Enhanced heparosan biosynthesis in Corynebacterium glutamicum by systematic metabolic engineering and translation-level fine-tuning.

As a widely used anticoagulant, heparin is industrially produced chiefly via animal tissue extraction, which suffers from unstable supply and potential safety hazards. Heparosan shares a similar polysaccharide backbone with heparin and can be converted into heparin under mild enzymatic catalysis. In addition, heparosan exhibits favorable biocompatibility and non-immunogenicity, rendering its efficient, eco-friendly biosynthesis essential. In this study, we systematically engineered Corynebacterium glutamicum, a Generally Recognized as Safe (GRAS) microorganism, to synthesize heparosan via two

Draft genome and physiological characterization of a newly isolated L-arabinose-utilizing Corynebacterium glutamicum CS176.

This study reports the physiological and genomic characterization of Corynebacterium glutamicum CS176, a newly isolated strain exhibiting a rare combination of traits: efficient L-arabinose utilization and L-glutamate production without chemical induction, even under excess biotin conditions. Genome analysis of the 3.10 Mb draft sequence (54.20% GC) revealed a 7.8 kb L-arabinose utilization gene cluster highly similar to those of arabinose-assimilating strains, suggesting acquisition via horizontal gene transfer. To link genotype with phenotype, the effects of temperature, oxygen availability,

Metabolic engineering of Corynebacterium glutamicum for efficient production of optically pure (2R,3R)-2,3-butanediol.

BACKGROUND: 2,3-butanediol is an important platform compound which has a wide range of applications, involving in medicine, chemical industry, food and other fields. Especially the optically pure (2R,3R)-2,3-butanediol can be employed as an antifreeze agent and as the precursor for producing chiral compounds. However, some (2R,3R)-2,3-butanediol overproducing strains are pathogenic such as Enterobacter cloacae and Klebsiella oxytoca. RESULTS: In this study, a (3R)-acetoin overproducing C. glutamicum strain, CGS9, was engineered to produce optically pure (2R,3R)-2,3-butanediol efficiently. Fir

Reactions upstream of glycerate-1,3-bisphosphate drive Corynebacterium glutamicum (D)-lactate productivity under oxygen deprivation.

We previously demonstrated the simplicity of oxygen-deprived Corynebacterium glutamicum to produce D-lactate, a primary building block of next-generation biodegradable plastics, at very high optical purity by introducing heterologous D-ldhA gene from Lactobacillus delbrueckii. Here, we independently evaluated the effects of overexpressing each of genes encoding the ten glycolytic enzymes on D-lactate production in C. glutamicum. We consequently show that while the reactions catalyzed by glucokinase (GLK), glyceraldehyde 3-phosphate dehydrogenase (GAPDH), phosphofructokinase (PFK), triosephosph

Multi-omics analyses of evolved Corynebacterium glutamicum mutants reveal the molecular responses to formaldehyde stress.

Formaldehyde serves as a crucial intermediate metabolite during C1 compounds biotransformation. To optimize C1 utilization efficiency, it is essential to identify genes related to formaldehyde tolerance and enhance microbial resistance. Hence, we developed an evolved Corynebacterium glutamicum strain, FM-3, capable of withstanding 2.6 mM formaldehyde-a significant improvement over the parental strain. Integrated transcriptomic and proteomic analyses revealed that the enhanced formaldehyde tolerance correlated with the upregulation of cell wall biosynthesis proteins and DNA repair machinery. Ge

Newly isolated Corynebacterium glutamicum strain PP80 for use in high-temperature L-glutamic acid fermentation.

High-temperature fermentation is a strategy to increase the fermentation rates and process stability while reducing cooling costs; however, metabolic heat generated during large-scale cultivations can impair cell growth and product formation. To address this bottleneck, we sought high-temperature L-glutamic acid fermentation processes using naturally thermotolerant Corynebacterium strains as alternatives to experimentally evolved strains. We isolated Corynebacterium glutamicum PP80 from soil in Thailand and evaluated its performance in penicillin-triggered L-glutamic acid fermentation at eleva

A genome-reduced Corynebacterium glutamicum derivative discloses a hidden pathway relevant for 1,2-propanediol production.

