The role of previous exposure in the appetitive and consummatory effects of orexigenic neuropeptides.

The ingestion of foods is comprised of two distinct phases of behavior: appetitive and consummatory. While most food intake paradigms include both phases, the intraoral intake test emphasizes the stereotyped consummatory-phase by infusing a liquid food directly into the oral cavity. Several hypothalamic peptides have been shown to increase intake of chow in standard food intake paradigms and the current experiments sought to test whether these peptides would increase food intake in the intraoral intake paradigm. NPY, melanin-concentrating hormone (MCH) and orexin-A were infused into the third

[Hunger and satiety factors in the regulation of pleasure associated with feeding behavior].

Feeding is an instinctive behavior accompanied by rewarding feeling of pleasure during obtaining and ingesting food, corresponding to the preparatory and consummatory phases of motivated behavior, respectively. Perception of this emotional state together with alternating feelings of hunger and satiety drives the feeding behavior. Because alterations of feeding behavior including either overeating or anorexia may lead to obesity and cachexia, respectively, understanding the neurochemical mechanisms of regulation of feeding pleasure may help to develop new therapies of these diseases. The dopami

Development of an ethanol model using social insects: II. Effect of Antabuse on consumatory responses and learned behavior of the honey bee (Apis mellifera L.).

The ability of Antabuse (disulfiram) to influence ethanol consumption and learning in harnessed honey bees was investigated. In the first series of experiments a factorial design was used with 5 levels of ethanol concentration (0%, 1%, 5%, 10%, 20%), 4 doses of Antabuse (0, 37 microg/g, 3.7 microg/g, .37 microg/g), and 2 testing intervals (1 min., 10 min.). Animals were fed a single 1 microl dose of Antabuse and contact time with an ethanol solution measured. A second series of experiments investigated the influence of Antabuse on the formation of Pavlovian conditioning of the proboscis extens

Parental behavior in the mouse: effects of lesions in the entorhinal/piriform cortex.

The effects of bilateral radiofrequency lesions in the region of the entorhinal and piriform cortex (ENT/PIR cortex) on pup-retrieving and ultrasound-elicited pup-searching behavior were investigated in ovariectomized estrogen-treated female mice, which were sensitized to pups by co-caring for a litter for 5 days together with the mother (FoEBex), as well as in primiparous lactating females (Flact). A further group of FoEBex were rendered anosmic by an intranasal treatment with zinc sulfate-solution before the sensitization period and tests for parental motivation. Both pup-retrieving and pup-

The contributions of taste cues to ethanol responses in female and male rats.

Bitter-tasting stimuli, like ethanol (EtOH), are innately avoided yet variability in orosensory-driven behaviors may contribute to differences in ethanol intake. Experiment 1 was designed to test the hypothesis that there are sex differences in the taste qualities of ethanol. Female and male rats were presented 5 or 10 % EtOH followed by administration (i.p.) of LiCl to induce visceral malaise, or saline as a control. Generalization of the conditioned avoidance was assessed in a brief-access taste test (10-s trials; 30-min session) in which water, 0.03 and 0.3 M sucrose ("sweet"), 0.03 and 0.3

Infusions of bicuculline to the ventral tegmental area attenuates sexual, exploratory, and anti-anxiety behavior of proestrous rats.

Actions of 5alpha-pregnan-3alpha-ol-20-one (3alpha,5alpha-THP), in the midbrain ventral tegmental area (VTA) modulate sexual receptivity of female rats. Actions of 3alpha,5alpha-THP at GABAergic substrates in the VTA are known to modulate consummatory aspects of sexual behavior among rodents, such as lordosis. However, the extent to which GABA(A) receptors in the VTA are important for appetitive (exploratory, anti-anxiety, social) aspects of sexual receptivity is not well-understood. Proestrous rats were bilaterally-infused with saline or bicuculline (100 ng), a GABA(A) receptor antagonist, to

Bremelanotide: an overview of preclinical CNS effects on female sexual function.

