The fibroblast is the target cell in the connective tissue manifestations of Graves' disease.

A controversy exists concerning whether the extraocular muscle cell or the fibroblast is the autoimmune target in Graves' ophthalmopathy (GO). Although the extraocular muscle bodies are grossly enlarged in GO, the muscle cells themselves are histologically intact. Within the muscle bodies, as well as in the fatty connective tissue of the orbit, there is an accumulation of glycosaminoglycans (GAG). These hydrophilic mucopolysaccharides are made by fibroblasts. In this commentary, I will present evidence that the fibroblast, and not the extraocular muscle cell, is the autoimmune target in GO and

Natural killer cell activity in the systemic connective tissue diseases.

By doing 51Cr release assays with K562 cells and single natural killer (NK) cell assays with and without interleukin-2 (IL-2) containing supernatants, we were able to establish differences and similarities in NK cell function of patients with systemic connective tissue diseases. Decreased NK cell activity in systemic lupus erythematosus was found to be due to a paucity of active NK cells, with increased proportions of inactive and IL-2 unresponsive NK cells. The few active NK cells responded well to IL-2 by increasing their recycling capacity. Patients with mixed connective tissue disease had

Giant Cell Hepatitis - A Rare Association with Connective Tissue Disease.

A 68-year-old gentleman presented to hepatology department with asymptomatic year-long history of stably deranged liver function tests. His peak alkaline phosphatase (ALP), was 828 with alanine transaminase (ALT) of 141. Full liver workup was negative; hence, a liver biopsy was organised, which confirmed giant cell hepatitis (GCH). A computed tomography (CT) scan revealed non-specific interstitial pneumonitis (NSIP) pattern interstitial lung disease supported by lung function tests. Antibody testing showed strongly positive antinuclear antibody (ANA) with anti-polymyositis/scleroderma (anti-PM

Intestinal Stromal Cells in the Turmoil of Inflammation and Defective Connective Tissue Remodeling in Inflammatory Bowel Disease.

In steady state, intestinal subepithelial myofibroblasts form a thin layer below the basement membrane. Unlike the rest of the stromal cells in the lamina propria, they express tensile proteins, guide epithelial regeneration, and sense luminal microbiota. Upon inflammation in inflammatory bowel disease (IBD), they express activation markers, accept trophic signaling by infiltrating neutrophils and macrophages, and are activated by cytokines from helper T cells to produce a narrow spectrum of cytokines and a wider spectrum of chemokines, attract cells of innate and adaptive immunity, orchestrat

PDGFR Signaling Mediates Hyperproliferation and Fibrotic Responses of Subsynovial Connective Tissue Cells in Idiopathic Carpal Tunnel Syndrome.

Fibrosis of the subsynovial connective tissue (SSCT) is a pathognomonic change in carpal tunnel syndrome (CTS). Identification of molecular targets and anti-fibrotic therapies could provide new treatment strategies for CTS. The contribution of SSCT cells to fibrosis and the signaling pathways that initiate and aggravate fibrosis in CTS remain unknown. Here we report that platelet-derived growth factor receptor alpha (PDGFRα) positive ( + ) cells accumulate in CTS SSCT and that the presence of fibrotic growth factor, PDGF-AA, results in increased proliferation of PDGFRα+ cells via PI3K/Akt sign

mTOR Complexes Repress Hypertrophic Agonist-Stimulated Expression of Connective Tissue Growth Factor in Adult Cardiac Muscle Cells.

Connective tissue growth factor (CTGF) is a fibrogenic cytokine that promotes fibrosis in various organs. In the heart, both cardiomyocytes (CM) and cardiac fibroblasts have been reported as a source of CTGF expression, aiding cardiac fibrosis. Although the mammalian target of rapamycin (mTOR) forms 2 distinct complexes, mTORC1 and mTORC2, and plays a central role in integrating biochemical signals for protein synthesis and cellular homeostasis, we explored its role in CTGF expression in adult feline CM. CM were stimulated with 10 μM phenylephrine (PE), 200 nM angiotensin (Ang), or 100 nM insu

CD25 is expressed by canine cutaneous mast cell tumors but not by cutaneous connective tissue mast cells.

