Longitudinal profiling of oligomeric Aβ in human nasal discharge reflecting cognitive decline in probable Alzheimer's disease.

Despite clinical evidence indicating a close relationship between olfactory dysfunction and Alzheimer's disease (AD), further investigations are warranted to determine the diagnostic potential of nasal surrogate biomarkers for AD. In this study, we first identified soluble amyloid-β (Aβ), the key biomarker of AD, in patient nasal discharge using proteomic analysis. Then, we profiled the significant differences in Aβ oligomers level between patient groups with mild or moderate cognitive decline (n = 39) and an age-matched normal control group (n = 21) by immunoblot analysis and comparing the le

Visual Short-Term Memory Activity in Parietal Lobe Reflects Cognitive Processes beyond Attentional Selection.

Visual short-term memory (VSTM) and attention are distinct yet interrelated processes. While both require selection of information across the visual field, memory additionally requires the maintenance of information across time and distraction. VSTM recruits areas within human (male and female) dorsal and ventral parietal cortex that are also implicated in spatial selection; therefore, it is important to determine whether overlapping activation might reflect shared attentional demands. Here, identical stimuli and controlled sustained attention across both tasks were used to ask whether fMRI si

CSF tau368/total-tau ratio reflects cognitive performance and neocortical tau better compared to p-tau181 and p-tau217 in cognitively impaired individuals.

INTRODUCTION: Cerebrospinal fluid (CSF) tau biomarkers are reliable diagnostic markers for Alzheimer's disease (AD). However, their strong association with amyloid pathology may limit their reliability as specific markers of tau neurofibrillary tangles. A recent study showed evidence that a ratio of CSF C-terminally truncated tau (tau368, a tangle-enriched tau species), especially in ratio with total tau (t-tau), correlates strongly with tau PET tracer uptake. In this study, we set to evaluate the performance of the tau368/t-tau ratio in capturing tangle pathology, as indexed by a high-affinit

Left dorsomedial prefrontal cortex hyperactivation reflects early compensation for emerging social cognition difficulties in multiple sclerosis.

BACKGROUND: Social cognition (SC) has emerged as a relevant neurocognitive domain in people with Multiple Sclerosis (pwMS), given its impact on everyday functioning. However, the neural mechanisms supporting SC performance in pwMS remain underexplored, highlighting the need for ecologically valid assessment paradigms capable of capturing both behavioral and neural aspects of social-cognitive functioning. In this study, we addressed this gap by investigating the clinical application of an fMRI-adapted version of the Edinburgh Social Cognition Test (ESCoT) to explore the neural correlates of SC

Altered temporal dynamics of prefrontal ERP responses reflecting neural adaptation in patients with amnestic mild cognitive impairment.

BACKGROUND: With increasing life expectancy, age-related cognitive disorders including mild cognitive impairment (MCI) represent major public health challenges, and amnestic MCI (aMCI) has the greatest risk of progression to dementia due to Alzheimer's disease (AD). Event-related potentials (ERPs), particularly the P200 component, have been studied as potential biomarkers, but conventional grand-averaged approaches often yield inconsistent results. This study examined the temporal dynamics of prefrontal ERPs in a large cohort of older adults to assess neural adaptation processes associated wit

Contrasting cognitive, behavioral, and physiological responses to breathwork vs. naturalistic stimuli in reflective chamber and VR headset environments.

The MindGym, a novel immersive technology utilizing a reflective chamber environment, was developed to create standardized experiential content, including anxiolytic experiences. This study examined whether therapeutic experiences originally created in the MindGym could maintain their efficacy when delivered via 360-degree recordings through virtual reality (VR) headsets. A randomized controlled trial (N=126) compared anxiety reduction, cognitive performance, and physiological responses across four conditions: MindGym and VR platforms, each delivering either breathwork or rain stimuli. Neural Correlates of Stepping in Healthy Elderly: Parietal and Prefrontal Cortex Activation Reflects Cognitive-Motor Interference Effects.

