Sphingomyelin synthase 1-generated sphingomyelin plays an important role in transferrin trafficking and cell proliferation.

Transferrin (Tf) endocytosis and recycling are essential for iron uptake and the regulation of cell proliferation. Tf and Tf receptor (TfR) complexes are internalized via clathrin-coated pits composed of a variety of proteins and lipids and pass through early endosomes to recycling endosomes. We investigated the role of sphingomyelin (SM) synthases (SMS1 and SMS2) in clathrin-dependent trafficking of Tf and cell proliferation. We employed SM-deficient lymphoma cells that lacked SMSs and that failed to proliferate in response to Tf. Transfection of SMS1, but not SMS2, enabled these cells to inc

APOE4 and INPP5D converge on membrane mechanics to regulate endocytosis in human astrocytes.

Disrupted endocytosis is an early feature of Alzheimer's disease (AD), but how genetic risk factors functionally impact this pathway remains unclear. Using isogenic human iPSC-derived astrocytes, we show that the AD risk variant, APOE4, impairs clathrin-mediated membrane curvature, clathrin-mediated endocytosis, and alters plasma membrane lipid saturation and tension. Compared to APOE3 astrocytes, APOE4 cells display an accumulation of flat clathrin structures, reduced maturation of clathrin-coated pits, decreased early endosomes, and reduced endocytic uptake. We then identify the AD risk gene

Mannose 6-phosphate receptor in porcine thyroid follicle cells. Localization and possible implications for the intracellular transport of thyroglobulin.

Thyroglobulin has been shown to be phosphorylated and to carry the mannose 6-phosphate (M6P) signal in terminal position. In order to investigate whether the cation-independent mannose 6-phosphate receptor (CI-MPR) can possibly play a role in the transport of thyroglobulin the localization of the receptor was analyzed in thyroid follicle cells. The immunocytochemical observations showed that the CI-MPR is primarily located in elements of the endocytic pathway such as coated pits and endosomes. This localization of the CI-MPR in thyrocytes differs from the receptor sites in other cell types by

Retrograde transport of the transmembrane estrogen receptor, G-protein-coupled-receptor-30 (GPR30/GPER) from the plasma membrane towards the nucleus.

G-protein-coupled receptor 30 (GPR30/GPER) belongs to the seven transmembrane receptor (7TMR) superfamily, the most common class of surface receptor with approximately 800 known members. GPER promotes estrogen binding and rapid signaling via membrane-associated enzymes resulting in increased cAMP and release of heparan bound epidermal growth factor (proHB-EGF) from breast cancer cells. However, GPER is predominately localized intracellularly in breast cancer cells with minor amounts of receptor on the cell surface, an observation that has caused some controversy regarding its potential role as

Reframing clathrin-mediated viral entry: evidence standards, context dependence and host-directed vulnerabilities.

Viral entry is the first obligatory step of productive infection and a rational point for host-directed antiviral intervention. This structured narrative review synthesizes evidence from database searches and citation tracking performed up to 18 June 2026. Clathrin-mediated endocytosis (CME) can internalize virus-receptor complexes and deliver them to endosomal compartments that support penetration, fusion, capsid remodeling, uncoating or genome release. However, CME should be interpreted as a series of experimentally separable checkpoints rather than as a single pathway label. Viral use of CM

Identification of proteins involved in microglial endocytosis of alpha-synuclein.

Aggregated alpha-synuclein, a protein playing pivotal roles in the pathogenesis of Parkinson disease (PD) and related synucleinopathy, has been shown to activate microglia, the key cells in neuroinflammation. However, the mechanisms by which aggregated alpha-synuclein enters microglia remain uncharacterized. In this study, we first replicated our previous results with a modified protocol that generated aggregated alpha-synuclein more efficiently. Next, using two recently developed proteomic techniques, SILAC (Stable Isotope Labeling of Amino acid in Cell cultures) and PROCEED (PROteome of Cell

Transferrin receptors on the surfaces of retinal pigment epithelial cells are associated with the cytoskeleton.