Canine cutaneous mast cell tumors (MCT) of different histological grades have distinct biological behaviors. However, little is known about underlying molecular mechanisms that lead to tumor development and increasing malignancy with higher tumor grade. Recent studies have identified the interleukin-2 receptor (IL-2R) subunits CD25 and CD2 as markers that distinguish nonneoplastic from neoplastic mast cells in human systemic mastocytosis. In this study, their potential as a marker for canine MCT and their possible impact on MCT carcinogenesis were evaluated. mRNA expression levels of both gene

[Transformation of connective tissue cells by pathological altered synovial fluid as demonstrated by experimental studies (author's transl)].

Joint cavities and tendon sheaths are fissurations in the mesenchyme. The coating synovial lining cell layer is caused through the environment of the synovial fluid. The pathologically altered synovial fluid initiates the transformation and proliferation of the marginal connective tissue cells. Evidence for this thesis are the synovial transformatory processes of the cutaneous connective tissue in the area of a synovial fistula and the formation of synovial like structures in the subcutaneous fatty tissues of the rat following the application of synovial fluid from patients with rheumatoid art

Connective tissue growth factor mediates transforming growth factor β-induced collagen expression in human endometrial stromal cells.

BACKGROUND: Adenomyosis is a medical condition defined by the abnormal presence of endometrial tissue within the myometrium, in which fibrosis occurs with new collagen deposition and myofibroblast differentiation. In this study, the effect of several mediators and growth factors on collagen expression was investigated on human endometrial stromal cells (fibroblasts) derived from adenomyotic endometrium. EXPERIMENTAL APPROACH: RT-PCR, Western blot analysis, pharmacological interventions and siRNA interference were applied to primary cultured human endometrial stromal cells (fibroblasts). Immun

Connective-Tissue Growth Factor (CTGF/CCN2) Induces Astrogenesis and Fibronectin Expression of Embryonic Neural Cells In Vitro.

Connective-tissue growth factor (CTGF) is a modular secreted protein implicated in multiple cellular events such as chondrogenesis, skeletogenesis, angiogenesis and wound healing. CTGF contains four different structural modules. This modular organization is characteristic of members of the CCN family. The acronym was derived from the first three members discovered, cysteine-rich 61 (CYR61), CTGF and nephroblastoma overexpressed (NOV). CTGF is implicated as a mediator of important cell processes such as adhesion, migration, proliferation and differentiation. Extensive data have shown that CTGF

Mechanisms of low-density lipoprotein-induced expression of connective tissue growth factor in human aortic endothelial cells.

Hyperlipidemia is a recognized risk factor for atherosclerotic vascular disease. The underlying mechanisms that link lipoproteins and vascular disease are undefined. Connective tissue growth factor (CTGF) is emerging as a key determinant of progressive fibrotic diseases, and its expression is upregulated by diabetes. To define the mechanisms through which low-density lipoproteins (LDL) promote vascular injury, we evaluated whether LDL can modulate the expression of CTGF and collagen IV in human aortic endothelial cells (HAECs). Treatment of HAECs with LDL (50 microg/ml) for 24 h produced a sig

Increased expression of connective tissue growth factor in fibrotic human liver and in activated hepatic stellate cells.

BACKGROUND/AIMS: Connective tissue growth factor is a recently described mitogenic protein implicated in a variety of fibrotic disorders. Connective tissue growth factor may be a downstream mediator of the pro-fibrotic and mitogenic actions of transforming growth factor-beta, promoting extracellular matrix deposition and fibrogenesis. As transforming growth factor-beta is considered important to the pathogenesis of hepatic fibrosis, we examined the possible contribution of connective tissue growth factor to this process. METHODS: Connective tissue growth factor expression was examined in norm

[Studies on von Willebrand factor antigen in endothelial cells and sera of patients with connective tissue diseases].

