Invasive Curvularia Infection in Pediatric Patients With Hematologic Malignancy Identified by Fungal Sequencing.

Curvularia is a saprophytic dematiaceous mold and a rare human pathogen. Here, we report three severely immunocompromised pediatric patients who developed invasive Curvularia infection. Diagnosis was achieved or confirmed in all cases by fungal ribosome sequencing, which hastened species identification and targeted treatment for the patients reported. There are no treatment guidelines for invasive Curvularia infection, though we report three patients who were cured of their infection through a combination of surgical resection and various anti-fungal therapies, indicating a relatively low viru

Invasive phaeohyphomycosis co-infection with Alternaria spp. and Curvularia spp. in a neutropenic host.

Phaeohyphomycoses are infections caused by dark-walled dematiaceous fungi. Alternaria and Curvularia are two genera of dematiaceous molds known to cause invasive fungal rhinosinusitis, particularly in immunocompromised patients. Co-infection with two dematiaceous fungi is rarely reported in the literature. This report describes a case of biopsy proven invasive fungal rhinosinusitis with Alternaria spp. and Curvularia spp. co-infection in a neutropenic host. The infection characteristics, microbiologic findings, and treatment are described.

The stability and steady-state kinetics of vanadium chloroperoxidase from the fungus Curvularia inaequalis.

In this article we report on the steady-state kinetics of the chlorination and the stability of the vanadium chloroperoxidase from the fungus Curvularia inaequalis. The data show that the kinetics of this enzyme resemble that of the vanadium bromoperoxidase from the seaweed Ascophyllum nodosum. At low pH, chloride inhibited the enzyme, but the inhibition was of a dual nature. At pH 4.1 a mixed type of inhibition by chloride with respect to hydrogen peroxide was observed whereas at pH 3.1 the nature of the inhibition became competitive. The log Km for hydrogen peroxide decreased linearly with p

Tenckhoff catheter obstruction resulting from invasion by Curvularia lunata in the absence of peritonitis.

A 60-year-old black woman on continuous ambulatory peritoneal dialysis (CAPD) presented with Tenckhoff catheter malfunction. She exhibited no signs or symptoms of acute peritonitis. Darkly pigmented plaques were adherent to the inner wall of the catheter and exchange tubing focally throughout its length. Similar material was adherent to the protein coagulum in the dialysis bag. The Tenckhoff catheter and tubing were removed, and hemodialysis was initiated. Cultures of the catheter and the dialysate protein coagulum grew the saprophytic soil fungus, Curvularia lunata. Histologic examination of

A Special Tinea Nigra Caused by Curvularia lunata: Case Report and Literature Review.

Tinea nigra is a superficial fungal infection usually caused by Hortaea werneckii (H. werneckii). We report a special case of tinea nigra in an immunocompetent child who developed a unilateral, rapidly growing pigmented lesion on her palm. Interestingly, Curvularia lunata (C. lunata) was isolated from the lesion scrapes and was identified by both morphological features and molecular biology methods. The lesion was completely cleared by topical naftifine hydrochloride and ketoconazole cream. We present-to the best of our knowledge-the first case of tinea nigra where the causative pathogen was i

Histopathologic Insights Into Curvularia-Induced Chronic Granulomatous Fungal Sinusitis: A Guide for Surgical Pathologists.

Chronic granulomatous fungal sinusitis is a rare, debilitating infection characterized by an enlarging mass affecting the orbit, nose, palate, and paranasal sinuses, with occasional intraorbital and intracranial extension. Most cases are reported in subtropical regions, with rare reports from the United States and Europe. Globally, Aspergillus flavus is the primary causative agent in >60% of cases, followed by other Aspergillus species and dematiaceous molds. This study highlights the histopathologic, microbiologic, and molecular diagnostic features of 6 cases with chronic granulomatous fungal

Acetylcholinesterase inhibiting withanolides from Curvularia pseudobrachyspora, an endophytic fungus of Withania somnifera (L.) Dunal.

