Avian circoviruses of the genus Circovirus: A potential trigger in Pigeon breeder's lung (PBL)/Bird fancier's lung (BFL).

Pigeon breeder's lung (PBL) or Bird fancier's lung (BFL) is one of the most common extrinsic allergic alveolitis or hypersensitivity pneumonitis. It is caused after prolonged inhalation of avian antigens and provokes a hypersensitivity reaction in the lungs of sensitised people. Although the pathogenic mechanism is unclear, the epidemiology of BFL shows that it occurs worldwide, and has been described in adults keeping birds and also in their children. Laboratory findings associated with the disease classified as a type III immunologic reaction that produces blood precipitin antibodies against

Postweaning multisystemic wasting syndrome: a review of aetiology, diagnosis and pathology.

This review paper concentrates on the aetiology, diagnosis, and pathological aspects of postweaning multisystemic wasting syndrome (PMWS). PMWS was first recognized in Canada in 1996 as a new emerging disease which caused wasting in postweaned pigs. Since then, PMWS has been recognized in pigs in many countries. The syndrome is caused by a DNA virus referred to as porcine circovirus 2 (PCV2), which is classified in the family Circoviridae. PMWS primarily occurs in pigs between 25 and 120 days of age with the highest number of cases occurring between 60 and 80 days of age. The diagnosis of PMWS

Detection of circovirus and herpesvirus co-infection in suddenly dead.

This article investigates the causes of the sudden death of six common ravens (Corvus corax). All six birds were found on the same day in central Poland. The ravens were necropsied, and samples were collected for bacteriological, histopathological, and toxicological examinations. The presence of genetic material from common viral and some bacterial pathogens was examined using molecular methods. The ravens were found to be in good nutritional condition. Two birds had enlarged spleens with petechiae on their surface. Histopathological examination revealed multifocal hemorrhages and inflammatory

Co-infection of canine parvovirus and circovirus in fatal gastroenteritis outbreak among service dogs in Kazakhstan, 2023.

INTRODUCTION: Between November 2023 and January 2024, a severe gastroenteritis outbreak with high mortality occurred among working dogs based in the Almaty region of Kazakhstan. The epidemic was characterized by an acute onset, rapid progression, and resulted in the death of over 100 juveniles (under 12-month-old) and several vaccinated adult dogs. In this study, we investigated the co-occurrence of canine circovirus and canine parvovirus DNAs in clinical samples from affected dogs, performed genetic characterization of the identified viruses, and evaluated their role in the outbreak. METHODS

Coinfection with porcine circovirus type 2 and Streptococcus suis serotype 2 enhances pathogenicity by dysregulation of the immune responses in piglets.

Porcine circovirus type 2 (PCV-2) and Streptococcus suis (S. suis) are common pathogens in pigs. Both pathogens are associated with the porcine respiratory disease complex. Clinically, coinfection of PCV-2 and S. suis are often detected in pigs with respiratory symptoms, while interactions between the two pathogens during coinfection and the coinfection pathogenesis are poorly understood. In this study, a piglet model coinfected with PCV-2 and Streptococcus suis serotype 2 (SS2) was established; coinfection of piglets increased the contents of SS2 in blood, and piglets showed more severe pneum

Influence of porcine circovirus type 2 vaccination on the level of antimicrobial consumption on 65 Austrian pig farms.

The administration of antibiotics to farm animals is an important contemporary topic. Veterinarians, pig producers, politicians, retailers and consumers all have a vested interest in reducing antimicrobial use on farm, while ensuring adequate health and welfare of food-producing animals. Vaccination programmes may be used to reduce the overall level of clinical disease in a population, subsequently leading to a decline in antimicrobial use. In 2008, a vaccination programme against porcine circovirus type 2 (PCV-2) was initiated in Austria. In the retrospective observational study presented her

Pollen-inspired multifunctional nanocarrier for porcine circovirus 2 vaccine delivery.

Traditional vaccines targeting porcine circovirus type 2 (PCV2) have several limitations, such as low antigen delivery efficiency and inadequate immunogenicity. To address these challenges, this study developed a bionic nanocarrier (Fe₃O₄@MnSiO₃) that synergistically integrates antigen enrichment and immune stimulation functions. Inspired by the hierarchical structure of sunflower pollen, the design constructs composite particles featuring a superparamagnetic Fe₃O₄ core and a mesoporous MnSiO₃ shell, allowing efficient loading and delivery of the PCV2 Cap protein. Owing to its sophisticated bi

Porcine Circovirus Type 3: Diagnostics, Genotyping, and Challenges in Vaccine Development.

Porcine circovirus (PCV) comprises four distinct species, namely, PCV1-4, with considerable differences between them, resulting in limited crossprotection. PCV2 inflicts substantial economic losses on the swine industry. PCV3 was found to have been circulating before its discovery in 2015. PCV3 is suspected of having a comparable impact to PCV2; however, its characterization studies are still underway. The symptoms of PCV3 infection are similar to those of PCV2 infection. However, PCV2 and PCV3 share a maximum amino acid identity of only 37%, which partially explains the inadequacy of the PCV2

Porcine Circovirus Type 2 Hijacks Host IPO5 to Sustain the Intracytoplasmic Stability of Its Capsid Protein.

