Screening of Reference Genes for Quantitative Real-Time PCR Analysis in Tissues and during Testis Development, and Application to Analyze the Expression of kifc1 in Hemibarbus labeo (Teleostei, Cypriniformes, Cyprinidae).

The selection of proper reference genes is vital for ensuring precise quantitative real-time PCR (qPCR) assays. This study evaluates the stability of the expression of nine candidate reference genes in different tissues and during testicular development in H. labeo. The results show that eef1a is recommended as a reference gene for qPCR analysis in tissues and during testicular development. Furthermore, we evaluated the optimal number of reference genes needed when calculating gene expression levels using the geomean method, revealing that two reference genes are sufficient. Specifically, eef1

Two new species of Myxobolus Bütschli, 1882 (Cnidaria: Bivalvulida: Myxobolidae) infecting the gill of the black redhorse, Moxostoma duquesnei (Lesueur) (Cypriniformes: Catostomidae) in the Little Tennessee River Basin, North Carolina.

Two new species of Myxobolus Bütschli, 1882 (Bivalvulida: Myxobolidae) are described from the gill of the black redhorse (Moxostoma duquesnei [Leueur][Cypriniformes: Catostomidae]) from the Little Tennessee River Basin, North Carolina, United States. Myxobolus branchiofilum n. sp. infects lumen of the lamellar arterioles and Myxobolus branchiopecten n. sp. infects the bone and cartilage at the tip of the gill rakers. They differ from all congeners by a combination of myxospore dimensions and the presence or absence of an iodinophilic vacuole in the sporoplasm, mucous envelope, intercapsular pr

De Novo transcriptome assembly of Ligula intestinalis (Linnaeus, 1758) Gmelin, 1790 (Cestoda: Diphyllobothriidae) plerocercoids and their host, the common bream Abramis Brama L. (Cypriniformes: Leuciscidae): model for studying host-parasite interactions.

BACKGROUND: Ligula intestinalisis a parasitic tapeworm with a complex life cycle. During the procercoid stage, they parasitize copepods. Later, the cestodes develop into plerocercoids in the body cavity of plankton-feeding fish before finally maturing within the intestines of piscivorous birds. The impact of plerocercoid infection on hosts may be dramatic, including organ compression, disrupted maturation and reproduction, and alter behavior. These effects, which are mediated by neuroendocrine and immunomodulatory mechanisms, can also lead to a mass fish kill. L. intestinalis and their host fi

Plagioporus wataugaensis n. sp. (Digenea: Opecoelidae) infecting intestine of northern hogsucker, Hypentelium nigricans, and white sucker, Catostomus commersonii, (Cypriniformes: Catostomidae) from the eastern USA, including an emended diagnosis, key to Nearctic congeners, and phylogenetic analysis.

We describe a new species of Plagioporus Stafford, 1904 infecting the intestine of two catostomids in the eastern USA. We emend Plagioporus to account for Nearctic congeners having ceca terminating at the level of the testes (previously diagnosed as having ceca terminating in the post-testicular space only) and testes in the posterior body extremity (a feature not previously considered as having generic importance). Of the accepted Nearctic species, Plagioporus wataugaensis n. sp. resembles Plagioporus serotinus Stafford, 1904, Plagioporus hypentelii Hendrix, 1973, and Plagioporus hageli Fayto

Convergent genome streamlining accompanies independent miniaturization in the world's smallest fishes.

Miniaturization, the reduction of adult body size to an extreme degree, has evolved repeatedly across vertebrates. Yet its genomic underpinnings remain poorly understood. Cypriniformes, the most species-rich order of freshwater fishes, contains multiple miniaturized lineages that have evolved via contrasting developmental processes. Proportioned dwarfs are tiny-bodied but otherwise morphologically similar to larger relatives, while progenetic miniatures exhibit developmental truncation and retain larval-like anatomical features into adulthood. Using a new time-calibrated phylogeny of 309 cypri

Genome size estimation and its associations with body length, chromosome number and evolution in teleost fishes.

