Dietary Supplementation with Cholic Acid Reduces Insulin Secretion in Response to Intraperitoneal Glucose Administration in Rats.

The major characteristic of type 2 diabetes is insulin resistance, which is associated with plasma level of 12-hydroxylated bile acids (BAs) in humans. In this study, we investigated whether the rise of enterohepatic 12-hydroxylated BAs associates with glucose tolerance and/or insulin secretion using rats fed a diet supplemented with cholic acid (CA) at a level of 0.5 g/kg diet. Almost no increase was observed in plasma insulin in response to the intraperitoneal glucose administration in the CA-fed rats despite the significant increase of plasma insulin in control with the same treatment. In c

Cholic Acid-Derived Amphiphile which Combats Gram-Positive Bacteria-Mediated Infections via Disintegration of Lipid Clusters.

Inappropriate and uncontrolled use of antibiotics results in the emergence of antibiotic resistance, thereby threatening the present clinical regimens to treat infectious diseases. Therefore, new antimicrobial agents that can prevent bacteria from developing drug resistance are urgently needed. Selective disruption of bacterial membranes is the most effective strategy for combating microbial infections as accumulation of genetic mutations will not allow for the emergence of drug resistance against these antimicrobials. In this work, we tested cholic acid (CA) derived amphiphiles tethered with

Oral Delivery of Cholic Acid-Derived Amphiphile Helps in Combating Salmonella-Mediated Gut Infection and Inflammation.

A major impediment to developing effective antimicrobials against Gram-negative bacteria like Salmonella is the ability of the bacteria to develop resistance against existing antibiotics and the inability of the antimicrobials to clear the intracellular bacteria residing in the gastrointestinal tract. As the critical balance of charge and hydrophobicity is required for effective membrane-targeting antimicrobials without causing any toxicity to mammalian cells, herein we report the synthesis and antibacterial properties of cholic acid-derived amphiphiles conjugated with alkyl chains of varied h

Methylprednisolone administration in primary biliary cirrhosis increases cholic acid turnover, synthesis, and deoxycholate concentration in bile.

As immunosuppressive agents, corticosteroids may be considered an appropriate treatment for primary biliary cirrhosis, even if bone loss and other side effects may occur. We studied biliary lipid metabolism in 10 nonicteric patients, with histologically proven primary biliary cirrhosis (stage I-IV). We administered methylprednisolone (24 mg daily) for 30 days to ascertain its effects on biliary lipid metabolism, which are largely still unknown. All patients underwent a 30-day drug-washout period before entering the trial. The following parameters were studied before and after methylprednisolon

Gut microbiota-derived cholic acid mediates neonatal brain immaturity and white matter injury under chronic hypoxia.

Chronic hypoxia, common in neonates, disrupts gut microbiota balance, which is crucial for brain development. This study utilized cyanotic congenital heart disease (CCHD) patients and a neonatal hypoxic rat model to explore the association. Both hypoxic rats and CCHD infants exhibited brain immaturity, white matter injury (WMI), brain inflammation, and motor/learning deficits. Through 16s rRNA sequencing and metabolomic analysis, a reduction in B. thetaiotaomicron and P. distasonis was identified, leading to cholic acid accumulation. This accumulation triggered M1 microglial activation and inf

Differences in iron balance observed with dietary cholic acid supplementation and marginal iron deficiency in rats.

We investigated whether a cholic acid (CA)-supplemented diet and marginal iron deficiency (MID) diet influence hepatic lipid accumulation and iron balance in rats for 2 weeks. The CA diet enhanced hepatic lipid accumulation and modulated iron metabolism such as enhancement of fecal iron excretion, reduction in iron absorption, and no alteration in plasma iron levels. The MID diet did not alter hepatic lipid concentrations with reduced iron concentration in the liver and plasma. In combination, influence of the CA supplementation on the hepatic iron concentration was opposite between iron-suffi

The novel Listeria monocytogenes bile sensor BrtA controls expression of the cholic acid efflux pump MdrT.