Nuclear entrance and stability of porcine circovirus type 2 (PCV2), the smallest virus in mammals, are crucial for its infection and replication. However, the mechanisms are not fully understood. Here, we found that the PCV2 virion maintains self-stability via the host importin 5 (IPO5) during infection. Coimmunoprecipitation combined with mass spectrometry and glutathione S-transferase pulldown assays showed that the capsid protein (Cap) of PCV2 binds directly to IPO5. Fine identification demonstrated that the N-terminal residue arginine24 of Cap is the most critical to efficient binding to t

Investigation and sequence analysis of psittacine beak and feather disease virus and avian polyomavirus from companion birds in Windhoek, Namibia.

The commercial farming and trading of parrots and ornamental birds as companion animals are important economic activities in many countries. Some of the bird species farmed/traded are captured from the wild or are closely related to wild birds and therefore represent a risk of pathogen exchange/introduction. Beak and feather disease virus (BFDV) and avian poliomavirus (APV) are among the viruses with the biggest impact on companion bird populations and have been detected in different hosts worldwide. Despite their relevance for both domesticated and wild birds, our knowledge of BFDV and APV ep

Characterization of porcine circovirus-like virus P1 replication and lesions in BALB/c mice.

Five-week-old male BALB/c mice were inoculated intraperitoneally with a single (sP1) or multiple doses (mP1) of porcine circovirus-like virus P1 or cell culture medium. None of the mice exhibited clinical signs or gross lesions throughout the study. However, the body weights of the mP1 mice were significantly decreased, and the mice inoculated with P1 exhibited viral replication, seroconversion, and microscopic lesions. P1 nucleic acid was detected in the heart, liver, spleen, lung, bladder, testis, brain, thymus, and pancreatic tissues. Special P1 antibody was found in the P1-inoculated mice.

Viral metagenomics in Brazilian Pekin ducks identifies two gyrovirus, including a new species, and the potentially pathogenic duck circovirus.

Viral metagenomics coupled to high-throughput sequencing has provided a powerful tool for large-scale detection of known and unknown viruses associated to distinct hosts and environments. Using this approach, known and novel viruses have been characterized from sylvatic and commercial avian hosts, increasing our understanding of the viral diversity in these species. In the present work we applied an exploratory viral metagenomics on organs (spleen, liver and bursa of Fabricious) of Pekin ducks from Southern Brazil. The virome contained sequences related to a known duck pathogen (duck circoviru

Identification of microRNAs in PCV2 subclinically infected pigs by high throughput sequencing.

Porcine circovirus type 2 (PCV2) is the essential etiological infectious agent of PCV2-systemic disease and has been associated with other swine diseases, all of them collectively known as porcine circovirus diseases. MicroRNAs (miRNAs) are a new class of small non-coding RNAs that regulate gene expression post-transcriptionally. miRNAs play an increasing role in many biological processes. The study of miRNA-mediated host-pathogen interactions has emerged in the last decade due to the important role that miRNAs play in antiviral defense. The objective of this study was to identify the miRNA ex

Clinical beak and feather disease virus infection in wild juvenile eastern rosellas of New Zealand; biosecurity implications for wildlife care facilities.

CASE HISTORY: Four juvenile eastern rosellas (Platycercus eximius) were admitted to two separate wildlife care facilities in the Auckland region by members of the public. They had missing or dystrophic wing and tail feathers that rendered them flightless, suggestive of beak and feather disease virus (BFDV) infection. Two were subject to euthanasia after failing to re-grow their feathers, with samples taken for histopathology and PCR analysis. Blood samples were obtained from the other two birds at the time of examination, however these individuals were lost to follow up. PATHOLOGICAL AND MOLE

Identification of porcine circovirus in tissues of pigs with porcine dermatitis and nephropathy syndrome.

Thirty-three pigs affected by porcine dermatitis and nephropathy syndrome, 30 from Spain and three from the USA, were investigated in order to detect porcine circovirus (PCV) in their tissues. A standard in situ hybridisation technique using a specific DNA 317-bp probe based on a well-conserved sequence of PCV (which recognises both PCV-1 and PCV-2) was applied to formalin-fixed, paraffin-embedded tissues. Twenty-eight of the 30 Spanish pigs and all three American pigs had PCV in at least one tissue. Viral nucleic acid was detected mainly in lymphoid organs, and especially the lymph nodes. The

Pathogenesis of NGPV and DuCV CO-infection pathogenicity analysis of co-infection with novel goose parvovirus and duck circovirus in Cherry Valley meat ducks.

The prevalence of short beak and dwarfism syndrome (SBDS) in China is increasing, causing much economic losses to the waterfowl industry. Novel goose parvovirus (NGPV) has been identified as the primary pathogen; however, it is difficult to reproduce SBDS with NGPV infection alone. Duck circovirus (DuCV) orchestrates immunosuppression in ducks, rendering them more prone to other pathogenic infection. Notably, a high co-infection rate between DuCV and NGPV has been observed. Given this context, the present study utilized the Cherry Valley meat duck, the largest breed in China, as the experiment