Retinal pigment epithelial cells, derived from human donor eyes, have been grown in culture as monolayers on membrane filters or plastic surfaces and shown to possess transferrin receptors with a monomeric molecular mass of 93,000. These receptors internalize 125I-labelled transferrin and recycle it to the surrounding medium in a similar manner to other cell types. Scatchard analyses show that there are about 100,000 high-affinity receptors on the surface of each cell and most of these receptors are associated with the cytoskeleton. In total cell extracts, there are additional low-affinity bin

Spermatogenic cells do internalize Sertoli androgen-binding protein: a transmission electron microscopy autoradiographic study in the rat.

Specific binding sites for androgen-binding protein (ABP) have been demonstrated recently to be present on germ cells from the rat and on membrane-enriched fractions from rat germ cells. The present study was undertaken to test if such receptors could lead to the activation of a specific internalization pathway as in other cells. Isolated rat germ cells and in situ rat germ cells, maintained within the seminiferous epithelium with either an intact or a bypassed blood testis barrier, were exposed to culture medium containing 12,000 cpm/ml [3H] delta 6-testosterone (2.5 pg) photoaffinity-labeled

Identification of membrane-bound carbonic anhydrase in white matter coated vesicles: the fate of carbonic anhydrase and other white matter coated vesicle proteins in triethyl tin-induced leukoencephalopathy.

We have extended our studies on the content of white matter derived coated vesicles (WMCVs) to show that they are enriched in membrane-bound carbonic anhydrase. Within the myelin complex membrane-bound carbonic anhydrase is concentrated in the periaxolemmal domain; however, this protein is enriched almost sevenfold in the bilayer of coated vesicles even relative to this myelin membrane region. These data suggest that some vesicles are derived from a site at which this enzyme is highly localized. The enrichment observed for membrane-bound carbonic anhydrase is unique since other periaxolemmal p

Coated vesicles from the thyroid gland: isolation, characterization, and a search for a possible role in thyroglobulin transport.

We report the first isolation of purified coated vesicles (CVs) from thyroid gland. Bovine thyroid CVs were isolated by differential centrifugation, including a step through sucrose-D2O, using a modification of the method described by Nandi et al. (1) for bovine brain CVs. The CVs were characterized by electron microscopy, sedimentation properties, and SDS-PAGE of the protein components. Thyroglobulin (Tg) was found to be associated with the purified CVs. When the thyroid CVs were exposed to conditions known to remove the protein coat from brain CVs, such as low ionic strength at pH 8.5, most

Inhibitors of clathrin-dependent endocytosis enhance TGFbeta signaling and responses.

Clathrin-dependent endocytosis is believed to be involved in TGFbeta-stimulated cellular responses, but the subcellular locus at which TGFbeta induces signaling remains unclear. Here, we demonstrate that inhibitors of clathrin-dependent endocytosis, which are known to arrest the progression of endocytosis at coated-pit stages, inhibit internalization of cell-surface-bound TGFbeta and promote colocalization and accumulation of TbetaR-I and SARA at the plasma membrane. These inhibitors enhance TGFbeta-induced signaling and cellular responses (Smad2 phosphorylation/nuclear localization and expres

Molecular mechanisms of receptor-mediated endocytosis in the renal proximal tubular epithelium.

Receptor-mediated endocytosis is a pivotal function of renal proximal tubule epithelial cells (PTECs) to reabsorb and metabolize substantial amounts of proteins and other substances in glomerular filtrates. The function accounts for the conservation of nutrients, including carrier-bound vitamins and trace elements, filtered by glomeruli. Impairment of the process results in a loss of such substances and development of proteinuria, an important clinical sign of kidney disease and a risk marker for cardiovascular disease. Megalin is a multiligand endocytic receptor expressed at clathrin-coated p

Glycosyl-phosphatidylinositol-anchored membrane proteins.

Many proteins of eukaryotic cells are anchored to membranes by covalent linkage to glycosyl-phosphatidylinositol (GPI). These proteins lack a transmembrane domain, have no cytoplasmic tail, and are, therefore, located exclusively on the extracellular side of the plasma membrane. GPI-anchored proteins form a diverse family of molecules that includes membrane-associated enzymes, adhesion molecules, activation antigens, differentiation markers, protozoan coat components, and other miscellaneous glycoproteins. In the kidney, several GPI-anchored proteins have been identified, including uromodulin

Purification and properties of 100-kd proteins from coated vesicles and their reconstitution with clathrin.

