Interrelations between sleep and chronobiology as been studied in isolated experimental situations. A succession of hormonal regulations has been described to explain these mechanisms. Some disruptions of these regulations might be at the beginning of a lot of sleep pathologies (jet lag syndrome, burn out syndrome, insomnia...).

BACKGROUND: Chronobiology is devoted to the study of biological rhythms. It is possible that a given medication may be therapeutic and safe when administered at some time, but subtherapeutic or poorly tolerated at another.
METHODS: We focused on some classes of drugs, widely used by the internists, performing a PubMed search with the single drugs associated with the MeSH terms "Chronotherapy", "Circadian rhythm", and "Chronobiology, phenomena". Among the studies found, we considered only those provided with discrete numerosity or clearly stated methodological characteristics.

The objective of this study was to examine the chronobiology of visual hallucinations in Parkinson's disease (PD) patients to test whether hallucinations are more severe during darker months (winter) than summer months. We extracted longitudinal data on the UPDRS Thought Disorder (TD) score, taken as part of regular patient management on sequential visits through a 1-year cycle in hallucinating PD patients. To reduce the confounding effects of medication adjustments, analysis was restricted to patients with a baseline evaluation that did not involve a medication change. The primary outcome was

BACKGROUND: Nocturia is a common symptom among adults and is often associated with disrupted sleep and diminished quality of life. In many cases, it is driven by nocturnal polyuria, an imbalance in nighttime urine production. Emerging evidence suggests that circadian rhythm disturbances, which regulate hormone secretion, renal function, and blood pressure, may play a role in this altered diuresis. This systematic review evaluated the contribution of circadian dysregulation to the pathophysiology of two distinct nocturnal voiding patterns: nocturia (defined by voiding frequency) and nocturnal p

BACKGROUND: Circadian rhythm dysregulation may contribute to sleep-wake disorders and cognitive impairment. In the elderly, circadian abnormalities have been observed in Mild Cognitive Impairment (MCI), suggesting a possible association between neurocognitive deficits and circadian dysfunctions.
OBJECTIVE: To analyze the association between circadian rhythm disturbances and cognitive decline in the population aged ≥ 60 years.
METHODS: This systematic review included studies evaluating the association between circadian rhythm disturbances and cognitive decline in elderly individuals aged 60 y

Mitochondria are fundamental for life and require balanced ion exchange to maintain proper functioning. The mitochondrial cation exchanger LETM1 sparks interest because of its pathophysiological role in seizures in the Wolf Hirschhorn Syndrome (WHS). Despite observation of sleep disorganization in epileptic WHS patients, and growing studies linking mitochondria and epilepsy to circadian rhythms, LETM1 has not been studied from the chronobiological perspective. Here we established a viable letm1 knock-out, using the diurnal vertebrate Danio rerio to study the metabolic and chronobiological cons

Traveling through several time zones results in a constellation of symptoms known as jet lag. These include reduced alertness, daytime fatigue, loss of appetite, reduced cognitive skills, and disruption of the sleep/wake cycle. In susceptible air travel passengers, jet lag may exacerbate affective illness and result in psychiatric morbidity. Dysregulation of circadian rhythms and melatonin secretion represent the common underlying factor in jet lag and other circadian disorders. Recent studies have established the effectiveness of strategically timed administration of melatonin and appropriate

Women experience a high prevalence of mood and anxiety disorders, and comorbidity of mood and anxiety disorders is highly prevalent. Both mood and anxiety disorders disturb sleep, attention and, thereby, cognitive function. They result in a variety of somatic complaints. The mood disorder continuum includes minor depression, dysthymia, major depression and bipolar disorder. Chronobiological disorders, such as seasonal affective disorder as well as premenstrual dysphoric disorder, occur in some women, with comorbid seasonal affective disorder and premenstrual dysphoric disorder in just under ha

Nighttime eating is categorized as either night eating syndrome (NES) or the sleep-related eating disorder (SRED). Both diseases are often connected with an increase of the body mass, obesity, and with psychiatric disturbances. NES is characterized by evening hyperphagia, abnormally increased food intake after the evening meal, nocturnal awakings with ingestions, morning anorexia, and insomnia. Patients suffering from NES are aware of their nocturnal ingestions. It is suggested that NES is an abnormality in the circadian rhythm of meal timing that occurs in people with normal circadian rhythm

Among other factors, bipolar disorder is characterized by disturbances in sleep and biological rhythms that typically cycle over a 24-h, or circadian period. Indeed, almost all of the functions that constitute symptoms of depression and mania (changes in mood, energy, sleep, interest, appetite, capacity for concentration, etc.) show relatively regular variation over the circadian period. Thus, it would appear logical to search for clues to the pathophysiology of bipolar disorder in the function of the circadian timing system (Wirz-Justice in Int Clin Psychopharmacol 21:S11-S15, 2006). In this

