Phytoplankton Toxins and Their Potential Therapeutic Applications: A Journey toward the Quest for Potent Pharmaceuticals.

Phytoplankton are prominent organisms that contain numerous bioactive substances and secondary metabolites, including toxins, which can be valuable to pharmaceutical, nutraceutical, and biotechnological industries. Studies on toxins produced by phytoplankton such as cyanobacteria, diatoms, and dinoflagellates have become more prevalent in recent years and have sparked much interest in this field of research. Because of their richness and complexity, they have great potential as medicinal remedies and biological exploratory probes. Unfortunately, such toxins are still at the preclinical and cli

Harmful cyanobacteria-diatom/dinoflagellate blooms and their cyanotoxins in freshwaters: A nonnegligible chronic health and ecological hazard.

Human and ecological health depends on the vitality of freshwater systems, but these are increasingly threatened by cyanotoxins released from harmful algal blooms (HABs). Periodic cyanotoxin production, although undesirable, may be tolerable when there is enough time for cyanotoxins to degrade and dissipate in the environment, but the year-round presence of these toxins will be a chronic health for humans and ecosystems. The purpose of this critical review is to document the seasonal shifts of algal species and their ecophysiological acclimatation to dynamic environmental conditions. We discus

Aerosolized Cyanobacterial Harmful Algal Bloom Toxins: Microcystin Congeners Quantified in the Atmosphere.

Cyanobacterial harmful algal blooms (cHABs) have the potential to adversely affect public health through the production of toxins such as microcystins, which consist of numerous molecularly distinct congeners. Microcystins have been observed in the atmosphere after emission from freshwater lakes, but little is known about the health effects of inhaling microcystins and the factors contributing to microcystin aerosolization. This study quantified total microcystin concentrations in water and aerosol samples collected around Grand Lake St. Marys (GLSM), Ohio. Microcystin concentrations in water

Exposure to Aerosolized Algal Toxins in South Florida Increases Short- and Long-Term Health Risk in Drosophila Model of Aging.

Harmful algal blooms (HABs) are a rising health and environmental concern in the United States, particularly in South Florida. Skin contact and the ingestion of contaminated water or fish and other seafood have been proven to have severe toxicity to humans in some cases. However, the impact of aerosolized HAB toxins is poorly understood. In particular, knowledge regarding either the immediate or long-term effects of exposure to aerosolized cyanotoxins produced by freshwater blue-green algae does not exist. The aim of this study was to probe the toxicity of aerosolized cyanobacterial blooms usi

PSP toxin release from the cyanobacterium Raphidiopsis brookii D9 (Nostocales) can be induced by sodium and potassium ions.

Paralytic shellfish poisoning (PSP) toxins are a group of naturally occurring neurotoxic alkaloids produced among several genera of primarily freshwater cyanobacteria and marine dinoflagellates. Although saxitoxin (STX) and analogs are all potent Na(+) channel blockers in vertebrate cells, the functional role of these compounds for the toxigenic microorganisms is unknown. Based upon the known importance of monovalent cations (such as sodium) in the maintenance of cellular homeostasis and ion channel function, we examined the effect of high extracellular concentrations of these ions on growth,

Cyanotoxin and domoic acid occurrence, relation with salinity, and potential recreational health risks in U.S. coasts in the 2015 US EPA National Coastal Condition Assessment.

In the first nationwide study of cyanotoxins in U.S. estuaries, algal toxins, cyanotoxins, chlorophyll, and salinity were measured in samples collected during the National Coastal Condition Assessment 2015. Anatoxin-a (ANAA), cylindrospermopsin (CYLS), domoic acid (DMAC), and microcystins (MCs) were detected by LC/MS/MS in 0.6, 0.9, 8.3, and 2.0 % of samples with mean concentrations of detections of 0.13, 0.13, 0.53, and 0.49 µg/L, respectively. MCs by ELISA were also evaluated, and 4.0 % of samples had measurable MCs with a mean of 0.78 µg/L. While ANAA and CYLS were detected south of 40° lat

Cyanotoxins and Food Contamination in Developing Countries: Review of Their Types, Toxicity, Analysis, Occurrence and Mitigation Strategies.

Cyanotoxins have gained global public interest due to their potential to bioaccumulate in food, which threatens human health. Bloom formation is usually enhanced under Mediterranean, subtropical and tropical climates which are the dominant climate types in developing countries. In this context, we present an up-to-date overview of cyanotoxins (types, toxic effects, analysis, occurrence, and mitigation) with a special focus on their contamination in (sea)food from all the developing countries in Africa, Asia, and Latin America as this has received less attention. A total of 65 publications have

Potential Endocrine Disruption of Cyanobacterial Toxins, Microcystins and Cylindrospermopsin: A Review.