Gait analysis involving cognitive-motor dual task (DT) is a diagnostic tool in geriatrics. Cognitive-motor interference effects during DT, such as decreased walking speed and increased step-to-step variability, have a high predictive value for fall risk and cognitive decline. Previously we showed the feasibility of DT during functional magnetic resonance imaging (fMRI) using an MRI-compatible stepping device. Here, we improved the DT-fMRI protocol with respect to task difficulty and signal robustness, making it more suitable for individualized analysis to better understand the neuronal substra

Intra-erythrocytes magnesium deficiency could reflect cognitive impairment status due to vascular disease: a pilot study.

BACKGROUND AND AIMS: Magnesium is a fundamental cation that regulates neuronal transmission, protein synthesis, energy metabolism. Magnesium deficiency mostly affects nervous and cardiovascular systems determining weakness, tremors, seizure and arrhythmias. This condition retains also a role in memory function and neuronal plasticity. Importantly magnesium deficiency could remain latent and asymptomatic resulting a risk factor for cardiovascular disease. In this sense we aim to determine magnesium status in patient presenting cognitive impairment of vascular origin. METHODS: 21 healthy subjec

Client reflections on confirmation and disconfirmation of expectations in cognitive behavioral therapy for generalized anxiety disorder with and without motivational interviewing.

Objective: Addressing methodological shortcomings of prior work on process expectations, this study examined client process expectations both prospectively and retrospectively following treatment. Differences between clients receiving cognitive behavioral therapy (CBT) versus motivational interviewing integrated with CBT (MI-CBT) were also examined. Method: Grounded theory analysis was used to study narratives of 10 participants (N = 5 CBT, 5 MI-CBT) who completed treatment for severe generalized anxiety disorder as part of a larger randomized controlled trial. Results: Clients in both groups

Frontal EEG activation asymmetry reflects cognitive biases in anxiety: evidence from an emotional face Stroop task.

Electroencephalography (EEG) has been extensively used in studies of the frontal asymmetry of emotion and motivation. This study investigated the midfrontal EEG activation, heart rate and skin conductance during an emotional face analog of the Stroop task, in anxious and non-anxious participants. In this task, the participants were asked to identify the expression of calm, fearful and happy faces that had either a congruent or incongruent emotion name written across them. Anxious participants displayed a cognitive bias characterized by facilitated attentional engagement with fearful faces. Fea

Regional Tau Deposition Reflects Different Pathways of Subsequent Neurodegeneration and Memory Decline in Cognitively Normal Older Adults.

OBJECTIVE: Tau pathology is recognized as a primary contributor to neurodegeneration and clinical symptoms in Alzheimer's disease (AD). This study aims to localize the early tau pathology in cognitively normal older people that is predictive of subsequent neurodegeneration and memory decline, and delineate factors underlying tau-related memory decline in individuals with and without β-amyloid (Aβ). METHODS: A total of 138 cognitively normal older individuals from the Berkeley Aging Cohort Study underwent 11 C-Pittsburgh Compound-B (PiB) positron emission tomography (PET) to determine Aβ posit

Serum Daytime Melatonin Levels Reflect Cerebrospinal Fluid Melatonin Levels in Alzheimer's Disease but Are Not Correlated with Cognitive Decline.

BACKGROUND: Nocturnal cerebrospinal fluid (CSF) and blood melatonin levels are altered in Alzheimer's disease (AD). However, literature remains inconclusive on daytime blood melatonin levels. A positive correlation between melatonin levels and Mini-Mental State Examination (MMSE) scores in AD subjects has been evidenced following cross-sectional analyses. Whereas a correlation between serum and spinal CSF melatonin has been shown in healthy volunteers, an equal investigation in AD patients still has to be undertaken. OBJECTIVE: 1) To evaluate whether serum melatonin levels correlate with spin

The APOE paradox: do attentional control differences in mid-adulthood reflect risk of late-life cognitive decline.

