A biologically inspired repair mechanism for neuronal reconstructions with a focus on human dendrites.

Investigating and modelling the functionality of human neurons remains challenging due to the technical limitations, resulting in scarce and incomplete 3D anatomical reconstructions. Here we used a morphological modelling approach based on optimal wiring to repair the parts of a dendritic morphology that were lost due to incomplete tissue samples. In Drosophila, where dendritic regrowth has been studied experimentally using laser ablation, we found that modelling the regrowth reproduced a bimodal distribution between regeneration of cut branches and invasion by neighbouring branches. Ethanol-related increases in degenerating bodies in the Purkinje neuron dendrites of aging rats.

Chronic ethanol consumption in aging rats results in regression of Purkinje neuron (PN) dendritic arbors ([Pentney, 1995 Measurements of dendritic pathlengths provide evidence that ethanol-induced lengthening of terminal dendritic segments may result from dendritic regression. Non-allometric expansion and enhanced compartmentalization of Purkinje cell dendrites in the human cerebellum.

Purkinje cell (PC) dendrites are optimized to integrate the vast cerebellar input array and drive the sole cortical output. PCs are classically seen as stereotypical computational units, yet mouse PCs are morphologically diverse and those with multi-branched structure can receive non-canonical climbing fiber (CF) multi-innervation that confers independent compartment-specific signaling. While otherwise uncharacterized, human PCs are universally multi-branched. Do they exceed allometry to achieve enhanced integrative capacities relative to mouse PCs? To answer this, we used several comparative

Developing tough, fatigue-resistant and conductive hydrogels via in situ growth of metal dendrites.

Developing hydrogels with high conductivity and toughness via a facile strategy is important yet challenging. Herein, we proposed a new strategy to develop conductive hydrogels by growing metal dendrites. Water-soluble Sn2+ ions were soaked into the gel and then converted to Sn dendrites via an electrochemical reaction; the excessive Sn2+ ions were finally removed by water dialysis, accompanied by dramatic shrinkage of the gel. Based on in situ transformation from metal ions to dendrites, the method integrated the advantages of ionic conductive fillers, such as LiCl (uniform dispersion), and e

Reduced lysosomal density in neuronal dendrites mediates deficits in synaptic plasticity in Huntington's disease.

Huntington's disease (HD) usually causes cognitive disorders, including learning difficulties, that emerge before motor symptoms. Mutations related to lysosomal trafficking are linked to the pathogenesis of neurological diseases, whereas the cellular mechanisms remain elusive. Here, we discover a reduction in the dendritic density of lysosomes in the hippocampus that correlates with deficits in synaptic plasticity and spatial learning in early CAG-140 HD model mice. We directly manipulate intraneuronal lysosomal positioning with light-induced CRY2:CIB1 dimerization and demonstrate that lysosom

Local pruning of dendrites and spines by caspase-3-dependent and proteasome-limited mechanisms.

Synapse loss occurs normally during development and pathologically during neurodegenerative disease. Long-term depression, a proposed physiological correlate of synapse elimination, requires caspase-3 and the mitochondrial pathway of apoptosis. Here, we show that caspase-3 activity is essential--and can act locally within neurons--for regulation of spine density and dendrite morphology. By photostimulation of Mito-KillerRed, we induced caspase-3 activity in defined dendritic regions of cultured neurons. Within the photostimulated region, local elimination of dendritic spines and dendrite retra

Staphylococcus aureus activates dendrite elongation in dendritic cells.

Dendritic cells (DCs) are thought to extend dendrites to enhance the efficiency of antigen uptake and presentation. We previously reported that short-chain fatty acids (SCFAs), such as butyrate and valerate, promote dendrite extension in DCs. In this study, we found that the human pathogen Staphylococcus aureus also induces dendrite extension in DCs and investigated the underlying mechanisms. Dendrite extension in DC2.4 cells was induced not only by live S. aureus but also by heat-killed bacteria and purified peptidoglycan (PGN). DC2.4 cells lacking TLR2 or its adaptor protein MyD88 extend den

Structural alterations in osteocyte dendrites and lacunar-canalicular networks of the mandible of ovariectomized mice.

