Secretory protein-I (SP-I, also referred to as chromogranin A) is a major acidic, sulfated glycoprotein of the parathyroid gland secretory granule. It is cosecreted with parathormone under calcium regulation. SP-I is similar if not identical to chromogranin A of the adrenal medulla chromaffin cell and has been found in most endocrine cells. Its function is unknown, but it may play a role in the packaging of secreted substances. We have examined the calcium-binding properties of purified bovine SP-I and find that it binds 14 or more Ca2+ at low affinity. Measurements of sedimentation equilibriu

Antisera against bovine chromogranin A, B, and C were used to identify these proteins in human tissues by one- and two-dimensional immunoblotting and by immunohistochemistry. Human chromaffin granules contain all three chromogranins. Their molecular weight differs slightly from the bovine proteins. Chromogranin B and to a lesser degree chromogranin A are apparently processed by endogenous proteases yielding several smaller proteins which cross-react immunologically. In human granules the two families of chromogranins A and B represent the major protein groups, whereas in bovine granules the ch

Pheochromocytoma is a rare adrenal medullary neoplasm in cats, with limited published cases and diagnostic challenges due to variable clinical presentation. An 8-year-old neutered male domestic shorthair cat presented with severe watery diarrhea, lethargy, and anorexia. Diagnostic evaluation revealed chronic kidney disease, pyelonephritis, and a large cystic right adrenal mass identified on ultrasonography and computed tomography. Endocrine testing did not support hyperadrenocorticism or hyperaldosteronism, and functional assessment of catecholamine excess was not performed. Surgical adrenalec

The ciliate protozoan Paramecium tetraurelia produces secretory granules (trichocysts) which release needle-like structures composed of small, acidic proteins. Using antibodies against isolated chromogranin A (CGA) and against trichocyst proteins, we found cross-reactive proteins in chromaffin granules and trichocysts. Four independently derived sera against isolated CGA stained bands of the Mr 15,000-25,000 family of trichocyst proteins on immunoblots. A positive response was also obtained with antiserum against chemically synthesized peptides (PL26 and GE25) corresponding to defined regions

Chromogranin A (CgA) is a prohormone and a granulogenic factor that regulates secretory pathways in neuroendocrine tissues. In β-cells of the endocrine pancreas, CgA is a major cargo in insulin secretory vesicles. The impact of CgA deficiency on the formation and exocytosis of insulin vesicles is yet to be investigated. In addition, no literature exists on the impact of CgA on mitochondrial function in β-cells. Using three different antibodies, we demonstrate that CgA is processed to vasostatin- and catestatin-containing fragments in pancreatic islet cells. CgA deficiency in Chga-KO islets lea

The aim of the study was to investigate the presence and distribution of PE-11, a peptide derived from chromogranin B, in the rat eye. For this purpose, newborn rats were injected with a single dosage of 50mg/kg capsaicin subcutaneously under the neck fold and after three months, particular eye tissues were dissected and the concentration of PE-11-like immunoreactivity was determined by radioimmunoassay. Furthermore, PE-11-like immunoreactivities were characterized in an extract of the rat eye by reversed phase HPLC. Then, the distribution pattern of PE-11 was investigated in the rat eye and r

Chromogranin A like immunoreactivity (CGA-IR) was measured by a specific radioimmunoassay using N-terminal specific antiserum R-0763 in respiratory tissues of guinea pigs. All guinea pig were actively immunized by recurrent Ovalbumin (OA)-inhalation and divided into two groups; one was challenged by OA, and the other inhaled saline as a control. These groups were studied for respiratory resistance and sacrificed for measurement of CGA-IR concentrations in the trachea, major bronchus and lower lung. In the control group, CGA-IR level was 0.4-2 pmol/g wet weight of tissue, and its distribution o

Chromogranin A (CGA), the most abundant protein in bovine adrenal chromaffin granules, is a high-capacity, low-affinity Ca(2+)-binding protein found in most neuroendocrine cells, and binds calmodulin (CaM) in a Ca(2+)-dependent manner. The binding of chromogranin A to calmodulin was determined by measuring the intrinsic tryptophan fluorescence of chromogranin A in the presence and absence of Ca2+. Binding was specifically Ca(2+)-dependent; neither Mg2+ nor Mn2+ could substitute for Ca2+. Chelation of Ca2+ by EGTA completely eliminated the chromogranin A-calmodulin interaction. CaM binding was

Pancreastatin is a carboxyl-terminal amidated peptide derived from chromogranin A (CgA) corresponding to porcine CgA residues 240-288 (CgA240-288). CgA, in turn, is an acidic glycoprotein of about 450 amino acids that is costored and cosecreted with PTH in the parathyroid gland and with native hormones of many endocrine tissues. Among its effects on various endocrine glands, pancreastatin strongly inhibits the secretion of PTH and CgA by the parathyroid. The present report evaluates the action of pancreastatin on the regulation of cellular levels of PTH messenger RNA (mRNA) and CgA mRNA. After

