Morphology and phylogeny of the centrohelid heliozoans Raphidocystidae and their ability to consume cyanobacteria.

The centrohelid family Raphidocystidae is a large group of heliozoans whose morphology and biology are poorly understood and whose taxonomy and phylogenetic relationships are currently confusing. Here, we report the results of a study of clonal cultures of raphidocystids belonging to the species Raphidocystis tubifera, R. ambigua, R. marginata and R. symmetrica. The cells were examined using light microscopy as well as scanning and transmission electron microscopy. New data on the morphology of the studied species were obtained and compared with other representatives of Raphidocystidae. For th

Impact of cyanobacteria on zinc and carbon sequestration in hydrous magnesium carbonates.

Mineral carbonation has the potential to permanently sequester CO2 in stable carbonate minerals, mimicking the natural precipitation of Ca- or Mg-rich carbonates. These minerals are found in ultramafic rocks and mine wastes, which also contain metals that are crucial resources for renewable energy technologies. This study aims to better characterize the mechanisms and quantify the potential of cyanobacterium (Gloeocapsa sp.) for hydrous Mg carbonate (HMC) precipitation sequestering both CO2 and heavy metals such as Zn. The results show that Zn removal mechanisms depend on solution pH and bacte

A Minority of Desert Cyanobacteria and Algae Is Responsible for the Bulk of CO2 Fixation.

Cyanobacteria and algae are the major photosynthetic organisms in deserts because they survive desiccation, high solar radiation and extreme temperature fluctuations better than other plants. Under favourable conditions, desert cyanobacteria and algae evidently photosynthesise. However, our understanding of whether each group modulates this metabolic process in response to preceding harsh conditions remains limited. To find out the effect of aridity on the photosynthetic activity of desert cyanobacteria and algae, we compared their cellular biovolume-specific carbon dioxide (CO2) fixation in t

Screening of Lipid-Reducing Activity and Cytotoxicity of the Exometabolome from Cyanobacteria.

Cyanobacteria are rich producers of secondary metabolites, excreting some of these to the culture media. However, the exometabolome of cyanobacteria has been poorly studied, and few studies have dwelled on its characterization and bioactivity assessment. In this work, exometabolomes of 56 cyanobacterial strains were characterized by HR-ESI-LC-MS/MS. Cytotoxicity was assessed on two carcinoma cell lines, HepG2 and HCT116, while the reduction in lipids was tested in zebrafish larvae and in a steatosis model with fatty acid-overloaded human liver cells. The exometabolome analysis using GNPS revea

An improved method for extraction of polar and charged metabolites from cyanobacteria.

A key requirement for 13C Metabolic flux analysis (13C-MFA), a widely used technique to estimate intracellular metabolic fluxes, is an efficient method for the extraction of intermediate metabolites for analysis via liquid chromatography mass spectrometry (LC/MS). The 13C isotopic labeling results in further distribution of an already sparse pool of intermediate metabolites into isotopologues, each appearing as a separate chromatographic feature. We examined some of the reported solvent systems for the extraction of polar intracellular metabolites from three strains of cyanobacteria of the gen

Low-level concentrations of aminoglycoside antibiotics induce the aggregation of cyanobacteria.

The interactions between antibiotics and microorganisms have attracted enormous research attentions. In this study, we investigated the effects of two typical aminoglycoside antibiotics on the aggregation of the model cyanobacterium, Synechococcus elongatus, and the dominating strain in algal blooms, Microcystis aeruginosa, via the analysis of zeta potentials, hydrophobicity, and extracellular polymeric substances (EPS) secretion. The results showed that low-level antibiotics promoted the aggregation of S. elongatus and M. aeruginosa by 40 and 18% under 0.10 and 0.02 μg/mL of kanamycin, respec

Distributions of iron, phosphorus and sulfur along trichomes of the cyanobacteria Trichodesmium.

The nonheterocystous cyanobacterium Trichodesmium fixes C and N concurrently during the light period in tropical and subtropical oceans. Synchrotron mapping of Fe, P and S in trichomes of Trichodesmium erythraeum Erhenberg IMS 101 (CCMP 1985) collected during exponential and senescent growth revealed that 16% of trichomes contained sections of up to 25 cells with ca. 2-fold elevated Fe and S but ca. 2-fold lowered P in comparison to neighboring trichome sections. The correlation between Fe and S in these trichomes was moderate to strongly positive (R > 0.35), while the correlation between Fe a

Optimization of isopropanol production by engineered cyanobacteria with a synthetic metabolic pathway.