Microcystins (MCs) and cylindrospermopsin (CYN), although classified as hepatotoxins and cytotoxins, respectively, have been shown to also induce toxic effects in many other systems and organs. Among them, their potential endocrine disruption (ED) activity has been scarcely investigated. Considering the increasing relevance of ED on humans, mammals, and aquatic organisms, this work aimed to review the state-of-the-art regarding the toxic effects of MCs and CYN at this level. It has been evidenced that MCs have been more extensively investigated than CYN. Reported results are contradictory, wit

Effects of rainfall patterns on toxic cyanobacterial blooms in a changing climate: between simplistic scenarios and complex dynamics.

Toxic cyanobacterial blooms represent a serious hazard to environmental and human health, and the management and restoration of affected waterbodies can be challenging. While cyanobacterial blooms are already a frequent occurrence, in the future their incidence and severity are predicted to increase due to climate change. Climate change is predicted to lead to increased temperature and changes in rainfall patterns, which will both have a significant impact on inland water resources. While many studies indicate that a higher temperature will favour cyanobacterial bloom occurrences, the impact o

Are known cyanotoxins involved in the toxicity of picoplanktonic and filamentous North Atlantic marine cyanobacteria?

Eight marine cyanobacteria strains of the genera Cyanobium, Leptolyngbya, Oscillatoria, Phormidium, and Synechococcus were isolated from rocky beaches along the Atlantic Portuguese central coast and tested for ecotoxicity. Strains were identified by morphological characteristics and by the amplification and sequentiation of the 16S rDNA. Bioactivity of dichloromethane, methanol and aqueous extracts was assessed by the Artemia salina bioassay. Peptide toxin production was screened by matrix assisted laser desorption/ionization time of flight mass spectrometry. Molecular analysis of the genes in

A review and assessment of cyanobacterial toxins as cardiovascular health hazards.

Eutrophicated waters frequently support bloom-forming cyanobacteria, many of which produce potent cyanobacterial toxins (cyanotoxins). Cyanotoxins can cause adverse health effects in a wide range of organisms where the toxins may target the liver, other internal organs, mucous surfaces and the skin and nervous system. This review surveyed more than 100 studies concerning the cardiovascular toxicity of cyanotoxins and related topics. Over 60 studies have described various negative effects on the cardiovascular system by seven major types of cyanotoxins, i.e. the microcystin (MC), nodularin (NOD

Cyanobacterial blooms: statistical models describing risk factors for national-scale lake assessment and lake management.

Cyanobacterial toxins constitute one of the most high risk categories of waterborne toxic biological substances. For this reason there is a clear need to know which freshwater environments are most susceptible to the development of large populations of cyanobacteria. Phytoplankton data from 134 UK lakes were used to develop a series of Generalised Additive Models and Generalised Additive Mixed Models to describe which kinds of lakes may be susceptible to cyanobacterial blooms using widely available explanatory variables. Models were developed for log cyanobacterial biovolume. Water colour and

Is the future blue-green? A review of the current model predictions of how climate change could affect pelagic freshwater cyanobacteria.

There is increasing evidence that recent changes in climate have had an effect on lake phytoplankton communities and it has been suggested that it is likely that Cyanobacteria will increase in relative abundance under the predicted future climate. However, testing such a qualitative prediction is challenging and usually requires some form of numerical computer model. Therefore, the lake modelling literature was reviewed for studies that examined the impact of climate change upon Cyanobacteria. These studies, taken collectively, generally show an increase in relative Cyanobacteria abundance wit

Matrix Effect of Diverse Biological Samples Extracted with Different Extraction Ratios on the Detection of β-N-Methylamino-L-Alanine by Two Common LC-MS/MS Analysis Methods.

Neurotoxin β-N-methylamino-L-alanine (BMAA) is hypothesized as an important pathogenic factor for neurodegenerative diseases such as amyotrophic lateral sclerosis/parkinsonism-dementia complex (ALS-PDC). Comparative study on the accuracy of BMAA analyzed by the regular LC-MS/MS methods is still limited for different biological matrices. In this study, a free-BMAA sample of cyanobacterium and BMAA-containing positive samples of diatom, mussel, scallop, and oyster were extracted with varied extraction ratios (ER) ranging from 1:20 to 1:2000. These extracts were then purified by MCX cartridges. A