Chromogranin A (CgA), mostly known as a nonspecific neuroendocrine tumor marker, was the first glycoprotein from the granin family characterized as a prohormone for various bioactive peptides including vasostatin I/II (VS-I, VS-II), catestatin (CST), chromofungin (CHR), pancreastatin (PST), WE-14, and others. CgA and its derivatives present various functions, often antagonistic, in maintaining body homeostasis and influencing the immune system. This review aims to summarize the not fully understood role of CgA and its derivatives in inflammation, autoimmunity, and infections. CgA seems to be i

Chromogranin A (CGA) is a ubiquitous 48-kDa secretory protein present in adrenal medulla, anterior pituitary, central and peripheral nervous system, endocrine gut, thyroid, parathyroid, and endocrine pancreas. Recently, we have demonstrated that the protein could be a precursor of bioactive peptides capable of modulating catecholamine secretion from cultured adrenal medullary chromaffin cells. Here we cleaved CGA purified from bovine chromaffin granules with endoproteinase Lys-C, and we isolated and partially sequenced the peptide inhibiting catecholamine secretion from cultured chromaffin cel

Chromogranin A, co-stored and co-released with catecholamines from adrenal medullary and sympathetic neuronal vesicles, is elevated in the plasma of patients with pheochromocytoma. The usefulness of the hormone in the differential diagnosis of hypertension is examined. An elevated level of chromogranin A had comparable diagnostic sensitivity (83%, 24/29) to, but greater diagnostic specificity (96%, 86/90) than the level of plasma catecholamines when subjects with pheochromocytoma (n = 29) were evaluated in comparison to several reference groups, including normotensive controls (n = 49), subjec

BACKGROUND: Chromogranin B (CGB) regulates B-type natriuretic peptide (BNP) production. Circulating CGB levels are elevated in heart failure (HF) animal models and HF patients, but also increase in healthy individuals in response to physical activity. Therefore, CGB seems to integrate information from myocardial stress and systemic neuro-endocrine activation. Substantial gaps remain in our understanding of CGB regulation in HF.
METHODS AND RESULTS: We conducted a retrospective registry study including 372 patients. CGB and N-terminal pro-BNP (NT-proBNP) plasma levels were assessed in acute HF

Chromogranin A (CgA), a neuroendocrine pro-hormone, undergoes proteolytic cleavage to yield bioactive peptides, notably catestatin (CST) and pancreastatin (PST), which exert opposing effects on metabolic and inflammatory processes. Using CgA and CST knockout (KO) mice, this study investigated their roles in pancreatic endocrine function, morphology, neurotransmitter dynamics, and systemic glucose homeostasis. CST deficiency induced insulin resistance, altered islet architecture, and heightened catecholamine levels, whereas CgA-KO mice lacking both CST and PST exhibited improved insulin sensiti

BACKGROUND/AIMS: Chromogranin A is an important tool in the diagnosis of neuroendocrine tumors. Autoimmune gastritis is an autoimmune disorder marked by hypergastrinemia, which stimulates enterochromaffin-like cell proliferation. Chromogranin A is also elevated in autoimmune gastritis patients with a different level of increase in each patient. The goal of this study is to explore constituents that influence serum chromogranin A levels in autoimmune gastritis patients.
MATERIALS AND METHODS: One hundred and eighty-eight autoimmune gastritis patients and 20 patients with type I gastric carcino

BACKGROUND: Salivary stress-related biomarkers in connection with periodontal disease have not been extensively studied. In addition to cortisol as a well-known marker of stress loading, chromogranin A (CgA) and α-amylase (AA) are supposed to link the activity of the neuroendocrine system to local and systemic immune functions and to be related to periodontitis. This study aims to determine CgA and AA in saliva and serum in periodontal health and disease to assess their potential relationship to periodontitis.
METHODS: Patients with aggressive (AgP) (n = 24) and chronic periodontitis (CP) (n

In a previous study, chromogranin A (CgA) cell density in the colon of patients with irritable bowel syndrome (IBS) was found to be reduced. It has been suggested that intestinal CgA cell density may be used as a marker for the diagnosis of IBS. The rectum harbours a larger number of large intestinal endocrine cells and is more accessible for biopsies than the colon. The present study aimed at determining the CgA cell density in the rectum of IBS patients. A total of 47 patients with IBS that fulfilled the Rome Criteria III (39 females and 8 males; average age, 38 years) were included. A total
