Proteomic profiling reveals novel insights into hypercortisolism adrenal adenomas.

Adrenal cortisol-producing adenomas (CPAs) represent the most common endocrine disorder among adrenal incidentalomas and are classified as either subclinical Cushing's syndrome (SCS) or Cushing's syndrome (CS) based on the degree of cortisol hypersecretion. The molecular mechanisms driving hypercortisolism remain poorly understood. To address this, we perform comprehensive proteomic profiling of 9 CS, 10 SCS, and 10 non-functioning adenomas (NFAs). Our analysis reveals a central role for cholesterol metabolic reprogramming in the pathogenesis of CPAs. We observe a progressive gradient of dysre

Consensus clinical recommendations for the management of plasma lipid disorders in the Middle East.

BACKGROUND: Plasma lipid disorders are key risk factors for the development of atherosclerotic cardiovascular disease (ASCVD) and are prevalent in the Middle East, with rates increasing in recent decades. Despite this, no region-specific guidelines for managing plasma lipids exist and there is a lack of use of guidelines developed in other regions. METHODS: A multidisciplinary panel of regional experts was convened to develop consensus clinical recommendations for the management of plasma lipids in the Middle East. The panel considered existing international guidelines and regional clinical e

Efficacy and outcome of expanded newborn screening for metabolic diseases--report of 10 years from South-West Germany.

BACKGROUND: National newborn screening programmes based on tandem-mass spectrometry (MS/MS) and other newborn screening (NBS) technologies show a substantial variation in number and types of disorders included in the screening panel. Once established, these methods offer the opportunity to extend newborn screening panels without significant investment and cost. However, systematic evaluations of newborn screening programmes are rare, most often only describing parts of the whole process from taking blood samples to long-term evaluation of outcome. METHODS: In a prospective single screening ce

Orchestration of a blood-derived and ADARB1-centred network in Alzheimer's and Parkinson's disease.

The peripheral immune system is thought to influence the pathogenesis of the central nervous system in Alzheimer's disease (AD) and Parkinson's disease (PD). This study aimed to investigate the characteristics of peripheral leukocytes in AD and PD by comprehensively analyzing the transcriptomic and metabolic features in the blood (NCONTROL = 15; NAD = 11; NPD = 13). The study found an ADARB1-centered module that was associated with diagnosis, phenethylamine, and glutamate. The module consisted of ADARB1, a vital RNA-editing enzyme, LINC02960, and 109 miRNAs. The study also predicted that the A

[Chinese multi-disciplinary consensus on the diagnosis and treatment of hyperuricemia and its related diseases].

The prevalence of hyperuricemia (HUA) in China has been increasing rapidly during the past decade and this disease has stepped up to the second most common metabolic disease following diabetes mellitus. Different disciplinary panels have developed guidelines and consensus on hyperuricemia and gout in respective fields. However, hyperuricemia has been well illustrated to be related to multiple organ disorders such as kidney injuries, endocrine and metabolic abnormalities, and cardiocerebrovascular diseases etc. A multi-disciplinary expert consensus on hyperuricemia and its related diseases will

Hypophosphatasia: Results of a Country-Wide Selective Screening Program Using NGS Technology as a First-Tier Test.

Hypophosphatasia (HPP) is a rare hereditary metabolic disorder caused by nucleotide variants (NVs) in the ALPL gene, leading to a deficiency of tissue-nonspecific alkaline phosphatase (TNSALP). HPP has a variable clinical presentation (such as impaired mineralization of bones and teeth, respiratory dysfunctions, muscle weakness, muscle and bone pain, growth deficiency, nephrocalcinosis, seizures, encephalopathy, intracranial hypertension, and intellectual disability) overlapping with other disorders which often delays a correct diagnosis. In this study, we present the results of a two-center (

Discriminative Plasma Lipidomic Signatures of Dementia with Lewy Bodies and Alzheimer's Disease: A Targeted Mass Spectrometry and Machine Learning Approach.

BACKGROUND: Dementia with Lewy bodies (DLB) exhibits a more aggressive progression and poorer prognosis than Alzheimer's disease (AD), yet clinical differentiation remains challenging. Dysregulated lipid metabolism, implicated in α-synuclein aggregation and neuroinflammation, may offer specific biomarkers for distinguishing DLB and AD. METHODS: This cross-sectional study implemented targeted lipidomic profiling to comprehensively characterize plasma lipidomes in a cohort comprising 50 DLB patients and 56 AD patients. Integrated bioinformatics analysis reveals cytokine dysregulation, pathway alterations, and machine learning identification of diagnostic biomarkers in sepsis.

Sepsis, a life-threatening dysregulated host response to infection, involves complex cytokine signaling. Comprehensive bioinformatics analysis of cytokine activity, associated pathways, and immune alterations in sepsis is warranted. Using the sepsis dataset GSE26378 from GEO, we analyzed differential cytokine pathway activity with ssGSEA and identified differentially expressed genes (DEGs). Cytokine-related genes (CRGs) were extracted and overlapped with DEGs. Protein-Protein Interaction (PPI) network analysis and functional enrichment were performed on differentially expressed CRGs. Cytokine

Metabolic phenotyping of sepsis in large multicenter cohorts: identification of a reproducible high-risk subgroup.

BACKGROUND: Sepsis represents a profound metabolic crisis, yet the prognostic significance of glycolipid biomarkers remains unclear. Most prior studies examined single metabolic indicators, overlooking their combined effects on organ dysfunction and survival. This study aimed to identify glycolipid-related biomarkers predicting outcomes in sepsis and to define a reproducible metabolic phenotype linked to multi-organ dysfunction and mortality. METHODS: We conducted a multicenter retrospective cohort study of 2,970 adults with sepsis admitted to three tertiary hospitals in China (2015–2023), wit

Application of high-resolution mass spectrometry profiling towards the diagnosis and acute management of maple syrup urine disease.

The current approach for investigating patients with suspected inborn errors of metabolism (IEMs) involves traditional targeted biochemical assays such as amino/organic acid analyses. Although highly effective for confirmatory testing, they are less effective in identifying disorders not included in newborn screening (NBS) panels, and for patients with non-classical clinical presentations. Targeted assays analyze a narrow range of metabolites and are conducted across different analytical platforms, often requiring more than one specimen type. In contrast, comprehensive metabolic profiling usin
