Photoreceptors of the retina and pinealocytes of the pineal gland share common components of signal transduction.

Light absorbed by retinal photoreceptors triggers a cascade of reactions that initiate cGMP hydrolysis, cation channel closure and membrane hyperpolarization. Down-regulation of the cascade involves additional proteins that interfere with amplification along the cascade. Pinealocytes are activated by norepinephrine during the dark phase of the day/night cycle. Mature pinealocytes of the mammalian pineal express the known photoreceptor proteins that are implicated in down-regulation of the visual cascade, but the cascade components that produce cGMP hydrolysis and membrane hyperpolarization are

The cyclic dinucleotide c-di-AMP is an allosteric regulator of metabolic enzyme function.

Cyclic di-adenosine monophosphate (c-di-AMP) is a broadly conserved second messenger required for bacterial growth and infection. However, the molecular mechanisms of c-di-AMP signaling are still poorly understood. Using a chemical proteomics screen for c-di-AMP-interacting proteins in the pathogen Listeria monocytogenes, we identified several broadly conserved protein receptors, including the central metabolic enzyme pyruvate carboxylase (LmPC). Biochemical and crystallographic studies of the LmPC-c-di-AMP interaction revealed a previously unrecognized allosteric regulatory site 25 Å from the

Evidence for an A2 adenosine receptor in guinea pig lung.

Adenosine receptors in guinea pig lung were characterized by measurement of cyclic AMP formation and radioligand binding. 5'-N-Ethylcarboxamidoadenosine (NECA) increased cyclic AMP levels in lung slices about 4-fold over basal values with an EC50 of 0.32 mumol/l. N6-R-(-)-Phenylisopropyladenosine (R-PIA) was 5-fold less potent than NECA. 5'-N-Methylcarboxamidoadenosine (MECA) and 2-chloroadenosine had EC50-values of 0.29 and 2.6 mumol/l, whereas adenosine and inosine had no effect. The adenosine receptors in guinea pig lung can therefore be classified as A2 receptors. Several xanthine derivati

Regulation of delta-opioid receptor mRNA levels by receptor-mediated and direct activation of the adenylyl cyclase-protein kinase A pathway.

The effects of activation of the adenylyl cyclase-protein kinase A pathway on the expression of delta-opioid receptor mRNA in the NG108-15 neuroblastoma x glioma cell line has been investigated. Activation of prostaglandin E1 (PGE1) receptors, which are positively coupled to adenylyl cyclase, resulted in a reduction in delta-receptor messenger RNA levels. Direct stimulation of adenylyl cyclase by forskolin or treatment of cells with the cyclic AMP analogue dibutyryl cyclic AMP (db-cAMP) mimicked the effect of PGE1. Down-regulation in receptor protein levels, as measured by loss of radioligand

Absence of 1-Methyladenine Production in Follicle Cells Obtained from Starfish Ovaries in the Post-Spawning Season: (starfish follicle celis/1-MeAde/GSS/cAMP/post-spawning season).

Follicle cells from the ovaries of starfish Asterina pectinifera collected in the breeding season and then kept in an aquarium for three months did not produce 1-methyladenine (1-MeAde) in response to genad-stimulating substance (GSS). The cyclic AMP content of follicle cells of the ovaries was much lower in the post-spawning season than in the spawning season. In the post-spawning season, the cyclic AMP level was increased slightly by GSS, but nor insufficiently for production of 1-MeAde. Addition of 3-isobutyl-1-methylxanthine, a potent phosphodiesterase inhibitor, stimulated the productions

Positive inotropic and chronotropic effects of 8-substituted derivatives of cyclic AMP and activation of protein kinase A.