BACKGROUND: Postmenopausal osteoporosis (PMOP) leads to systemic bone metabolism disorders, significantly affecting oral bone health and complicating treatment. Osteocytes, the most abundant cells in bone tissue, play a crucial role in bone remodeling through cellular communication. However, the mineralized bone matrix surrounding osteocytes presents significant challenges for studying their dendritic networks. METHODS: Eight week old C57BL/6J female mice were randomly assigned to either a Sham group or an ovariectomized (OVX) group. The PMOP animal model was established via bilateral ovariec

Impact of 2D lanthanum sheets and iron oxide dendrites composites on phosphate recovery from wastewater.

Eutrophication and phosphorus depletion are the major concerns that have received increasing global attention. This study investigates the adsorption behaviour of phosphate onto α-Fe2O3 dendrites, composited with 2D lanthanum sheets adsorbents through comprehensive material characterization and kinetic experiments. The adsorbent was characterized using different analytical techniques to elucidate its structural, morphological, and surface properties. The impact of different lanthanum concentrations on phosphate sorption capacity was studied, and a sample with 0.6 g lanthanum nitrate (sample LF

Tau regulates Arc stability in neuronal dendrites via a proteasome-sensitive but ubiquitin-independent pathway.

Tauopathies are neurodegenerative disorders characterized by the deposition of aggregates of the microtubule-associated protein tau, a main component of neurofibrillary tangles. Alzheimer's disease (AD) is the most common type of tauopathy and dementia, with amyloid-beta pathology as an additional hallmark feature of the disease. Besides its role in stabilizing microtubules, tau is localized at postsynaptic sites and can regulate synaptic plasticity. The activity-regulated cytoskeleton-associated protein (Arc) is an immediate early gene that plays a key role in synaptic plasticity, learning, a

Na superionic conductor-type compounds as protective layers for dendrites-free aqueous Zn-ion batteries.

Aqueous rechargeable Zn-ion batteries (ARZIBs) have attracted much attention owing to their safety, high energy density and environmental friendliness. However, dendrite formation and corrosive reactions on Zn anode surface limit the development of ARZIBs. Here, Ga3+-doped NaV2(PO4)3 with Na superionic conductor (NASICON) structure [NVP-Ga(x), x = 0, 0.25, 0.5, 0.75] have been exploited as the high-efficiency artificial layer to stabilize Zn anode. The optimal NVP-Ga(0.5) layer can homogenize ion flux and promote uniform deposition of zinc, the dendrite growth and the parasitic reactions can b

Enhanced Electroreduction of Carbon Dioxide to Methanol Using Zinc Dendrites Pulse-Deposited on Silver Foam.

The electrocatalytic CO2 reduction reaction (CO2 RR) can dynamise the carbon cycle by lowering anthropogenic CO2 emissions and sustainably producing valuable fuels and chemical feedstocks. Methanol is arguably the most desirable C1 product of CO2 RR, although it typically forms in negligible amounts. In our search for efficient methanol-producing CO2 RR catalysts, we have engineered Ag-Zn catalysts by pulse-depositing Zn dendrites onto Ag foams (PD-Zn/Ag foam). By themselves, Zn and Ag cannot effectively reduce CO2 to CH3 OH, while their alloys produce CH3 OH with Faradaic efficiencies of appr

Detection of activity-dependent vasopressin release from neuronal dendrites and axon terminals using sniffer cells.

Our understanding of neuropeptide function within neural networks would be improved by methods allowing dynamic detection of peptide release in living tissue. We examined the usefulness of sniffer cells as biosensors to detect endogenous vasopressin (VP) release in rat hypothalamic slices and from isolated neurohypophyses. Human embryonic kidney cells were transfected to express the human V1a VP receptor (V1aR) and the genetically encoded calcium indicator GCaMP6m. The V1aR couples to Gq11, thus VP binding to this receptor causes an increase in intracellular [Ca2+] that can be detected by a ri

Retrograde plasticity and differential competition of bipolar cell dendrites and axons in the developing retina.