Cyanobacterium is an attractive host for the production of various chemicals and alternative fuels using solar energy and carbon dioxide. In previous study, we succeeded to produce isopropanol using engineered Synechococcus elongatus PCC 7942 under dark and anaerobic conditions (0.43 mM, 26.5 mg/l). In the present study, we report the further optimization of this isopropanol producing condition. We then optimized growth conditions for production of isopropanol by the engineered cyanobacteria, including the use of cells in early stationary phase and buffering of the production medium to neutral

Growth of Legionella pneumophila in association with blue-green algae (cyanobacteria).

Legionella pneumophila (Legionnaires disease bacterium) of serogroup 1 was isolated from an algal-bacterial mat community growing at 45 degrees C in a man-made thermal effluent. This isolate was grown in mineral salts medium at 45 degrees C in association with the blue-green alga (cyanobacterium) Fischerella sp. over a pH range of 6.9 to 7.6. L. pneumophila was apparently using algal extracellular products as its carbon and energy sources. These observations indicate that the temperature, pH, and nutritional requirements of L. pneumophila are not as stringent as those previously observed when

Titanium Dioxide Nanoparticle-Driven Metabolic and Molecular Reprogramming in Cyanobacteria.

Cyanobacteria are promising platforms for bioenergy, carbon sequestration, and bioproduct synthesis, but their photosynthetic efficiency is limited by suboptimal light utilization, electron transport constraints, and environmental stress. Titanium dioxide nanoparticles (n-TiO2) have emerged as powerful photocatalytic materials that can enhance light absorption, modulate electron transport, and influence the redox balance in biological systems. This review advances the concept of photocatalytic-biological coupling, in which n-TiO2 functions as artificial light amplifiers that augment cyanobacte

Life Course Exposure to Cyanobacteria and Amyotrophic Lateral Sclerosis Survival.

Cyanobacterial harmful algal blooms (cyanoHABs) occur worldwide and can cause ingestion and inhalation exposure to microcystin and other potent toxins. This study develops life course exposure measures for cyanobacteria for application in population studies and then associates these measures with the survival of individuals with amyotrophic lateral sclerosis (ALS). The exposure measures utilize an individual's residence history, date of disease onset, and satellite data from the Cyanobacteria Assessment Network. Residence duration for selected exposure windows referenced to disease onset date

Insertion Sequences show diverse recent activities in Cyanobacteria and Archaea.

BACKGROUND: Mobile genetic elements (MGEs) play an essential role in genome rearrangement and evolution, and are widely used as an important genetic tool. RESULTS: In this article, we present genetic maps of recently active Insertion Sequence (IS) elements, the simplest form of MGEs, for all sequenced cyanobacteria and archaea, predicted based on the previously identified ~1,500 IS elements. Our predicted IS maps are consistent with the NCBI annotations of the IS elements. By linking the predicted IS elements to various characteristics of the organisms under study and the organism's living co

Moisture-Driven CO2 Direct Air Capture and Delivery for Cultivating Cyanobacteria.

A system was developed and demonstrated that uses solid sorbents to capture CO2 passively from ambient air and to subsequently release CO2 when immersed in an alkaline medium. The system was used to cultivate the cyanobacterium Synechocystis sp. PCC 6803 at the flask (50 mL) and bench (12 L) scales. A small pilot-scale system installed in a 4.2-m2 outdoor raceway pond (840 L) was evaluated for over 300 days of outdoor wet/dry cycling and in cultivation trials over four seasons. Sorbent CO2-release capacity and kinetics were reduced due to competitive binding of nitrate and biofouling, and sorb

Molecular insights into phycobilisome assembly pathway reveal crystalline bodies in cyanobacteria.