Alzheimer's disease is the most prominent neurological illness, and the present investigation sets out to determine the impact of bioactive substances from endophytic fungi on acetylcholinesterase (AChE) inhibition. Medicinal plant Withania somnifera (L.) Dunal was used to isolate endophytic fungi, and WSE42 (out of 49 isolates) exhibited higher AChE inhibition (78.57%) and free radical scavenging activity (82.47%) in its methanolic crude extract. WSE42 was identified as Curvularia pseudobrachyspora (accession number-PP837926), and among all the fractions collected by column chromatography, tw

Curvularia infection presenting as recurrent conjunctival erosion overlying a scleral-fixated intraocular lens suture.

PURPOSE: To describe a case of an indolent, chronic Curvularia infection after a ruptured globe injury from an intraocular metallic foreign body in a 38-year old male. Following globe repair and vitrectomy, the patient underwent a sutured scleral-fixated lens. The patient subsequently presented with recurrent painless conjunctival erosion associated with small, pigmented skip lesions of the exposed suture. OBSERVATION: Indolent, chronic Curvularia infections can present with subtle signs, including small areas of suture exposure and pigmentation as well as and small ciliary body collections.

Curvularia--favorable response to oral itraconazole therapy in two patients with locally invasive phaeohyphomycosis.

Curvularia species are ubiquitous and occasionally lead to infections in humans. In immunosuppressed patients, infections are often serious, and systemic dissemination is not uncommon. The optimal antifungal therapy is unclear. I here present two cases, a healthy man with locally invasive, mulicentric paranasal fungal sinusitis, and a case of progressive verrucal distal onychomycosis that developed while the patient was undergoing accelerated chemotherapy for non-Hodgkin's lymphoma. Both patients showed excellent responses to treatment with itraconazole suspension. Oral itraconazole may provid

Phytotoxic and antifungal metabolites from Curvularia sp. FH01 isolated from the gut of Atractomorpha sinensis.

Two main phytotoxic and antifungal phthalic acid butyl isobutyl ester (1) and radicinin (2) were isolated from the culture of Curvularia sp. FH01, a fungus residing in the Atractomorpha sinensis gut. The structures of isolated metabolites were established on the basis of spectral analysis. Metabolites 1 and 2 exhibited significant phytotoxic activity against the radical growth of Echinochloa crusgalli with their IC(50) values of 61.9 and 5.9 μg/mL, respectively, which were comparable to that 2,4-dichlorophenoxyacetic acid (2.0 μg/mL) used as a positive control. The antifungal test results show

Invasive fungal infection caused by curvularia species in a patient with intranasal drug use: A case report.

Chronic invasive fungal sinusitis (CIFS) is an invasive fungal infection that can occur in immunocompetent individuals and is typically caused by Aspergillus species. Although many reported cases are unable to identify an etiology for the infection, certain risk factors such as chronic intranasal cocaine use can make patients susceptible to CIFS. This case report describes a unique case of CIFS secondary to Curvularia species in an immunocompetent patient with intranasal drug use.

Breakthrough mucormycosis after voriconazole use in a case of invasive fungal rhinosinusitis due to Curvularia lunata.

Invasive fungal rhinosinusitis (FRS) is a potentially fatal illness requiring early diagnosis and aggressive treatment with surgery and antifungals. We report a case of chronic FRS in a recently diagnosed diabetic individual due to Curvularia lunata. Imaging revealed extension into the right orbit and right basifrontal lobe. This was further complicated by development of nosocomial mucormycosis which was attributed to voriconazole therapy. The patient responded well to debridement and amphotericin B based therapy. To our knowledge, there are no reported cases of invasive FRS due to Curvularia

First report of human infection caused by Curvularia warraberensis, manifesting as invasive sinusitis with intracranial involvement.

Curvularia species are saprophytic dematiaceous fungi commonly isolated from environmental sources. Most often, they are responsible for allergic fungal rhinosinusitis, an intense, allergic inflammatory sinus disease in immunocompetent individuals. Though invasive infections are rare and more commonly observed in immunocompromised patients, recent reports indicate an increasing trend of invasive sinusitis caused by Curvularia species in immunocompetent hosts. Over the past few years, new species of the genus Curvularia are increasingly being recognized as human pathogens. Here, we report the f
