SipD and IpaD induce a cross-protection against Shigella and Salmonella infections.

Salmonella and Shigella species are food- and water-borne pathogens that are responsible for enteric infections in both humans and animals and are still the major cause of morbidity and mortality in the emerging countries. The existence of multiple Salmonella and Shigella serotypes as well as the emergence of strains resistant to antibiotics require the development of broadly protective therapies. Those bacteria utilize a Type III Secretion System (T3SS), necessary for their pathogenicity. The structural proteins composing the T3SS are common to all virulent Salmonella and Shigella spp., parti

Cross-protection and cross-neutralization capacity of ancestral and VOC-matched SARS-CoV-2 adenoviral vector-based vaccines.

COVID-19 vaccines were originally designed based on the ancestral Spike protein, but immune escape of emergent Variants of Concern (VOC) jeopardized their efficacy, warranting variant-proof vaccines. Here, we used preclinical rodent models to establish the cross-protective and cross-neutralizing capacity of adenoviral-vectored vaccines expressing VOC-matched Spike. CoroVaxG.3-D.FR, matched to Delta Plus Spike, displayed the highest levels of nAb to the matched VOC and mismatched variants. Cross-protection against viral infection in aged K18-hACE2 mice showed dramatic differences among the diff

Convalescent Immunity to Guinea Pig Cytomegalovirus Induces Limited Cross Strain Protection against Re-Infection but High-Level Protection against Congenital Disease.

The guinea pig is the only small animal model for congenital cytomegalovirus (cCMV) but requires guinea pig cytomegalovirus (GPCMV). Current GPCMV research utilizes prototype strain 22122, which limits the translational impact of GPCMV as numerous human CMV strains exist and cCMV is possible in the setting of re-infection. A novel strain of GPCMV (TAMYC) exhibited differences to 22122 in various glycoproteins with GP74 (gO homolog) the most variable (25% difference). Antibody ELISAs for TAMYC-convalescent animals evoked similar immune response to viral glycoprotein complexes (gB, gH/gL, gM/gN,

Sun protection as a protective factor for actinic cheilitis: Cross-sectional population-based study.

OBJECTIVE: To determine whether sun protection is associated with lower occurrence of actinic cheilitis in adults living in a city from southern Brazil. MATERIALS AND METHODS: A multi-stage proportional sample of 404 individuals 18 years and older was obtained. Interviews and clinical examinations were conducted in participants' households. Four categories of self-reported use of sun protection were determined. Multivariable Poisson regression was used to assess the associations. RESULTS: Prevalence of actinic cheilitis was 47.1%. In the first main-effects multivariable model, AC was signifi

Immunogenic cross-reactivity between different serotypes novel duck reoviruses: Evaluation of cross-protection provided by mono or bivalent vaccine.

To investigate the immunogenic cross-reactivity among different novel duck reovirus isolates, animals immunized with monovalent inactivated duck reovirus were challenged with homologous and heterologous viruses, and serum samples were collected after immunization for cross-neutralization test. The results showed that the virus neutralizing titer against homologous virus was significantly higher than that against heterologous virus(p < 0.000001), which indicated that there was an important difference in antigenicity between DE13 and WL01 strains, which affected the specificity of protective vir

Programmable trivalent nanocage vaccine confers durable cross-species protection against Bordetella bronchiseptica infection.

Bordetella bronchiseptica (B. bronchiseptica), a respiratory pathogen endemic in companion animals, poses an escalating zoonotic threat to humans due to intensified human-pet contact, particularly among immunocompromised individuals. Here, we report a programmable trivalent nanocage vaccine, mi3-F1P2Fim2, featuring immunodominant regions of FHA, Prn, and Fim2 covalently fused into a single-chain construct and displayed on a self-assembling mi3 nanocage via the SpyTag003/SpyCatcher003 system, enabling high-density and multivalent antigen presentation. Compared with its soluble monomeric counter

A nanoemulsion-adjuvanted trivalent neuraminidase vaccine confers cross-protection against divergent influenza viruses.

