Chromosome abnormalities in dogs with disorders of sex development (DSD).

Disorders of sex development (DSD) caused by chromosome abnormalities are rarely diagnosed in dogs. In this report, there is a focus on five DSD cases in which the dogs had abnormal karyotypes. All animals were recognized by owners as females, however, these dogs had a large number of reproductive defects. Among these were abnormal external genitalia such as an enlarged clitoris, abnormal development of the labia, abnormal location of the vulva and urethral orifice, and other abnormalities were observed in four dogs. Gonadal histology assessments were conducted on three dogs and there were dia

A common molecular basis for rearrangement disorders on chromosome 22q11.

The chromosome 22q11 region is susceptible to rearrangements that are associated with congenital anomaly disorders and malignant tumors. Three congenital anomaly disorders, cat-eye syndrome, der() syndrome and velo-cardio-facial syndrome/DiGeorge syndrome (VCFS/DGS) are associated with tetrasomy, trisomy or monosomy, respectively, for part of chromosome 22q11. VCFS/DGS is the most common syndrome associated with 22q11 rearrangements. In order to determine whether there are particular regions on 22q11 that are prone to rearrangements, the deletion end-points in a large number of VCFS/DGS patien

The Philadelphia chromosome negative chronic myeloproliferative disorders: a practical overview.

The chronic myeloid disorders are collectively characterized by a stem cell origin of the clonal process and a variable tendency to undergo indigenous disease transformation and leukemic conversion. Classification of the chronic myeloid processes is based primarily on the presence or absence of the Philadelphia chromosome (bcr-abl translocation) and secondarily on the morphologic picture of the bone marrow in conjunction with the clinical manifestation. Essential thrombocythemia (ET), polycythemia vera (PV), and agnogenic myeloid metaplasia (AMM) constitute the classical group of bcr-abl negat

Clinical significance of the del(20q) chromosome in hematologic disorders.

Patients with hematologic neoplasias often have chromosomal aberrations in the cells of their bone marrow or unstimulated blood. One recurrent abnormality is a deletion of the long arm of chromosome 20, primarily described in polycythemia vera, but later seen in a range of hematologic disorders. We have studied 32 patients with del(20q) as the sole chromosomal aberration, investigating significance of this aberration for the clinical diagnoses, hematologic parameters, and prognoses within this patient group. According to our results, del(20q) is primarily associated with myeloid disorders, but

[Familial, structural aberration of the Y chromosome with fertility disorders].

Cytogenetic studies on a patient with Klinefelter's syndrome revealed an inherited, structural aberration of the Y-chromosome which has not been described before. The aberrant Y-chromosome was characterized by eight different banding methods. The value of individual staining techniques in studies on Y-heterochromatin aberrations is emphasized. Analysis of the cytogenetic studies (banding methods, restriction endonuclease of DNA, and measurement of the length of the Y-chromosome) permits an interpretation to be made on how the aberrant Y-chromosome originated. The functions of the Y-chromosome

Obsessive-compulsive symptoms in two patients with chromosomal disorders involving the X chromosome.

INTRODUCTION: The etio-pathophysiology of obsessive-compulsive disorder (OCD) can be explained using a biopsychosocial model. Little is known about obsessive-compulsive symptoms (OCS) in the context of chromosomal disorders involving the X chromosome. METHODS: Case studies of two patients with chromosomal disorders involving the X chromosome (Patient 1 with a variant of Turner syndrome and Patient 2 with triple X syndrome). RESULTS: Both patients were treated due to severe OCS. In the research MRI analysis, the most pronounced MRI change in both patients was a gray matter volume loss in the

The Impact of an Extra Chromosome on Language: Developmental Language Disorder in Sex Chromosome Trisomies.

PURPOSE: This study investigated the language and cognitive skills of preschool children with sex chromosome trisomies (SCTs) with and without a co-diagnosis of developmental language disorder (DLD), considering possible differences between SCT types (i.e., XXX, XXY, and XYY). METHOD: Fifty-five children with a prenatally diagnosed SCT (19 XXX, 25 XXY, and 11 XYY) aged 48-64 months were included in the study. The presence of a DLD diagnosis or other neurodevelopmental disorder was established by structured interviews, while cognitive and language skills were assessed using a test battery incl

Coagulation Disorders in Philadelphia Chromosome-negative Myeloproliferative Neoplasms-A Guide for Specialists in Coagulation Disorders.

ABSTRACT: Philadelphia chromosome-negative myeloproliferative neoplasms (MPNs) are acquired clonal disorders of hematopoiesis that typically cause thromboembolic and hemorrhagic complications. These include an increased risk of both arterial and venous thromboembolic events as well as a heightened bleeding tendency. Venous thromboembolic events frequently manifest at atypical sites and characteristically present as splanchnic vein thrombosis. A common cause of the elevated bleeding risk is acquired von Willebrand syndrome. Clinical indicators of an underlying MPN include thromboembolic events

Evaluating pregnancy termination rates for fetal chromosome and single gene disorders.