Bullock brain coated vesicles contain a family of at least six 100-kd polypeptides which have the property of promoting clathrin assembly. These proteins have been purified from Triton X-100-extracted coated vesicles by a combination of gel filtration and chromatography on hydroxylapatite and DE-52 cellulose. Three major 100-kd species occur as complexes with a stoichiometric amount of a 50-kd polypeptide. On cross-linking these complexes, the chief products appear to contain two polypeptides of 100 kd and two of 50 kd. These 100-kd/50-kd complexes will polymerise with low concentrations of cl

Howell-Jolly body counting as a measure of splenic function. A reassessment.

Non-surgical and surgical asplenia predisposes to fatal infections; therefore, simple, non-invasive and repeatable tests for assessing splenic function are required, even in non-specialized medical institutions. Howell-Jolly bodies are the most characteristic peripheral blood abnormality after splenectomy, but their counting is not considered a reliable measure of splenic function. In this study, in a group of splenectomized subjects and of patients with non-surgical hyposplenism, we have compared counting of Howell-Jolly bodies, stained by both the May-Grünwald/Giemsa method and the Feulgen r

Molecular requirements for the internalisation step of endocytosis: insights from yeast.

Molecular genetic studies of endocytosis using the unicellular eukaryote Saccharomyces cerevisiae (budding yeast) have led to the identification of many cellular components, both proteins and lipids, required for this process. While initially, many of these requirements (e.g. for actin, various actin-associated proteins, the ubiquitin conjugation system, and for ergosterol and sphingolipids) appeared to differ from known requirements for endocytosis in higher eukaryotes (e.g. clathrin, AP-2, dynamin), it now seems that endocytosis in higher and lower eukaryotes share many requirements. Often,

Induction of thyroiditis in guinea-pigs by intravenous injection of rabbit anti-guinea-pig thyroglobulin serum. 3. Electron microscopic study.

The ultrastructure of the thyroids of guinea-pigs receiving rabbit anti-guinea-pig thyroglobulin serum intravenously has been studied. Rounded, moderately electron-dense deposits were found in the intercellular spaces and between the basal plasma membrane and the basement membrane of the follicle cells. The deposits increased in size with time, and they probably correspond to the rounded droplets of γ-globulin noted in a previous fluorescence microscopic study of material from the same animals. The intracellular changes in the follicle cells were restricted to an increase in the size and the n

Use of colloidal gold cytochemistry in the study of the basic cell biology of cancer.

We are currently investigating the morphologic aspects of two areas of the basic cell biology of cancer: tumor-specific surface antigens as targets for immunotoxins, and the phenomenon of multidrug resistance in chemotherapy of human tumors. Colloidal gold cytochemistry has provided a useful method for the electron-microscopic cytochemical detection of materials endocytosed by cells in culture. This technique has been used to study the internalization pathway of ligands bound to the surface of cancer cells, particularly antibodies for use as immunologic targeting reagents for the construction

Mechanism of the metal-mediated endocytosis of the prion protein.

The cellular form of the prion protein, PrP(c), is critically required for the establishment of prion diseases, such as Creutzfeldt-Jakob disease. Within the N-terminal half of PrP(c) are four octapeptide repeats that bind Cu(2+). Exposure of neuronal cells expressing PrP(c) to Cu(2+) results in the rapid endocytosis of the protein. First, PrP(c) translocates laterally out of detergent-resistant lipid rafts into detergent-soluble regions of the plasma membrane, then it is internalized through clathrin-coated pits. The extreme N-terminal region of PrP(c) is critically required for its endocytos

Furrow-specific endocytosis during cytokinesis of zebrafish blastomeres.

Mutations affecting endocytosis, such as those in clathrin and dynamin, unexpectedly cause defects in cytokinesis in a number of organisms. To explore the relationship between endocytosis and cytokinesis, we used the relatively large cells of the transparent zebrafish embryo. Using fluorescent markers for fluid-phase as well as plasma membrane uptake, we demonstrate that cytokinesis involves furrow-specific endocytosis. Clathrin-coated pits are visible near the furrow in ultrathin sections, while immunolabeling demonstrates that clathrin and caveolin are localized to the cleavage furrow. Hence
