Photoregulation in prokaryotes.

The spectroscopic identification of sensory rhodopsin I by Bogomolni and Spudich in 1982 provided a molecular link between the light environment and phototaxis in Halobacterium salinarum, and thus laid the foundation for the study of signal transducing photosensors in prokaryotes. In recent years, a number of new prokaryotic photosensory receptors have been discovered across a broad range of taxa, including dozens in chemotrophic species. Among these photoreceptors are new classes of rhodopsins, BLUF-domain proteins, bacteriophytochromes, cryptochromes, and LOV-family photosensors. Genetic and

Optical Activation of TrkB Signaling.

Brain-derived neurotrophic factor, via activation of tropomyosin receptor kinase B (TrkB), plays a critical role in neuronal proliferation, differentiation, survival, and death. Dysregulation of TrkB signaling is implicated in neurodegenerative disorders and cancers. Precise activation of TrkB signaling with spatial and temporal resolution is greatly desired to study the dynamic nature of TrkB signaling and its role in related diseases. Here we develop different optogenetic approaches that use light to activate TrkB signaling. Utilizing the photosensitive protein Arabidopsis thaliana cryptochr

Magnetically Stimulated Myogenesis Recruits a CRY2-TRPC1 Photosensitive Signaling Axis.

The cryptochromes are flavoproteins that either individually or synergistically respond to light and magnetic field directionality as well as are implicated in circadian rhythm entrainment and development. Single brief exposures (10 min) to low energy (1.5 mT) pulsed electromagnetic fields (PEMFs) were previously shown to enhance myogenesis by stimulating transient receptor potential canonical 1 (TRPC1)-mediated Ca2+ entry, whereby downwardly directed fields produced greater myogenic enhancement than upwardly directed fields. Here, we show that growth in the dark results in myoblasts losing th

Postnatal constant light compensates Cryptochrome1 and 2 double deficiency for disruption of circadian behavioral rhythms in mice under constant dark.

Clock genes Cryptochrome (Cry1) and Cry2 are essential for expression of circadian rhythms in mice under constant darkness (DD). However, circadian rhythms in clock gene Per1 expression or clock protein PER2 are detected in the cultured suprachiasmatic nucleus (SCN) of neonatal Cry1 and Cry2 double deficient (Cry1 (-/-)/Cry2 (-/-)) mice. A lack of circadian rhythms in adult Cry1 (-/-)/Cry2 (-/-) mice is most likely due to developmentally disorganized cellular coupling of oscillating neurons in the SCN. On the other hand, neonatal rats exposed to constant light (LL) developed a tenable circadia

Homing pigeon navigation relies on superparamagnetic macrophages under overcast conditions.

Birds use a variety of navigational strategies, including the geomagnetic field, especially when other cues are not available, such as under overcast or nocturnal conditions. Magnetite particles in the beak, cryptochromes in the eye, cellular ion-channel alterations, and changes in the vestibular system have been proposed to explain magnetoreception, but the exact mechanisms remain debated. Here, we used physical, morphological, functional, and genomic assays to identify the presence of superparamagnetic macrophages in the liver. We found that after macrophage depletion, pigeons flying under o

Evaluation of the Digital Ventilated Cage® system for circadian phenotyping.

The study of circadian rhythms has been critically dependent upon analysing mouse home cage activity, typically employing wheel running activity under different lighting conditions. Here we assess a novel method, the Digital Ventilated Cage (DVC®, Tecniplast SpA, Italy), for circadian phenotyping. Based upon capacitive sensors mounted under black individually ventilated cages with inbuilt LED lighting, each cage becomes an independent light-controlled chamber. Home cage activity in C57BL/6J mice was recorded under a range of lighting conditions, along with circadian clock-deficient cryptochrom

5-Methyltetrahydrofolate inhibits photosensitization reactions and strand breaks in DNA.

The known functions of folate are to support one-carbon metabolism and to serve as photoreceptors for cryptochromes and photolyases. We demonstrate that 5-methyltetrahydrofolate (5-MTHF, the predominant folate in plasma) is also a potent, near diffusion limited, scavenger of singlet oxygen and quencher of excited photosensitizers. Both pathways result in decomposition of 5-MTHF, although ascorbate can protect against this loss. In the absence of photosensitizers, 5-MTHF is directly decomposed only very slowly by UVA or UVB. Although synthetic folic acid can promote DNA damage by UVA, submicrom

Brown fat activity deepens depression: True or false?

