Substrate-dependent differential splicing of introns in the regions encoding the cellulose binding domains of two exocellobiohydrolase I-like genes in Phanerochaete chrysosporium.

Recently, we have shown differential splicing of an intron in the cbhI.2 gene of Phanerochaete chrysosporium ME446; this intron lies within the region of the gene encoding the cellulose binding domain (P.F.G. Sims, M. S. Soares-Felipe, Q. Wang, M.E. Gent, C. Tempelaars, and P. Broda, Mol. Microbiol. 12:209-216, 1994). Here, we show that such differential splicing occurs in the cbhI.1 gene of this fungus as well as in the cbhI.2 gene and that this phenomenon is substrate dependent. Avicel elicits the synthesis of both classes of mRNA transcripts from both of these genes. In contrast, carboxymet

A calmodulin inhibitor, W-7 influences the effect of cyclic adenosine 3', 5'-monophosphate signaling on ligninolytic enzyme gene expression in Phanerochaete chrysosporium.

The capacity of white-rot fungi to degrade wood lignin may be highly applicable to the development of novel bioreactor systems, but the mechanisms underlying this function are not yet fully understood. Lignin peroxidase (LiP) and manganese peroxidase (MnP), which are thought to be very important for the ligninolytic property, demonstrated increased activity in Phanerochaete chrysosporium RP-78 (FGSC #9002, ATCC MYA-4764™) cultures following exposure to 5 mM cyclic adenosine 3', 5'-monophosphate (cAMP) and 500 μM 3'-isobutyl-1-methylxanthine (IBMX), a phosphodiesterase inhibitor. Algicidal Molecular Mechanism and Toxicological Degradation of Microcystis aeruginosa by White-Rot Fungi.

Current research on the inhibition of Microcystis aeruginosa growth is primarily focused on algae-lysing bacteria, and few studies have investigated the inhibitory mechanisms by which fungi affect it at the molecular level. A comparative analysis of the effects of Phanerochaete chrysosporium on the expression of the algal cell antioxidant protease synthesis gene prx, the biological macromolecule damage and repair genes recA, grpE, and fabZ, and the photosynthesis system-related genes psaB, psbD1 and rbcL, as well as genes for algal toxin synthesis mcyB, were performed to elucidate the molecula

The effectiveness of arbuscular-mycorrhizal fungi and Aspergillus niger or Phanerochaete chrysosporium treated organic amendments from olive residues upon plant growth in a semi-arid degraded soil.

Arbuscular mycorrhizal (AM) fungi and a residue from dry olive cake (DOC) supplemented with rock phosphate (RP) and treated with either Aspergillus niger (DOC-A) or Phanerochaete chrysosporium (DOC-P), were assayed in a natural, semi-arid soil using Trifolium repens or Dorycnium pentaphyllum plants. The effects of the AM fungi and/or DOC-A were compared with P-fertilisation (P) over eleven successive harvests to evaluate the persistence of the effectiveness of the treatments. The biomass of dually-treated plants after four successive harvests was greater than that obtained for non-treated plan

Geomyces pannorum as a possible causative agent of dermatomycosis and onychomycosis in two patients.

Chrysosporium pannorum (Link) Hughes is a soil keratinophilic fungus present in organic residues, on human skin surface and in the general environment of human beings. Clear evidence for the pathogenicity of this fungus for human beings was lacking. In 1999, a case of fungal infection in a chow-chow dog and its owner was published, where Chrysosporium pannorum (Link) Hughes was determined as the only possible infection trigger. The State Hygiene Institute in Bratislava repeatedly confirmed the cultivation results. Chrysosporium pannorum (Link) Hughes was detected in the material provided by a

[Effect of heavy metals on degradation of BDE-209 by white-rot fungus].