In oxygenic photosynthetic organisms, light energy is converted to chemical energy to drive CO2 fixation reactions and sustain life on Earth. Cyanobacteria contain phycobilisome (PBS) complexes that play critical roles in light harvesting and directing light energy to the photosystem I and II reaction centers. The proper assembly of PBS components is an intricate process that is required for their activity and association with photosystem I and II. To understand the complex mechanisms regulating the PBS assembly, we knocked out the terminal emitter apcE, which forms the structural scaffold for

Inorganic carbon levels regulate growth via SigC signaling cascade in cyanobacteria.

Cyanobacterial growth depends on inorganic carbon (Ci; CO2 and bicarbonate) concentration, but mechanism(s) adjusting photosynthesis and growth according to Ci remain unclear. ΔrpoZ cells lacking the ω subunit of the RNA polymerase (RNAP) show a unique high-CO2 lethal phenotype in Synechocystis sp. PCC 6803. Bioinformatics, biochemical and 3D modeling studies were used to reveal how suppressor mutations rescue ΔrpoZ cells in 3% CO2. Suppressor mutations were mapped to the ssr1600 gene. Ssr1600 was shown to function as an anti-σ factor antagonist. The Slr1861 protein was identified as an anti-σ

Vertical Immobilization Method for Time-Lapse Microscopy Analysis in Filamentous Cyanobacteria.

A main event in bacterial cell division is the septation process, where the protein FtsZ is the key element. FtsZ polymerizes forming a ring-like structure (Z-ring) in the middle of the cell that serves as a scaffold for other division proteins. Super-resolution microscopy in bacterial models Escherichia coli and Bacillus subtilis showed that the Z-ring is discontinuous, while live cell imaging studies demonstrated that FtsZ moves along the ring by a mechanism known as treadmilling. To study the dynamics of FtsZ in vivo, a special cell placement in a vertical position is necessary for imaging

Antiviral activity of five filamentous cyanobacteria against coxsackievirus B3 and rotavirus.

Coxsackievirus B3 (CVB3) and rotavirus (RV) are pathogens of some chronic human diseases. The aim of this study was to determine in vitro antiviral activity of some cyanobacteria against RV and CVB3 infections. Five cyanobacteria were collected from Egypt, identified, and analyzed biochemically. Then, the inhibition of the cytopathic effect of RV and CVB3 viruses by cyanobacterial extracts was examined. Methanol extract of the cyanobacterial isolates showed high antiviral activity against CVB3 with Therapeutic index (TI) of 50.0, 30.0, 27.6, 16.6, and 20.0 for Leptolyngbya boryana, Arthrospira

The SOS response and functional diversification of the transcription factor LexA in cyanobacteria.

The SOS response is a widespread bacterial mechanism for coping with DNA-damaging stress. The key regulator of the process is the transcriptional repressor LexA, which represses expression of DNA-repair-related genes under nonstress conditions and undergoes self-cleavage to induce them under DNA-damaging conditions. Here, we gained insights into the SOS response and functional diversification of LexA in cyanobacteria by characterizing 2 model species, Synechocystis sp. PCC 6803 (S.6803) and Anabaena sp. PCC 7120 (A.7120), together with an early branching species Gloeobacter violaceus PCC 7421

Content and bioavailability of iron from cyanobacteria and microalgae as food sources.

The consumption of microalgae as a novel food in diets holds significant potential as a sustainable approach to addressing iron deficiency anaemia. The present study investigated the mineral content and bioavailability of iron from different cyanobacteria and microalgae samples. The bioaccessibility of the samples was analysed using in vitro simulated gastrointestinal digestion and quantified using inductively coupled plasma optical emission spectrometry (ICP-OES). The bioavailability of iron was measured using the Caco-2 cell model via inductively coupled plasma mass spectrometry (ICP-MS) and

Interaction of cyanobacteria and microplastics polystyrene spiked with pharmaceutical drug-paracetamol.

The microplastics polymer-polystyrene, polyethylene, polyvinyl chloride, etc. are now recognized as potent threats to the aquatic system due to the Trojan horse effect i.e., they adsorb other pollutants such as pharmaceutical drugs, organic solvents, metals etc. and act as a vector or carrier. Polystyrene (PS) is one of the most usable plastics worldwide that abundantly contaminates the aquatic body. However, to date, only a few studies have focused on the eco-toxic effects of polystyrene in combination with other pollutants. Therefore, in the present study, the effect of polystyrene (pristine