Mammalian bile has potent anti-microbial activity, yet bacterial pathogens of the gastrointestinal tract and hepatobiliary system nonetheless persist and replicate within bile-rich environments. Listeria monocytogenes, a Gram-positive pathogen, encounters bile at three stages throughout its infectious cycle in vivo: in the gut during initial infection, in the liver where it replicates robustly and in the gallbladder, from which it can return to the intestine and thence to the environment. The mechanisms by which L. monocytogenes senses mammalian bile and counteracts its bactericidal effects ar

Cholic Acid-Induced Akkermansia Expansion and FGF-15 Upregulation Improve Diabetes While Exacerbating Steatohepatitis in TSOD Mice.

Type 2 diabetes mellitus (T2DM) and metabolic dysfunction-associated steatohepatitis (MASH) frequently coexist and are mechanistically linked via insulin resistance, lipotoxicity, and chronic inflammation. Cholic acid (CA), a primary bile acid (BA), modulates metabolic and immune pathways by influencing BA composition and the gut microbiota. In this study, we examined the role of dietary CA in the modulation of T2DM and MASH in Tsumura Suzuki obese diabetes (TSOD) mice, a model of spontaneous obesity and diabetes. Mice were fed a high-fat, high-cholesterol diet with or without CA (iHFC and CA(

Bezafibrate and Cholic Acid-Mediated Lipid Reduction Improves the Cryotolerance of Vitrified In Vivo-Derived Porcine Embryos.

BACKGROUND: High lipid accumulation limits the cryotolerance of porcine embryos. This study aimed to identify lipid-lowering combinations during in vivo porcine morulae developing into blastocysts and evaluates their effects on post-warming embryonic development and embryo transfer (ET) pregnancy. METHODS: A stable method of in vivo embryo production was established in Meishan pigs to obtain morulae. Morulae were treated with bezafibrate and cholic acid alone, in combination, or with either compound replaced by L-carnitine or niacin, respectively. Treated embryos were vitrified-warmed, follow

Cholic acid inhibits ovarian steroid hormone synthesis and follicular development through farnesoid X receptor signaling in mice.

This study investigated the effects of cholic acid (CA) on steroid hormone synthesis and follicular development in mouse ovaries and the regulatory mechanism of CA on the expression of steroidogenesis-related genes in granulosa cells. The mice were divided into control and CA groups, and serum and ovarian samples were collected after 1, 2, and 4 months of treatment, respectively. The results showed that CA treatment for 1, 2, and 4 months reduced ovarian weights, disrupted the estrous cycle, decreased the numbers of antral follicles and corpora lutea, and lowered the serum levels of progestero

Synthesis of non-steroidal cholic acid analogues as potent and selective TGR5 allosteric agonists with antidiabetic effects.

In this study, we designed and synthesized 12 novel non-steroidal cholic acid (CA) analogues as potential Takeda G protein coupled receptor 5 (TGR5) allosteric agonists based on scaffold hopping strategy. Biological results showed that all final compounds were able to activate the receptor both in human TGR5 (hTGR5) and mouse TGR5 (mTGR5) by Glosensor cAMP accumulation assay. Remarkably, lactone A1 and lactam A10 displayed 48- and 24-fold higher potencies than that of CA in hTGR5 cAMP accumulation, respectively. Furthermore, target compounds exhibited higher selectivity for cAMP formation comp

Cholic acid activation of GPBAR1 does not induce or exacerbate acute pancreatitis but promotes exocrine pancreatic secretion.

Obstruction of bile ducts due to gallstones can lead to biliary acute pancreatitis (BAP). According to Perides et al., G protein-coupled bile acid receptor-1 (GPBAR1) mediates BAP. However, Zi's findings suggest that GPR39, rather than GPBAR1, mediates TLCAS-induced increases in cytosolic calcium and acinar cell necrosis, casting doubt on the role of GPBAR1 in BAP. Numerous G protein-coupled receptors on pancreatic acinar cells utilize Ca2+ and cyclic adenosine monophosphate (cAMP) as second messengers to manage pancreatic exocrine secretion, with significant cross-talk between these signals.

mPEG-Cholic acid/TPGS mixed micelles for combined delivery of paclitaxel and bufalin to treat hepatocellular carcinoma.

