The effect of a microfilament-disrupting drug, cytochalasin B, on 6-hourly and daily eruption rates of the rat mandibular incisor.

The drug did not affect 6-hourly and daily eruption rates of the unimpeded mandibular incisor of the rat. It is suggested that the tooth eruption is not inhibited by the disruption of microfilaments because the disruption was not associated with the motility of the cells and/or that the motility of the cells is not associated with the motive force of tooth eruption. Further studies may be necessary to confirm that the effects of cytochalasin B are on both microfilaments and motility of the periodontal fibroblasts.

Difference in polynucleation of cultured cells from human mammary tumors and normal mammary glands on treatment with cytochalasin B.

In order to study the biological nature of various mammary tumors, differences in the formation of polynuclear cells after the administration of cytochalasin B were investigated in cultures of human mammary tumors and normal mammary gland. When cytochalasin B was applied to the cultures, polynuclear cells increased in all cancer cases (6.1% on average), but relatively little effect was seen in cases of benign tumors and normal mammary gland (less than 1.1% on average). From these results, it appears that the difference in polynucleation on treatment with cytochalasin B may be useful as a biolo

Superoxide anion-generating activities of macrophages as studied by using cytochalasin E and lectins as synergistic stimulants for superoxide release.

Treatment of macrophages with cytochalasin E in combination with a lectin was found to stimulate the generation of superoxide anions (O2-) very efficiently. The macrophages stimulated with concanavalin A, phytohemagglutinin or wheat germ agglutinin released superoxide, but cells pretreated with cytochalasin E released much greater amounts of superoxide, without notable lag time, upon stimulation with the lectin. Wheat germ agglutinin was found to be the most efficient stimulant among the lectins tested. Superoxide generation in guinea pig macrophages was shown to be dependent largely on cytopl

Multifunctional theranostic nanocarrier platform for microglia-selective cytochalasin D delivery to modulate neuroinflammation after acute brain injuries.

Targeting microglia to modulate neuroinflammation after acute brain injuries (ABIs) is promising but limited by poor blood-brain barrier (BBB) penetration and systemic toxicity of candidate agents. In addition, multifunctional platforms combining targeted therapy with real-time traceability remain limited. We developed and characterized a multifunctional theranostic, folate-functionalized, polydopamine (PDA)-coated mesoporous silica nanoparticle (FA-NP) system for microglia-selective delivery and intracellular release of cytoskeleton inhibitor Cytochalasin D (CytoD). FA-NPs exhibited ideal phy

Cytoskeleton-disrupting agent cytochalasin B reduces oxidative stress caused by high glucose in the human arterial smooth muscle.

The role of cytoskeleton dynamics in the oxidative stress toward human vasculature has been unclear. The current study examined whether the cytoskeleton-disrupting agent cytochalasin B reduces oxidative stress caused by high glucose in the human arterial smooth muscle. All experiments in the human omental arteries without endothelium or the cultured human coronary artery smooth muscle cells were performed in d-glucose (5.5 mmol/L). The exposure toward d-glucose (20 mmol/L) for 60 min reduced the relaxation or hyperpolarization to an ATP sensitive K+ channel (KATP) opener levcromakalim (10-8 to

Somatostatin inhibition of glucose-, tolbutamide-, theophylline, cytochalasin B-, and calcium-stimulated insulin release in monolayer cultures of rat endocrine pancreas.

Somatostatin inhibited insulin secretion stimulated by glucose, tolbutamide, glucose-theophylline, glucose-cytochalasin B, and calcium in monolayer cell cultures of neonatal rat endocrine pancreas. Both 2-deoxyglucose-inhibited glucose-induced insulin release and basal insulin secretion occurring at glucose 1.7 mM were further reduced by somatostatin. In the presence of somatostatin, 1.0 mug/ml, insulin secretion due to glucose, tolbutamide, or glucose-cytochalasin B were inhibited to levels below the basal secretion seen with glucose 1.7 mM. However, insulin secretion stimulated by calcium, a

The effect of organophosphorus inhibitors, p-nitrophenol and cytochalasin B on cytotoxic killing of tumour cells by immune spleen cells, and the effect of shaking.