Developing universal influenza vaccines (UIVs) capable of eliciting broad protection remains a critical public health priority. Neuraminidase (NA), a relatively conserved surface antigen of influenza viruses, is a promising target for UIV design. Here, we produced full-length trivalent NA proteins via a baculovirus-insect cell expression system, matching the WHO-recommended strains (H1N1, H3N2, and B/Victoria) for the 2024-2025 season. Immunogenicity evaluation of N2NA (derived from H3N2) in mice demonstrated that nanoemulsion (NE) adjuvant significantly enhanced NA-inhibiting (NI) antibody ti

Therapeutic Potential of Saikosaponins in MASLD and Cross-Organ Protection: A Research Update.

Metabolic dysfunction-associated steatotic liver disease (MASLD), formerly known as non-alcoholic fatty liver disease (NAFLD), has emerged as the most prevalent chronic liver disorder in China, posing a significant threat to public health. While current Western pharmacological therapies demonstrate efficacy, they don't comprehensively ameliorate pathological components of MASLD and are often associated with substantial adverse effects. Saikosaponins (SSs), as the main active components of Bupleurum chinense, a traditional Chinese medicine (TCM), have been demonstrated to exert potential therap

β-Glucan Chitosan Particle Provides Cross-Protection Against Multi-Drug- Resistant Candida auris.

Candida auris is a multidrug-resistant fungal pathogen that can survive outside the host, easily spread, and colonize the healthcare environment, medical devices, and human skin. C. auris causes serious, life-threatening infections with mortality rates of ~ 60% in immunosuppressed patients. Some isolates of C. auris are resistant to virtually all clinically available antifungal drugs. Therefore, alternative therapeutic approaches are urgently needed. C. auris cell wall contains β-glucan similar to non-pathogenic yeasts such as Saccharomyces cerevisiae. Moreover, cellular proteins are also heav

Multistage protective anti-CelTOS monoclonal antibodies with cross-species sterile protection against malaria.

CelTOS is a malaria vaccine antigen that is conserved in Plasmodium and other apicomplexan parasites and plays a role in cell-traversal. The structural basis and mechanisms of CelTOS-induced protective immunity to parasites are unknown. Here, CelTOS-specific monoclonal antibodies (mAbs) 7g7 and 4h12 demonstrated multistage activity, protecting against liver infection and preventing parasite transmission to mosquitoes. Both mAbs demonstrated cross-species activity with sterile protection against in vivo challenge with transgenic parasites containing either P. falciparum or P. vivax CelTOS, and

A bacterial pigment provides cross-species protection from H2O2- and neutrophil-mediated killing.

Bacterial infections are often polymicrobial. Pseudomonas aeruginosa and Staphylococcus aureus cause chronic co-infections, which are more problematic than mono-species infections. Understanding the mechanisms of their interactions is crucial for treating co-infections. Staphyloxanthin (STX), a yellow pigment synthesized by the S. aureus crt operon, promotes S. aureus resistance to oxidative stress and neutrophil-mediated killing. We found that STX production by S. aureus, either as surface-grown macrocolonies or planktonic cultures, was elevated when exposed to the P. aeruginosa exoproduct, 2

Infectious Bronchitis Virus Vaccination at Day 1 of Age Further Limits Cross Protection.

Cross-protection and immune responses elicited by infectious bronchitis virus (IBV) vaccination on Day 1 of age or at later time points were examined. Specific-pathogen-free chickens were vaccinated with a Massachusetts-type vaccine and heterologous challenge was performed with an Arkansas (Ark) -type virulent strain. In Trial 1, chickens vaccinated on Day 1 or Day 10 of age were challenged 21 days after vaccination. Analysis of tracheal histopathology and viral load demonstrated less cross protection when vaccination was performed on Day 1 of age. In Trial 2, chickens were vaccinated on Day 1

BCG-Induced Cross-Protection and Development of Trained Immunity: Implication for Vaccine Design.

