Cognitive enhancers as a treatment for heroin relapse and addiction.

Heroin addiction is a disorder that stems from maladaptive plasticity within neural circuits and produces broad cognitive deficits. Despite considerable advances in psychotherapy and pharmacotherapy for heroin relapse and addiction, effective treatments for heroin use disorder are still lacking. Increasing preclinical evidence indicates that heroin seeking behavior is persistent after withdrawal, while cognitive dysfunctions associated with chronic heroin use are an important contributing factor to risk of heroin relapse and addiction. Cognitive enhancers may be used to stimulate treatment suc

[The 1998 domestic state of development of cognitive enhancers].

Recently, there is a great social problem that geriatric disorders, especially senile dementia, are growing rapidly with the increasing percentage of aged individuals in the total population. However, there is yet no drug that has reliable effects on senile dementia on the market. Therefore, society requires the development of new drugs that can support patients so that they can smoothly live their daily lives by themselves. In this study, we attempted to investigate the 1998 domestic state of development of cognitive enhancers by summarizing many publications and by gathering questionnaires f

Efficacy of an internet-delivered cognitive enhancement intervention for subjective residual cognitive deficits in remitted major depressive disorder: A randomized crossover trial.

BACKGROUND: Cognitive deficits such as difficulties with attention, memory, and executive functions are frequently reported during remission from depression and relates to adverse functioning in daily life and risk of relapse. There is therefore a need for interventions targeting cognitive deficits after depression. However, few randomized controlled trials have investigated the efficacy of interventions targeting subjective residual cognitive deficits in adults remitted from depression. METHODS: This randomized crossover trial aimed to investigate the efficacy of an internet-delivered cognit

Multimodal colour modulation for cognitive enhancement in intelligent rehabilitation: a systematic review and translational guidance.

BACKGROUND: Colour modulation has been investigated as a non-invasive means of supporting cognitive performance, yet translation to rehabilitation-oriented applications has been constrained by fragmented evidence, inconsistent parameterisation, and heterogeneous assessment strategies. MAIN BODY: This systematic review synthesises 75 studies published between 2011 and 2025 through a Colour-Modality-Cognition framework that links stimulus design, measurement strategy, and cognitive goals. Across the evidence base, a graded and context-dependent pattern emerged. Contrast-related variables, parti

6,7-Dihalo-Benzothiadiazines as Potent and Selective AMPA Receptor Modulators for Cognitive Enhancement and Neuroprotection.

α-Amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors are tetrameric ionotropic glutamate receptors that mediate fast excitatory synaptic transmission in the brain and represent important therapeutic targets for neurological disorders. Positive allosteric modulators (PAMs) of AMPA receptors enhance rapid excitatory signaling by increasing receptor's sensitivity to glutamate and have been widely explored as agents to improve cognitive function in central nervous system (CNS) diseases. Structural modification of 3,4-dihydro-2H-1,2,4-benzothiadiazine 1,1-dioxides (BTDs) analogs is

Psychotherapeutic treatments for generalized anxiety disorder: cognitive and behavioral therapies, enhancement strategies, and emerging efforts.

INTRODUCTION: Generalized anxiety disorder (GAD) is common and disabling. Different versions of cognitive behavioral therapy (CBT) have been tested, but no treatment works for everyone. Therefore, researchers have attempted approaches to enhance CBT. AREAS COVERED: The current narrative review examines meta-analyses and individual trials of CBT-based treatments for GAD. We focus on CBT and its cognitive and behavioral components as well as efforts to enhance CBT and its dissemination and generalizability. Enhancement efforts included interpersonal and emotional processing therapy, mindfulness

Neurostimulation for cognitive enhancement in Alzheimer's disease (the NICE-AD study): a randomized clinical trial.

New therapies for symptoms in Alzheimer's disease (AD) are urgently needed. Prior studies suggest that transcranial direct current stimulation (tDCS), a noninvasive neuromodulatory method, may be a safe and potentially effective treatment, but conclusions have been limited by small-sample sizes and brief stimulation protocols. This double-blind randomized trial involving 100 older adults with mild-to-moderate AD examines effects of 6 months of at-home active tDCS or sham delivered over the dorsolateral prefrontal cortex. The primary outcome is global cognitive performance. Secondary outcomes i

Cognitive enhancement of healthy young adults with hyperbaric oxygen: A preliminary resting-state fMRI study.

OBJECTIVE: To date, no study has examined the effect of hyperbaric oxygen (HBO) on the cognitive performance and spontaneous brain activity in healthy adults using resting-state functional magnetic resonance imaging (rsfMRI). Our aim was to reveal the neural mechanism underlying the change in cognitive performance caused by increased oxygen. METHODS: In this study, we acquired fMRI data from 20 healthy young adults and used placebo-controlled (PBO) rsfMRI to identify the effect of HBO on the cognitive measures and the regional homogeneity (ReHo) in healthy adults. RESULTS: Compared to the PB

Smart drugs and synthetic androgens for cognitive and physical enhancement: revolving doors of cosmetic neurology.