INTRODUCTION: Bremelanotide is an analogue of the naturally occurring peptide alpha-melanocyte-stimulating hormone (alpha-MSH). It stimulates erection in men and male rats, and is currently in clinical trials for the treatment of erectile dysfunction. AIM: To review the effects of bremelanotide, an analogue of the naturally occurring peptide alpha-MSH, on the preclinical indices of sexual desire in female rats, and where in the brain these actions may occur. MAIN OUTCOME MEASURES: Appetitive sexual behaviors, such as solicitations, hops and darts, and pacing, were assessed along with consumm

The effects of chronic administration of testosterone propionate with or without estradiol on the sexual behavior and plasma steroid levels of aged female rats.

Low sexual desire concomitant with feelings of distress is reported in naturally and surgically menopausal women. A combination of estradiol (E2) and testosterone (T) restores sexual desire and interest in these women. The central mechanisms by which E2 and T act to restore desire are poorly understood. Here we examined the effect of chronic treatment with testosterone propionate (TP) administered by a sc SILASTIC brand capsule in aged ovary-intact female rats. Females were first treated with TP alone, followed by a second phase when TP was administered in combination with estradiol benzoate (

Dissociation of consummatory and vocal components of feeding in squirrel monkeys treated with benzodiazepines and alcohol.

The primary aim of the current experiments was to develop methods that engender vocalizations associated with positive social situations comprising affiliative behavior and feeding that could be quantified under controlled laboratory conditions and were sensitive to anxiolytic drugs. Classical conditioning procedures were used to elicit vocalizations during presentation of stimulus lights (i.e., CS condition) previously paired with either preferred foods (e.g., grapes, peanuts, bananas) or standard foods (e.g., monkey chow) as well as during presentation of both food types (i.e., UCS condition

Ontogeny of thirst in the infant rat.

The development of water drinking in response to thirst stimuli was found to be precocious and sequential in the neonatal rat. It was studied in suckling rats with a technique that took advantage of their vigorous consummatory reflexes and minimized appetitive responding. Responsiveness to the following thirst challenges appeared in the suckling rat with a clear timetable: cellular dehydration at 2 days of age, hypovolemia induced by colloid dialysis at 4 days, and beta-adrenergic activation at 6 days. Responsiveness to deprivation from mother's milk was vigorous at the earliest day tested (2

Adrenoreceptor modulation of oromotor pathways in the rat medulla.

Regulation of feeding behavior involves the integration of multiple physiological and neurological pathways that control both nutrient-seeking and consummatory behaviors. The consummatory phase of ingestion includes stereotyped oromotor movements of the tongue and jaw that are controlled through brain stem pathways. These pathways encompass not only cranial nerve sensory and motor nuclei for processing feeding-related afferent signals and supplying the oromotor musculature but also reticular neurons for orchestrating ingestion and coordinating it with other behaviors that utilize the same musc

Delineating anticipatory arousal from reward consumption: evaluating fixed-intervals in cocaine seeking-taking response chains.

RATIONALE: Anticipation is a critical antecedent to drug use, in which the prospect of imminent drug availability can potently motivate instrumental actions directed to procure it. Models that capture the behavioral dynamics that precede drug access may allow for the dissociation of key neural mechanisms underlying appetitive or consummatory processes in drug self-administration. OBJECTIVES: We aimed to isolate measurements attributed to the procurement and consumption of a reward by defining distinct actions for each using a chain-schedule of reinforcement. METHODS: Male Long-Evans rats wer

Animal models of anxiety based on classical conditioning: the conditioned emotional response (CER) and the fear-potentiated startle effect.

Stimuli consistently paired with shock become capable of suppressing ongoing operant or consummatory behavior (the conditioned emotional response--CER) or elevating the amplitude of the startle reflex (fear-potentiated startle). These changes are used to infer a central state of fear which involves the central nucleus of the amygdala and its efferent projections to the brainstem. The present paper reviews how psychoactive drugs affect these measures. Both the CER and fear-potentiated startle are reduced by benzodiazepines, barbiturates and opiates. Advantages and disadvantages of these animal

RFamide-related Peptide-3 and the Trade-off between Reproductive and Ingestive Behavior.

