Comparative Evaluation of Routine and Special Staining Techniques in Fine Needle Aspiration Cytology of Lymph Nodes.

BACKGROUND AND OBJECTIVE: Fine needle aspiration cytology (FNAC) is a fast, inexpensive, and least invasive form of diagnosis, which is commonly used in the assessment of lymphadenopathy. In developing nations such as India, malignancies and tuberculosis (TB) are some of the most frequent causes of the enlargement of the lymph nodes. Although regular cytology stains are commonly used, they are not always sensitive to some infections and neoplastic diseases. The objectives of this research were to compare and evaluate diagnostic using routine and special staining methods in FNAC of lymph nodes.

A Novel Manual "Centrifuged-Enhanced" Cytosmear Technique for Improving Hypocellular Cytology in the Diagnosis of Vitreoretinal Lymphoma.

PURPOSE: To develop a manual "centrifuged-enhanced cytosmear" technique for cytologic and immunohistochemical analysis of hypocellular vitreous biopsy specimens in vitreoretinal lymphoma (VRL). METHODS: Diffuse large B-cell lymphoma cells were diluted to simulate vitreous biopsy yields (1000-50,000 cells/100 µL) and fixed in PreservCyt. Samples were centrifuged in a standard laboratory centrifuge and deposited into 1000 cells/100 µL PAP pen-defined circles on charged slides. Smears were air-dried, methanol-fixed, and stained with toluidine blue and CD20 immunohistochemistry (IHC). Cell densit

Use of a double-lumen cytology brush catheter to allow double-guidewire technique for endoscopic interventions.

BACKGROUND AND AIMS: Serial stent placement may be necessary during endoscopic interventions, but the passage of a guidewire alongside an initial stent can be challenging, time-consuming, and sometimes unsuccessful. We describe a modification of a cytology brush catheter to allow simultaneous placement of 2 guidewires to facilitate serial stent placement and demonstrate its application in different scenarios. METHODS: This is a retrospective series of 3 patients with different conditions (acute cholecystitis, pancreas pseudocyst, and severe biliary stricture) in whom placement of a second gui

The Role of Liquid Based Cytology and Ancillary Techniques in the Peritoneal Washing Analysis: Our Institutional Experience.

BACKGROUND: The cytological analysis of peritoneal effusions serves as a diagnostic and prognostic aid for either primary or metastatic diseases. Among the different cytological preparations, liquid based cytology (LBC) represents a feasible and reliable method ensuring also the application of ancillary techniques (i.e immunocytochemistry-ICC and molecular testing). METHODS: We recorded 10348 LBC peritoneal effusions between January 2000 and December 2014. They were classified as non-diagnostic (ND), negative for malignancy-NM, atypical-suspicious for malignancy-SM and positive for malignancy

Cell tube block: a new technique to produce cell blocks from fluid cytology samples.

BACKGROUND: Cell blocks are widely used in human cytopathology. Several techniques have been proposed to convert fluid specimens into solid material, which after embedding in paraffin can be used for histochemistry, immunohistochemistry, or molecular testing. In contrast, only in the last few years, have cell blocks begun to be used in the veterinary field. OBJECTIVES: The purpose of the study was to present the production and features of cell tube blocks (CTB) with veterinary liquid samples. METHODS: Liquid samples including cerebrospinal fluid, cutaneous cyst fluid, pericardial and pleural

Cytological diagnosis of deep-seated cellular hemangioma of the parotid gland by using cell button technique.

Intraparotid hemangioma of the children is a rare neoplasm, posing diagnostic dilemma to the diagnosticians as well as treating clinicians. A 2-month-old male infant presented with a diffuse swelling in the parotid region since birth that was gradually increasing in size. The ultrasonography (USG) report was suggestive of a right intraparotid mass of uncertain etiology; whereas magnetic resonance imaging (MRI) report inclined toward a mass associated with chronic inflammatory pathology. Fine-needle aspiration cytology (FNAC) suggested two differentials - a vascular neoplasm of the parotid glan

The role of fine needle aspiration cytology (FNAC) in the investigation of superficial lymphadenopathy; uses and limitations of the technique.

