Particulate matter contamination in the corneal stroma of severe eye burns in humans.

Corneal buttons obtained from keratoplasty were examined by energy dispersive x-ray analysis (EDXA) combined with scanning electron microscopy (SEM). This method enables to assay the mineral composition of minute parts of tissue samples identified in SEM images. Samples were cut from paraffin embedded corneae, deparaffinized in xylol, dried in aceton, critical-point desiccated, covered by evaporating with a thin layer of carbon and examined by SEM. In healthy human donor eyes, only some iron particles had been found. In the 22 patients samples high amounts of different particles were identifie

Latent-Transforming growth factor - β Binding protein 1 (LTBP1) in corneal stroma.

We aim to determine the presence and function of LTBP1 in the corneal stroma. We investigated the temporal and spatial corneal expression of LTBP1, and microfibril-associated glycoprotein 1 (MAGP1), known components of the elastic system and regulators of TGF-β storage and latency. Protein quantification -blotting and proteomics-, immunofluorescence microscopy, mRNA analysis and a luciferase assay were used to study the regulation of transforming growth factor β (TGF-β) by LTBP1 in cornea derived fibroblasts and myofibroblasts. Expression of LTBP1 and MAGP1 was found in adult corneas, and abse

Rejection of Acellular Porcine Corneal Stroma Transplantation During Coronavirus Disease 2019 Pandemic.

To report 2 successfully managed cases of graft rejection with acellular porcine corneal stroma (APCS) transplantation in patients with fungal corneal ulcer. Two patients were diagnosed with fungal corneal ulcer and received APCS transplantation. Graft rejection developed due to the lost follow-up during the period of coronavirus disease 2019 outbreak. Amniotic membranes transplantation and cauterization of neovascularization was performed, respectively. The graft failure resolved successfully after the procedure. To the best of our knowledge, amniotic membranes transplantation and cauterizati

EDC/NHS cross-linked collagen foams as scaffolds for artificial corneal stroma.

In this study, a highly porous collagen-based biodegradable scaffold was developed as an alternative to synthetic, non-degradable corneal implants. The developed method involved lyophilization and subsequent stabilization through N-ethyl-N'-[3-dimethylaminopropyl] carbodiimide/N-hydroxy succinimide (EDC/NHS) cross-linking to yield longer lasting, porous scaffolds with a thickness similar to that of native cornea (500 microm). For collagen-based scaffolds, cross-linking is essential; however, it has direct effects on physical characteristics crucial for optimum cell behavior. Hence, the effect

Corneal Opacification and Spontaneous Recovery following Injection of Healon5 into the Corneal Stroma during Intervention for Postoperative Hypotony.

We report the self-limited nature of corneal opacification after accidental injection of Healon5 into the corneal stroma. A 52-year-old male with a new diagnosis of severe stage, primary open-angle glaucoma underwent successful trabeculectomy OS, which was complicated by ocular hypotony and shallow anterior chamber (AC) on postoperative day 1. Healon5, a hyaluronic acid-containing viscoelastic device, was accidentally introduced into the corneal stroma during attempted injection into the AC. The cornea hydrodissected and opacified, leading to precipitous loss of best-corrected visual acuity (B

Impaired healing in an incision wound in corneal stroma in a lumican-null mouse.

PURPOSE: We investigated healing pattern of an incisional wound in corneal stroma of lumican-null (KO) mice. METHODS: C57BL/6 mice (wild-type, WT) and lumican-null (knockout, KO) mice were used. A linear full-thickness incision was produced in one cornea of each mouse. After intervals of healing, the corneas were processed for the following analyses. Histology was employed to measure the distance between each edge of the disrupted Descemet's membrane at the center of the cornea. Immunohistochemistry and real-time RT-PCR were employed to evaluate the expression of wound healing-related compone

rad21 Is Involved in Corneal Stroma Development by Regulating Neural Crest Migration.

Previously, we identified RAD21R450C from a peripheral sclerocornea pedigree. Injection of this rad21 variant mRNA into Xenopus laevis embryos disrupted the organization of corneal stroma fibrils. To understand the mechanisms of RAD21-mediated corneal stroma defects, gene expression and chromosome conformation analysis were performed using cells from family members affected by peripheral sclerocornea. Both gene expression and chromosome conformation of cell adhesion genes were affected in cells carrying the heterozygous rad21 variant. Since cell migration is essential in early embryonic develo

Amyloidosis of corneal stroma in patients with trachoma. A clinicopathologic study of 62 cases.

Sixty-two cases of corneal amyloidosis were studied. The median age of the 48 men and 14 women was 66 years. The patients had bilateral, diffuse corneal opacity extending to the limbus and severe visual impairment. The corneal condition was not familial. Advanced stages of trachoma were present in all individuals. Climatic droplet keratopathy (CDK) was also observed clinically in 19 patients. Histopathologically, the Congo red-positive, birefringent and dichroic amyloid deposits in the stroma were similar to that of lattice corneal dystrophy. Bowman's membrane was mostly absent, and peculiar m

Sphere-forming corneal cells repopulate dystrophic keratoconic stroma: Implications for potential therapy.