We report pregnancy termination rates following a variety of fetal diagnoses and determine which factors may influence this decision. We conducted a retrospective chart review of pregnancies diagnosed with a genetic abnormality at a single institution from January 2012 to April 2023. The type of diagnosis, termination status, and thirteen demographic factors were collected. The primary outcome assessed was whether or not an individual terminated the pregnancy. Data analysis consisted of multivariable logistic regression. Of the 2120 patients who underwent prenatal diagnostic testing, 332 recei

[Chromosome disorders and immunoreactivity in golden hamsters infected with Opisthorchis felineus].

Opisthorchosis of golden hamster causes the disarrangement of the number and structure of chromosomes of bone marrow cells 15 days after the infection. Analogous changes take place in man's leucocytes under the influence of water-salt extract from adult trematodes. The injury of the chromosomal apparatus advances with the increasing period of the infection and is coordinated with modified immunoreactivity of the organism.

Covert Brain Infarcts in Patients with Philadelphia Chromosome-Negative Myeloproliferative Disorders.

BACKGROUNDS AND PURPOSE: Philadelphia chromosome-negative myeloproliferative disorders (Ph-negative MPD) are a rare group of hematological diseases, including three distinct pathologies: essential thrombocythemia (ET), polycythemia vera (PV), and primary myelofibrosis (PMF). They most often manifest with thrombotic complications, including cerebrovascular events. Covert brain infarcts (CBIs) are defin ed as predominantly small ischemic cerebral lesions that are detected using magnetic resonance imaging (MRI) in the absence of clinical stroke events. The relationship between MPD and CBIs remain

Subtelomeric region of chromosome 2 in patients with autism spectrum disorders.

Autism spectrum disorders are severe psychiatric diseases commonly identified in the population. They are diagnosed during childhood and the etiology has been much debated due to their variations and complexity. Onset is early and characterized as communication and social interaction disorders and as repetitive and stereotyped behavior. Autistic disorders may occur together with various genetic and chromosomal diseases. Several chromosomal regions and genes are implicated in the predisposition for these diseases, in particular those with products expressed in the central nervous system. There

No association between chromosome-18 markers and lithium-responsive affective disorders.

An allelic association study of excellent responders to lithium was conducted with a candidate gene (Golf, a G-protein receptor gene) and five other chromosome-18p markers. Golf is of special interest because it maps to a region of chromosome 18 where two independent groups (Berrettini et al., 1994; Stine et al., 1995) have found linkage to bipolar disorder. It has been proposed that G proteins are involved in the pathogenesis of bipolar disorder, and lithium, an effective prophylactic agent, is known to impair G-protein activation. To reduce heterogeneity--a common obstacle to genetic investi

Inactivated X chromosome fold in human leukemia and related clonal disorders.

A disturbance of the X chromosome inactivation process has been demonstrated in some human tumors. In order to study this phenomenon in leukemia, the site of unusually frequent folding in Xq13-q21 was used as a morphological marker. A total of 858 GTG-banded mitoses from 61 women with different hematological malignancies were analyzed, 35 of them with normal karyotype and 26 with chromosome aberrations. The folding patterns and the fold percentage remained unchanged in our patients. Using this marker, X-inactivation could easily be verified in leukemia and related disorders on routine preparat

Molecular characterization of chromosome 7 long arm deletions in myeloid disorders.

Partial deletion of the long arm of chromosome 7 is a common abnormality in the bone marrow cells of patients with myelodysplastic syndrome (MDS) or acute nonlymphocytic leukemia (ANLL). This study was undertaken to characterize the chromosome breakpoints in molecular terms and to determine if hemizygosity or submicroscopic deletions occur in patients without any cytogenetically detectable abnormality of chromosome 7. We studied restriction fragment length polymorphisms with 10 chromosome 7-specific DNA probes in separated WBC fractions. No molecular abnormalities occurred in lymphocyte-derive

Uniparental disomy is a chromosomic disorder in the first place.

BACKGROUND: Uniparental disomy (UPD) is well-known to be closely intermingled with imprinting disorders. Besides, UPD can lead to a disease by 'activation' of a recessive gene mutation or due to incomplete (cryptic) trisomic rescue. Corresponding to all common theories how UPD forms, it takes place as a consequence of a "chromosomic problem", like an aneuploidy or a chromosomal rearrangement. Nonetheless, UPD is rarely considered as a cytogenetic, but most often as a molecular genetic problem. RESULTS: Here a review on the ~ 4900 published UPD-cases is provided, and even though being biased a

Autosomal ring chromosomes in human genetic disorders.

Ring chromosomes arise following breakage and rejoining in both chromosome arms. They are heterogeneous with variable size and genetic content and can originate from any chromosome. Phenotypes associated with ring chromosomes are highly variable as apart from any deletion caused by ring formation, imbalances from ring instability can also occur. Of interest is ring chromosome 20 which has a significant association with epilepsy with seizure onset in early childhood. Severe growth deficiency without major malformations is a common finding in the ring chromosome carrier. This phenotype associate
