Disseminated aspergillosis in dogs has been associated with Aspergillus terreus or A. deflectus infection. We report a case of disseminated A. versicolor infection presenting as diskospondylitis, osteomyelitis, and pyelonephritis. The diagnosis was made based on clinical, radiographic, and pathological findings. The etiologic agent was identified by fungal culture and internal transcribed spacer (ITS) ribosomal DNA (rDNA) sequencing. This is the first description of canine aspergillosis caused by A. versicolor.

To develop a rapid and quantitative diagnostic technique for the detection and identification of a wide range of fungi, an improved molecular method based on real-time PCR and the analysis of its products that targets the internal transcribed spacer (ITS) 2 region was established. The real-time PCR could quantitatively and specifically detect the ITS2 region from all 24 tested pathogenic fungal species at between 10(1) and 10(7) copies per test without amplification of bacterial or human DNA. The sequences of the primer-binding sites are conserved in the registered sequences of 34 other pathog

This study aimed to detect filarial parasites in blood samples of Japanese black bears (Ursus thibetanus japonicus) collected from Iwate Prefecture, Japan. Positive amplicons were obtained from 26 out of 30 samples by nested PCR targeting 18S ribosomal RNA gene and first internal transcribed spacer regions. DNA sequences of Mansonella sp. close to M. ozzardi and Dirofilaria sp. were detected for eight and 11 positive amplicons, respectively. Co-infection was detected for the remaining seven amplicons. Dirofilaria sp. was identified as D. ursi by further genetic analysis of 5S ribosomal RNA gen

During a survey of endophytic fungi associated with medicinal plants in Iran, three novel species of the genus Xenodidymella including X. euphorbiae, X. hamanae and X. kopetdagensis were isolated from asymptomatic tissues of Euphorbia kopetdaghi Prokh and Thymus transcaspicus Klokov. Species delimitation was conducted using a combination of morphological characterisation and multilocus phylogenetic analyses based on the ITS rDNA, partial beta-tubulin (tub2), and RNA polymerase II second largest subunit (rpb2) gene regions. Both Bayesian inference and Maximum Likelihood phylogenetic analyses re

We investigated Malassezia species and genotypes colonizing dogs, comparing those recovered from the ear canal with those from other anatomical body sites, as well as from diseased and healthy skin. The Malassezia isolates were obtained from four types of skin samples, i.e., diseased ear, diseased skin, healthy ear, and healthy skin. Sequences of the 26S ribosomal DNA region, the intergenic spacer 1 (IGS-1) and the internal transcribed spacer 1 (ITS-1) DNA region were analyzed. These confirmed the presence of Malassezia pachydermatis, which could be separated into three main sequence genotype

An examination of one specimen of Nile crocodile, Crocodylus niloticus (Laurenti, 1768), from Lake Turkana (Kenya), revealed the presence of two ascaridoid nematodes belonging to the genus Dujardinascaris Baylis, 1947. Dujardinascaris madagascariensis Chabaud & Caballero, 1966 was studied by scanning electron microscopy, redescribed, and differentiated from D. dujardini (Travassos, 1920). Dujardinascaris madagascariencsis is the second of the genus to be sequenced. An internal fragment of the small ribosomal subunit and nuclear ribosomal DNA internal transcribed spacer 2 region were amplified-

Madurella mycetomatis is the commonest cause of eumycetoma in Sudan and other countries in tropical Africa. Currently, the early diagnosis of mycetoma is difficult. In attempting to improve the identification of M. mycetomatis and, consequently, the diagnosis of mycetoma, we have developed specific oligonucleotide primers based on the sequence of the internal transcribed spacer (ITS) regions spacing the genes encoding the fungal ribosomal RNAs. The ITS regions were amplified with universal primers and sequenced, and then two sets of species-specific primers were designed which specifically amp

In this study, DNA was sequenced from adults of Patagifer Dietz, 1909 and Nephrostomum Dietz, 1909 (Digenea: Echinostomatidae) collected from avian definitive hosts in Nearctic, Neotropic, Palearctic and Afrotropic regions. Phylogenetic analyses of entire mitochondrial genomes, nuclear rDNA operons, and partial 28S, ITS2, cox1, and nad1 all indicate that Nephrostomum is synonymous with Patagifer, which is given priority, and that head-collar characters formerly used to separate these genera represent traits that vary among closely related congeneric species. We report four cases of scalp white piedra (SWP) in Brazilian female children. Morphological and physiological approaches gave inconsistent results for identifying Trichosporon to species level, while the sequencing of the intergenic spacer 1 region of ribosomal DNA accurately identified the agent of SWP as T. inkin. These cases emphasize the occurrence of this species causing this type of infection. The molecular identification of the suspected agent is needed for appropriate epidemiological surveillance of superficial mycoses caused by Trichosporon species.

Bryoria (Parmeliaceae, Ascomycota) is one of the dominant genera of hair lichens in western North America and is characteristic of high-elevation conifer forest ecosystems. In areas where Bryoria is abundant, it is common to find thalli in which the thalline filaments become conglutinated, forming brittle dead zones. After sampling Bryoria thalli across western Canada and the northwestern United States at different times of the year, we found that this dieback phenomenon is associated with the winter growth of a mold-forming basidiomycete. We report that this fungus belongs to Athelia (Athelia

Cecidophyopsis mites are important pests in all cultivation regions of Ribes causing bud galls and sterility. Despite their economic importance, the knowledge on Cecidophyopsis species infesting Ribes in various areas of the world is still deficient. The present study was carried out to identify Cecidophyopsis species occurring in Latvia on cultivated and wild Ribes, to assess their host range and gain insight into the genetic diversity of these insufficiently studied pests by use of multiplex PCR, rDNA sequences and morphological characters. Cecidophyopsis alpina, C. aurea, C. spicata and C.

The genus Taenia includes several species of veterinary and public health importance, but diagnosis of the etiological agent in definitive and intermediate hosts often relies on labor intensive and few specific morphometric criteria, especially in immature worms and underdeveloped metacestodes. In the present study, a multiplex PCR, based on five primers targeting the 18S rDNA and ITS2 sequences, produced a species-specific banding patterns for a range of Taenia spp. Species typing by the multiplex PCR was compared to morphological identification and sequencing of cox1 and/or 12S rDNA genes. Despite the fact that the non-human primates are our closest relatives and represent a species-rich mammalian group, little is known about their intestinal protistan parasites/commensals. Particularly, the intestinal trichomonads represent a neglected part of the fauna of the primate digestive system. We have established 30 trichomonad strains isolated from feces of 11 primate species kept in 3 Czech zoos and performed an analysis of their SSU rDNA and ITS1-5·8S rDNA-ITS2. Our results showed that intestinal trichomonads are rather common among non-human primates. Molecular phylogenetic analysi