BACKGROUND: Keratoconus is a degenerative corneal disease characterised by aberrant cell behaviour and loss of matrix that can result in vision loss. Cells extracted from peripheral corneas can form stem cell-enriched spheres, which have shown the potential to repopulate the normal peripheral corneal stroma in vitro upon sphere implantation but have not been previously studied in keratoconic tissue. AIM: To investigate the therapeutic potential of stem cell-enriched spheres formed from extracted peripheral human corneal cells when introduced to keratoconic tissue. METHODS: Stem cell-enriched

Efficient lentiviral gene transfer into corneal stroma cells using a femtosecond laser.

We investigated a new procedure for gene transfer into the stroma of pig cornea for the delivery of therapeutic factors. A delimited space was created at 110 mum depth with a LDV femtosecond laser in pig corneas, and a HIV1-derived lentiviral vector expressing green fluorescent protein (GFP) (LV-CMV-GFP) was injected into the pocket. Corneas were subsequently dissected and kept in culture as explants. After 5 days, histological analysis of the explants revealed that the corneal pockets had closed and that the gene transfer procedure was efficient over the whole pocket area. Almost all the kera

Thiol-functionalized collagen patch channelizes TGFβ1- JNK trafficking and reverses corneal stroma opacity.

Pro-regenerative patches are appealing for cornea injuries owing to their robustness, and ease of use in clinical settings. Collagen-based synthetic patches are building blocks for corneal repair, but their cross-linker toxicity, less optical clarity, and deformation instability remain unresolved. In response to these challenges, the study introduces disulfide crosslinked thiolated collagen patches (CSH). Human umbilical cord collagen (UC) comprises abundant lysine; thus, more primary amines are available for thiolation. UC was thiol functionalized, whereby the degree of amine substitution is

An Incidentally Detected Synthetic Fiber Embedded in the Corneal Stroma: A Report of a Rare Case.

Corneal foreign bodies (CFBs) are typically caused by the penetration of external materials such as metallic particles, concrete fragments, or plant matter into the corneal tissue and are usually accompanied by intense pain or a foreign-body sensation. We report an extremely rare case of an intrastromal CFB composed of synthetic fiber, which was discovered incidentally in an asymptomatic patient and confirmed histopathologically. A 54-year-old woman presented with bilateral visual impairment due to cataract. Slit-lamp examination revealed bilateral cataracts and a curved, white, linear foreign

Development, structure, and bioengineering of the human corneal stroma: A review of collagen-based implants.

Bio-engineering technologies are currently used to produce biomimetic artificial corneas that should present structural, chemical, optical, and biomechanical properties close to the native tissue. These properties are mainly supported by the corneal stroma which accounts for 90% of corneal thickness and is mainly made of collagen type I. The stromal collagen fibrils are arranged in lamellae that have a plywood-like organization. The fibril diameter is between 25 and 35 nm and the interfibrillar space about 57 nm. The number of lamellae in the central stroma is estimated to be 300. In the anter

[Consequences of different ablation rates in Bowman's membrane and corneal stroma for photorefractive keratectomy].

If photorefractive keratectomy (PRK) is performed with an ArF-excimer-laser, the existence of different ablation rates in Bowman's membrane and corneal stroma is of crucial importance, as ablation rates are considered to be constant in PRK. Consequently, if photoablation cannot be confined to Bowman's layer alone, at least two new surfaces with different refractive powers would result. For the correction of myopia with PRK, different ablation rates would mean that: (1) the central corneal surface with the desired refractive power is smaller than the total ablation zone. The lower the ablation

Proteomic profiling of human corneal stroma reveals molecular signatures associated with myopia progression.

Myopia progression entails complex molecular changes in the ocular tissues, yet the stage-specific proteomic alterations in the human corneal stroma across different myopia stages remain largely uncharacterized. This study aimed to investigate proteomic profiles in corneal stroma samples obtained from patients undergoing small incision lenticule extraction (SMILE), including 19 low-myopia, 19 moderate-myopia, and 20 high-myopia cases. Using four-dimensional data-independent acquisition mass spec-trometry (4D-DIA-MS), we identified 3493 proteins, with 3248 common to all groups. Pairwise compari

Short-term results of acellular porcine corneal stroma keratoplasty for herpes simplex keratitis.

BACKGROUND: Corneal transplantation is a common surgical intervention for restoring vision loss due to corneal damages. However, for cultural reasons, there is a huge shortage of donor corneas in China. Acellular porcine corneal stromas (APCSs) can be used as corneal substitutes in lamellar keratoplasty for corneal ulcers. This study was conducted to analyze the results of APCS use for herpes simplex keratitis (HSK). METHODS: The study involved HSK patients who underwent keratoplasty with APCSs from February 2016 to October 2017 in the second affiliated hospital of Zhejiang University. Patien