Effects and mechanisms of heavy metals Cu, Cd and Pb on the growth of white-rot fungus P. chrysosporium and its ability of BDE-209 degradation were studied. The results showed that low concentrations of heavy metals (< or = 1 mg x L(-1)) stimulated the growth of P. chrysosporium with the order of Cd > Pb > Cu when 1 mg x L(-1) of each heavy metal was concerned, while high concentrations (> 1 mg x L(-1)) depressed it. P. chrysosporium degraded BDE-209 efficiently with degradation efficiency reaching 69.7% after 7 days. The presence of heavy metals significantly influenced the capability of P. c

The cytotoxicities in prokaryote and eukaryote varied for CdSe and CdSe/ZnS quantum dots and differed from cadmium ions.

The present study focused on the bioaccumulation and cytotoxicities of Cd2+, CdSe quantum dots (QDs) and CdSe/ZnS QDs in Escherichia coli (E. coli, represents prokaryotic system) and Phanerochaete chrysosporium (P. chrysosporium, represents eukaryotic system), respectively. Two types of QDs were characterized by transmission electron microscopy (TEM) and dynamic light scattering. The inductively coupled plasma optical emission spectrometer results showed that the bioaccumulation amounts of CdSe QDs by E. coli and P. chrysosporium were larger than those of CdSe/ZnS QDs due to the smaller partic

Adiaspiromycosis in a European beaver from Sweden.

An infection with the rare mycosis Chrysosporium parvum was diagnosed in a European beaver (Castor fiber) shot in northern Sweden. The animal was in normal body condition and no signs of disease were observed. In the lungs a large number of nodules, up to 5 mm diameter, were observed. A large number of adiaspores were observed in the interstitium of the lungs and in the mediastinal lymph node. A chronic inflammatory reaction dominated by mononuclear leukocytes and giant cells was observed around the spores. This is the first report of adiaspiromycosis (Chrysosporium parvum) in the European bea

Novel strategy to improve the colonizing ability of Irpex lacteus in non-sterile wheat straw for enhanced rumen and enzymatic digestibility.

Pretreatment with white rot fungi is a promising method to enhance the digestibility of lignocelluloses; however, sterilization of feedstocks prior to inoculation is one of the costliest steps. To improve the colonizing ability of white rot fungi under non-sterile condition, Irpex lacteus, Pleurotus ostreatus, and Phanerochaete chrysosporium were inoculated in the wheat straw ensiled for 28 days and incubated for 56 days to determine the changes in microbe counts, organic acid content, chemical composition, and rumen and enzymatic digestibility. Results showed that ensiling produced abundant o

[Gilchrist's disease and fungal taxonomy: a thorny problem].

The study of iconographical data and of six american and african strains of B. dermatitidis allows us to conclude that there are several discriminating characters between the members of the Blastomyces dermatitidis complex. The african strains are distinguished by the lack of sexual forms and old strains show the lost of the M lead to Y conversion. An identical character prevails among the strains of the two continents: tuberculate conidia which are obtained on corn meal agar. The authors are insisting on the identical characters encountered on Blastomyces and Histoplasma. All the mycotic stra

Evaluation of Novel Filtration System Utilizing Biochar, Zero-Valent Iron, and White Rot Fungi to Inhibit Foodborne Pathogenic Bacteria, Viruses, and Protozoan Parasites.

Agricultural water of appropriate microbiological criteria for intended use is crucial for a number of practices, including irrigation, and the possibility of contamination of agricultural water by foodborne pathogens, has raised concerns regarding food safety and the potential for crop contamination, which leads to foodborne illness. To address these concerns, novel water treatment systems are being explored. For example, in this study, a novel combination filtration system that utilizes zero-valent iron, sand, biochar, and white-rot fungi is being assessed. This study was conducted to evalua

Aberrant Histoplasma capsulatum. Confirmation of identity by a chemiluminescence-labeled DNA probe.

A cottony, light tan, filamentous fungus with pear-shaped microconidia and lacking tuberculated macroconidia was isolated from a bronchial lavage specimen. Subculture on several media at 37 degrees C failed to convert the fungus to a yeast form after several weeks; attempts at in vivo conversion in mice were also unsuccessful. Sera obtained several months apart showed M bands with Histoplasma capsulatum (HC) antigen by immunodiffusion and an increase in complement fixation titers with mycelial and yeast phase antigens of HC. OSIP1 is a self-assembling DUF3129 protein required to protect fungal cells from toxins and stressors.

