Case report and review of Chromobacterium sepsis--a gram-negative sepsis mimicking melioidosis.

Chromobacterium violaceum has recently been recognised as a pathogen which can cause life-threatening disease. It is the only Chromobacterium species which is pathogenic to humans. Due to its unfamiliarity, clinicians often do not appreciate its importance when it is isolated in sterile cultures and may dismiss it as a "containment". It is therefore important for us to be aware of this infection and its clinical spectrum since it is a disease of the tropics. We report a paediatric case of documented Chromobacterium sepsis in Singapore who presented like septicaemic melioidosis associated with

Sepsis and Hepatapostema Secondary to Chromobacterium Violaceum Infection on Lower Limb Skin: A Case Report.

BACKGROUND: Chromobacterium violaceum (C. violaceum) is a Gram-negative bacterium capable of causing severe infections in both humans and specific animals. Despite its infrequency, C. violaceum infections exhibit a notably high mortality rate. The timely and precise detection of this pathogen stands as a critical factor in achieving effective diagnosis and treatment. Traditional diagnostic approaches possess limitations, particularly in terms of their time-consuming nature and the range of pathogens they can identify. Metagenomic next-generation sequencing (mNGS) testing has emerged as a highl

Sepsis caused by Chromobacterium violaceum - probably the first case in Europe, or Macbeth read anew.

Rare diseases, almost by definition, present us with diagnostic as well as therapeutic difficulties as. They also include infectious diseases outside endemic areas. Without expecting them, we are not preparing to fight them. Like Macbeth, we feel safe, convinced that tropical diseases do not reach us, like Birnam forest towards his castle. Nevertheless, the forest moved according to the prophecy of the three witches, and in a similar way tropical flora is moving towards us according to the predictions of environmentalists. This is illustrated by the history of the presented patient, who was ad

A Chemical Counterpunch: Chromobacterium violaceum ATCC 31532 Produces Violacein in Response to Translation-Inhibiting Antibiotics.

Antibiotics produced by bacteria play important roles in microbial interactions and competition Antibiosis can induce resistance mechanisms in target organisms, and at sublethal doses, antibiotics have been shown to globally alter gene expression patterns. Here, we show that hygromycin A from Streptomyces sp. strain 2AW. induces Chromobacterium violaceum ATCC 31532 to produce the purple antibiotic violacein. Sublethal doses of other antibiotics that similarly target the polypeptide elongation step of translation likewise induced violacein production, unlike antibiotics with different targets.

Incidence of carbapenem non-susceptible Chromobacterium violaceum harboring blaOXA-232 from clinical-environmental interface.

PURPOSE: Chromobacterium violaceum is an opportunistic environmental pathogen, which act as a potential source of antimicrobial resistance genes. Acquisition of such genes by non-susceptible organisms, aided by mobile genetic elements possess a serious clinical threat. With the rise and expansion of carbapenem resistance worldwide, the present study investigates two carbapenem non-susceptible Chromobacterium violaceum isolates through whole genome sequencing. METHODS: The study isolates were assessed for ESBLs and carbapenemases production using combined disc diffusion and Carba NP tests resp

Genomic analysis of Chromobacterium haemolyticum: insights into the species resistome, virulence determinants and genome plasticity.

The increasing number of Chromobacterium haemolyticum human infection reports, especially in tropical regions and connected with environmental sources, resulted in an urge to better describe this species. This study aimed to characterize the C. haemolyticum resistome, virulence determinants and genetic platforms related with genome plasticity. A comparative genomic analysis was conducted between clinical C. haemolyticum genomes publicly available and the genome of an environmental isolate obtained in this study. The pangenome of C. haemolyticum was calculated and a total of 3378 core genes wer

A Nonlive Preparation of Chromobacterium sp. Panama (Csp_P) Is a Highly Effective Larval Mosquito Biopesticide.

Given the continued high prevalence of mosquito-transmitted diseases, there is a clear need to develop novel disease and vector control strategies. Biopesticides of microbial origin represent a promising source of new approaches to target disease-transmitting mosquito populations. Here, we describe the development and characterization of a novel mosquito biopesticide, derived from an air-dried, nonlive preparation of the bacterium Chromobacterium sp. Panama (family: Neisseriaceae). This preparation rapidly and effectively kills the larvae of prominent mosquito vectors, including the dengue and

EmrR-Dependent Upregulation of the Efflux Pump EmrCAB Contributes to Antibiotic Resistance in Chromobacterium violaceum.

Chromobacterium violaceum is an environmental Gram-negative bacterium that causes infections in humans. Treatment of C. violaceum infections is difficult and little is known about the mechanisms of antibiotic resistance in this bacterium. In this work, we identified mutations in the MarR family transcription factor EmrR and in the protein GyrA as key determinants of quinolone resistance in C. violaceum, and we defined EmrR as a repressor of the MFS-type efflux pump EmrCAB. Null deletion of emrR caused increased resistance to nalidixic acid, but not to other quinolones or antibiotics of differe

Novel RAG1 mutation and the occurrence of mycobacterial and Chromobacterium violaceum infections in a case of leaky SCID.