BACKGROUND: 1,2-propanediol (1,2-PDO) is widely used in the cosmetic, food, and drug industries with a worldwide consumption of over 1.5 million metric tons per year. Although efforts have been made to engineer microbial hosts such as Corynebacterium glutamicum to produce 1,2-PDO from renewable resources, the performance of such strains is still improvable to be competitive with existing petrochemical production routes. RESULTS: In this study, we enabled 1,2-PDO production in the genome-reduced strain C. glutamicum PC2 by introducing previously described modifications. The resulting strain sh

Evolutionary specialization of MscCG, an MscS-like mechanosensitive channel, in amino acid transport in Corynebacterium glutamicum.

MscCG, a mechanosensitive channel of Corynebacterium glutamicum provides a major export mechanism for glutamate in this Gram-positive bacterium, which has for many years been used for industrial production of glutamate and other amino acids. The functional characterization of MscCG is therefore, of great significance to understand its conductive properties for different amino acids. Here we report the first successful giant spheroplast preparation of C. glutamicum amenable to the patch clamp technique, which enabled us to investigate mechanosensitive channel activities of MscCG in the native m

Expansion of the substrate range of the gentisate 1,2-dioxygenase from Corynebacterium glutamicum for the conversion of monohydroxylated benzoates.

The gentisate 1,2-dioxygenases (GDOs) from Corynebacterium glutamicum and various other organisms oxidatively cleave the aromatic nucleus of gentisate (2,5-dihydroxybenzoate), but are not able to convert salicylate (2-hydroxybenzoate). In contrast, the α-proteobacterium Pseudaminobacter salicylatoxidans synthesises an enzyme ('salicylate dioxygenase', SDO) which cleaves gentisate, but also (substituted) salicylate(s). Sequence comparisons showed that the SDO belongs to a group of GDOs mainly originating from Gram-positive bacteria which also include the GDO from C. glutamicum ATCC 13032. The c

Requirement of de novo synthesis of the OdhI protein in penicillin-induced glutamate production by Corynebacterium glutamicum.

We found that penicillin-induced glutamate production by Corynebacterium glutamicum is inhibited when a de novo protein synthesis inhibitor, chloramphenicol, is added simultaneously with penicillin. When chloramphenicol was added 4 h after penicillin addition, glutamate production was essentially unaffected. (3)H-Leucine incorporation experiments revealed that protein synthesis continued for 1 h after penicillin addition and then gradually decreased. These results suggest that de novo protein synthesis within 4 h of penicillin treatment is required for the induction of glutamate production. To

Corynebacterium glutamicum alleviated colitis by downregulating the TNF signaling pathway mediated by cIAP1/2 in mice.

The gut microbiota and its associated micro-ecosystem are closely related to the onset and development of ulcerative colitis (UC). It is known that Corynebacterium glutamicum (C. glutamicum) helps rebuild gut eubiosis from diabetes dysbiosis; however, its effects on UC remain unknown. This study aims to investigate the therapeutic effects and mechanisms of C. glutamicum on UC. In this study, C. glutamicum was encapsulated with thiolated hyaluronic acid (HA-SH) to form a hydrogel, termed as CG-HA-SH. The adhesion and distribution of CG-HA-SH in the intestine were evaluated, along with its thera

Efficacy of supplemental amino acids with Corynebacterium glutamicum cell mass on growth and health of nursery pigs.

Removing filtration and crystallization provides amino acids with reduced purity because Corynebacterium glutamicum cell mass (CGCM) is not removed. This study aimed to evaluate the nutritional and functional values of granulated Threonine (Thr) and Valine (Val) with CGCM in diets for growth performance, jejunal mucosa-associated microbiota, nutrient digestibility, and health of nursery pigs. Seventy-two newly weaned pigs (28 d-of-age; initial body weight (BW) of 8.2 ± 0.4 kg) were allotted to 9 treatments (n = 8) based on a randomized complete block design with sex and initial BW as blocks. Development of a growth-coupled selection platform for directed evolution of heme biosynthetic enzymes in Corynebacterium glutamicum.