Organophosphorus (OP) agents are irreversible inhibitors of OP sensitive esterases. Their effect on the cytotoxic killing of 51Cr-labelled mastocytoma cells by immune spleen cells was assessed by the release of 51Cr. OP fluoridates inhibited cytotoxic killing whether used as a pretreatment before the addition of the labelled target cells or as a concurrent treatment. The effect of pretreatment was time, temperature and concentration dependent and was abolished by hydrolysis. It occurred at concentrations which did not inhibit the incorporation of [14C] leucine and uridine. It is tentatively su

Spontaneous rosette formation by lymphocytes. Differential effect of cytochalasin B on thymocytes and thymus-related peripheral blood lymphocytes in the guinea-pig.

In the present investigation the differences in the capacity of thymocytes (Th) and thymus-related peripheral blood lymphocytes (TBL) of the guinea-pig to form spontaneous rosettes with rabbit erythrocytes (RRBC) were further evaluated. Cytochalasin B (CB) inhibited TBL rosette formation but was largely ineffective with respect to Th. This suggests that membrane modulation and microfilament function are requirements for the formation of TBL but not of Th rosettes. A certain critical RRBC receptor density on the lymphocyte may be necessary for a stable rosette. Our results suggest that differen

Involvement of Ras/extracellular signal-regulated kinase, but not Akt pathway in risedronate-induced apoptosis of U937 cells and its suppression by cytochalasin B.

Although risedronate, a nitrogen containing bisphosphonate (BPs), strongly inhibits bone resorption by enhanced apoptosis of osteoclasts, its mechanism remained unclear. In this study, we investigated the molecular mechanism of risedronate-induced apoptosis of U937 cells, with a focus on extracellular signal-regulated kinase 1/2 (ERK 1/2) and protein kinase B (Akt) pathways, mitochondria-mediated apoptosis, and the effect of disruption of the actin cytoskeleton. Risedronate facilitated the relocation of Ras from membrane to cytosol through inhibited isoprenylation. Accordingly, risedronate sup

Effects of Clostridium botulinum C2 toxin and cytochalasin D on in vitro invasiveness, motility and F-actin content of a murine T-lymphoma cell line.

In order to investigate the role of microfilaments in the crawling movements of lymphoid cells, we have analyzed the effects of botulinum C2 toxin and of cytochalasin D (cytoD) on the actin cytoskeleton and on the motility of a BW5147 T-lymphoma-derived cell line. Actin was ADP-ribosylated by C2 toxin in the living cells, and this resulted in a time and dose-dependent disappearance of F-actin, as assessed by staining with labeled phalloidin. CytoD did not affect the amount of polymerized actin, but rather changed its distribution from a diffuse peripheral network to focal accumulations on one

Endophytic Xylaria sp. from Syzygium myrtifolium as a novel source of cytochalasin D with antimalarial activity against Plasmodium falciparum and anticancer activity against MCF-7 cells.

Endophytic fungi associated with Syzygium myrtifolium have significant potential as rapid sources of bioactive compounds for drug discovery. This study aimed to identify antimalarial and anticancer compounds from an endophytic fungal isolate Xylaria sp., associated with S. myrtifolium. The Xylaria sp. SmPdDh-4 isolate was identified using morphological characterisation and phylogenetic analysis. Metabolite profiling was performed using LC-HRMS and further confirmed by NMR spectroscopy, identifying cytochalasin D as the major compound, which was also confirmed by X-ray diffraction examination.

Simultaneous detection of X-chromosome loss and non-disjunction in cytokinesis-blocked human lymphocytes by in situ hybridization with a centromeric DNA probe; implications for the human lymphocyte in vitro micronucleus assay using cytochalasin B.

