Coagulase Activity by Staphylococcus aureus: A Potential Target for Therapy?

Staphylococcus aureus is a leading cause of skin and soft tissue infections, foreign body infections, and infective endocarditis. In case of endovascular infection with S. aureus, higher rates of cardiac valve destruction, embolic complications, severe sepsis, and death occur. The unique capacity of S. aureus to induce clotting has been known for over a century; however, its role in virulence has long been controversial. S. aureus secretes two coagulases, staphylocoagulase and von Willebrand factor binding protein that both activate prothrombin to generate fibrin. A better understanding of the

Teicoplanin in the treatment of infections caused by coagulase-negative staphylococci.

A total of 88 patients were treated with teicoplanin for infections caused by coagulase-negative staphylococci in open clinical studies in France and the UK. Teicoplanin was administered once daily, with a mean dose of 323 mg, for a mean duration of 16 days. Thirty-nine patients received teicoplanin alone while 49 received combination treatment. Clinical cure or improvement occurred in 79 of 82 evaluable cases (96.3%) and bacteriological elimination in 82 cases (95.1%). Treatment with teicoplanin alone was clinically successful in all cases, including 20 septicaemias, and the elimination rate

Identification of coagulase-negative staphylococci with the API staph system.

A kit for the identification of staphylococci based on the biochemical criteria proposed by Kloos and Schleifer (W.E. Kloos and K.H. Schleifer, J. Clin. Microbiol., 1:82-88, 1975) is now available commercially. The system was used to identify 100 strains of coagulase-negative staphylococci isolated from various body sites as the primary etiological agent of clinical infection. The increasing importance of staphylococci and their resistance to antibiotics provided the rationale for such an investigation. Over 90% of the Staphylococcus isolates were easily identified as to their species on the b

Role of coagulase-negative staphylococci and micrococci in ocular disease.

Forty-nine strains of coagulase-negative staphylococci and 26 strains of micrococci isolated from human eyes were classified by the Baird-Parker system of 1966. The staphylococci belonged to subgroups II, III, IV, and V with one strain each in subgroups III and IV. However, when biotyped by Baird-Parker's system of 1974, less than 50% (23/49) of strains were typable and they belonged to biotypes 1 and 2. With our proposed modified classification all strains could be classified into biotypes 1, 2, and 3. All the 26 strains of micrococci belonged to subgroup VII. Staphylococci isolated from dive

Mucosa or skin as source of coagulase-negative staphylococcal bacteraemia?

Nosocomial bacteraemia is associated with significant morbidity, mortality, and cost worldwide, and is most commonly caused by coagulase-negative staphylococci (CONS). Establishing the source of CONS bacteraemia is therefore important in the prevention and management of this infection. CONS infections are presumed to originate at the cutaneous sites of central venous catheters (CVCs), a belief that has led to prevention strategies that focus almost exclusively on the skin. However, mucosal colonisation by CONS is well established, suggesting that mucosal sites might be an important source of C

Relationship of maternal to neonatal colonization with coagulase-negative staphylococci.

Infections with coagulase-negative staphylococci (C-S) have become problems of major clinical importance among very low birthweight infants in neonatal intensive care units. Colonization with slime-producing strains of C-S may be a risk factor for these infants in the development of invasive infections. The present study evaluated the maternal genital tract as a possible source for neonatal colonization with C-S. Specific objectives were to determine the incidence of vaginal colonization with C-S in pregnant women with special reference to slime-producing strains and whether transmission of C-

Baerveldt Scleral Patch Graft Abscess Secondary to Coagulase-Negative Staphylococcus.

We report the case of a 69-year-old female who underwent a Baerveldt implant placement for severe-stage primary open-angle glaucoma and developed a bacterial infection of the conjunctiva and abscess of the scleral patch graft with subsequent tube exposure. The infection was identified 3 weeks postoperatively and a topical antibiotic was immediately initiated. A concurrent systemic staphylococcal infection was discovered by an outside physician and oral cephalexin was initiated. Despite antibiotic treatment, the conjunctival erosion progressed, and tube revision was required. Culture of the abs

Drug-resistant coagulase-negative staphylococcal endophthalmitis following dexamethasone intravitreal implant.