Cognitive enhancement can be defined as the use of drugs and/or other means with the aim to improve the cognitive functions of healthy subjects in particular memory, attention, creativity and intelligence in the absence of any medical indication. Currently, it represents one of the most debated topics in the neuroscience community. Human beings always wanted to use substances to improve their cognitive functions, from the use of hallucinogens in ancient civilizations in an attempt to allow them to better communicate with their gods, to the widespread use of caffeine under various forms (energy

Posttraining androgens' enhancement of cognitive performance is temporally distinct from androgens' increases in affective behavior.

Steroid hormone-induced variations in spatial learning and memory tasks have been reported. In this study, androgens' effects in various cognitive and affective tasks were investigated in order to determine whether any observed differences in cognitive performance could be due to affective changes produced by the hormones. Ovariectomized rats (N = 72) received 0.0, 3.0, or 7.5 mg/kg subcutaneously, of testosterone (T), dihydrotestosterone (DHT), or 5 alpha-androstane-3 alpha,17 beta-diol (3 alpha-Diol) suspended in 10% ethanol/sesame oil v/v. Prescribing trends in cognition enhancing drugs in Australia.

BACKGROUND: This study sought to examine the trends in the prescribing of subsidized and unsubsidized cognition enhancing drugs (CEDs) in Australia over five years from 2002 to 2007. Subsidized cholinesterase inhibitor medication could be prescribed to people with mild to moderate Alzheimer's disease (AD) once a specialist physician had confirmed this diagnosis. Memantine was available for use in moderately severe AD but not subsidized. METHODS: We analyzed the Medicare Australia and Drug Utilisation Sub-Committee databases for CED prescription data, 2002-2007, by gender, age and prescriber c

Task-specific theta enhancement and domain-general alpha/beta suppression as oscillatory signatures of cognitive flexibility.

Cognitive flexibility-the ability to adapt to changing situational or task demands-is a fundamental aspect of human cognition and a potential source of individual differences in higher-order cognitive abilities. To further understand the neural mechanisms underlying this ability, we examined event-related spectral perturbations (ERSP) in three cued switching tasks (parity/magnitude, global/local, number/letter) in a sample of 148 adults. Employing a data-driven approach that combined mass-univariate time-frequency analysis, cluster-based permutation testing, and latent change structural equati

Cognitive Level Enhancement through Vision Exams and Refraction (CLEVER): study protocol for a randomised controlled trial.

BACKGROUND: Longitudinal observational studies have found an association between vision impairment and accelerated decline in cognition. However, no randomised trials have assessed the possible benefit of vision correction on cognitive change. We present the protocol for a three-year randomised controlled trial designed to assess the impact of spectacles for distance and/or near vision correction on cognitive change among community-dwelling elderly participants in India. METHODS: Cognitive Level Enhancement through Vision Exams and Refraction (CLEVER) is a single-centre, open-label, parallel-

Combined physical and cognitive training enhances telomere length in mild cognitive impairment patients.

BACKGROUND: Environmental enrichment training may delay cognitive decline in mild cognitive impairment (MCI) by reducing telomere shortening, a cellular marker of aging. We investigated whether combined physical/cognitive training affects leukocyte telomere length (LTL) in the randomized trial “Train the Brain”. METHODS: LTL was assessed in 94 MCI (75.1 ± 5.1 years) subjects and 37 non-MCI subjects (73.2 ± 4.6 years). MCI patients were assigned to receive either 7 months of training or standard care. After 7 months, both groups were reassessed for changes in LTL and the Alzheimer's Disease Ass

How can we enhance cognitive bias modification techniques? The effects of prospective cognition.

BACKGROUND AND OBJECTIVES: Cognitive bias modification for interpretation, a computerized program which manipulates biased interpretations, has shown therapeutic promise, including evidence that negatively biased interpretations can be reduced, leading to corresponding improvements in symptoms. Cognitive bias modification for cognitive errors (CBM-errors) is a second generation CBM-I procedure which manipulates seven types of cognitive error and is especially relevant to depressive cognition. The aim of this study was to investigate whether the effects of the CBM-errors manipulation would be e

Cognitive and brain function enhancement in Gen X group after personalized, AI supervised EEG-neurofeedback training.

Objective.Interventions supporting medical care and enhancing quality of life in neurodegenerative or age-related cognitive decline are strongly needed. Electroencephalographic (EEG) neurofeedback can enable users to modulate their brain activity through real-time feedback. However, evidence for its clinical effectiveness remains inconclusive, partly due to limited personalization and insufficient task relevance of existing protocols. We tested whether personalized EEG neurofeedback supervised by deep neural networks (DNNs) can enhance cognitive performance in older adults (Gen X).Approach.Fif