Most neurons function in the context of pathways that process and propagate information through a series of stages, e.g., from the sensory periphery to cerebral cortex. Because activity at each stage of a neural pathway depends on connectivity at the preceding one, we hypothesized that during development, axonal output of a neuron may regulate synaptic development of its dendrites (i.e., retrograde plasticity). Within pathways, neurons often receive input from multiple partners and provide output to targets shared with other neurons (i.e., convergence). Converging axons can intermingle or occu

Normal distribution of VGLUT1 synapses on spinal motoneuron dendrites and their reorganization after nerve injury.

Peripheral nerve injury induces permanent alterations in spinal cord circuitries that are not reversed by regeneration. Nerve injury provokes the loss of many proprioceptive IA afferent synapses (VGLUT1-IR boutons) from motoneurons, the reduction of IA EPSPs in motoneurons, and the disappearance of stretch reflexes. After motor and sensory axons successfully reinnervate muscle, lost IA VGLUT1 synapses are not re-established and the stretch reflex does not recover; however, electrically evoked EPSPs do recover. The reasons why remaining IA synapses can evoke EPSPs on motoneurons, but fail to tr

Vacant postsynaptic densities on supraoptic dendrites of adult rats diminish in number with chronic stimuli.

In earlier ultrastructural studies of the supraoptic nucleus in adult rats we noted "free" and incompletely covered postsynaptic densities (collectively referred to here as vacant postsynaptic densities) on dendritic shafts. Free postsynaptic densities have been reported in other parts of the central nervous system of normal rodents. We investigated the possibility that physiological activation of the supraoptic cells, which produces changes in many aspects of their morphology, would alter the incidence of the free or incompletely covered postsynaptic densities on dendrites in the supraoptic b

Dendritic transport. I. Colchicine stimulates the transport of lysosomal enzymes from cell bodies to dendrites.

Injection of colchicine into the lateral cerebral ventricle of the rat was found to induce a paradoxical translocation of two lysosomal enzymes, dipeptidyl peptidase II (Dpp II) and acid phosphatase, from the soma of neurons to the dendrites. Following a single injection of colchicine, neuronal somata, which normally contain the bulk of these lysosomal enzymatic activities, become depleted of these enzymes, whereas dendrites become abnormally enriched. All neurons which contained these enzymes, except those of the mesencephalic nucleus of the trigeminal nerve, displayed this phenomenon. Lysoso

Development of single-molecule ubiquitination mediated fluorescence complementation to visualize protein ubiquitination dynamics in dendrites.

The ubiquitination/proteasome system is important for the spatiotemporal control of protein synthesis and degradation at synapses, while dysregulation may underlie autism spectrum disorders (ASDs). However, methods allowing direct visualization of the subcellular localization and temporal dynamics of protein ubiquitination are lacking. Here we report the development of Single-Molecule Ubiquitin Mediated Fluorescence Complementation (SM-UbFC) as a method to visualize and quantify the dynamics of protein ubiquitination in dendrites of live neurons in culture. Using SM-UbFC, we demonstrate that t

In Vivo Two-photon Calcium Imaging in Dendrites of Rabies Virus-labeled V1 Corticothalamic Neurons.

Monitoring neuronal activity in vivo is critical to understanding the physiological or pathological functions of the brain. Two-photon Ca2+ imaging in vivo using a cranial window and specific neuronal labeling enables real-time, in situ, and long-term imaging of the living brain. Here, we constructed a recombinant rabies virus containing the Ca2+ indicator GCaMP6s along with the fluorescent protein DsRed2 as a baseline reference to ensure GCaMP6s signal reliability. This functional tracer was applied to retrogradely label specific V1-thalamus circuits and detect spontaneous Ca2+ activity in th

Tick-borne flaviviruses alter membrane structure and replicate in dendrites of primary mouse neuronal cultures.

Neurological diseases caused by encephalitic flaviviruses are severe and associated with high levels of mortality. However, detailed mechanisms of viral replication in the brain and features of viral pathogenesis remain poorly understood. We carried out a comparative analysis of replication of neurotropic flaviviruses: West Nile virus, Japanese encephalitis virus and tick-borne encephalitis virus (TBEV), in primary cultures of mouse brain neurons. All the flaviviruses multiplied well in primary neuronal cultures from the hippocampus, cerebral cortex and cerebellum. The distribution of viral-sp
