Al Alloy Melting Behavior and Interfacial Reactions with Steel Under Natural Convection.

The Al alloy melting behavior and interfacial reactions during the steelmaking process of high-Al automotive steel were investigated in this study. The total dissolution time of Al bars (20 × 20 × 80 mm) in molten steel was quite short, decreasing from 21.4 s to 10.0 s with an increase in bath temperature from 1580 to 1620 °C. The Al alloy melting process at the molten steel temperature of 1600 °C included the formation of a solidified steel layer, the latter's rapid melting, and Al alloy normal melting, while at 1600 °C, the process included a second increase in the thickness of the solidifie

Intraparenchymal convection enhanced delivery of AAV in sheep to treat Mucopolysaccharidosis IIIC.

BACKGROUND: Mucopolysaccharidosis IIIC (MPSIIIC) is one of four Sanfilippo diseases sharing clinical symptoms of severe cognitive decline and shortened lifespan. The missing enzyme, heparan sulfate acetyl-CoA: α-glucosaminide-N-acetyltransferase (HGSNAT), is bound to the lysosomal membrane, therefore cannot cross the blood-brain barrier or diffuse between cells. We previously demonstrated disease correction in MPSIIIC mice using an Adeno-Associated Vector (AAV) delivering HGSNAT via intraparenchymal brain injections using an AAV2 derived AAV-truetype (AAV-TT) serotype with improved distributio

Clinical tolerance of corticospinal tracts in convection-enhanced delivery to the brainstem.

OBJECTIVE: Convection-enhanced delivery (CED) has been explored as a therapeutic strategy for diffuse intrinsic pontine glioma (DIPG). Variables that may affect tolerance include infusate volume, infusion rate, catheter trajectory, and target position. Supratentorial approaches for catheter placement and infusate distribution patterns may conflict with corticospinal tracts (CSTs). The clinical relevance of these anatomical constraints has not been described. The authors report their experience using CED in the brainstem as it relates to anatomical CST conflict and association with clinical tol

Divergent urban storm response to convective, frontal and tropical systems.

Urbanization modifies precipitation1,2, yet previous studies have reported inconsistent results, with some cities experiencing rainfall enhancement and others showing suppression3. To reconcile these discrepancies, we examine how urban impacts vary across storm types using an event-based analysis. With three-dimensional radar reflectivity data (1995-2017), we identify more than 40,000 warm-season storms across four Texas cities (Dallas, Austin, San Antonio and Houston). Here we show that classifying storms into five types reveals distinct urban influences linked to storm scales and dynamics. L

Influence of sedimentation on the threshold for Soret-driven convection in colloidal suspensions.

The onset of Soret-driven convection in a horizontal layer of a colloidal suspension is investigated by considering a particulate medium model. We consider a dilute suspension of spherical solid particles being subjected to convection in a Rayleigh-Bénard geometry setup. The mathematical model takes into account the effects of thermophoresis, particle sedimentation, and Brownian diffusion. The equations governing the convective motion consist of the momentum equation which includes an extra body force term to account for the thermophoretic force effect, the conservation of particles equation w

Effects of gas bubbling and other forms of convection on platelets in vitro.

Citrated platelet-rich human plasma was subjected to one of three experimental treatments at 37 degrees C for 15 min: stirring, bubbling (with stirring), and gentle agitation achieved by a rocking motion. The last two were "equiconvective" as judged by equilibration rates with CO2 and O2 but presumably differed in the shear stress they imposed on the cells. Stirring platelets in normal air or 5% CO2-air caused no significant aggregation. Bubbling air through platelet-rich plasma increased its pH and marked aggregation occurred. Bubbling CO2-air caused the platelet-rich plasma pH to attain its

Convective mixing mechanisms in high frequency intermittent jet ventilation.

A liquid flow visualization technique was used to identify the location of neutrally buoyant bead clouds injected into airway models during flows simulating high frequency intermittent jet ventilation (HFIJV) in neonatal lungs. The motions of these bead clouds show that the convective or bulk mixing that occurs during HFIJV is made up of two parts; a turbulent convective exchange with the atmosphere caused by the jet in the trachea and a streaming motion along the airways driven by an interaction between the jet and the expansion and contraction of the airways due to their compliance. These co

Global stability and uniform persistence of the reaction-convection-diffusion cholera epidemic model.

We study the global stability issue of the reaction-convection-diffusion cholera epidemic PDE model and show that the basic reproduction number serves as a threshold parameter that predicts whether cholera will persist or become globally extinct. Specifically, when the basic reproduction number is beneath one, we show that the disease-free-equilibrium is globally attractive. On the other hand, when the basic reproduction number exceeds one, if the infectious hosts or the concentration of bacteria in the contaminated water are not initially identically zero, we prove the uniform persistence res

Entry points for catheter placement in convection-enhanced delivery--a retrospective anatomic analysis.

INTRODUCTION: Planning the trajectories for catheter positioning in convection-enhanced delivery (CED) is delicate. The bulk flow follows the path of least resistance. Additional factors such as the varying tumor location and the functional relevance of the surrounding brain areas add to complicate the trajectory planning even further. Therefore several criteria have been established, dealing with the correct general entry and target point position. The goal of our study was to analyze the trajectories for CED catheters placed in our department to find preferable entry point positions. METHOD

Effect of Convection Rolls in Motility-Induced Phase Separation of Active Janus Particles.

We numerically study motility-induced phase separation of active particles in two-dimensional convection rolls. We analyze local packing-fraction distributions, density fluctuations, the corresponding phase diagrams, and diffusivity curves to characterize the interplay between self-propulsion, global packing fraction, and advection strength. In the weak-flow regime, the system exhibits phase separation characterized by bimodal density distributions, slowly decaying density fluctuations, and a sharp reduction in diffusivity. Increasing advection suppresses clustering by enhancing particle trans

Convection-Enhanced Delivery of Auristatin-Conjugated Layer-by-Layer Nanoparticles for Glioblastoma Treatment.