Inotropic and chronotropic effects of 8-substituted derivatives of cyclic AMP (8-SH, 8-SCH2C6H5, 8-N3, 8-SCH3, 8-Br, 8-N(CH3)2, 8-OCH3) were studied using guinea pig atrial and ventricular muscle preparations and correlated with the activation of the protein kinase A derived from the bovine myocardium. All the compounds produced positive inotropic and chronotropic effects. A good correlation was found between the chronotropic effect and the activation of the enzyme, while such a good correlation was not found between the enzyme activation and the positive inotropic effect. However, after treat

Increased cholera toxin-, and forskolin-induced cyclic AMP accumulations in psoriatic involved versus uninvolved or normal human epidermis.

Psoriatic involved epidermis reveals variously altered receptor-adenylate cyclase responses; among them the most prominent is defective beta-adrenergic adenylate cyclase response, which is normally the major receptor-adenylate cyclase system of human epidermis. It is known that activation of hormone-stimulated adenylate cyclase, a membrane-bound enzyme complex, requires functional coupling of at least 3 distinct subunits: 1) receptor subunit (R), 2) guanine nucleotide binding protein (G), and 3) catalytic subunit (C). The precise nature of the beta-adrenergic defect in the psoriatic epidermis,

Sensing a problem here: nucleotide signaling in immunity.

Cyclic nucleotides are shared messengers of antiviral immunity across bacteria and animals, yet the same chemical signal can produce strikingly different cellular outcomes. Recent studies show that bacteria generate the mammalian cGAS (cyclic GMP-AMP synthase product), 2'-5',3'-5'-cyclic GMP-AMP, during phage infection and use it to activate membrane-disrupting SAVED (SMODS-associated and fused to various effector domains)-family effectors. Structural work further reveals how ligand binding drives receptor oligomerization, filament formation, and vertical membrane shearing. In parallel, viral

Forskolin stimulates adenylate cyclase and iodine metabolism in thyroid.

Forskolin is a potent activator of the cyclic AMP-generating system in many tissues. In dog thyroid slices, the enhancement of cyclic AMP level was rapid, sustained in the presence of forskolin, but easily reversible after its withdrawal. Contrary to TSH, forskolin induced little apparent desensitization. Forskolin potentiated the effects of TSH, PGE1 and cholera toxin. However, the forskolin-induced cyclic AMP accumulation was still sensitive to inhibitors of dog thyroid adenylate cyclase such as iodide, norepinephrine and adenosine. As fluoride, but contrary to TSH and PGE1, forskolin stimul

Differential adrenergic regulation of rat pineal cyclic AMP production and N-acetyltransferase activity during postnatal development: involvement of G alpha s and G alpha i1-2 proteins.

We have studied why rat pineal N-acetyltransferase (NAT) activity is relatively insensitive to isoproterenol in young rats when compared with adult rats. We report that activation by isoproterenol of pineal cyclic AMP production and NAT activity is higher in adult than in 2-week-old rats. However, the effect of dibutyryl cyclic AMP, which enters the pinealocyte and duplicates the effect of cyclic AMP, on NAT activity was similar at both ages. Moreover, we found that both alpha- and beta-adrenergic receptors are highly specific at both ages, since the binding of the specific radioligands used t

Multiple signal transduction pathways lead to extracellular ATP-stimulated mitogenesis in mammalian cells: II. A pathway involving arachidonic acid release, prostaglandin synthesis, and cyclic AMP accumulation.

We have previously shown that extracellular ATP acts as a mitogen via protein kinase C (PKC)-dependent and independent pathways (Wang, D., Huang, N., Gonzalez, F.A., and Heppel, L.A. Multiple signal transduction pathways lead to extracellular ATP-stimulated mitogenesis in mammalian cells. I. Involvement of protein kinase C-dependent and independent pathways in the mitogenic response of mammalian cells to extracellular ATP. J. Cell. Physiol., 1991). The present aim was to determine if metabolism of arachidonic acid, resulting in prostaglandin E2 (PGE2) synthesis and elevation of cAMP levels, pl

Dopamine D1 receptor modulates the voltage-gated sodium current in rat striatal neurones through a protein kinase A.