Circadian rhythm sleep disorders (CRSD) result from a disturbed endogenous clock (intrinsic CRSD) or from a misalignment between the biological clock and an imposed environment (extrinsic CRSD). Among intrinsic CRSD, one distinguishes the delayed sleep-wake phase disorder, the advanced sleep-wake phase disorder, the irregular sleep-wake rhythm disorder and the non-24-hour sleep-wake rhythm disorder. Shift work disorder, jet lag disorder and circadian sleep-wake disorder not otherwise specified are extrinsic CRSD. Prevalences of the different CRSD remain largely unknown. Some CRSD are particula

Non-24-h sleep-wake disorder in blind patients without light perception is an orphan circadian rhythm sleep-wake disorder and is extremely rare in sighted people. Non-24-h sleep-wake disorder is characterized by insomnia and daytime sleepiness alternating with asymptomatic episodes. The frequency of symptomatic periods depends on the daily desynchronization of endogenous circadian pattern of each patient. Diagnosis requires anamnesis, a sleep diary, and actigraphy, if possible; in addition, repeated 24-h measures of circadian markers such as melatonin secretion are also required. Treatment con

OBJECTIVES: Two recent scientific breakthroughs may alter the treatment of mental illness, as discussed in this narrative review. The first was the invention of white light-emitting diodes (LEDs), which enabled an ongoing, rapid transition to energy-efficient LEDs for lighting, and the use of LEDs to backlight digital devices. The second was the discovery of melanopsin-expressing photosensitive retinal ganglion cells, which detect environmental irradiance and mediate non-image forming (NIF) functions including circadian entrainment, melatonin secretion, alertness, sleep regulation and the pupi

Despite a complex relationship between mood, sleep and rhythm, the impact of circadian disruptions on bipolar disorder (BD) has not been clarified. The purpose of this systematic review was to define current evidence regarding chronotype and circadian rhythm patterns in BD patients. 42 studies were included, involving 3432 BD patients. Disruption of the biological rhythm was identified, even in drug-naïve BD patients and independently of mood status. Daily profiles of melatonin levels and cortisol indicated a delayed phase. Depression was more frequently associated with circadian alterations t

BACKGROUND: Attention-Deficit/Hyperactivity Disorder (ADHD) is increasingly recognized as a neurodevelopmental condition involving both dopaminergic dysfunction and circadian misalignment. While these mechanisms have traditionally been studied independently, their combined impact on symptom expression and comorbidity-particularly with mood disorders, substance use disorders, and obesity-remains insufficiently understood. This review aims to integrate current evidence on the interaction between circadian and dopaminergic dysfunction in ADHD and to evaluate the therapeutic potential of chronobio

Seasonal affective disorder or major depressive episode (MDE) with winter seasonal pattern is a severe form of recurrent depressive disorder persisting for at least 40 % of the year. This seasonal vulnerability may involve specific chronobiological and sleep regulation disturbances, however, actigraphy and polysomnography studies reported conflicting and inconclusive findings. In this context, we hypothesized that individuals with MDE who exhibit higher vulnerability to seasonal changes display distinct chronobiological and sleep regulation disturbances. We investigated how sleep and circadian

Epidemiological trends towards a 'graying' population make the issue of insomnia in the elderly an increasingly important research and clinical topic. It is often challenging to determine how much of a psychiatric dimension there is to a clinical condition that is best viewed as both as a symptom and a true psychosomatic entity in its own right. To categorize insomnia as either psychiatric or medically based risks oversimplification of the complexities of sleep disruption in the elderly. Normal senescence-related changes in sleep architecture and circadian rhythms must be considered, as well a

The human pineal gland is a neuroendocrine transducer that forms an integral part of the brain. Through the nocturnally elevated synthesis and release of the neurohormone melatonin, the pineal gland encodes and disseminates information on circadian time, thus coupling the outside world to the biochemical and physiological internal demands of the body. Approaches to better understand molecular details behind the rhythmic signalling in the human pineal gland are limited but implicitly warranted, as human chronobiological dysfunctions are often associated with alterations in melatonin synthesis.

A review of EEG sleep physiology and disorders of sleep is presented from a developmental perspective. After sleep physiology is reviewed in relation to the aging process, with various sleep changes considered as possible signs of normal or pathological aging, sleep disorder characteristics of different parts of the life cycle and/or the ontogenetic aspects of sleep disorders per se are also described. This review highlights the need for four diverse but related areas of research: (1) longitudinal studies to describe the natural history of primary sleep disorders; (2) controlled treatment and

BACKGROUND: While the research and treatment focus of biological aspects of depression has traditionally centered on neutrotransmitters disturbances, there has been relatively little attention paid to the chronobiological aspects of depression that offer rapid acting chronotherapeutis and from recently also an innovative circadian rhythms resynchronizing antidepressant.
OBJECTIVE: This article discusses chronobiological aspects of psychiatric treatment, particularly related to depression. It is concerned with chronotherapeutics and pharmacological interventions to resychronize circadian rhyth