Circadian clocks have evolved a slowing-down mechanism. Temperature may be the original and universal time-giver to the organism. Brown adipose tissue generates heat and guides the circadian rhythm of core body temperature. The cryptochrome proteins regulate the temperature entrainability, and their dysfunction may let the activation of brown adipose tissue affect the brain more easily. Therefore, the activity of brown adipose tissue may compromise the slowing-down mechanism and thereby contribute to the emergence of mood disorders and the increase in suicide mortality around the time of puber

Light in the Fungal World: From Photoreception to Gene Transcription and Beyond.

Fungi see light of different colors by using photoreceptors such as the White Collar proteins and cryptochromes for blue light, opsins for green light, and phytochromes for red light. Light regulates fungal development, promotes the accumulation of protective pigments and proteins, and regulates tropic growth. The White Collar complex (WCC) is a photoreceptor and a transcription factor that is responsible for regulating transcription after exposure to blue light. In Neurospora crassa, light promotes the interaction of WCCs and their binding to the promoters to activate transcription. In Asperg

Quantitative Trait Locus Determining the Time of Blood Feeding in Culex pipiens (Diptera: Culicidae).

Mosquitoes and other blood feeding arthropods are vectors of pathogens causing serious human diseases, such as Plasmodium spp. (malaria), Wuchereria bancrofti (lymphatic filariasis), Borrelia burgdorferi (Lyme disease), and viruses causing dengue, Zika, West Nile, chikungunya, and yellow fever. Among the most effective strategies for the prevention of vector-borne diseases are those aimed at reducing human-vector interactions, such as insecticide applications and insecticide-treated bed nets (ITNs). In some areas where ITNs are widely used, behavioral adaptations have resulted in mosquitoes sh

Cry1 expression in the chicken pineal gland: effects of changes in the light/dark conditions.

Cryptochromes (Cry) are core components in the gene regulation of circadian rhythmic processes. It was shown earlier, that Cry1 mRNA content of the avian pineal gland was increased after a 4h exposure to light during subjective night; however, a 30min exposure was ineffective. In this study, changes in pineal Cry1 expression were detected in chickens during and after being placed into reversed light/dark environment. Cry1 mRNA content was higher if light was on during the night; however, in the first 2h of light exposure at night, Cry1 mRNA contents were decreased. Following the first overnigh

CRY2 is associated with rapid cycling in bipolar disorder patients.

BACKGROUND: Bipolar disorder patients often display abnormalities in circadian rhythm, and they are sensitive to irregular diurnal rhythms. CRY2 participates in the core clock that generates circadian rhythms. CRY2 mRNA expression in blood mononuclear cells was recently shown to display a marked diurnal variation and to respond to total sleep deprivation in healthy human volunteers. It was also shown that bipolar patients in a depressive state had lower CRY2 mRNA levels, nonresponsive to total sleep deprivation, compared to healthy controls, and that CRY2 gene variation was associated with win

Basidiomycete fruiting body morphology correlates with the convergent loss of photolyase-related proteins.

Cryptochromes and photolyase-related (CRY/PHR) proteins are flavoproteins involved in blue-light signaling and/or the light-dependent repair of UV-induced DNA damage across all domains of life. Although well-studied in many eukaryotes, their evolution and function in mushroom-forming fungi (Agaricomycetes and Basidiomycota) remain poorly understood. By analyzing 655 genomes, we show that fungi ancestrally possessed four CRY/PHR subfamilies, of which class I cyclobutane pyrimidine dimer (CPD) photolyases became broadly conserved across Dikarya. Class I CPD photolyase-related proteins have been

Integrated Analysis of CRY1 Gene Expression and InDel Polymorphism Reveals Associations with Ovarian Morphological Traits in Chinese Holstein Dairy Cattle.