Ingestive and sex behaviors are important for individual survival and reproductive success, but when environmental energy availability is limited, individuals of many different species make a trade-off, forfeiting sex for ingestive behavior. For example, food-deprived female Syrian hamsters (Mesocricetus auratus) forego vaginal scent marking and lordosis (sex behaviors) in favor of foraging, hoarding, and eating food (ingestive behavior). Reproductive processes tend to be energetically costly, and individual survival requires homeostasis in metabolic energy. Thus, during energetic challenges,

Dopamine and preparatory behavior: III. Effects of metoclopramide and thioridazine.

The effects of metoclopramide and thioridazine on feeding behaviors of male hooded rats were investigated in 2 experiments. Metoclopramide (2.5-7.5 mg/kg) attenuated conditioned preparatory responses to a conditional stimulus signaling delivery of a meal. However, consummatory responses were affected only by the highest dose. Similar effects had previously been observed following administration of pimozide (Blackburn, Phillips, & Fibiger, 1987). Thioridazine (10-30 mg/kg) had no significant effect on any measure of feeding behavior. The different effects of the 2 drugs may be related to their

Amphetamine stereotypy, the basal ganglia, and the "selection problem".

Amphetamine and other stimulant drugs induce stereotyped head movements in rats, which interfere with normal goal-directed behavior. However, rats given access to food while intoxicated learn to suppress these movements in order to feed. This suggests that the suppression of stereotypy is an instrumentally learned response reinforced by the ingestion of food. Consistent with this interpretation, rats learn to suppress stereotyped head movements when intraoral infusions of milk are made contingent on maintaining a stationary head position, but not when such infusions are given noncontingently.

Effect of chlorpromazine and haloperidol on negative contrast.

Rats shifted from 32 to 4% sucrose consume substantially less of the 4% solution than animals that have not had prior experience with the 32% sucrose. This negative contrast effect was not substantially influenced by chlorpromazine (1, 3, and 5 mg/kg) or haloperidol (0.1, 0.5, and 1.0 mg/kg). Haloperidol decreased overall lick frequency, but this decrease occurred proportionately in shifted and unshifted rats, leaving contrast intact. The benzodiazepine flurazepam (5, 10, and 20 mg/kg), included as a positive control, reduced contrast at the two highest doses. The results suggest that neurolep

Sex- and reward-dependent effects of early life scarcity-adversity on adolescent behavioral responses to natural rewards.

Early life adversity (ELA) increases risk for multiple psychiatric disorders that are characterized by reward-related dysfunction. Disrupted reward-related processes are commonly observed in humans and rodents following ELA. Rodent studies have shown sex differences in response to natural and drug rewards at baseline, following ELA, and in rodent models of psychiatric diseases that are potentiated by ELA. Yet, less is known regarding the development of ELA-induced alterations in reward-related responses, including how these may differ by sex. To this end, we tested behavioral responses to cons

Behavioral Effects of a Novel Benzofuranyl-Piperazine Serotonin-2C Receptor Agonist Suggest a Potential Therapeutic Application in the Treatment of Obsessive-Compulsive Disorder.

Selective serotonin reuptake inhibitors (SSRIs) are the only effective pharmacological treatments for obsessive-compulsive disorder (OCD). Nonetheless, their generally limited efficacy, side-effects, and delayed onset of action require improved medications for this highly prevalent disorder. Preclinical and clinical findings have suggested serotonin2C (5-HT2C) receptors as a potential drug target. Data in rats and mice are presented here on the effects of a novel 5-HT2C receptor agonist ((3S)-3-Methyl-1-[4-(trifluoromethyl)-7-benzofuranyl]-piperazine) (CPD 1) with high potency and full efficac

Exendin-4 decreases amphetamine-induced locomotor activity.

Glucagon-like peptide-1 (GLP-1) is released in response to nutrient ingestion and is a regulator of energy metabolism and consummatory behaviors through both peripheral and central mechanisms. The GLP-1 receptor (GLP-1R) is widely distributed in the central nervous system, however little is known about how GLP-1Rs regulate ambulatory behavior. The abused psychostimulant amphetamine (AMPH) promotes behavioral locomotor activity primarily by inducing the release of the neurotransmitter dopamine. Here, we identify the GLP-1R agonist exendin-4 (Ex-4) as a modulator of behavioral activation by AMPH
