on the active cutaneous anaphylaxis (ACA) reaction induced by intradermal injection of Ascaris suum antigen in naturally sensitized dogs were investigated. Chlorpheniramine (10 mg/kg, intraduodenally (i.d.)) almost abolished the ACA reaction. NCO-650 (100 mg/kg, i.d.) had no inhibitory effect, while tranilast (300 mg/kg, i.d.) showed a weak inhibitory effect. These findings show that the ACA reaction is almost totally mediated by histamine and ACA reaction is considerably resistant to antiallergic drugs such as tranilast and NCO-650.

A Randomized Controlled Pilot Trial to Test the Efficacy of Intranasal Chlorpheniramine Maleate With Xylitol for the Treatment of Allergic Rhinitis.

The prevalence of allergic rhinitis (AR), including symptoms of sneezing, nasal itching, airflow obstruction, and nasal discharge caused by histamine and immunoglobulin E (IgE)-mediated reactions, is ~30% in the U.S. Recent studies seem to suggest that the allergic inflammatory processes in AR may be induced by the interaction between an allergen (trigger) and the nasal microbiome (substrate). In this study, we have identified two agents with antihistaminic and microbiome-modulating characteristics that can be administered intranasally, namely, chlorpheniramine maleate (CPM) and xylitol (X). Effects of chlorpheniramine on emergence agitation after general anesthesia for ureteroscopic stone surgery: a retrospective cohort study.

BACKGROUND: The presence of a urinary catheter, postoperative pain, and postoperative nausea and vomiting are risk factors for emergence agitation (EA). Antimuscarinic agents are primary agents used in the prevention and treatment of urinary catheter-related bladder discomfort. Chlorpheniramine has antimuscarinic, antinociceptive, and antiemetic effects. This retrospective study investigated the role of chlorpheniramine in EA prevention following ureteroscopic stone surgery. METHODS: Of 110 adult patients who underwent ureteroscopic stone surgery under general anesthesia between January and D

Chlorpheniramine is no more effective than placebo in relieving the symptoms of childhood atopic dermatitis with a nocturnal itching and scratching component.

BACKGROUND: Childhood atopic dermatitis (AD) is a common disorder affecting 15% of children aged over 18 months. AD is associated with intense nocturnal itching. The central sedative effect of antihistamines is thought to be useful in interrupting the itching cycle and may prevent exacerbations. OBJECTIVE: A multi-centred, double-blind, placebo-controlled study was carried out in 155 children to evaluate chlorpheniramine in alleviating symptoms of AD. METHODS: Assessments were carried out over a 4-week study period consisting of 3 visits to out-patient clinics. During the visits the severity