Clock genes, such as cryptochrome 1 (CRY1), exhibit rhythmic expression in the reproductive organs. This gene is a key component of the circadian clock and is involved in various physiological processes, including reproduction, suggesting that it may be linked to ovarian activity via neuroendocrine pathways. However, the association between CRY1 variants and ovarian traits in dairy cows is unclear. In this study we used qRT-PCR to assay the mRNA expression of this gene, and found that CRY1 expression was highest in oocytes in several tissues; subsequently, we found that CRY1 expression was hig

Magnetosensitivity of Model Flavin-Tryptophan Radical Pairs in a Dynamic Protein Environment.

Light-induced radical pairs in cryptochrome proteins located in the retina are thought to be the receptors at the heart of the magnetic compass sense of migratory songbirds. Reliable simulations of the performance of such sensors face several fundamental challenges. The quantum spin dynamics of large spin systems must be modeled for periods in excess of a microsecond including realistic local magnetic interactions that fluctuate on a picosecond to microsecond time scale as a result of thermal motion. Here we employ newly developed computational methods that combine explicitly time-dependent in

CRY2 (rs11605924) and G6PC2 (rs560887) single nucleotide polymorphisms increase risk of type 2 diabetes mellitus.

Single nucleotide polymorphisms (SNPs) are genetic risk factors for type 2 diabetes (T2D). SNPs of the cryptochrome circadian regulator 2 (CRY2), hepatocyte nuclear factor 1α, prospero homeobox 1 and glucose-6-phosphatase catalytic subunit (G6PC2) genes are associated with T2D. To the best of our knowledge, however, the association is unclear in African patients and the burden of T2D is growing rapidly in Africa. The present study aimed to investigate SNPs of cryptochrome circadian regulator 2 (CRY2), hepatocyte nuclear factor 1α, prospero homeobox 1 and glucose-6-phosphatase catalytic subunit

The LOV domain: a chromophore module servicing multiple photoreceptors.

Three different families of blue-light receptors have been characterized from higher plants: three cryptochromes, two phototropins, and the three members of the ZTL/ADO family. Phototropins and the ZTL/ADO proteins have chromophore modules, designated LOV domains, that bind flavin mononucleotide and undergo formation of a C(4a) flavin-cysteinyl adduct. All contain the highly conserved amino acid motif GXNCRFLQ. Over 90 prokaryote proteins also contain LOV domains with this motif upstream from one of several different functional groups. All of these that have been investigated to date act as ph

Effects of light on cyanide-resistant respiration and alternative oxidase function in Arabidopsis seedlings.

Mitochondrial alternative oxidase (AOX), the unique respiratory terminal oxidase in plants, catalyzes the energy wasteful cyanide (CN)-resistant respiration and plays a role in optimizing photosynthesis. Although it has been demonstrated that leaf AOX is upregulated after illumination, the in vivo mechanism of AOX upregulation by light and its physiological significance are still unknown. In this report, red light and blue light-induced AOX (especially AOX1a) expressions were characterized. Phytochromes, phototropins and cryptochromes, all these photoreceptors mediate the light-response of AOX

"Time" for obesity-related cancer: The role of the circadian rhythm in cancer pathogenesis and treatment.

The circadian rhythm is regulated by an intrinsic time-tracking system, composed both of a central and a peripheral clock, which influences the cycles of activities and sleep of an individual over 24 h. At the molecular level, the circadian rhythm begins when two basic helix-loop-helix/Per-ARNT-SIM (bHLH-PAS) proteins, BMAL-1 and CLOCK, interact with each other to produce BMAL-1/CLOCK heterodimers in the cytoplasm. The BMAL-1/CLOCK target genes encode for the repressor components of the clock, cryptochrome (Cry1 and Cry2) and the Period proteins (Per1, Per2 and Per3). It has been recently demo

Clock genes rescue nphp mutations in zebrafish.

The zebrafish pronephros model, using morpholino oligonucleotides (MO) to deplete target genes, has been extensively used to characterize human ciliopathy phenotypes. Recently, discrepancies between MO and genetically defined mutants have questioned this approach. We analyzed zebrafish with mutations in the nphp1-4-8 module to determine the validity of MO-based results. While MO-mediated depletion resulted in glomerular cyst and cloaca malformation, these ciliopathy-typical manifestations were observed at a much lower frequency in zebrafish embryos with defined nphp mutations. All nphp1-4-8 mu