Glioblastoma (GBM) has limited treatment options, as the restrictive blood-brain barrier (BBB) prevents most therapeutics from accumulating at sufficient levels in the brain. Convection-enhanced delivery (CED) offers a method for administering therapeutics directly into brain tumor tissue, but free drugs can be cleared rapidly and may be toxic to off-target cells. Drug-loaded nanoparticles (NPs) are a promising platform to prolong the residence time and improve cellular targeting of therapeutics. We designed drug-conjugated NPs comprising a liposomal core modified with a layer-by-layer (LbL) p

Direct Characterization of Free Solutal Convection in Porous Rocks for CO2 Storage Applications.

Free solutal convection refers to the mixing process induced and sustained by local density differences arising from solute dissolution. This process underpins the long-term storage of carbon dioxide (CO2) following its injection and dissolution in the formation brine of subsurface rock formations, such as saline aquifers. Direct experimental evidence of free solutal convection in porous rocks is to-date still lacking, leaving large uncertainties on the realized rate of CO2 dissolution and its contribution toward storage. Using an analogue solute-solvent pair and 4D X-ray computed tomography,

Convection-Enhanced Delivery of Muscimol Into the Bilateral Subthalamic Nuclei of Nonhuman Primates.

BACKGROUND: Muscimol is a gamma-aminobutyric acid receptor agonist that selectively and temporarily inhibits neurons. Local bolus injection of muscimol has been used experimentally to inhibit neuronal populations within discrete anatomical structures and discern their physiological function. OBJECTIVE: To determine the safety and behavioral effects of convection-enhanced delivery of muscimol into the bilateral subthalamic nuclei (STN) of nonhuman primate rhesus macaques (NHPs). METHODS: Six awake NHPs underwent co-infusion of gadolinium-diethylenetriamine pentaacetic acid (Gd-DTPA), a surrog

The distribution, clearance, and brainstem toxicity of panobinostat administered by convection-enhanced delivery.

OBJECTIVE The pan-histone deacetylase inhibitor panobinostat has preclinical efficacy against diffuse intrinsic pontine glioma (DIPG), and the oral formulation has entered a Phase I clinical trial. However, panobinostat does not cross the blood-brain barrier in humans. Convection-enhanced delivery (CED) is a novel neurosurgical drug delivery technique that bypasses the blood-brain barrier and is of considerable clinical interest in the treatment of DIPG. METHODS The authors investigated the toxicity, distribution, and clearance of a water-soluble formulation of panobinostat (MTX110) in a small

Time-reversal acoustics and ultrasound-assisted convection-enhanced drug delivery to the brain.

Time-reversal acoustics is an effective way of focusing ultrasound deep inside heterogeneous media such as biological tissues. Convection-enhanced delivery is a method of delivering drugs into the brain by infusing them directly into the brain interstitium. These two technologies are combined in a focusing system that uses a "smart needle" to simultaneously infuse fluid into the brain and provide the necessary feedback for focusing ultrasound using time-reversal acoustics. The effects of time-reversal acoustics-focused ultrasound on the spatial distribution of infused low- and high-molecular w

Safety of intraparenchymal convection-enhanced delivery of cintredekin besudotox in early-phase studies.

OBJECT: Convection-enhanced delivery (CED) is an increasingly used novel local/regional delivery method targeted directly to tissue. It relies on a continuous pressure gradient for distribution of therapeutic agents into the interstitial space, with administration of the infusate over a few days. Cintredekin besudotox (also known as IL13- PE38QQR) is a recombinant chimeric cytotoxin consisting of interleukin-13 and a truncated exotoxin produced by the Pseudomonas aeruginosa bacterium, which targets malignant glioma cells. METHODS: Cintredekin besudotox was administered via intraparenchymal CE

Mechanical alterations of electrospun poly(ϵ-caprolactone) in response to convective thermobonding.

Vascular graft failure has persisted as a major clinical problem. Mechanical, structural, and transport properties of vascular grafts are critical factors that substantially affect their function and thus the outcome of implantation. The manufacturing method, post-processing technique, and material of choice have a significant impact on these properties. The goal of this work is to use thermal treatment to modulate the transport properties of PCL-based vascular engineered constructs. To this end, we electrospun PCL tubular constructs and thermally bonded the electrospun fibers in a convective

Image-guided, direct convective delivery of glucocerebrosidase for neuronopathic Gaucher disease.

OBJECTIVE: To determine if convection-enhanced delivery (CED) of glucocerebrosidase could be used to treat targeted sites of disease progression in the brain and brainstem of a patient with neuronopathic Gaucher disease while monitoring enzyme distribution using MRI. METHODS: A CED paradigm in rodents (n = 8) and primates (n = 5) that employs co-infusion of a surrogate MRI tracer (gadolinium diethylenetriamine penta-acetic acid [Gd-DTPA]) with glucocerebrosidase to permit real-time monitoring of distribution was developed. The safety and feasibility of this delivery and monitoring paradigm we

Estimation of sieving coefficients of convective absorption of drugs in perfused rat jejunum.

Intestinal absorption of many hydrophilic drugs cannot be explained solely in terms of pH-partition and solvent-drag effects have been described in a number of cases. However, quantitative estimates of sieving coefficient (phi) for drug molecules have tended to be variable. In the present work an in situ perfused intestinal loop preparation in the rat has been used to measure the disappearance of five hydrophilic drugs from the intestinal lumen and a mathematical model of drug absorption in the presence of net and unidirectional fluid fluxes has been developed. The model allows separate estima