Possession of an Apolipoprotein E (APOE) e4 allele is an established risk factor for Alzheimer's disease, whereas the less commonly studied e2 variant is premised to offer some protection. This research explores the purported deleterious-protective dichotomy of APOE variants on attentional control in mid-adulthood. Sixty-six volunteers, aged 45-55 years, completed 3 tasks that provided complementary measures of attentional control: prospective memory, sustained attention, and inhibition. Performance was compared between e2 carriers, e4 carriers, and e3 homozygotes (the population norm). Carrie

A feasibility study of a new version of self-practice/self-reflective (SP/SR) groupwork for cognitive behaviour therapy training.

Clients receiving cognitive behaviour therapy (CBT) may disengage from therapy for different reasons, including limited self-awareness and interpersonal skills among the modifiable therapist factors. In this feasibility study, we evaluated recruitment, retention, acceptability, utility of outcome measures, adverse events, and cost of a self-practice/self-reflective (SP/SR) groupwork intervention facilitated by Institute of Group Analysis (IGA)-trained practitioners, alongside outcomes. CBT trainees undertook six SP/SR sessions over 12 months delivered by IGA-trained facilitators. Emotional-rel

MRI-Based Medial Temporal Atrophy May Reflect Amyloid PET Positivity in Cognitively Normal Individuals: A Preliminary Study.

This study investigated whether magnetic resonance imaging (MRI)-based medial temporal morphometry can predict amyloid positron emission tomography (PET) positivity in cognitively normal individuals. Sixteen consecutive participants (eight amyloid PET-positive and eight PET-negative) were retrospectively identified, all within the normal cognitive range (Mini-Mental State Examination 28-30). Amyloid PET was performed using florbetapir, with visual interpretation as the primary diagnostic criterion and Centiloid values as supportive measures. MRI-derived volumetric analysis was conducted using

Alterations in heart-brain interactions under mild stress during a cognitive task are reflected in entropy of heart rate dynamics.

Many people experience mild stress in modern society which raises the need for an improved understanding of psychophysiological responses to stressors. Heart rate variability (HRV) may be associated with a flexible network of intricate neural structures which are dynamically organized to cope with diverse challenges. HRV was obtained in thirty-three healthy participants performing a cognitive task both with and without added stressors. Markers of neural autonomic control and neurovisceral complexity (entropy) were computed from HRV time series. Based on individual anxiety responses to the expe

Relationship between primitive reflexes, extra-pyramidal signs, reflective apraxia and severity of cognitive impairment in dementia of the Alzheimer type.

Controversy exists in the literature about the significance of primitive reflexes (PR) and extrapyramidal signs (EP) as diffuse cortical dysfunction signs and their relationship to age and cognitive impairment. A sample of 91 patients with a dementia of the Alzheimer type were examined with a standardized technique to assess the relationship between the finding of PR, EP and severity of cognitive impairment measured by Mini-Mental Status Examination. The value of a short cognitive test, the reflective apraxia (i.e. imitation of meaningless gestures), were also assessed. A significantly lower M

[Strategies and reflections on the construction of a comprehensive management system for dementia in the elderly: based on the practice of cognitive disorder specialty capacity building].

With the acceleration of population aging, the number of elderly individuals suffering from dementia continues to rise sharply, becoming a significant social issue that affects public health and urgently requires a comprehensive management model throughout the disease course to address it. However, China still faces numerous challenges in the diagnosis and treatment of senile dementia, including insufficient public awareness, limited screening capabilities at the grassroots level, a shortage of specialist physicians, a lack of continuity in management systems, and inadequate rehabilitation and

Frontal midline theta power during the cue-target-interval reflects increased cognitive effort in rewarded task-switching.

Cognitive performance largely depends on how much effort is invested during task-execution. This also means that we rarely perform as good as we could. Cognitive effort is adjusted to the expected outcome of performance, meaning that it is driven by motivation. The results from recent studies suggest that the expenditure of cognitive control is particularly prone to being affected by modulations of cognitive effort. Although recent EEG studies investigated the neural underpinnings of the interaction of effort and control, reports on how cognitive effort is reflected by oscillatory activity of