A 60-year-old female came to our hospital with defective vision due to persistent diabetic macular edema and was treated with intravitreal ozurdex implant in the right eye. Three days later, the patient presented with diminution of vision, ocular pain, congestion, and hypopyon with no fundus view. B-scan ultrasonography showed vitritis, and diagnosis of endophthalmitis was made. Subsequently, 25-gauge pars plana vitrectomy was performed along with intravitreal vancomycin and amikacin and removal of implant. On culture, there was growth of coagulase-negative staphylococcus which was resistant t

Treatment Options for Persistent Coagulase Negative Staphylococcal Bacteremia in Neonates.

BACKGROUND: Coagulase-negative staphylococci (CoNS) are the leading cause of late-onset sepsis in neonatal intensive-care units (NICUs). Despite adequate therapeutic levels and in vivo susceptibility, vancomycin is not always adequate in clearing CoNS bacteremias and other theurapeutic regimens are warranted especially when dealing with persistent CoNS bacteremias. METHODS: Retrospective analysis of neonates with persistent CoNS bacteremias in a tertiary NICU in two study periods (January 2006 to December 2007 and January 2011 to December 2012). Demographics, clinical characteristics and anti

[Clinical consideration of coagulase negative Staphylococci isolated in blood culture].

Despite blood culture's usefulness in antimicrobial therapy, fewer blood cultures and the infrequency of more than 1 set in cultures appear to be problems in Japan. Since June 2007 infection control team (ICT) recommended more than 1 set in blood sampling and intervention in positive blood culture, coagulase negative Staphylococci (CNS) has frequently been isolated from blood culture and its clinical significance is often difficult to judge. To determine the effect of ICT intervention, we evaluated the number of blood culture specimens, the frequency of more than 1 set in all blood culture spe

Retrospective study of necropsy-associated coagulase-positive staphylococci in horses.

BACKGROUND/AIM: Prostate cancer is the second most common malignancy among men worldwide, with progression to castration-resistant prostate cancer (CRPC) posing significant therapeutic challenges. Enzalutamide, a second-generation androgen receptor antagonist, initially demonstrates efficacy in treating metastatic CRPC; however, resistance inevitably develops. Dysregulation of the epidermal growth factor receptor (EGFR) signaling pathway has been implicated in therapy resistance and metastatic progression. Secretory carrier membrane protein 3 (SCAMP3) and epidermal growth factor receptor subst

Clinical validation for oxacillin susceptibility testing of coagulase negative staphylococci.

Oxacillin susceptibility of coagulase negative staphylococci as assessed by conventional methods was confirmed by a modified Etest method, extended to detect heteroresistance. Verification of susceptibility was followed by successful treatment for six consecutive children with deep seated infections. Physicians' trust in such a validated method will contribute to the appropriate use of antibiotics.

Nosocomial coagulase negative Staphylococci bacteremia: five year prospective data collection.

UNLABELLED: Coagulase-negative Staphylococcus (CoNS) species, as a group, constitute a major component of the normal microflora of the human skin and mucous membranes. Over the last 20 years, there has been an increase in the documentation of infections due to CoNS, especially with S. epidermidis species, the most common cause of nosocomial primary bloodstream infections. OBJECTIVE: To determine the frequency of CoNS isolates in blood cultures, to evaluate the meaning of this isolation (contaminant or pathogen), and to determine their epidemiologic and susceptibility patterns (oxacillin, cipr

Risk of endocarditis among patients with coagulase-negative Staphylococcus bacteremia.

Coagulase-negative staphylococci (CoNS) are currently considered typical microorganisms causing infective endocarditis (IE) in patients with prosthetic valves. The objective was to determine variables associated with IE in patients with CoNS bacteremia. We performed an analysis of the clinical characteristics of patients with CoNS bacteremia admitted to a university hospital in Madrid (Spain) from 2021 to December 2022 according to the occurrence of IE. This study is an evaluation of a bacteremia registry. During the study period, 106 patients with CoNS bacteremia were detected. In 85 patients