Precise estimation of genome size (GS) is vital for various genomic studies, such as deciding genome sequencing depth, genome assembly, biodiversity documentation, evolution, genetic disorders studies, duplication events etc. Animal Genome Size Database provides GS of over 2050 fish species, which ranges from 0.35 pg in pufferfish (Tetraodon nigroviridis) to 132.83 pg in marbled lungfish (Protopterus aethiopicus). The GS of majority of the fishes inhabiting waters of Indian subcontinent are still missing. In present study, we estimated GS of 51 freshwater teleost (31 commercially important, 7

Danionella cerebrum.

Model organisms are bargains with biology. Squid gave neurophysiology an oversized cable. Arabidopsis gave plant biology a weed that grew too fast to ignore. Planarians gave regeneration biology a body that could be cut into questions. Danionella cerebrum offers a different bargain still: a sexually mature vertebrate whose brain and body remain tiny, transparent and accessible.

Identification of larval Gnathostoma obtained from imported Chinese loaches.

We observed the morphological characteristics and identified the species of gnathostome larvae obtained from the imported Chinese loaches. The early third-stage larvae (EL3) were collected from viscera of the loaches and a part of them were infected to mice. The advanced third-stage larvae [AdL3] were recovered from the mice at 4 weeks post-infection, both larval worms were fixed with 10% formalin, cleared in alcohol-glycerin solution, mounted with glycerin-jelly, and observed. A total of 369 EL3 were collected from viscera of 9,493 Chinese loaches. The whole body of EL3 was covered with about

Zebrafish (Danio rerio) as a Model for Understanding the Process of Caudal Fin Regeneration.

After its introduction for scientific investigation in the 1950s, the cypriniform zebrafish, Danio rerio, has become a valuable model for the study of regenerative processes and mechanisms. Zebrafish exhibit epimorphic regeneration, in which a nondifferentiated cell mass formed after amputation is able to fully regenerate lost tissue such as limbs, heart muscle, brain, retina, and spinal cord. The process of limb regeneration in zebrafish comprises several stages characterized by the activation of specific signaling pathways and gene expression. We review current research on key factors in lim

Enobarbus is a synonym of Lepidocephalichthys (Osteichthyes, Cobitidae).

External and internal morphological characters of the holotype of Enobarbus maculatus were examined and compared with those of other cobitid species to verify whether Enobarbus is a distinct genus. Analyses of radiographs demonstrated that the neural spines and the pterygiophores of the dorsal fin are of irregular shape, often doubled, reduced or bent. The holotype of E. maculatus bears an ossified structure on the last pectoral-fin ray, a structure otherwise known only from males of the genus Lepidocephalichthys. In Lepidocephalichthys, the shape of the ossified structure is species-specific,

Eidinemacheilus, a new generic name for Noemacheilus smithi Greenwood (Teleostei; Nemacheilidae).

Eidinemacheilus, new genus, is described for the troglomorphic Noemacheilus smithi Greenwood. The sole species of the genus is endemic to an aquifer in the Karun River drainage in the Iranian Zagros Mountains. Eidinemacheilus smithi was identified as a species in Paracobitis as both genera share an elevated dorsal adipose crest, a character state being widespread in nemacheilid loaches. Eidinemacheilus is distinguished from all other genera of Nemacheilidae in the Middle East by having 7+7 branched caudal-fin rays, a strongly reduced head canal system, no pigmentation and no eyes. The genome assemblies of the Tui chub, Siphateles bicolor, and Arroyo chub, Gila orcuttii.