Heme is an important tetrapyrrole compound, and has been widely applied in food and medicine industries. Although microbial production of heme has been developed with metabolic engineering strategies during the past 20 years, the production levels are relatively low due to the multistep enzymatic processes and complicated regulatory mechanisms of microbes. Previous studies mainly adopted the strategies of strengthening precursor supply and product transportation to engineer microbes for improving heme biosynthesis. Few studies focused on the engineering and screening of efficient enzymes invol

Integrated Analysis of the Transcriptome and Metabolome of Corynebacterium glutamicum during Penicillin-Induced Glutamic Acid Production.

Corynebacterium glutamicum is known for its ability to produce glutamic acid and has been utilized for the fermentative production of various amino acids. Glutamic acid production in C. glutamicum is induced by penicillin. In this study, the transcriptome and metabolome of C. glutamicum is analyzed to understand the mechanism of penicillin-induced glutamic acid production. Transcriptomic analysis with DNA microarray revealed that expression of some glycolysis- and TCA cycle-related genes, which include those encoding the enzymes involved in conversion of glucose to 2-oxoglutaric acid, is upreg

L-Glutamine as a nitrogen source for Corynebacterium glutamicum: derepression of the AmtR regulon and implications for nitrogen sensing.

Corynebacterium glutamicum, a Gram-positive soil bacterium employed in the industrial production of various amino acids, is able to use a number of different nitrogen sources, such as ammonium, urea or creatinine. This study shows that l-glutamine serves as an excellent nitrogen source for C. glutamicum and allows similar growth rates in glucose minimal medium to those in ammonium. A transcriptome comparison revealed that the nitrogen starvation response was elicited when glutamine served as the sole nitrogen source, meaning that the target genes of the global nitrogen regulator AmtR were dere

A bioactive lysate of Corynebacterium glutamicum orchestrates intestinal barrier repair mainly through L-arginine-mediated FOXO pathway.

Intestinal barrier dysfunction is a core pathological feature of inflammatory bowel disease (IBD). Postbiotics have emerged as promising alternatives to probiotics due to their superior stability and safety. C. glutamicum is an industrial microorganism being Generally Recognized as Safe (GRAS); however, the efficacy and mechanism of its lysate in repairing the intestinal barrier remain largely unexplored. In this study, a dextran sulfate sodium (DSS)-induced colitis mouse model and a lipopolysaccharide (LPS)-induced Caco-2 cell inflammatory barrier model were employed to evaluate the barrier-r

Two-Component MprAB System Regulates the Expression of Genes Involved in Cell Envelope Biosynthesis in Corynebacterium glutamicum.

To accurately detect internal and environmental cues, bacteria have evolved signal transduction pathways such as two-component systems (TCSs) to reprogram appropriate genetic and physiological functions for adaptation and survival. The MprAB TCS is commonly found in actinobacteria and has been associated with important processes such as mycobacterial virulence, nutrient starvation, and environmental stress, particularly cell envelope stress. However, a comprehensive investigation of the function and response network of the MprAB TCS in corynebacteria remains to be carried out. In this study, w

From Aquaculture to Aquaculture: Production of the Fish Feed Additive Astaxanthin by Corynebacterium glutamicum Using Aquaculture Sidestream.

Circular economy holds great potential to minimize the use of finite resources, and reduce waste formation by the creation of closed-loop systems. This also pertains to the utilization of sidestreams in large-scale biotechnological processes. A flexible feedstock concept has been established for the industrially relevant Corynebacterium glutamicum, which naturally synthesizes the yellow C50 carotenoid decaprenoxanthin. In this study, we aimed to use a preprocessed aquaculture sidestream for production of carotenoids, including the fish feed ingredient astaxanthin by C. glutamicum. The addition

Heterologous expression of hydroxylase coupled with pathway optimization enables highly efficient ʟ-DOPA production in Corynebacterium glutamicum.

ʟ-3,4-dihydroxyphenylalanine (ʟ-DOPA), the direct precursor of the neurotransmitter dopamine, is a cornerstone therapeutic agent for Parkinson's disease. In this study, Corynebacterium glutamicum was used as a microbial chassis to systematically evaluate its potential as a cell factory for ʟ-DOPA production. The strain was initially evaluated for ʟ-DOPA tolerance to confirm its suitability for production. Key hydroxylases, HpaB and HpaC, were introduced, and promoter and ribosome-binding site (RBS) optimization was applied to establish a de novo ʟ-DOPA biosynthetic pathway. To minimize flux to