In this study, amphiphilic block copolymer mPEG-cholic acid was synthesized in conjunction with TPGS as stabilizer to prepare multifunctional micelles. The formed polymeric micelles (PCTm) were used for the delivery of paclitaxel (PTX) and bufalin (BF). PEG group could enhance solubility and extend circulation time, while cholic acid groups achieved the liver targeted function. Combinations of these approaches could realize a synergistic therapeutic effect in the treatment of advanced hepatocellular carcinoma. CLSM in vitro results demonstrated that drug capsulation into PCTm could enhance cel

Dietary fat, cholesterol, and cholic acid affect the histopathologic severity of nonalcoholic steatohepatitis in Sprague-Dawley rats.

Understanding of the pathogenesis of nonalcoholic steatohepatitis (NASH)-associated fibrosis has been hampered by the lack of a comprehensive and physiological small animal model of NASH with fibrosis. Feeding a high-fat and high-cholesterol (HFC) diet supplemented with cholic acid to rats is known to replicate human NASH pathology, and it induces fibrosis earlier than with an HFC diet alone. In the present study, physiological and histopathological observations from 65 Sprague-Dawley (SD) rats fed an HFC diet with or without cholic acid for 9 or 18 weeks in our laboratory between January 2013

Strain-Dependent Effects of Dietary Cholic Acid on Liver Fibrogenesis and Gut Microbiota in TSNO and TSOD Mice.

Background: Metabolic dysfunction-associated steatohepatitis (MASH) is defined by hepatocellular damage accompanied by inflammation and fibrotic changes. Bile acids (BAs) and gut microbiota play pivotal roles in disease progression. However, the contribution of dietary cholic acid (CA), a primary BA, remains unclear. Methods: We investigated the effect of dietary CA supplementation in Tsumura-Suzuki obese diabetic (TSOD) and Tsumura-Suzuki non-obese (TSNO) mouse strains with distinct metabolic phenotypes. The mice were fed normal diet (ND) or 0.5% CA-supplemented ND. Liver injury, fibrosis, an

Structure-guided discovery of cholic acid derivatives as potent and selective TGR5 allosteric agonists for the treatment of diabetes.

In this work, we designed and synthesized 48 new cholic acid (CA) derivatives as potential Takeda G protein coupled receptor 5 (TGR5) positive allosteric modulators (PAMs). The GloSensor cyclic adenosine monophosphate (cAMP) accumulation assay was used to evaluate the potency and mechanism of action of target compounds. Pharmacological results showed that all final compounds were capable of activating the TGR5. Interestingly, compound B1, selective methylation of 7-hydroxyl group in CA, displayed 80-fold higher potency for TGR5 than its lead compound CA. Furthermore, B1 positively modulated th

Serum cholic acid and cecal Faecalibaculum increase in a male-specific manner in a murine hepatocellular carcinoma model.

Hepatocellular carcinoma (HCC) has a higher incidence in males and is a leading cause of cancer-related deaths, which lacks effective therapies and surveillance markers. Using a murine model of bile acid excess (farnesoid X receptor and small heterodimer partner double knockout, DKO), which phenocopies many aspects of HCC, including sex differences, we investigated the links between sex, bile acid metabolism, and microbial composition. Unexpectedly, the increase in the ratios of carcinogenic deoxycholic acid (DCA) was similar between DKO males with HCC and cancer-resistant DKO females. However

Disruption of the sterol 27-hydroxylase gene in mice results in hepatomegaly and hypertriglyceridemia. Reversal by cholic acid feeding.

Sterol 27-hydroxylase (CYP27) participates in the conversion of cholesterol to bile acids. We examined lipid metabolism in mice lacking the Cyp27 gene. On normal rodent chow, Cyp27(-/-) mice have 40% larger livers, 45% larger adrenals, 2-fold higher hepatic and plasma triacylglycerol concentrations, a 70% higher rate of hepatic fatty acid synthesis, and a 70% increase in the ratio of oleic to stearic acid in the liver versus Cyp27(+/+) controls. In Cyp27(-/-) mice, cholesterol 7alpha-hydroxylase activity is increased 5-fold, but bile acid synthesis and pool size are 47 and 27%, respectively, o