We present genome assemblies for two cyprinoid fishes, the tui chub (Siphateles bicolor) and the arroyo chub (Gila orcuttii). These fishes are ecologically important representatives of native fish assemblages in the western United States and are both species of conservation concern. The two species hybridize where introductions bring them into contact, with potentially important ecological and evolutionary implications that have not yet been thoroughly examined from a genomic perspective. We present de novo assemblies for both species, representing the first scaffold-level genomes within their

Neuroanatomical relationships between FMRFamide-immunoreactive components of the nervus terminalis and the topology of olfactory bulbs in teleost fish.

The nervus terminalis (NT) is the most anterior of the vertebrate cranial nerves. In teleost fish, the NT runs across all olfactory components and shows high morphological variability within this taxon. We compare the anatomical distribution, average number and size of the FMRFamide-immunoreactive (ir) NT cells of fourteen teleost species with different positions of olfactory bulbs (OBs) with respect to the ventral telencephalic area. Based on the topology of the OBs, three different neuroanatomical organizations of the telencephalon can be defined, viz., fish having sessile (Type I), pseudose

Transcriptomic analysis of intermuscular bone development in barbel steed (Hemibarbus labeo).

Intermuscular bones (IBs), which are little, bony spicules in muscle, are embedded in lower teleosts' myosepta. Despite the importance of studying IB development in freshwater aquaculture species, the genes associated with IB development need to be further explored. In the present study, we identified four stages of IB development in barbel steed (Hemibarbus labeo), namely stage 1: IBs have not emerged, stage 2: a few small IBs have emerged in the tail, stage 3: longer IBs gradually emerged in the tail and stage 4: all of the IBs in the tail are mature and long, via Alizarin red staining. Developmental stages and germ cell lineage of the loach (Misgurnus anguillicaudatus).

The staging of embryonic and larval development, and the germ cell lineage of the loach, Misgurnus anguillicaudatus, are described. Fertilized eggs were obtained by artificial insemination. For the convenience of detailed observation and photography of the external appearance, we use dechorionated embryos. Through a series of operations, these embryos were cultured at 20 degrees C in an incubator. Embryonic and larval development of the loach was divided into five periods: cleavage, blastula, gastrula, segmentation, and hatching. Stages were assigned within each of these periods. A cyclical switch of gametogenic pathways in hybrids depends on the ploidy level.

The cellular and molecular mechanisms governing sexual reproduction are conserved across eukaryotes. Nevertheless, hybridization can disrupt these mechanisms, leading to asexual reproduction, often accompanied by polyploidy. In this study, we investigate how ploidy level and ratio of parental genomes in hybrids affect their reproductive mode. We analyze the gametogenesis of sexual species and their diploid and triploid hybrids from the freshwater fish family Cobitidae, using newly developed cytogenetic markers. We find that diploid hybrid females possess oogonia and oocytes with original (dipl

The chromosome-level genome and key genes associated with mud-dwelling behavior and adaptations of hypoxia and noxious environments in loach (Misgurnus anguillicaudatus).

BACKGROUND: The loach (Misgurnus anguillicaudatus), the most widely distributed species of the family Cobitidae, displays a mud-dwelling behavior and intestinal air-breathing, inhabiting the muddy bottom of extensive freshwater habitats. However, lack of high-quality reference genome seriously limits the interpretation of the genetic basis of specialized adaptations of the loach to the adverse environments including but not limited to the extreme water temperature, hypoxic and noxious mud environment. RESULTS: This study generated a 1.10-Gb high-quality, chromosome-anchored genome assembly, w

Revision of the Schistura cincticauda species group (Teleostei, Nemacheilidae) using molecular and morphological markers.

To approach the taxonomy of large and complex animal groups it is of advantage to focus on species groups with shared derived character state. We investigate the composition, morphological characteristics and relationships of and within the Schistura cincticauda species group, whose members are small freshwater fishes that inhabit streams and rivers in eastern Myanmar and western and southern Thailand. A phylogenetic analysis using molecular genetic markers demonstrated the monophyly of this group; a combined genetic and morphological analysis revealed the inclusion of at least twelve species.