Von Willebrand factor antigen (vWF: Ag) is known to be produced and excreted by endothelial cells (EC). The influences of interferon (IFN) on surface, excreted and intracellular vWF: Ag amounts of EC were studied by flow cytometry and ELISA. Serum levels of vWF: Ag in patients with connective tissue diseases were also studied by ELISA. Experiments in vitro showed that IFN increased vWF: Ag amounts of EC. Serum levels of vWF: Ag in patients with mixed connective tissue disease (MCTD), systemic lupus erythematosus, progressive systemic sclerosis, polymyositis/dermatomyositis or rheumatoid arthri

Raised serum hyaluronate levels in scleroderma: an effect of growth factor induced activation of connective tissue cells?

The circulating levels of hyaluronate were determined in 36 patients with scleroderma and in 36 control subjects matched for age and sex. The mean serum hyaluronate concentration in patients with progressive systemic sclerosis (n = 25) was 131 +/- 67 (SD) microgram/l and significantly greater (p less than 0.001) than that of the controls (mean level 49 +/- 21 (SD) microgram/l). Hyaluronate levels in patients with localised scleroderma (n = 4) were 141 +/- 47 (SEM) microgram/l and in patients with scleroderma-associated overlap syndromes (n = 7) 202 +/- 54 (SEM) microgram/l. Pathogenic Roles of Autoantibodies and Aberrant Epigenetic Regulation of Immune and Connective Tissue Cells in the Tissue Fibrosis of Patients with Systemic Sclerosis.

Systemic sclerosis (SSc) is a multi-system autoimmune disease with tissue fibrosis prominent in the skin and lung. In this review, we briefly describe the autoimmune features (mainly autoantibody production and cytokine profiles) and the potential pathogenic contributors including genetic/epigenetic predisposition, and environmental factors. We look in detail at the cellular and molecular bases underlying tissue-fibrosis which include trans-differentiation of fibroblasts (FBs) to myofibroblasts (MFBs). We also state comprehensively the pro-inflammatory and pro-fibrotic cytokines relevant to MF

Cell adhesion molecule 1 is upregulated in connective tissue mast cells and potentially contributes in IgE-mediated degranulation.

Mast cells are heterogeneous tissue-resident immune cells, with connective tissue mast cells (CTMCs) and mucosal mast cells in rodents exhibiting distinct phenotypes and activation profiles. Cell adhesion molecule 1 (CADM1), an immunoglobulin superfamily adhesion molecule, has been implicated in mast cell-nerve and mast cell-stromal interactions as well as in the pathogenesis of atopic dermatitis. Here, we investigated CADM1 function in CTMCs using a monoclonal antibody against the extracellular domain of CADM1, termed 3E1. CTMCs were differentiated from bone marrow-derived mast cells (BMMCs)

Proliferating bile duct epithelial cells are a major source of connective tissue growth factor in rat biliary fibrosis.

Connective tissue growth factor (CTGF) is a downstream mediator of transforming growth factor-beta1 (TGF-beta1) and thus a potential target for antifibrotic treatment strategies. CTGF is up-regulated in disorders such as atherosclerosis, scleroderma, and fibrosis of kidneys and lungs. We investigated the temporospatial expression patterns of CTGF and TGF-beta1 mRNA in rat livers with acute fibrogenesis (after a single dose of CCl(4)) and with advanced fibrosis (6 weeks after complete bile duct occlusion). Multiprobe ribonuclease protection assay revealed increasing TGF-beta1 and CTGF mRNA leve

Musculoskeletal connective tissue cells with muscle: expression of muscle actin in and contraction of fibroblasts, chondrocytes, and osteoblasts.

The expression of the gene for a muscle actin in certain nonmuscle cells and the contraction of these connective tissue cells has been associated with several important physiological and pathological processes; the contraction of healing skin wounds and the contracture in Dupuytren's disease being two notable examples. Studies in recent years have shown that a much wider variety of connective tissue cells than previously considered, including cells in many of the musculoskeletal tissues, e.g., chondrocytes and osteoblasts, can express the gene for alpha-smooth muscle actin and can display cont