A methodology for the simultaneous detection of chromosome loss and gain in mammalian cells has been developed which is based upon the analysis of chromosome distribution in daughter nuclei of binucleated human lymphocytes. X-chromosome distribution was followed by in situ hybridization, using a commercial biotinylated DNA probe specific for the centromeric alphoid sequences of human X-chromosome. In order to optimize the experimental protocol for the use of cytokinesis-blocked lymphocytes in aneuploidy assays, the effect of harvest time and cytochalasin B (Cyt B) dosage upon chromosome distri

An important role of actin polymerization in the human zona pellucida-induced acrosome reaction.

The effects of inhibitors of actin polymerization and depolymerization, cytochalasins and phalloidin, on the human zona pellucida (ZP)-induced acrosome reaction (AR) were investigated. Motile spermatozoa, selected by swim-up technique from normozoospermic men, were incubated in medium with or without the actin modulators. Oocytes (four per test) which had failed to fertilize in vitro were added and incubation continued for 2 h. The spermatozoa bound to the ZP were dislodged by repeatedly aspirating the oocytes with a small-bore pipette and the status of the acrosomes was determined by fluoresc

Effect of thrombopoietin alone and a combination of cytochalasin B and ethylene glycol bis(beta-aminoethyl ether) N,N'-tetraacetic acid-AM on the survival and function of autologous baboon platelets stored at 4 degrees C for as long as 5 days.

BACKGROUND: PLTs stored at 22 degrees C have the potential for bacterial contamination, a problem that could be reduced by 4 degrees C storage. Nevertheless, PLTs stored at 4 degrees C exhibit a significantly reduced life span. This study was performed to determine whether treatment of PLTs with thrombopoietin or cytochalasin B plus ethylene glycol bis(beta-aminoethyl ether) N,N'-tetraacetic acid (EGTA)-AM could prevent exponential loss of PLTs stored at 4 degrees C. STUDY DESIGN AND METHODS: Autologous baboon PLTs were stored at 22 or 4 degrees C. The 4 degrees C stored PLTs were treated wit

Relationship of actin filament assembly to clearance of fibrinogen gold, GPIIb-IIIa complexes on spread platelets.

The present study has evaluated the influence of high concentrations of cytochalasins B and E on the detergent-resistant actin levels in fully spread platelets by PAGE gel electrophoresis, and the effects of the two inhibitors of new actin filament assembly on translocation of fibrinogen gold (Fgn/Au) labelled GPIIb-IIIa receptors on the surface-activated cells. Concentrations of 10(-4) M and 10(-5) M cytochalasin B and E reduced detergent-resistant actin in fully spread platelets to levels present in resting discoid platelets in suspension. Despite reduction of actin filaments to levels in re

Implications for treatment of variations in length of the first metacarpal in different types of triphalangeal thumbs.

BACKGROUND/AIM: Cytochalasin B is a mycogenic toxin that preferentially damages malignant cells through multiple mechanisms. The microfilament-disrupting agent inhibits cytokinesis, producing enlarged and multinucleated neoplastic cells without enlarging or producing multinucleated normal cells. In addition, cytochalasin B has been shown to induce apoptosis and to increase the mitochondrial activity of malignant cells. In spite of these pharmacological properties potentially exploitable in cancer chemotherapy, no cytochalasin congener or derivative and indeed no microfilament-directed agent ha

Effects of medium calcium, and agents affecting cytoskeletal function, on cellular volume and morphology in liver tissue in vitro.

The possible role of an exocytotic, vesicular mechanism in cellular volume regulation under iso-osmotic conditions has been studied in slices of rat liver. The effects of incubation conditions and agents affecting the actin cytoskeleton were examined for changes of water, ionic composition, and ultrastructure. Slices were pre-incubated at 1°C in an iso-osmotic buffered medium to induce swelling. Upon restoration to 37°C in the same medium, tissue lost water. The Na+-K+ adenosine triphosphatase (ATPase) inhibitor ouabain inhibited water extrusion of about 50%, an effect that was accompanied by