1. Whole-cell recordings were made from striatal neurones obtained from neonatal rats and maintained in primary cultures. The effects of dopamine D1 receptor activation were studied on the voltage-gated sodium current. 2. Bath application of a specific D1 agonist, SKF38393 (1 microM), reduced the neuronal excitability recorded in current-clamp by increasing the threshold for generation of action potentials. 3. In voltage-clamp recordings, SKF38393 (1 microM) reversibly reduced the peak amplitude of the sodium current by 37.8 +/- 4.95%. This effect was reversed by the D1 antagonist SCH23390 and

cAMP pathway alterations from the cell surface to the nucleus in adrenocortical tumors.

The cyclic AMP (cAMP) pathway plays a major role in the development of endocrine tissues and various molecular defects of key components of this pathway (G protein, receptors, PKA, ...) have been observed in endocrine tumors. Hypersecretion of adrenocorticotropin hormone (ACTH), the key activator of the cAMP pathway in adrenal cortex, is associated with adrenocortical hyperplasia and cortisol oversecretion (Cushing's syndrome). The best example of "illegitimate" membrane receptors expression reported is the abnormal expression of the adenylyl cyclase activating gastric inhibitory peptide recep

Actions of adenosine A1 and A2 receptor antagonists on CFTR antibody-inhibited beta-adrenergic mucin secretion response.

1. The cystic fibrosis gene protein, the cystic fibrosis transmembrane conductance regulator (CFTR) acts as a chloride channel and is a key regulator of mucin secretion. The mechanism by which 3-isobutyl-1-methylxanthine (IBMX) corrects the defect in CFTR mediated beta-adrenergic stimulation of mucin secretion has not been determined. The present study has investigated the actions of adenosine A1 and A2 receptor antagonists to determine whether ability to stimulate mucin secretion correlates with correction of CFTR antibody inhibited beta-adrenergic response and whether excessive cyclic AMP ri

Cyclic Nucleotide-Directed Protein Kinases in Cardiovascular Inflammation and Growth.

Cardiovascular disease (CVD), including myocardial infarction (MI) and peripheral or coronary artery disease (PAD, CAD), remains the number one killer of individuals in the United States and worldwide, accounting for nearly 18 million (>30%) global deaths annually. Despite considerable basic science and clinical investigation aimed at identifying key etiologic components of and potential therapeutic targets for CVD, the number of individuals afflicted with these dreaded diseases continues to rise. Of the many biochemical, molecular, and cellular elements and processes characterized to date tha

The ATP-induced inward current in mouse lacrimal acinar cells is potentiated by isoprenaline and GTP.

1. ATP activates calcium (Ca2+) influx in mouse lacrimal acinar cells in the absence of phosphoinositide hydrolysis. Extracellular ATP (1 mM) activates receptor-operated cation channels, promoting entry of Na+ and Ca2+ (inward current). This Ca2+ influx in turn activates K+ channels resulting in a delayed, outward, current component. The present study uses patch-clamp current recording techniques to investigate the role of beta-adrenoceptor mechanisms, intracellular cyclic AMP and GTP in the regulation of the ATP-induced inward currents. 2. The beta-adrenoceptor agonist, isoprenaline (1 microM

Puerarin restores testicular steroidogenesis via VDR-cyclic AMP/PKA signaling in a chronic unpredictable stress (CUS) rat model.

Chronic psychological stress is a well-established factor contributing to male infertility by disrupting testicular function through oxidative stress, inflammation, and hormonal imbalance. This study aimed to investigate the protective effects of puerarin, a natural isoflavonoid, on chronic unpredictable stress (CUS)-induced testicular damage, with a specific focus on the vitamin D receptor (VDR)-mediated cyclic AMP/PKA signaling pathway. Adult male rats were divided into four groups (n = 6/group): Control, CUS, CUS + Puerarin (80 mg/kg; T1), and CUS + Puerarin (120 mg/kg; T2). CUS was given f