Aspirates (n = 163) from 157 patients with enlarged superficial lymph nodes were obtained over a 5-year period in a combined surgical/FNAC clinic. A definitive diagnosis was achieved in over 77% of the cases: benign 52.7%, malignant 25.1%. The diagnostic accuracy was 94.4%, sensitivity 85.4%, and specificity 100%. The false-negative rate was 12.5% but decreased to 3.5% when lymphoma cases were excluded. There were 36 cases of metastatic disease, the majority of which were from a primary breast carcinoma. The main diagnostic difficulty was in distinguishing low-grade lymphoma from reactive hype

Cytologic evaluation of conjunctival epithelium using Cytobrush-S: value of slide preparation by ThinPrep technique.

Recent clinical trials have indicated that an automated smear apparatus (ThinPrep process) of sample preparation has great diagnostic sensitivity. In this study, conjunctival brush cytology prepared using the ThinPrep method was applied in ocular surface disorders especially for dry eye status. To assess its diagnostic value in cellular samples, 17 patients with keratoconjunctivitis sicca (KCS) and 10 normal volunteer patients were examined using this technique. Conjunctival cells from normal controls revealed fine chromatin and polyhedral cytoplasm without having keratinized cytoplasm. On the

Brush cytology technique in the detection of oesophageal carcinoma in the asymptomatic, high risk subject; a pilot survey.

This study assesses the reliability of abrasive brush cytology in the diagnosis of early cancer of the oesophagus among asymptomatic rural Ciskeians. An inexpensive locally manufactured brush biopsy capsule was used to obtain cytological material from 1336 subjects; 59% of them were brushed a second time. The technique appears to have a high sensitivity (90%) and specificity (99.9%). Three of five asymptomatic subjects diagnosed as having early oesophageal cancer refused further treatment, indicating the importance of health education as a prerequisite to the introduction of a screening progra

Tools, techniques, and challenges in preparing cytology specimens for ancillary studies: results of the ASC Optimizing Cytology and Small Biopsy Specimen Processing for Ancillary Studies task force survey.

INTRODUCTION: Ancillary testing on cytopathology and other small biopsy specimens is crucial for diagnosis and provides critical information to clinicians. Testing is dependent on preanalytic factors and would benefit from standardization of specimen collection protocols across laboratories. To assess institutional practices and areas of need for evidence-based standards, we surveyed current practices across cytopathology laboratories. MATERIALS AND METHODS: A twelve-question electronic survey was distributed to American Society of Cytopathology (ASC) members through email, social media, and

Evaluation of precision of guidance techniques in image guided fine needle aspiration cytology of thoracic mass lesions.

BACKGROUND: Transthoracic fine needle aspiration cytology (FNAC) is an established and safe technique for diagnosis of thoracic mass lesions. Computed tomography (CT) scan depicts clear anatomical details and provides access to any area of the body. It is, however, expensive and the needle is not passed in real time. Ultrasound is cheaper, radiation free, and allows real time monitoring. Its limitations are obscurement of lesions by aerated lung, smaller, deep seated, and cavitary lesions. AIMS: This study aims to compare sensitivity and specificity of CT scan and ultrasonography (USG) in tho

Immunophenotypic analysis of non-Hodgkin's lymphomas in cytologic specimens: a correlative study of immunocytochemical and flow cytometric techniques.

Most Non-Hodgkin's lymphomas(NHL) can be accurately diagnosed and classified based on morphologic and immunophenotypic findings on cytologic specimens. Immunophenotyping can be accomplished via immunocytochemistry (IC) or flow cytometry (FC). We reviewed our experience with 98 cytology specimens (70 fine-needle aspirates [FNA] and 28 effusions) that were submitted for immunophenotyping utilizing both IC and FC between January 1992 and December 1997 for the diagnosis of NHL. Eighty-five percent of the cases were immunophenotyped by both techniques. Among these there were only two discrepancies

Cytology Techniques Can Provide Insight into Human Placental Structure Including Syncytiotrophoblast Nuclear Spatial Organisation.

The aim of this study was to provide the first systematic description of human placental cytology appearances and to investigate syncytiotrophoblast nuclear organisation patterns using cytology techniques. Term placentas from normal pregnancies were sampled using fine-needle aspiration (FNA) and direct scrapes. Standard histological examination was also performed to exclude pathological changes in the placentas being studied. Both Papanicolaou-stained cytospin preparations and air-dried Giemsa slides from FNA provided high-quality material for cytological assessment with good cellularity. Amon