Secreted proteins are key players in fungal physiology and cell protection against external stressing agents and antifungals. Oak stress-induced protein 1 (OSIP1) is a fungal-specific protein with unknown function. By using Podospora anserina and Phanerochaete chrysosporium as models, we combined both in vivo functional approaches and biophysical characterization of OSIP1 recombinant protein. The P. anserina OSIP1Δ mutant showed an increased sensitivity to the antifungal caspofungin compared to the wild type. This correlated with the production of a weakened extracellular exopolysaccharide/pro

Sonochemical synthesis of ZnS nanolayers on the surface of microbial cells and their application in the removal of heavy metals.

In order to improve the adsorption performance of microorganisms, we synthesized a novel material - phanerochaete chrysosporium cells covered with a layer of ZnS nanoparticles (ZnS-cells). The preparation of the ZnS-cells is based on the Sonochemical method to synthesize the ZnS nanoparticle layer on the surface of the microbial cells. The ZnS-cells were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), energy dispersive X-ray spectroscopy (EDS) and Fourier transform infrared spectroscopy (FTIR). Characterization results showe

Effective bioremediation of clarithromycin and diclofenac in wastewater by microbes and Arundo donax L.

Bioremediation of pharmaceuticals has gained large research efforts, but there is still a need to improve the performance of bioremediation systems by selecting effective organisms. In this study, we characterized the capability to remove clarithromycin (CLA) and diclofenac (DCF) by the bacterium Streptomyces rochei, and the fungi Phanerochaete chrysosporium and Trametes versicolor. The macrolide antibiotic CLA and the non-steroid anti-inflammatory DCF were selected because these are two of the most frequently detected drugs in water bodies. Growth and content of the PhCs and a DCF metabolite

Adiaspiromycosis in a wild European rabbit, and a review of the literature.

Adiaspiromycosis is a mycotic infection caused by thermally dimorphic fungi classified as Emmonsia parva and E. crescens (formerly Chrysosporium spp.) until recently, when new classifications were proposed. We document the pathologic findings in a severe case of adiaspiromycosis, with lymph node involvement, in a wild European rabbit ( Oryctolagus cuniculus). The rabbit exhibited granulomatous pneumonia with tracheobronchial lymph node enlargement. Histopathologically, the lung was expanded by myriad, densely cellular, heterophilic and granulomatous foci, surrounding bi- to trilaminar adiaspor

Description of a case of rhinosporidiosis in Spain.

Rhinosporidiosis is reviewed, and the first autochthonous case in Spain is presented (site: in the nasal cavity of a 19-year-old male from a rural background). Diagnosis was established morphologically after eliminating the possibilities of Cryptococcus neoformans, Coccidioides immitis, and Chrysosporium crescens. Clinico-pathological features are described. Preparations were stained with hematoxylin-eosin, PAS, and methenamine silver, and studied for fluorescence. Certain aspects of the epidemiology and diagnosis are commented upon.

Production of fungal biomass immobilized loofa sponge (FBILS)-discs for the removal of heavy metal ions and chlorinated compounds from aqueous solution.

A white rot basidiomycete, Phanerochaete chrysosporium, was immobilized on loofa sponge (FBILS) discs. It removed ca. 37 and 71 mg Cd (II) g(-1) from 50 and 200 mg l(-1) aqueous solutions and up to 89% of 4-chloroanisole from a 10 mg l(-1) aqueous solution. FBILS are physically strong and chemically recalcitrant, resisting temperature, mechanical agitation, and variations in pH without alteration to shape, structure or texture.

Lung parasites of least weasels in Finland.

Because of their constant exposure to normal rodent definitive hosts, least weasels (Mustela nivalis) were trapped in southern Finland in late fall 1994 and examined for lung parasites. Histological examination showed that 46% of the weasels (n = 46) were infected with adiaspores identified as Chrysosporium sp. Granulomas surrounding the adiaspores consisted of mostly unorganized layers of mononuclear cells. The adiaspores from least weasels were much smaller than those reported from their prey animals. Infection with Pneumocystis carinii also was found in two weasels.