Severe combined immunodeficiency (SCID) is a potentially fatal primary immunodeficiency (PID) that is caused by mutations in genes such as IL2RG, JAK3, IL7RA, RAG1, RAG2, and ADA. The products of these genes are involved in the development of several immune cells such as T, B and natural killer (NK) cells. Most of the SCID forms are autosomal recessive with the exception of IL2RG defects that cause an X-linked SCID. Among the different SCID types, there is a rare SCID form called leaky SCID, which is less severe when compared to the other classical SCID phenotypes. Leaky SCID can be caused by

The respiratory system of Chromobacterium violaceum grown under conditions of high and low cyanide evolution.

The particulate fraction of disrupted Chromobacterium violaceum grown under cyanide-evolving conditions was unable to oxidize ascorbate plus N,N,N',N'-tetra-methyl-p-phenylenediamine (TMPD), but oxidized NADH and succinate by a linear respiratory pathway which was very resistant to inhibition by cyanide. When the bacteria were grown under conditions where little cyanide evolution occurred, particulate fractions developed the ability to oxidize ascorbate-TMPD by a pathway highly sensitive to cyanide inhibition; respiratory activity with NADH and succinate proceeded via both the cyanide-sensitiv

Genomic and phenotypic characterization of indigenous violacein-producing Chromobacterium amazonense SAM215 from lake sediment in Bangladesh.

The genus Chromobacterium, found in diverse ecosystems, is well known for its production of violacein, quorum-sensing ability, and considerable biotechnological importance. While C. violaceum has been extensively studied, other species, including C. amazonense, have received relatively little attention. This study reports phenotypic characterization and genomic analysis of the indigenous Chromobacterium amazonense strain SAM215, isolated from Bijoypur Lake, Netrokona, Bangladesh. The violacein-producing purple colony was selected and characterized, exhibiting high salinity tolerance (up to 12.

Chromobacterium spp. mediate their anti-Plasmodium activity through secretion of the histone deacetylase inhibitor romidepsin.

The Chromobacterium sp. Panama bacterium has in vivo and in vitro anti-Plasmodium properties. To assess the nature of the Chromobacterium-produced anti-Plasmodium factors, chemical partition was conducted by bioassay-guided fractionation where different fractions were assayed for activity against asexual stages of P. falciparum. The isolated compounds were further partitioned by reversed-phase FPLC followed by size-exclusion chromatography; high resolution UPLC and ESI/MS data were then collected and revealed that the most active fraction contained a cyclic depsipeptide, which was identified a

Fulminant Chromobacterium violaceum Infection With Suspected Septic Arthritis and a Concurrent Dengue Virus Infection: A Case Report.

Chromobacterium violaceum is a rare but highly virulent Gram-negative organism found in tropical and subtropical environments, associated with rapidly progressive sepsis and high mortality. Musculoskeletal involvement, particularly septic arthritis, is exceedingly uncommon. We report a case of a previously healthy 23-year-old male who presented with progressive left ankle pain, swelling, and fever. Initial evaluation revealed thrombocytopenia, markedly elevated inflammatory markers, and positive serology for Dengue virus infection, suggestive of a possible concurrent infection. Blood cultures

Severe Systemic Chromobacterium violaceum Infection: A Case Study of a German Long-Term Resident in French Guyana.

Chromobacterium violaceum is a Gram-negative, facultative anaerobe proteobacterium. Its natural habitat is water and soil in tropical and subtropical regions. Human infections are characterized by rapid dissemination that can lead to high fatality rates. Here, we describe the first case of a C. violaceum infection reported from Germany. A German national with permanent residence in French Guyana contracted a C. violaceum infection presumably while bathing in a barrier lake in Brazil. The patient presented with a high fever and a crusty, erythematous skin lesion at an emergency department in Ha

Chromobacterium violaceum Septicaemia and Urinary Tract Infection: Case Reports from a Tertiary Care Hospital in South India.

Chromobacterium violaceum is a gram negative oxidase positive bacillus that causes human infections infrequently. It is a normal inhabitant of soil and stagnant water of the tropical and subtropical areas. In humans, it can cause infections ranging from life threatening sepsis with metastatic abscesses to skin infections and urinary tract infections. The organism is notoriously resistant to most cephalosporins and Ampicillin. Fluoroquinolones and aminoglycosides show good in vitro susceptibility. High mortality rates associated with these infections necessitate prompt diagnosis and appropriate

Attenuating effect of plumbagin on Chromobacterium violaceum quorum sensing and biofilm formation: an in-vitro and in-silico approach.

The increasing problem of antibiotic resistance over recent decades calls for alternative methods to reduce bacterial pathogenicity. Targeting quorum sensing (QS) is gaining attention as a promising alternative treatment. This study investigates the potential of plumbagin, a natural naphthoquinone derived from Plumbago species, to inhibit quorum sensing and biofilm formation in Chromobacterium violaceum using both in vitro and in silico methods. In vitro assays revealed that sub-minimum inhibitory concentrations of plumbagin significantly suppressed QS-regulated traits compared to controls. Th

Bioefficacy of Chromobacterium piscinae against the corn leafhopper Dalbulus maidis and genomic insights into virulence factors.

Chromobacterium subtsugae is the only species in this genus exploited for biological pest control. In this study, we demonstrated the insecticidal properties of Chromobacterium piscinae ESALQ6200 against Dalbulus maidis, even at low concentrations, using treatments with only bacterial cells and cell-free filtered supernatant containing secreted metabolites. Notably, we did not observe increased mortality with higher concentrations of C. piscinae ESALQ6200 cells (3.5 × 104 to 6.7 × 109 cells/mL) or cell-free filtered supernatant (1%, 25%, 100%). Mortality rates observed in the cell treatments w