The Bacillus Calmette-Guérin (BCG) is a live attenuated tuberculosis vaccine that has the ability to induce non-specific cross-protection against pathogens that might be unrelated to the target disease. Vaccination with BCG reduces mortality in newborns and induces an improved innate immune response against microorganisms other than Mycobacterium tuberculosis, such as Candida albicans and Staphylococcus aureus. Innate immune cells, including monocytes and natural killer (NK) cells, contribute to this non-specific immune protection in a way that is independent of memory T or B cells. This pheno

Partial cross-protection between Japanese encephalitis virus genotype I and III in mice.

Genotype III (GIII) Japanese encephalitis virus (JEV) predominance has gradually been replaced by genotype I (GI) over the last 20 years in many Asian countries. This genotype shift raises concerns about the protective efficacy of Japanese encephalitis (JE) vaccines, as all of the currently licensed JE vaccines are derived from GIII strains. In this study, we conducted vaccination-challenge protection assays to evaluate the cross-protective efficacy of GI- or GIII-derived vaccines against the challenge of a heterologous genotype using a mouse challenge model. Titration of the neutralizing anti

Th17-Mediated Cross Protection against Pneumococcal Carriage by Vaccination with a Variable Antigen.

Serotype-specific protection against Streptococcus pneumoniae is an important limitation of the current polysaccharide-based vaccines. To prevent serotype replacement, reduce transmission, and limit the emergence of new variants, it is essential to induce broad protection and restrict pneumococcal colonization. In this study, we used a prototype vaccine formulation consisting of lipopolysaccharide (LPS)-detoxified outer membrane vesicles (OMVs) from Salmonella enterica serovar Typhimurium displaying the variable N terminus of PspA (α1α2) for intranasal vaccination, which induced strong Th17 im

Cross-protection conferred by immunization with an rOmpH-based intranasal fowl cholera vaccine.

A previous study demonstrated that a recombinant outer membrane protein H (rOmpH)-based intranasal fowl cholera vaccine elicited efficient homologous protection against the Pasteurella multocida strain X-73 (A:1) in chickens. The present study aimed to determine the cross-protectivity against heterologous P. multocida strains. The rOmpH was purified via electroelution and formulated with two kinds of adjuvants. The vaccine formulations in a total volume of 100 µl were 100 µg rOmpH with 3 µg of Escherichia coli enterotoxin B or 10 µg of CpG ODN2007. Chickens were assigned to three experimental

Single- and multiple-clade influenza A H5N1 vaccines induce cross protection in ferrets.

The rapid evolution, genetic diversity, broad host range, and increasing human infection with avian influenza A (H5N1) viruses highlight the need for an efficacious cross-clade vaccine. Using the ferret model, we compared induction of cross-reactive immunity and protective efficacy of three single-clade H5N1 vaccines and a novel multiple-clade H5N1 vaccine, with and without MF59 adjuvant. Reverse genetics (rg) was used to generate vaccine viruses containing the hemagglutinin (HA) and neuraminidase genes of wild-type H5N1 viruses. Ferrets received two doses of inactivated whole-virus vaccine se

Cross-protection of hepatitis E virus genotypes 1 and 4 in rhesus macaques.

The purpose of this study was to determine cross-protection between HEV genotypes 1 and 4, which are prevalent in China. Fecal suspensions of genotypes 1 and 4 from patients, as well as genotype 4 from swine, were inoculated intravenously into rhesus macaques. Each inoculum contained 5 x 10(4) genome equivalents of HEV. After infection, serum and fecal samples were collected serially and the levels of alanine aminotransferase (ALT) and anti-HEV IgG and IgM in sera, and HEV RNA in fecal samples, were measured. Liver biopsies were carried out. All the infected monkeys (12/12) developed anti-HEV
